This article considers a two-phase estimation for the areal extent of K land categories partitioning a study region and a three-phase estimation for the biomass of W forest categories out of the K. In the first phase, a sample of N points is selected according to the unaligned systematic sampling. In the second phase, the selected points are partitioned into L strata on the basis of aerial photos. Then, a total sample of n < N points is selected by stratified sampling and the selected points are visited on the ground and correctly classified into one of K categories. The information achieved in the second phase is sufficient for obtaining an unbiased estimator of the areal extent vector together with a conservative estimator of its variance-covariance matrix. As to the estimation of the biomass of the W forest categories, in the third phase the second-phase sample is further partitioned into substrata on the basis of ground information. Finally, a total sample of m < n points is selected by stratified sampling. Then a plot of adequate radius centered at each point is considered and the biomass is recorded within. An unbiased estimator of the biomass vector is derived together with a conservative estimator of its variance-covariance matrix. The proposed strategy also makes it possible to obtain the calibrated estimator of the areal extent vector as well as estimators for the sums or ratios of the areal extents and biomasses. The application of the strategy in the Italian National Forest Inventory is considered.  相似文献

Acacia dealbata Link is an Australian woody legume that has become a serious environmental problem in Northwest Spain where it forms dense monospecific patches modifying the structure of different native ecosystems and threatening native aboveground biodiversity. In spite of the dramatic changes observed in the vegetation of invaded sites little is known about the consequences of invasion for soil microorganisms. To investigate the effect of A. dealbata invasion on the structure of soil fungi and bacteria communities, samples were taken from invaded and non-invaded areas from three different ecosystems in Northwest Spain: pine forest, shrubland and grassland. In each ecosystem type, soil samples were taken in areas of native vegetation, areas invaded by A. dealbata and in the transition zone between native and invaded vegetation. Soil microorganisms were analyzed in the different samples by PCR-DGGE using general primers for eubacteria and fungi. Soil analyses were also performed to evaluate the effect of A. dealbata invasion on soil fertility.The invasion by A. dealbata consistently increased soil N, C, organic matter and exchangeable P content in the three studied ecosystems. A clear effect of the invasion on the overall structure of microorganism communities was only observed in the shrubland where soil fungal communities in the invaded and transition areas clustered together and apart from the native soil. Significant differences in soil microorganisms richness and diversity between invaded and not invaded soils were only found in the grassland. Grassland invasion by A. dealbata lead to a significant increase of bacterial richness and to a significant reduction in fungal richness and diversity. Our results show that although the changes on soil chemistry due to A. dealbata invasion are consistent among the studied ecosystems, the effect on soil microorganisms depends on the ecosystem type affected by the invasion.  相似文献

The amino acid composition of 53 honey samples from Spain, consisting of 39 floral, 5 honeydew, and 9 blend honeys, has been determined. Physicochemical characteristics, polyphenolic content, amino acid composition, and estimation of the radical scavenging capacity against the stable free radical DPPH of the honey samples were analyzed. The resulting data have been statistically evaluated. The results showed that pH, acidity, net absorbance, electrical conductivity, and total polyphenolic contents of the honeys showed a strong correlation with the radical scavenging capacity. The correlation between the radical scavenging capacity of honey and amino acid contents was high with 18 of the 20 amino acids detected, with correlation values higher than those obtained for polyphenolic content. These results suggest that the amino acid composition of honey is an indicator of the sample's scavenging capacity.  相似文献

ABSTRACT The route of infection and the pattern of distribution of Prunus necrotic ringspot virus (PNRSV) in apricot pollen were studied. PNRSV was detected both within and on the surface of infected pollen grains. The virus invaded pollen during its early developmental stages, being detected in pollen mother cells. It was distributed uniformly within the cytoplasm of uni- and bicellular pollen grains and infected the generative cell. In mature pollen grains, characterized by their triangular shape, the virus was located mainly at the apertures, suggesting that PNRSV distribution follows the same pattern as the cellular components required for pollen tube germination and cell wall tube synthesis. PNRSV also was localized inside pollen tubes, especially in the growth zone. In vitro experiments demonstrated that infection with PNRSV decreases the germination percentage of pollen grains by more than half and delays the growth of pollen tubes by approximately 24 h. However, although PNRSV infection affected apricot pollen grain performance during germination, the presence of the virus did not completely prevent fertilization, because the infected apricot pollen tubes, once germinated, were able to reach the apricot embryo sacs, which, in the climatic conditions of southeastern Spain, mature later than in other climates. Thus, infected pollen still could play an important role in the vertical transmission of PNRSV in apricot.  相似文献

