Whole milk powder with an initial water content of 4.4% (w/w) and a water activity of 0.23 stored in hermetically sealed vials for up to 147 days below (37 and 45 degrees C) and above (55 degrees C) the glass transition temperature (T(g) determined to have the value 48 degrees C) showed a strong temperature dependence for quality deterioration corresponding to energies of activation close to 200 kJ/mol for most deteriorative processes. The glass transition was found not to cause any deviation from Arrhenius temperature dependence. Lactose crystallization, which occurred as a gradual process as monitored by isothermal calorimetry, is concluded to liberate bound water (a(w) increase to 0.46) with a modest time delay (approximately 2 days at 55 degrees C) and with concomitant surface browning as evidenced by an increasing Hunter b-value. Browning and formation of bound hydroxymethyl-furfural determined by HPLC seem to be coupled, while formation of another Maillard reaction product, furosine, occurred gradually and was initiated prior to crystallization. Initiation of lipid oxidation, as detected by lipid-derived radicals (high g-value ESR spectra), and progression of lipid oxidation, as detected by headspace GC, seem not to be affected by lactose crystallization and browning, and no indication of browning products acting as antioxidants could be determined.  相似文献

Induction of cytochrome P4501A (CYP1A) immunopositive protein was evaluated in the rat hepatoma cell line Fao, as a biomarker of organic pollution in extract of environmental soil samples, exposed to different sources and degrees of chemical contamination. Soil samples were collected in one area in Russia (Monchegorsk) and two areas in Southern Norway (Fiskaa and Birkenes). In addition, one reference soil sample was collected in Central Norway (Høoylandet). Contents of selected polycyclic aromatic hydrocarbons (PAHs) in the samples were also evaluated. To further evaluate the inducibility of the most potent soil extract (Fiskaa), S9 fraction of Fao cells, pretreated with this extract, was used as an activation system in the Ames Salmonella assay. The DNA adduct forming capacity of the soil extracts, analyzed by the 32P-postlabeling technique, was also investigated in Fao cells. The Fao cell line has previously been found to be a very sensitive biomonitoring system, that responds to environmentally relevant concentrations of planar model contaminants with increased level of CYP1A immunopositive protein and DNA adducts. In the present study the Fao cell line also showed its potential for use in evaluating the CYP1A inducing potency of environmental samples. All soil extracts induced CYP1A protein in the Fao cells, although the level of induction varied between the soil samples. The Fiskaa soil extract was the most potent CYP1A inducer and this extract also contained the highest level of PAHs. No significant correlation was observed between the level of the total of 16 PAHs and CYP1A protein level. However, a significant correlation was observed between CYP1A protein level and the level of Benzo[a]pyrene (B[a]P), which is a very potent CYP1A inducer. The S9 fraction of pretreated Fao cells activated B[a]P to mutagens in a concentration-dependent relationship, although the response was weak. No DNA adducts were detected in cells exposed to the soil extracts. This demonstrates the necessity of determining several biomarker parameters simultaneously as one single biomarker may fail to respond to potentially harmful compounds.  相似文献

Background

Staphylococcus aureus is commonly isolated from infected wounds both in animals and humans. It is known to be an excellent biofilm former and biofilms are present in as many as 60% of chronic wounds. Despite that the presence of biofilms in infections are common, antiseptics are usually qualified for in vivo testing according to their effect on planktonic cells. As it is well known that bacteria in biofilms are more tolerant to antiseptics than planktonic bacteria, biofilm infections can be difficult to treat. The aim of the study was to compare three different categories of antiseptics, biguanide (chlorhexidine), quaternary ammonium compound (QAC; Pyrisept) and iodine/iodophores (2% iodine liniment), with regards to efficacy in killing S. aureus in biofilm. If there was observed a difference in efficacy between these antiseptics, a second aim was to find the most effective of the three antiseptics.

Results

Large differences in the bactericidal effect of the different antiseptics against S. aureus in biofilm were observed in the present study. Iodine treatment was found to be the most effective followed by Pyrisept and chlorhexidine.

