BmHSP20.8 is Localized in the Mitochondria and has a Molecular Chaperone Function In Vitro

Heat shock proteins (HSPs) are abundant and ubiquitous in almost all organisms from bacteria to mammals. BmHSP20.8 is a small (sHSP) in Bombyx mori that contains a 561 bp open reading frame that encodes a protein of 186 amino acid residues with a predicted molecular mass of 20.8 kDa. The subcellular localization prediction indicated that BmHSP20.8 is likely distributed in the mitochondria with a 51% probability. To identify the subcellular localization of BmHSP20.8, three recombinant vectors were constructed and used to transfect BmN cells. The cytoplasmic and mitochondrial proteins were extracted 72 h after transfection. The Western blot showed that recombinant BmHSP20.8 exists only in the mitochondria. To locate the mitochondrial localization signal domain of BmHSP20.8 more accurately, we cloned four truncated recombinant vectors. The Western blot analysis of the cytoplasmic and mitochondrial proteins showed that the mitochondrial localization signal domain of BmHSP20.8 is located between amino acids 143 to 186. We constructed the pETduet-HIS-SUMO-BmHSP20.8 vector and a soluble BmHSP20.8 was expressed. In a citrate synthase (CS) thermal aggregation experiment, we found that the recombinant BmHSP20.8 protein can protect CS from aggregating at 43 and 48°C and thus exhibited molecular chaperone activity. Taken together, the results showed that BmHSP20.8 could be a mitochondrial protein and has a molecular chaperone activity, suggesting an important role in mitochondria.     

烟草废弃物生产有机肥对植烟土壤养分含量的影响

为探讨烟草废弃物生产有机肥对植烟土壤养分含量的影响,通过对烟草废弃物生产有机肥、烟草专用化学肥对植烟土壤影响的对比,以及不同有机肥配比量对改善植烟土壤的作用对比,分别测定了其对有机质含量、碱解氮、速效磷、速效钾含量以及全氮、全磷、全钾含量的影响。结果显示土壤碱解氮、速效磷、速效钾的含量与有机肥施用量呈正比例关系,同时有机肥的施用能增加土壤中有机质、全氮、全磷、全钾的含量,且当有机肥所占比例为50%时,植烟土壤中养分含量增加显著且符合优质烟叶生产的土壤条件,效果最为理想。实验表明烟草废弃物生产有机肥能有效改善土壤理化性质,增加土壤肥力,改善烟叶品质。同时在一定程度上可减少化肥的施用,进而保护土壤环境,实现烟草废弃物的循环利用。     

棉花抗盲蝽材料田间抗虫性和产量性状评估

在2008年抗盲蝽棉花材料筛选的基础上,选出4个对盲蝽抗性较好的棉花材料和2个感盲蝽的棉花材料,分析其田间抗盲蝽性,并对其生长势和产量构成因素与目前2个主栽棉花品种做了比较,以评估这些棉花材料的应用价值。试验结果表明,表现出良好抗盲蝽性的品种HH1、BJ2、HH2的田间生长势和产量构成参数与抗虫棉中棉所41相当,优于常规棉中棉所49不喷施化学农药田。     

超高效液相色谱-串联质谱法测定莲雾中多菌灵、噻菌灵和甲霜灵残留

建立了莲雾中多菌灵、噻菌灵和甲霜灵残留的超高效液相色谱-串联质谱(UPLC-MS/MS)的快速检测方法。样品直接用乙腈匀浆提取,过0.22μm滤膜后,在电喷雾正离子模式下以多反应监测(MRM)模式测定,基质匹配标准溶液外标法定量。结果表明:方法的线性范围为0.01~0.5μg/m L,决定系数(R2)0.99;在0.04、0.4和2 mg/kg 3个添加水平下,方法的平均回收率为75%~107%,相对标准偏差为2.0%~8.9%(n=5),定量限均为0.04 mg/kg。该方法样品前处理简单、快速,分析时间短,灵敏度、准确度和精密度均符合农药残留检测要求。     

施用保水剂对秋植大豆生长发育及产量的影响

为研究保水剂对秋植大豆在闽南丘陵旱地生长发育与产量的影响,在田间试验条件下,以不施保水剂为对照,研究了沟施不同施用量的保水剂对其发育及产量影响。结果表明：与对照相比,施用保水剂的大豆各生育时期都有所延长,全生育日数比对照延长了2~7天,并且提高了大豆单位面积产量与有效荚率,单株粒重增多,百粒重提高;且随施用量的增加,效果更为明显。沟施保水剂60 kg/hm²用量的增产增收效果最佳,可增加纯收入1503.00元/hm²,其投入产出比为1:2.67,适宜在闽南丘陵旱地及其同类地区旱地进行秋季种植大豆时使用。     

基于Web Services的分布式的林业科研档案数据管理系统的研究与设计

Web Services以其协议的通用性、完全的平台和语言独立性、高度松耦合性,逐步成为应用集成的重要方向。根据林科院科研档案数据分布式存储的特点,采用Web Services技术,提出了分布式林业科研档案数据管理系统的设计与实施,并在Windows.NET框架下实现了该系统的安全性、维护性和可扩展性。     

Effect of SS-toxin,a metabolite of <Emphasis Type="Italic">Stemphylium solani</Emphasis>, on H<Superscript>+</Superscript>-ATPase activity and standard redox system in plasma membranes from seedlings leaves of garlic (<Emphasis Type="Italic">Allium sativum</Emphasis>)

Leaf blight of garlic (Allium sativum) is a severe disease in garlic-growing regions. SS-toxin is a newly described non-proteinaceous toxin produced by the phytopathogenic fungus, Stemphylium solani, the cause of garlic leaf blight. In this study, the effects of SS-toxin on the H⁺-ATPase activity and standard redox activity in the plasma membranes isolated by aqueous polymer two-phase partitioning from garlic seedling leaves were studied in vitro. The H⁺-ATPase activity, NADH oxidation rate and Fe(CN)₆³⁻ reduction rate of the plasma membranes isolated from susceptible and resistant cultivars were all inhibited in a dose-dependent manner. Our results suggest that, under in vitro conditions, the plasma membrane H⁺-ATPase and standard redox system can be both the cellular targets of SS-toxin.
