Compositional transformation of South African semiarid grasslands and savannas owing to changes in soil nutrient status and fire-linked attributes is often reported. However, mechanisms of change are not fully understood. Currently, plant-derived smoke has attracted much attention as a fire-related cue responsible for stimulating germination and seedling growth. However, there is very little documentation on how these fire-linked factors, such as smoke, and soil macronutrients, such as nitrogen (N), phosphorus (P), and potassium (K), interact to effect seedling growth of grasses. In this study, smoke-responsive (Themeda triandra) and less smoke-responsive species (Eragrostis curvula and Panicum maximum) were tested with different concentrations and combinations of smoke-water and smoke-isolated butenolide with or without added N, P, or K under greenhouse conditions. In the absence of N, P, or K, smoke-water and butenolide treatments enhanced a number of seedling growth parameters of T. triandra. In contrast, exclusion of N from the nutrient solution significantly reduced shoot length, seedling weight, root volume, and vigor index of E. curvula at all tested concentrations of smoke-water and butenolide solutions compared to the control. In the presence of N, P, and K, smoke-water and butenolide suppressed seedling growth of P. maximum, whereas the absence of one of these macronutrients had a small promotory effect on some parameters. Isolation and characterisation of an Indian strain of Mycoplasma mycoides subsp. mycoides LC from a case of caprine arthritis is reported in the study. The isolate was identified based on biochemical, digitonin sensitivity and growth inhibition tests. The virulence of the organism was studied by pathogenicity test in mice and goat. The antigenic and genomic profile of the isolate was compared with that of the standard strain (Y-Goat). 以抗旱甘蔗品种ROC22为材料,调查水分胁迫及胁迫加喷施脱落酸(ABA)对甘蔗内源ABA合成及相关生理生化作用的影响。结果表明,干旱及干旱加外施ABA的条件下,甘蔗内源ABA合成水平上升,但干旱加ABA处理增幅更显著,甘蔗叶内的脯氨酸(Pro)、H2O2、丙二醛(MDA)含量增加,而ABA处理能缓解MDA的积累,使其含量处于低水平,ABA处理能防止叶绿素降解并对干旱引起的最大光能转化效率(Fv/Fm)、PSII实际量子效率(ΦPSII)下降有明显的缓解作用。干旱条件下,H2O2的积累伴随着抗氧化作用的酶CAT、GPX、GR和APX的活性提高,而ABA处理能进一步提高这些相关酶的活性而逐渐降低H2O2的含量,表明干旱条件下,外施ABA能增强甘蔗的抗氧化防护系统,提高抗旱性。 相似文献

Fire, natural or of anthropologic origin, is a recurrent phenomenon in South African mesic grassland. The species composition of these grasslands is sensitive to fire, particularly fire frequency. However, the mechanisms involved in influencing species composition are not fully understood. Currently there is a general suggestion that plant-derived smoke and smoke-isolated biologically active butenolide compound provide an important germination cue for a range of Poaceae species. Studies also show that these smoke solutions play a role in vegetative growth of many plants. We examined if this fire-response syndrome is related to the effect of plant-derived smoke-water (1:500 v/v) and smoke-derived butenolide compound (10-8 M) on seed germination and seedling growth of six major constituent species of the grassland. In addition, the interaction of the smoke solutions with temperature was examined by incubating seeds at a range of temperatures. Treating seeds with smoke-water and butenolide, the germination rate and final germination percentage were greater in three of the six species. Themeda triandra Forssk. and Tristachya leucothrix Trin. ex Nees showed the greatest response, with final germination increased from 43% to 67% and 35% to 63%, respectively. With increasing temperature (> 30°C), Aristida junciformis Trin. & Rupr., Hyparrhenia hirta (L.) Staph, and Panicum maximum Jacq. responded positively to the test solutions. In nearly all the species tested, smoke-water–treated seeds produced significantly longer shoots or roots. However, the degree of response varied from species to species and across different temperatures. Findings from this study suggest that plant-derived smoke and its interaction with temperature may significantly influence the germination and seedling growth of the South African mesic grassland species, which can further alter the grassland composition. 相似文献

As failure to control Rhyzopertha dominica (F.) with phosphine is a common problem in the grain-growing regions of Brazil, a study was undertaken to investigate the frequency, distribution and strength of phosphine resistance in R. dominica in Brazil. Nineteen samples of R. dominica were collected between 1991 and 2003 from central storages where phosphine fumigation had failed to control this species. Insects were cultured without selection until testing in 2005. Each sample was tested for resistance to phosphine on the basis of the response of adults to discriminating concentrations of phosphine (20 and 48 h exposures) and full dose-response assays (48 h exposure). Responses of the Brazilian R. dominica samples were compared with reference susceptible, weak-resistance and strong-resistance strains from Australia in parallel assays. All Brazilian population samples showed resistance to phosphine: five were diagnosed with weak resistance and 14 with strong resistance. Five samples showed levels of resistance similar to the reference strong-resistance strain. A representative highly resistant sample was characterised by exposing mixed-age cultures to a range of constant concentrations of phosphine for various exposure periods. Time to population extinction (TPE) and time to 99.9% suppression of population (LT(99.9)) values of this sample were generally similar to those of the reference strong-resistance strain. For example, at 0.1, 0.5 and 1.0 mg L(-1), LT(99.9) values for BR33 and the reference strong-resistance strain were respectively 21, 6.4 and 3.7 days and 17, 6.2 and 3.8 days. With both strains, doubling phosphine concentrations to 2 mg L(-1) resulted in increased LT(99.9) and TPE. High level and frequency of resistance in all population samples, some of which had been cultured without selection for up to 12 years, suggest little or no fitness deficit associated with phosphine resistance. The present research indicates that widespread phosphine resistance may be developing in Brazil. A laboratory study was undertaken to determine the persistence and efficacy of spinosad against Rhyzopertha dominica (F.) in wheat stored for 9 months at 30 degrees C and 55 and 70% relative humidity. The aim was to investigate the potential of spinosad for protecting wheat from R. dominica during long-term storage in warm climates. Wheat was treated with spinosad at 0.1, 0.5 and 1 mg kg(-1) grain and sampled after 0, 1.5, 3, 4.5, 6, 7.5 and 9 months of storage for bioassays and residue analyses. Residues were estimated to have declined by 30% during 9 months of storage at 30 degrees C and there was no effect of relative humidity. Spinosad applied at 0.5 or 1 mg kg(-1) was completely effective for 9 months, with 100% adult mortality after 14 days of exposure and no live F1 adults produced. Adult mortality was <100% in some samples of wheat treated with 0.1 mg kg(-1) of spinosad, and live progeny were produced in all samples treated at this level. The results show that spinosad is likely to be an effective grain protectant against R. dominica in wheat stored in warm climates. 相似文献

