Drought is one of the main abiotic stresses in agriculture worldwide. Drought could increase under the predicted scenario of climate change, particularly in the Mediterranean area. Breeding for drought tolerance requires screening of germplasm in order to identify sources of tolerance; therefore, were evaluated 51 diverse open-pollinated maize populations from various temperate regions under increasing levels of drought at germination, seedling establishment and early growth. There was genetic variability for drought tolerance among populations at all growth phases. Several populations from diverse origins and germplasm groups exhibited high germination across stress treatments. Some of those populations had high ability to sustain root development and showed differential performance depending on the stage of development and the phenotypic aspect considered. In general, BS17 showed high germination rate, fast seedling growth and early vigor under drought. Longlellow and Grano de trigo showed high germination and growth establishment rates, whereas AS3(HT)C3 showed high germination rate and early vigor under drought. The photosynthetic rate, stomatal conductance and transpiration of Enano Levantino/Hembrilla (ELH) and BS17 populations were not affected by drought. Water use efficiency of BS17, ELH, Northwestern Dent (NWD) and Viana was not affected by drought. Some of these populations are promising sources of drought tolerance which can provide different mechanisms of drought tolerance at different stages of plant development. Therefore, these results open new possibilities of breeding for drought tolerance by combining those mechanisms through crosses among potential donors. The main objective of this research was the evaluation of the variability present in a segregating wine grape population derived from a cross between Graciano × Tempranillo, two Spanish varieties, in order to select improved genotypes with potential for producing high-quality wines in a climate change scenario. For that purpose, the phenotypic segregation of 16 agronomic traits related to production and phenology and 11 enological traits related to technical and phenolic maturity was studied in the progeny for three consecutive years. All traits presented transgressive segregation and continuous variation. Year effect was significant for all traits except total, extractable and skin anthocyanins content. However, a high level of genotype consistency for enological traits was revealed by repeatabilities and correlations between years. Significant correlations among traits were observed but most associations were weak. Furthermore, the CAPS (Cleaved Amplified Polymorphic Sequence) marker for the VvmybA genotype was tested to determine whether it would be useful in indirect selection for berry anthocyanins content. The results showed that the number of homozygous and heterozygous genotypes for the functional colour allele adjusted to a 1:1 segregation ratio, and that homozygous genotypes had significantly higher anthocyanins content. Principal component analysis found eight variables that contributed up to 80 % of the phenotypic variability present in the population. Seven groups of hybrids were distinguished based on ripening time, cluster weight, berry weight and anthocyanins content by cluster analysis; and fourteen genotypes were pre-selected for further research. 相似文献

Six faba bean gene-pools were cultivated to develop locally adapted genetic material for low-input (LI) systems that take advantage of the observed benefit of LI agriculture in pollinator density and diversity. Bee pollinators intensify selection on floral traits important in attraction and mechanical-fit but may also select for traits that increase seed set through outcrossing and aided selfing. Our research is focused on examining the impact of natural selection in crop performance under different pollination management strategies, taking into account traits relevant for the inter-phase plant and pollinator. Architectural and floral patterns are the result of natural selection and they reflect local adaptation to factors of the environment and adaptations to differences in pollination management strategy. However, seed production patterns seem to be affected by natural selection in an inconsistent way. Principal Component models showed a large proportion of the temporal variation accounted by plant height and number of open flowers and by floral shape and sexual dimensions. Discriminant Analysis indicated that plant architecture and floral traits related to pollinators are also the result of selection for particular patterns in each pollination management strategy. Flowers with an increase in the zygomorphy were favored under the open-pollination. Our work confirms the need for careful management of plant-pollinator inter-phase in faba bean breeding for LI systems. 相似文献

