The lectin histochemical pattern (LHP) was characterized and compared in normal and cystic ovaries of sows. Six biotinylated lectins (PNA, SBA, WGA, RCA‐1, DBA and UEA‐1) were used on tissue sections. In the normal ovaries, the reaction to UEA‐1 and SBA was mild to moderate in mesothelial and endothelial cells. RCA‐1 staining was mild to moderate in theca interna of growing follicles, corpora luteum and mesothelium. In addition, this lectin presented strong reaction in endothelial cells, granulosa cells of atretic follicles, zona pellucida of growing follicles and plasma. DBA showed strong intensity in mesothelial and endothelial cells. There was mild to moderate reactivity to WGA in granulosa cells, corpus luteum and theca interna of follicles in development, and moderate in zona pellucida, in granulosa cells of atretic follicles and mesothelium. PNA staining was mild to moderate in oocytes and in the adventitia and media of medullary arteries. Changes in the LHP of the cystic ovaries were noted; however, there were no differences in these findings between the follicular and luteinized cysts. UEA‐1 reactivity in the cystic ovaries was moderately reduced in the mesothelial and endothelial cells, whereas there was mild reduction in the DBA staining in the granulosa cells. Reaction to RCA‐1 and WGA in the cysts also was decreased in theca interna, zona pellucida and granulosa cells of atretic follicles. Furthermore, endothelium and theca interna in the cystic ovaries presented mild reduction of marcation to SBA, whereas there was decreased reactivity to PNA in the oocytes and adventitia and media layers of the medullary arteries. The results of the current study show that cysts modify the LHP in swine ovaries. These changes of glycoconjugates in many ovarian structures could modify diverse process and may be one of the reasons for decreased fertility in sows.  相似文献

Background

Blood centrifugation and buffy coats are at the cornerstone of hematology. In mammals, the buffy coat has a layered disposition (from bottom to top) with neutrophils on top of erythrocytes, followed by monocytes/lymphocytes, and platelets. In nonmammals, this distribution is unknown. Recently, the cell tube block (CTB) technique was developed to study the buffy coat, but it was never applied to nonmammal buffy coats.

Objectives

This study aimed to evaluate using the CTB technique to study reptilian and avian buffy coats and to propose its use for clinical applications.

Methods

Blood from five birds and eight reptiles of different species was obtained to make CTBs that were processed for optical/electron microscopy. H&E, Sirius red, and immunohistochemistry staining against CD3 (to label T lymphocytes) were applied to the CTBs.

Results

In birds, the buffy coat had a layered appearance with the granulocyte layer containing granulocytes (heterophils and eosinophils) and nucleated erythrocytes followed by a mononuclear cell layer containing lymphocytes, monocytes, and thrombocytes. In some animals, a nucleated erythrocyte layer was observed admixed with the granulocyte/mononuclear cell layer. A small clot within the buffy coat was seen in seven reptiles, and less definition of layers occurred in reptiles, with only one or two layers. Lymphocytes appeared toward the top of the buffy coat.

Conclusions

From a comparative hematology perspective, the buffy coat of mammals differs from that of birds and more from that of reptiles. The CTB technique can be used to study these differences in avian and reptilian hematology, especially to study atypical circulating cells, hemoparasites, or blood cell proportions in health and disease.  相似文献

This study aimed to evaluate the effects of forage neutral detergent fiber (fNDF) levels on the voluntary feed intake, digestibility, ruminal fermentation and feeding behavior of goats fed diets with cactus pear. Five non‐lactating ruminally cannulated goats fed ad libitum were randomly assigned to a 5 × 5 Latin square design. Treatments consisted of levels of fNDF at 0, 109, 222, 339 and 463 g/kg of dry matter (DM) in cactus pear‐based diets. The intakes of DM and NDF were quadratically affected (p ≦ .045) by fNDF levels. Voluntary water intake (VWI) increased linearly as the fNDF levels increased in the diet. The digestibility coefficients of organic matter, NDF and ether extract and total digestible nutrients concentration were quadratically affected (p ≦ .048) by fNDF levels. The ruminal pH linearly increased (p = .001) with fNDF levels, ranging from 5.44 to 5.81 for diets containing 0 and 463 g fNDF/kg DM, respectively. The fNDF levels promoted a linear increase (p = .006) in chewing time, linearly decreased (p = .007) resting time and quadratically affected (p = .033) rumination time. The inclusion of fNDF in the diets provided favorable conditions for ruminal function, digestibility and feeding behavior in goats fed diets containing cactus pear.  相似文献

Strawberries represent the main source of ellagic acid derivatives in the Brazilian diet. They are also good sources of flavonoids, mainly anthocyanins, and phenolic acids, to which many beneficial effects have been attributed. However, as the fruit is not available all the year, the objective of this work was to determine whether the jams could also represent a good source of bioactive compounds. In the current study, five different commercially available strawberry jams were characterized in relation to flavonoids, total phenolics, free and total ellagic acid contents, and antioxidant capacity. Anthocyanins were detected only in two jams at very low content. Kaempferol glycosides were the main flavonoids present (from 0.38 to 1.05 mg/100 g fresh weight, FW), while quercetin glycosides were present in the range 0.14–1.20 mg/100 g FW. Free and total ellagic acid content ranged from 0.4 to 2.9 mg/100 g FW, and from 17.0 to 29.5 mg/100 g FW, respectively. Total phenolics varied from 58 to 136 mg/100 g FW, and the antioxidant capacity from 0.55 to 0.76 μmol BHT (Butylhydroxytoluene) equivalents/g FW. Overall, results indicated that jams can also represent a good source of antioxidant compounds, although compared to the fruit important losses seem to occur.  相似文献