Numerous strains of the psocid pest, Liposcelis decolor (Pearman) were collected from farms and central storages and interbred to form three representative strains from three major grain-growing states of Australia: Queensland, South Australia and New South Wales. These were tested against the grain protectants and structural treatments currently registered for use in Australia. Recently, L decolor has become an important pest of stored grain in Australia, particularly in the eastern and southern parts. There is no published information available on management of this pest and the current pest-management strategy, based predominantly on phosphine fumigation, has failed to control infestations of this pest in numerous grain storages in Australia. Alternative methods of control such as use of contact insecticides were explored in the present work to supplement phosphine fumigation to manage this new pest. From eight grain protectants tested as admixtures, only chlorpyrifos-methyl, bioresmethrin plus piperonyl butoxide, and fenitrothion were found to provide long term (3-9 months) protection against all three strains of L decolor. Chlorpyrifos-methyl gave the best protection, providing a minimum of 7.5 to a maximum of 9 months protection, depending on the strains tested. Three structural treatments, azamethiphos, azamethiphos plus carbaryl and permethrin provided long-term control (8-9 months) of all three strains of L decolor on galvanised steel surfaces, with permethrin delivering 9 months protection against all strains. However, all of these treatments failed to provide long-term control of any strains on concrete surfaces. We conclude that chlorpyrifos-methyl as a grain admixture can be incorporated into fumigation strategies to optimise the control of L decolor infestations. Purpose
The synthetic soil based bioremediation approach as reasonable and sustainable practice at the farming level where desired bioremediation could be established at lower cost.Materials and methods
Metal-tolerant bacteria from different environmental field samples, (a) a municipal dump site, (b) an agricultural field and (c) sludge of electro-plating industries, were screened and characterized. Bioremediation of metal contaminants through isolated bacteria was compared under two different conditions, synthetic soil and basic minimal media containing copper, cobalt and nickel.Results and discussion
The pollutants arising from industrial effluents are imparting a huge negative impact on agricultural land. Microbes are predominant in heavy metal-contaminated sites, which signifies as a potential opportunity for the researchers towards bioremediation. Three bacterial species showed high metal tolerance; 16S ribosomal DNA (rDNA) analysis revealed that the organisms were Proteus vulgaris strain, Stenotrophomonas sp. and Bacillus thuringiensis. Percentage removal of metals was also analysed under different concentrations and pH.Conclusions
The current tested methods are helpful in streamlining the natural compliance of fragile elements and its uptake into the microbial system under in vitro and in situ conditions.70.

Laboratory and greenhouse investigations were carried out with 65Zn-labeled sources to study the kinetics of desorption, transformation, and availability of Zn applied to soil as zinc-enriched
biosludge from distillery molasses (ZEMB) or as zinc sulfate heptahydrate (ZSH). Desorption (0.5 to 72 h) of added Zn by the
column method followed a biphasic kinetics with an initial (up to 12 h) faster phase followed by a slower desorption phase.
The desorption rate coefficient (K) of the latter phase and the amount of Zn desorbed during 12 to 72 h were significantly
higher with ZEMB than with ZSH. Sequential extraction of Zn added as ZEMB and ZSH showed that Zn added as ZEMB was present
in higher proportion as water soluble + exchangeable, carbonate bound, organically bound, and reducible fractions than Zn
applied as ZSH, which showed a higher proportion of residual fraction. Under greenhouse conditions, dry matter yield (35 days)
and total Zn uptake by rice fertilized with ZSH applied at 5 kg Zn ha−1 were statistically similar to those of rice treated with 2.5 kg Zn ha−1 supplied as ZEMB. The highest Zn uptake (167.08 μg pot−1) by rice was recorded in the treatment with 5 kg Zn ha−1 as ZEMB. For wheat plants grown after the harvest of rice, significantly higher dry matter yield over control was recorded
in the treatment with ZEMB applied at 5 kg Zn ha−1 to rice. Total Zn uptake by wheat was statistically similar for both ZEMB and ZSH treatments at 5 kg Zn ha−1 dose. Both zinc derived from fertilizer and the percent utilization of fertilizer Zn by rice and by the subsequent wheat
crop were significantly higher with ZEMB than with ZSH.