Two long primers of 19 (F17) and 20 (F13) nucleotides, respectively,were used in polymerase chain reactions to amplify DNA from differentcultivated barley accessions. These primers can distinguish closely relatedvarieties and, with a unique primer, all the barley accessions analysedshowed a characteristic fingerprint. Sixty per cent and 76% of thefragments generated using F13 and F17, respectively, were polymorphic.The genetic similarity values between accessions were estimated from F13and F17 data. The risk of undesirable by-products from chlorine disinfection in fresh-cut industries, together with its limited efficacy, has led to a search for alternative agents. The aim of this study was to test several alternative putative antimicrobial substances to reduce Escherichia coli O157:H7, Salmonella spp. and Listeria spp. populations on fresh-cut apple. Carvacrol, vanillin, peroxyacetic acid, hydrogen peroxide, N-acetyl-l-cysteine and Citrox were selected for their results in in vitro assays against E. coli O157:H7 and Listeria spp., to be tested on fresh-cut apple plugs. Apple flesh was inoculated by dipping in a suspension of a mix of the studied pathogens at 106 cfu mL?1, and then treated with the antimicrobial substances. All treatments were compared to deionized water and a standard sodium hypochlorite treatment (SH, 100 mg L?1, pH 6.5). Pathogen population on apple plugs was monitored for up to 6 days at 10 °C. Bacterial reductions obtained by peroxyacetic acid (80 and 120 mg L?1), vanillin (12 g L?1), hydrogen peroxide (5, 10, 20 mL L?1) and N-acetyl-l-cysteine (5 and 10 g L?1) were similar or higher than reduction obtained by SH. In addition, bacterial populations were maintained at low levels throughout storage. No cells of any of the pathogens were detected in the peroxyacetic acid, hydrogen peroxide, Citrox and SH washing solutions after apple treatment. Peroxyacetic acid, hydrogen peroxide and N-acetyl-l-cysteine could be potential disinfectants for the fresh-cut industry as an alternative to chlorine disinfection. detected. SSR pattern scores were used to produce an UPGMA dendrogram and a PCoA plot. A set of nine SSR loci, evenly dispersed

This article describes the treatment of single congenital portosystemic shunts (CPSs) (intrahepatic and extrahepatic) using an interventional radiology technique involving embolisation of anomalous vessels with percutaneous coils. Briefly, a multipurpose catheter was introduced into the caudal vena cava and then into the portosystemic shunt. An autoexpandable stent was placed in the caudal vena cava, next to the shunt, in order to avoid coil migrations, and a cobra-like vascular catheter was used to pass through the stent and to place the coils in the shunt. This technique was used for treatment of CPS in six dogs. The results indicate that percutaneous embolisation of a CPS using coils, a less invasive technique than the traditional surgical technique, may result in complete closure of the anomalous vessel without development of portal hypertension. 相似文献

The distribution of viral genome in the tissues of juvenile European seabass (Dicentrarchus labrax) during the course of a Red Spotted Grouper Nervous Necrosis Virus (RGNNV) infection has not yet been described. The present study addresses this and indicates which target organs may be involved in viral replication. This information should enable more accurate detection of virus in asymptomatic carriers, and in turn help to control the spread of the disease. The aim of this study was to examine the pattern of expression of viral genomic segments RNA1 and RNA2, using two absolute real-time PCRs (RT-qPCR), over the course of a RGNNV infection after administering the virus by intramuscular injection. In situ hybridization was also used to locate the RNA2 viral segment in different organs throughout the infection. The experimental challenge provoked an acute form of viral nervous necrosis (VNN), with a resulting cumulative mortality of 37%. The RT-qPCRs designed allowed the detection of both genomic segments in all the organs tested (nervous and non-nervous tissues) at all sampling times examined. The highest viral RNA copy number was found in eyes, although viral replication appeared to begin in the brain. Viral replication was also recorded in pooled internal organs and in caudal fin. However, the increase in the viral RNA copy number in these organs did not result in an increased viral titre, which may indicate that a productive infection does not take place in non-nervous tissues, possibly due to a failure in a viral post-replication step. 相似文献