The objective of this study was to evaluate the prevalence of infection with porcine circovirus-2 (PCV-2) and porcine reproductive and respiratory syndrome virus (PRRSV) through a longitudinal study in an integrated swine production system (7 farms) experiencing postweaning multisystemic wasting syndrome (PMWS). Risk factors for PCV-2 infection and for PCV-2 and PRRSV coinfection were also evaluated. Fifteen sows from each herd and 4 non-cross-fostered piglets from each sow were randomly selected at farrowing and ear-tagged at birth. Serum samples were analyzed for antibodies to PCV-2 and for detection of the PCV-2 and PRRSV genomes. Statistical analyses involved 2 approaches. The 1st approach characterized the dynamics of PCV-2 infection and their relationship with PRRSV infection. The 2nd approach analyzed the probability of being infected by PCV-2 or by both PCV-2 and PRRSV through a generalized linear mixed model incorporating sow and farm characteristics. At the 1st sampling time (1 wk of age), there was a significant relationship between sow PCV-2 infection and piglet PCV-2 infection (P < 0.0001). The risk of PCV-2 and PRRSV coinfection was 1.85 times greater in piglets from a sow with low titers of PCV-2 antibodies than in piglets from sows with medium to high titers (P = 0.03) and was 2.54, 2.40, and 2.02 times greater, respectively, in piglets from primiparous sows, PCV-2-infected sows, and farms in an area of high pig density than in piglets from sows of higher parity (P = 0.004), noninfected sows (P = 0.04), and farms in a low-density area (P = 0.09).  相似文献

In order to gain a better understanding of the pathogenesis of scrapie in sheep an experimental model was developed to characterise immune system cells in the minutes following inoculation with scrapie-brain homogenate. Four 1-year-old susceptible (ARQ/ARQ) sheep were inoculated via the subcutaneous route at four different peripheral lymph node (LNs) drainage sites, at specific time points, prior to euthanasia of the sheep. The LNs were removed post-mortem at 30, 90, 180 and 300 min after inoculation. Flow cytometric triple-labelling was carried out on the LN cells and indicated that inoculation of scrapie-brain homogenate adjacent to a lymph node may delay or even inhibit the number of host CD21+ B cells expressed within the first 5 h. Immunohistochemistry was used to attempt detection of the abnormal form of prion protein (PrPsc) in draining LNs adjacent to inoculation sites, with negative results at those time points.  相似文献

The influence of flaxseed (Linum usitatissimum L.) and its total non-digestible fraction (TNDF) on the expression of genes involved in azoxymethane (AOM)-induced colon cancer in Sprague Dawley rats was analyzed. The dose used in the animal model was two tablespoons of flaxseed per day, which is the dose recommended for humans. Flaxseed significantly decreased the crypt multiplicity (10.50?±?3.5) compared with the AOM treatment (34.00?±?11.0), which suggests that flaxseed exhibits a preventive effect against colon cancer. Both treatments (flaxseed and TNDF) influence the overexpression of genes involved in cell cycle arrest and mitochondrial apoptosis: p53, p21, bcl-2, bax and caspase-3. Flaxseed induced the expression of p53 and p21, whereas TNDF triggered the p21-independent expression of p53. This finding suggests that both of these treatments induced cell cycle arrest. In addition, TNDF induced mitochondrial apoptosis because the TNDF + AOM group exhibited the expression of caspase-3, decreased bcl-2 expression and increased bax expression. These results suggest that the expression of the analyzed genes is associated with the presence of dietary antioxidants linked to the cell wall of flaxseed.  相似文献

It has long been established that the sheep Prnp genotype influences the susceptibility to scrapie, and some studies suggest that it can also determine several aspects of the disease phenotype. Other studies, however, indicate that the source of infection may also play a role in such phenotype. To address this question an experiment was set up in which either of two different natural scrapie sources, AAS from AA136 Suffolk and VVC from VV136 Cheviot sheep, were inoculated into AA136, VA136 and VV136 sheep recipients (n = 52). The immunohistochemical (IHC) profile of disease-associated PrP (PrPd) accumulation in the brain of recipient sheep was highly consistent upon codon 136 homologous and semi-homologous transmission, but could be either similar to or different from those of the inoculum donors. In contrast, the IHC profiles were highly variable upon heterologous transmission (VVC to AA136 and AAS to VV136). Furthermore, sheep of the same Prnp genotype could exhibit different survival times and PrPd profiles depending on the source of infection, and a correlation was observed between IHC and Western blot profiles. It was found that additional polymorphisms at codons 112 or 141 of AA136 recipients resulted in a delayed appearance of clinical disease or even in protection from infection. The results of this study strongly suggest that the scrapie phenotype in sheep results from a complex interaction between source, donor and recipient factors, and that the Prnp genotype of the recipient sheep does not explain the variability observed upon codon 136 heterologous transmissions, arguing for other genetic factors to be involved.  相似文献
