Hawthorn (Crataegus monogyna) is one of the natural hosts of Cacopsylla melanoneura, the acknowledged vector of ‘Candidatus Phytoplasma mali’, the causal agent of Apple Proliferation disease, a serious and growing problem for apple production in Europe, particularly in northern Italy. Wild plants could be important sources of both insects and phytoplasmas, but their role in the epidemiology of phytoplasma diseases and their insect vectors has never been thoroughly examined. Cacopsylla melanoneura’s primary host is hawthorn, a plant closely related to apple which often grows wild near orchards. Other psyllid species feed on hawthorn, but no data are available on their possible role as phytoplasma vectors. We investigated the hawthorn’s psyllid fauna in northwestern Italy using yellow sticky traps, beat trays, and molecular analyses from 2003–2005, to study the relationship between hawthorn, the phytoplasma and the insect vector. Population dynamics were monitored, and insects and hawthorn samples were analysed by polymerase chain reaction (PCR), restriction fragment length polymorphism (RFLP), and DNA sequencing for the presence of phytoplasmas. Cacopsylla melanoneura was the dominant psyllid species, followed by C. peregrina, C. affinis and C. crataegi. PCR and RFLP analyses revealed the presence of different fruit tree phytoplasmas in hawthorn plants, and in all four psyllid species.  相似文献

Celiac disease is an autoimmune disorder caused by intolerance to gluten, which is found in wheat and similar proteins in barley, rye and oats. The present study was designed to examine the effects of the addition of gums, whey protein concentrate, dry egg, and water to corn and cassava starches on the rheological properties of a non-fermented dough used for the production of “empanadas” (a traditional meal in Latin América) and pie-crusts suitable for people with celiac disease. A 24 full factorial design was chosen. Viscoelastic measurements and texture analysis (puncture and elongation tests) were performed. The increase in gums content and the decrease in water level produced an increase in both moduli (G′ and G″) and a more elastic dough was obtained. Higher protein contents interfered with the formation of the three-dimensional gum network making the dough less ductile. Texture analysis led to similar conclusions to those obtained by dynamic rheological analysis. Formulations containing higher percentages of gums and lower water content led to an appropriate behavior for industrial production of these doughs.  相似文献

Brill (Scophthalmus rhombus L.) is a flatfish considered of special interest for aquaculture diversification, but the high mortality observed during the early larval rearing is the main obstacle to commercial culture. The objective of this study was to contribute to the knowledge of nutrient utilization of early‐hatched larvae, characterizing the changes in lipid and protein contents during embryogenesis and the yolk‐sac larval stage of S. rhombus. Total lipid, lipid classes and fatty acid contents remained constant during embryogenesis and yolk‐sac larval development, except for phosphatidylethanolamine and phosphatidylinositol, which increased in quantity during the yolk‐sac larval stage. On the other hand, total protein (including non‐protein nitrogen) and amino acids decreased their contents in both periods, especially at hatching. The decrease only in the serine, glutamic acid, proline and lysine contents during embryogenesis suggests a selective use of amino acids during this phase. Unlike embryogenesis, amino acids loss during hatching appears to be non‐selective, and almost all amino acids (essential and non‐essential) decreased. Our results suggest that there is higher catabolism of protein vs. lipid during embryogenesis and the yolk‐sac larval stage of S. rhombus.  相似文献

Abstract

This paper presents a study of operating costs in shrimp hatcheries. Size effects are presented for fixed, variable, unitary, and marginal costs. Cost structures by functional production department and by cost‐category are discussed for a hypothetical 100‐million postlarvae/month hatchery in an industrialized country, and compared to those of a hatchery in a developing country. Health management issues, mainly measures commonly used to manage bacterial diseases, and their economic impact are discussed, focusing on the trade off between survival rates and sanitation down‐time.

Salaries and benefits represent the largest cost‐component (37%) for hatcheries in industrialized countries, while feeds and operating supplies constitute the most important costs in developing countries (60%). The need to import hatchery feeds and other operating supplies into developing countries works against savings from lower labor costs typical of developing countries.

Total fixed costs depend on the size of a hatchery and are constant for all possible levels of output. Average fixed cost per unit (thousand postlarvae) can be reduced by producing high postlarval outputs. Hatcheries in areas with seasonal shrimp crops face the challenge of covering fixed costs with revenue from a short production season. Hatcheries with large fixed costs can operate in a cost‐efficient manner only by producing high levels of output. Average total cost and average variable cost curves follow a U‐shaped curve, in which costs first decline as output increases, then reach a minimum, and finally increase as output increases above a point of diminishing returns.

In hatcheries total costs are independent of larval survival, but lower output resulting from low survival increases unit costs. This effect is more pronounced at lower levels of output. Bacterial diseases decrease survival rates and increase unit costs.  相似文献

