A study was conducted to determine the water stress effect on yield and some physiological parameters including crop water stress index for drip irrigated second crop watermelon. Irrigations were scheduled based on replenishment of 100, 75, 50, 25, and 0% soil water depletion from 90 cm soil depth with 3-day irrigation interval. Seasonal crop evapotranspiration (ET) for I100, I75, I50, I25, and I0 were 660, 525, 396, 210, and 70 mm in 2003 and 677, 529, 405, 221, and 75 mm in 2004. Fruit yield was significantly lowered by irrigation water stress. Average water-yield response factor for both of the years was 1.14. The highest yield was obtained from full irrigated treatment as 34.5 and 38.2 t ha−1 in 2003 and 2004, respectively. Lower ET rates and irrigation amounts in water stress treatments resulted in reductions in all measured parameters, except water-soluble dry matter concentrations (SDM). Canopy dry weights, leaf relative water content, and total leaf chlorophyll content were significantly lowered by water stress. Yield and seasonal ET were linearly correlated with mean CWSI values. An average threshold CWSI value of 0.17 before irrigation produced the maximum yield and it could be used to initiate the irrigation for watermelon.  相似文献

Simple sequence repeats (SSRs) defined as sequence repeat units between 1 and 6 bp occur abundantly in both coding and non-coding regions in eukaryotic genomes and these repeats can affect gene expression. In this study, ESTs (expressed sequence tags) of Betula pendula (silver birch) were analyzed for in silico mining of EST-SSRs, protein annotation, open reading frames (ORFs), designing primers, and identifying codon repetitions. In B. pendula, the frequency of ESTs containing SSRs was 7.8 % with an average of 1SSR/4. 78 kb of EST sequences. A total of 188 SSRs was identified by using MISA software and di-nucleotide SSR motifs (65.9 %) were found to be the most abundant type of repeat motif followed by tri- (27.1 %), tetra- (4.8 %), and penta- (2.2 %) motifs. Based on ORF analysis, 175 of 178 sequences were predicted as ORFs and the most frequent SSRs were detected in 5′ UTR (58.43 %), followed by in ORF (31.46 %) and in 3′ UTR (8.43 %). 102 of 178 ESTs were annotated as ribosomal protein, transport protein, membrane protein, carrier protein, binding protein, and transferase protein. For a total of 102 SSRs (57.3 %) with significant matches, a set of 102 primers (100 %) with forward and reverse strands was designed by using Primer3 software. Serine (Ser, 19.6 %) was predominant in putative encoded amino acids and most of amino acids showed nonpolar (35.3 %) nature. Our data provide resources for B. pendula and can be useful for in silico comparative analyses of Betulaceae species, including SSR mining.  相似文献

The size of a fish is an important factor in its physiology, and metal uptake is affected by animal physiology. In this study, small and large tilapias (Oreochromis niloticus) differing approximately twofold in length and fivefold in weight were compared for their antioxidant response. Both groups were exposed to Cu or Cr (1.0 μg/mL) in a freshwater (?80 mg CaCO3/L, conductivity 1.77 mS/cm) using 2 exposure protocols (20 μM for 48 h and 10 μM for 6 days). Following the exposures, the antioxidant enzyme activities (superoxide dismutase, SOD; catalase, CAT; glutathione peroxidase, GPX; glutathione reductase, GR and glutathione S-transferase, GST) and glutathione (GSH) levels were measured in the liver of fish. Results showed that small fish was affected from exposure conditions much more than large ones as their antioxidant parameters significantly decreased even in controls. Metal exposures of small fish caused significant increases in SOD and CAT activity in acute Cu or Cr exposures. Subchronic Cr exposure of small fish also caused significant increases in CAT, GPx and GST activities, while there was no significant change in Cu-exposed ones. Large fish, however, showed different antioxidant responses as their levels mostly decreased. This study demonstrated that the response of antioxidant system in the liver of tilapia varied in relation to fish sizes and emphasized using different size groups in environmental monitoring and also in evaluation of fish biomarkers.  相似文献

This study investigated the bacterial agents causing sub-clinical mastitis and the mean somatic cell counts (SCC) of milk in Pirlak sheep at mid-lactation. The percentage of infected udder halves was 11.4% (53/464). The most frequently isolated species were coagulase-negative staphylococci (CNS) (64.2%), followed by Staphylococcus aureus (24.5%) and Escherichia coli (11.3%). Among the CNS, the most common species was Staphylococcus epidermidis (38.2%). The other species isolated from milk samples were Staphylococcus xylosus (17.7%), Staphylococcus chromogenes (14.7%), Staphylococcus simulans (8.8%) and Staphylococcus hyicus (8.8%). The mean SCC for culture positive and negative samples was 1742×10(3) and 161×10(3) cells/ml, respectively. A significant difference (p<0.05) was determined between with and without microbial growth groups in terms of the SCC values. Threshold limit for SCC was 374×10(3) cells/ml for Pirlak sheep. In conclusion, it was considered that SCC is an important predictor of sub-clinical mastitis in Pirlak sheep. This is the first study to describe the bacterial agents causing sub-clinical mastitis and threshold limit for SCC in Pirlak sheep in Turkey.  相似文献