The ELISA test was evaluated for the diagnosis of bovine cysticercosis using heterologous antigens from the larvae of T. solium and T. crassiceps, by using different types of positive and negative control sera, to allow a broader analysis of the results. The ELISA test showed low sensitivity under natural conditions of bovine cysticercosis manifestation, but high rates (up to 90%) under experimental conditions. The high specificity of the test (81-100%) made evident its capacity to differentiate cysticercosis from other bovine diseases. No difference in performance was found among the antigens studied. It was concluded that the ELISA test has deficiencies in detecting anti-cysticercosis antibodies of animals at slaughterhouse. However, it can be useful in detecting experimentally infected animals and differentiating cysticercosis from other bovine diseases.  相似文献

The organization of the lung's elastic fibres is amazingly uniform in all vertebrates, with the possible exception of birds, whose pulmonary architecture and air movement are unique. The overall goal of this work was to study and quantify elastic fibre distribution patterns and relative amounts in the parabronchi, during the incubation period until the 42nd day after hatching. Chick embryo lungs were examined on the 14th, 16th, 18th and 20th days of incubation and chick lungs on the 1st, 2nd, 7th, 14th, 35th and 42nd days after hatching. Four animals were used daily, and the observations were randomly performed on both lungs. A morphometric study was carried out focusing on the computerized image analysis of histological sections stained according to a modified Gomori technique. The values obtained for each day result from the observation and processing of 20 images. Complementary studies were performed using transmission electron microscopy, as on the 14th embryonic day the fibres were not visible on light microscopy. The results show that the area occupied by the elastic fibres increases gradually from the 16th day of incubation up till the 7th day after hatching and decreases slowly in the following days of the study. A prominent increase takes place before hatching, which points out to the adequate and essential structural roles played by the elastic fibres in the pulmonary maturation process.  相似文献

Triterpenic and other lipophilic components from industrial cork byproducts   总被引:1,自引:0,他引:1  
The detailed chemical composition of the lipophilic extractives of cork and cork byproducts generated throughout industrial processing has been investigated by gas chromatography-mass spectrometry. Triterpenes (cerine, friedeline, and betulinic acid) were the major components detected. Betulinic acid is the main triterpene (11.7 g/kg) identified in industrial cork powder, whereas in black condensates friedeline (95.3 g/kg), betuline (13.1 g/kg), and betulinic acid (12.1 g/kg) are the main triperpenes. Significant fractions of alpha-hydroxy fatty acids (115.1 g/kg) and alpha,omega-dicarboxylic acids (21.2 g/kg) were also detected in black condensate after alkaline hydrolysis. The results demonstrate that these two industrial byproducts can be considered as promising sources of bioactive chemicals or chemical intermediates for the synthesis of polymeric materials.  相似文献

Background: The Sysmex XT‐2000iV is a hematology analyzer that combines laser and impedance technology. Its usefulness for determining cell counts in canine and feline intracavitary effusions has not yet been studied. Objectives: The objectives of this study were to evaluate the analytical performance of the Sysmex XT‐2000iV for cell counts in effusions from dogs and cats, and to assess correlation with an impedance counter and concordance with diagnoses based on cytologic findings. Methods: Effusions (43 pleural, 23 peritoneal, 6 pericardial) were analyzed from 32 dogs and 34 cats. Total nucleated cell count (TNCC), HCT, and RBC count were determined on the Sysmex and compared with those obtained on an impedance counter (Hemat 8, SEAC). Imprecision, linearity, and limit of detection were determined for the Sysmex. An algorithm was designed using quantitative and qualitative data from the Sysmex to classify the effusions and the results were compared with diagnoses based on cytologic findings. Results: Intra‐assay and interassay coefficients of variation on the Sysmex were variable. Linearity of TNCC was ≥0.993 for dogs and cats, with the exception of effusions from cats with feline infectious peritonitis, which had delta (Δ) TNC values >3.0. In comparison with the Hemat 8, a proportional error was found for TNCC on the Sysmex. Effusion classification based on the algorithm was concordant with that obtained by cytologic examination in 43/72 (60%) samples. Discordant results usually were due to the misclassification of cells with similar morphology (such as mesothelial and carcinoma cells) in Sysmex scattergrams. Conclusion: The Sysmex XT‐2000iV provides a precise and accurate TNCC and has moderate concordance with cytologic findings for classifying canine and feline effusions. Although microscopic examination of effusions is necessary to achieve an accurate diagnosis, the Sysmex can provide preliminary information that may be helpful to cytopathologists.  相似文献

Zebu breeds play an important role in cattle production systems in Brazil. To assess the genetic variability from animals in the Herd Books of Nelore, Gir and Guzerat breeds, generation intervals, inbreeding, effective population size and parameters of gene origin (effective number of founders, ancestors and founder genomes) were calculated using pedigree records from 1938 to 1998. Breed subdivision was quantified by Wright's F -statistics. Calculations were separately carried out for consecutive 4-year intervals in the period 1979–98. Generation interval was around 8 years for the three breeds. Total inbreeding increased in all the breeds reaching values of 2.13%, 2.28% and 1.75%. Effective population size decreased from 85 to 68 in Nelore, from 70 to 45 in Gir and remained nearly constant around 104 in Guzerat. The quantities assessing the number of contributing ancestors decreased with time in all the breeds, and in the last analysed period the most important ancestor accounted for 14%, 3.1% and 4.1% in Nelore, Gir and Guzerat, respectively. Results indicate that the studied breeds are suffering from a loss of genetic variability which can result in negative effects on breeding and conservation purposes.  相似文献