Growth, biofilm formation, antagonism and residence time in green seawater tanks maintained under fish rearing conditions of Phaeobacter 27‐4 were studied in commercial biofilters made from plastic, sintered glass and ceramic. Phaeobacter reached 108–109 CFU cm?3 and formed rosettes in all materials, but a multilayer biofilm was only observed in the ceramic biofilters. In sterile seawater, plastic and ceramic biofilters reduced Vibrio anguillarum and V. splendidus concentration in one‐two Log after 24–48 h, showing 102–103 CFU mL?1. Sintered glass biofilters only inactivated V. anguillarum. In Marine Broth, sintered glass and ceramic biofilters inhibited V. anguillarum growth in two‐three Log, showing 104–105 CFU mL?1 after 24 h. Plastic biofilters reduced V. anguillarum concentration in one Log after 48 h. V. splendidus growth was only inhibited by sintered glass and ceramic biofilters in one‐two Log, showing 107 CFU mL?1 after 24 h. Phaeobacter also diminished biofilters colonization by the pathogens, both in seawater and in MB. Phaeobacter residence time in green seawater tanks maintained under fish rearing conditions was longer with sintered glass and ceramic biofilters. The latest showed the lowest detachment and, after 11 days, Phaeobacter (106 bacteria·cm?3) covered more than 80% of biofilters total culturable bacteria. DGGE profiles showed that Phaeobacter biofilters stabilizes the green seawater bacterial microbiota.  相似文献

The need to avert unacceptable and irreversible environmental change is the most urgent challenge facing society. Landscape ecology has the capacity to help address these challenges by providing spatially-explicit solutions to landscape sustainability problems. However, despite a large body of research, the real impact of landscape ecology on sustainable landscape management and planning is still limited. In this paper, we first outline a typology of landscape sustainability problems which serves to guide landscape ecologists in the problem-solving process. We then outline a formal problem-solving approach, whereby landscape ecologists can better bring about disciplinary integration, a consideration of multiple landscape functions over long time scales, and a focus on decision making. This framework explicitly considers multiple ecological objectives and socio-economic constraints, the spatial allocation of scarce resources to address these objectives, and the timing of the implementation of management actions. It aims to make explicit the problem-solving objectives, management options and the system understanding required to make sustainable landscape planning decisions. We propose that by adopting a more problem-solving approach, landscape ecologists can make a significant contribution towards realising sustainable future landscapes.  相似文献

In the present paper we report on the effects of the insertion of the Agrobacterium rhizogenes rolC gene in the tomato (Solanum lycopersicum L., formerly Lycopersicon esculentum Mill.) cultivar Tondino. Several transgenic lines were successfully obtained, between which two clones, rolC1 and rolC3, were chosen for the analysis of morpho-productive traits as well as of the endogenous levels of auxin and abscisic acid. Consistent with the known phenotypic effect of this gene, the transformed tomato plants were significantly shorter than the corresponding controls. On the other hand, even if yield was not affected by the transformation in terms of average number of fruits produced, fruit weight was significantly lower in the transgenics with respect to the controls. Therefore, insertion of the rolC gene does not lead to an improvement in plant productivity.  相似文献

The recent pandemic caused by human influenza virus A(H1N1) 2009 contains ancestral gene segments from North American and Eurasian swine lineages as well as from avian and human influenza lineages. The emergence of this A(H1N1) 2009 poses a potential global threat for human health and the fact that it can infect other species, like pigs, favours a possible encounter with other influenza viruses circulating in swine herds. In Europe, H1N1, H1N2 and H3N2 subtypes of swine influenza virus currently have a high prevalence in commercial farms. To better assess the risk posed by the A(H1N1) 2009 in the actual situation of swine farms, we sought to analyze whether a previous infection with a circulating European avian-like swine A/Swine/Spain/53207/2004 (H1N1) influenza virus (hereafter referred to as SwH1N1) generated or not cross-protective immunity against a subsequent infection with the new human pandemic A/Catalonia/63/2009 (H1N1) influenza virus (hereafter referred to as pH1N1) 21 days apart. Pigs infected only with pH1N1 had mild to moderate pathological findings, consisting on broncho-interstitial pneumonia. However, pigs inoculated with SwH1N1 virus and subsequently infected with pH1N1 had very mild lung lesions, apparently attributed to the remaining lesions caused by SwH1N1 infection. These later pigs also exhibited boosted levels of specific antibodies. Finally, animals firstly infected with SwH1N1 virus and latter infected with pH1N1 exhibited undetectable viral RNA load in nasal swabs and lungs after challenge with pH1N1, indicating a cross-protective effect between both strains.  相似文献

After detection of a high prevalence of scrapie in a large dairy goat herd, 72 infected animals were examined by immunohistochemistry with prion protein (PrP) antibody Bar224 to study the pathogenesis of the infection. Tissues examined included the brain and thoracic spinal cord (TSC), a wide selection of lymphoreticular system (LRS) tissues, the distal ileum and its enteric nervous system (ENS), and other organs, including the mammary gland. The whole open reading frame of the PRNP gene was sequenced and antibodies to caprine arthritis-encephalitis virus (CAEV) infection were determined. Unexpectedly, accumulation of disease-associated PrP (PrPd) in the brain was more frequent in methionine carriers at codon 142 (24/32, 75.0%) than amongst isoleucine homozygotes (14/40, 35.0%). The latter, however, showed significantly greater amounts of brain PrPd than the former (average scores of 9.3 and 3.0, respectively). A significant proportion of the 38 goats that were positive in brain were negative in the ENS (44.7%) or in the TSC (39.5%). These results, together with the early and consistent involvement of the circumventricular organs and the hypothalamus, point towards a significant contribution of the haematogenous route in the process of neuroinvasion. Chronic enteritis was observed in 98 of the 200 goats examined, with no association with either scrapie infection or presence of PrPd in the gut. Lymphoproliferative interstitial mastitis was observed in 13/31 CAEV-positive and scrapie-infected goats; PrPd in the mammary gland was detected in five of those 13 goats, suggesting a possible contribution of CAEV infection in scrapie transmission via milk.  相似文献