This study was carried out to investigate the protective effects of chitosan nanoparticles (CNP) against hydrogen peroxide (H2O2)-induced oxidative damage in murine macrophages RAW264.7 cells. After 24 h pre-incubation with CNP (25–200 μg/mL) and chitosan (CS) (50–200 μg/mL, as controls), the viability loss in RAW264.7 cells induced by H2O2 (500 μM) for 12 h was markedly restored in a concentration-dependent manner as measured by MTT assay (P < 0.05) and decreased in cellular LDH release (P < 0.05). Moreover, CNP also exerted preventive effects on suppressing the production of lipid peroxidation such as malondialdehyde (MDA) (P < 0.05), restoring activities of endogenous antioxidant including superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) (P < 0.05), along with increasing total antioxidant capacity (T-AOC) (P < 0.05). In addition, pre-incubation of CNP with RAW264.7 cells for 24 h resulted in the increase of the gene expression level of endogenous antioxidant enzymes, such as MnSOD and GSH-Px (P < 0.05). At the same concentration, CNP significantly decreased LDH release and MDA (P < 0.05) as well as increased MnSOD, GSH-Px, and T-AOC activities (P < 0.05) as compared to CS. Taken together, our findings suggest that CNP can more effectively protect RAW264.7 cells against oxidative stress by H2O2 as compared to CS, which might be used as a potential natural compound-based antioxidant in the functional food and pharmaceutical industries.  相似文献

In 2005 and 2006, a study was conducted to determine the effect of subsurface and surface drip irrigation systems and to determine optimum irrigation water using six different irrigation levels imposed on muskmelon (Cucumis Melo L. cv. Ananas F1) under semi-arid climatic conditions. Irrigation treatments received 0, 25, 50, 75, 100, and 125% of class A pan evaporation rates. In 2005, average yield from subsurface and surface drip irrigation systems ranged from 16.2 (I 0) to 31.1 (I 75) t ha−1 and from 16.2 (I 0) to 43.8 (I 75) t ha−1, respectively. While in 2006, fruit yields for the same systems ranged from 8.2 (I 0) to 40.4 (I 75) t ha−1 and from 8.2 (I 0) to 38.9 (I 100) t ha−1. Regression analysis of the yield data indicated no significant (P > 0.05) difference between years and irrigation systems. The highest muskmelon yields from subsurface and surface drip irrigation systems were obtained at 83 and 92% of class A pan. Bigger fruits were obtained with optimum irrigation amounts for both of the irrigation systems. However, there was no clear indication of irrigation water amounts on total soluble solid and flesh thickness of muskmelon fruits.  相似文献

The dyeability of poly(lactic acid) (PLA) fiber strongly depends on disperse dye structure due to the low dyeing temperature and the short dyeing time. Thus, the dye uptake value of PLA fiber is low for some disperse dyes and is needed to be improved. In the current study, the dyeability of PLA fiber is improved with the addition of N-Phenylaminopropyl polyhedral oligomeric silsesquioxane (AP-POSS) during melt spinning process. The effects of dyeing conditions including dyeing temperature and time, disperse dye type and AP-POSS concentrations are investigated on the dyeability properties of PLA fiber samples. The tensile, thermal and morphological properties of fiber samples are also characterized by tensile testing, differential scanning calorimeter (DSC), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). As the added amount of AP-POSS increases, the percent crystallinity increases and the tensile strength reduces. According to the dyeing results, AP-POSS is very effective for increasing the dyeability of PLA fiber especially for disperse dyes with low dye uptake values.  相似文献

Yeast-leavened dough processing is semicontinuous due to the requirement for fermentation at constant temperature and humidity. Also, new regulations on the emission of alcohols are becoming burdensome on the baking industry. Extrusion processing of dough with supercritical carbon dioxide (SC-CO(2)) is envisioned to alleviate emission problems and to decrease production time by eliminating fermentation. A bread dough formulation with 50% (w/w) moisture was leavened by injecting 1.5% (w/w) SC-CO(2) in a twin-screw extruder at 37 degrees C. Specific mechanical energy input was 260 kJ/kg. The operating apparent shear rate range was 60-260 s(-1). SCFX-leavened dough density (420-430 kg/m(3)) was in good agreement with values reported for similar doughs. The flow behavior index, obtained by an on-line slit rheometer, was 0.49 for the nonleavened control and 0.63 for the SCFX-leavened dough. Apparent viscosity of the SCFX-leavened dough varied from 37 to 23 Pa-s. This new continuous process offers attractive possibilities for industrial applications if further developed.  相似文献