Conclusions

The bactericidal effect of the different antiseptics used in the present study was found to vary significantly against S. aureus in biofilm. The present study gives valuable knowledge with regards to selecting the antiseptics that are most likely to be successful in treating biofilm infected wounds. This study also contributes to focus attention on the importance of qualifying antiseptics based on results using biofilm bacteria rather than planktonic bacteria.  相似文献

OBJECTIVE: To validate energy intake (EI) estimated from pre-coded food diaries against energy expenditure (EE) measured with a validated position-and-movement monitor (ActiReg) in groups of 13-year-old Norwegian schoolchildren. DESIGN: Two studies were conducted. In study 1 the monitoring period was 4 days; participants recorded their food intake for four consecutive weekdays using food diaries and wore the ActiReg during the same period. In study 2 the monitoring period was 7 days; participants recorded their food intake for four consecutive days but wore the ActiReg for a whole week.Settings: Participants were recruited from grade 8 in a school in and one outside Oslo (Norway).Subjects: Forty-one and 31 participants from study 1 and 2, respectively, completed the study. RESULTS: The group average EI was 34% lower than the measured EE in study 1 and 24% lower in study 2. The width of the 95% confidence limits of agreement in a Bland-Altman plot for EI and EE varied from -0.2 MJ to 8.2 MJ in study 1 and from -2.3 MJ to 6.9 MJ in study 2. The Pearson correlation coefficients between reported energy intake and expenditure were 0.47 (P=0.002) in study 1 and 0.74 (P<0.001) in study 2. CONCLUSION: The data showed that there was substantial variability in the accuracy of the food diary at the individual level. Furthermore, the diary underestimated the average energy intake. The ability of the food diary to rank individuals according to energy intake was found to be good in one of the studies and moderate in the other.  相似文献

With the purpose of analyzing the size and composition of enzyme-unextractable proteins in differently heat-treated soybean meals, a selection of extractants was screened for their ability to extract these proteins from enzyme-unextractable residues. The largest effects were obtained with urea, urea plus beta-mercaptoethanol, and dilute alkali; the latter extracted up to 87% of the enzyme-unextractable protein. Gel permeation chromatography indicated that a large proportion of the extracted material was of high molecular weight. However, the combined results from gel electrophoresis, LC-MS, and MALDI-ToF MS showed that the extracted protein material was composed of aggregated peptides. The largest aggregates were observed in the enzymatic residues originating from meals heat-treated at high humidity. Extracted aggregates were fully degraded upon subsequent proteolytic treatment.  相似文献

Voluntary schemes that offer payments to farmers who agree to desist from certain damaging operations or carrying out environmentally sensitive activities have become an integral part of current European agricultural policy. These schemes tend to operate from the presumption of an economically rational behaviour of the landowner obtaining the grant. Analysis of landowners using the Danish subsidy scheme for field afforestation (EU regulation 2080/92) has shown that there is a need to reconsider the perception of the implementation process of the scheme on the individual farm as based only on economic rationality. By using two continua (production-nature orientation and attachment to agriculture), five different types of landowners are identified: agricultural producers, non-agricultural producers, soft farmers, countryside residents, and amenity residents. By using the typology, it is found that when nature orientation is of increasing importance, planting independently of the subsidy gradually tends to become more likely. This means that not all applicants for the afforestation scheme fit into the pattern of economically rational actors. Instead, the applicants' activities reflect a wide range of practices and values concerning afforestation which we need to acknowledge if we are to fully achieve agri-environmental policies.  相似文献

Degradation of the potential photosensitizers, riboflavin, chlorophyll, and porphyrin, in Danbo cheese by monochromatic light of wavelength 366, 436, or 546 nm was studied. Three cheeses were investigated, two conventional (16% fat and 25% fat) and one "organic" (25% fat). The effect of illumination was measured by fluorescence spectroscopy and analyzed using multiway and multivariate data analysis. Riboflavin was found to degrade only by 436 nm light, whereas chlorophylls and porphyrins also were influenced by 436 and 546 nm light. The organic cheese had the largest chlorophyll content both before and after similar light exposure, and no change in chlorophyll of this cheese was observed for any of the illumination wavelengths. Upon light exposure of the cheeses, volatile compounds were formed, as analyzed by gas chromatography-mass spectrometry (GC-MS). The relative concentrations of methyl butanoate, 1-pentanol, benzaldehyde, 2-butanone, 2-heptanone, and butyl acetate were found to weakly correlate with the surface fluorescence intensity. 1-Pentanol and the ketones are secondary lipid oxidation products, consistent with a chemical coupling between photosensitizer degradation and formation of volatile lipid oxidation products.  相似文献

Abstract

Indicators of stress, behaviour in the pen, and lesions of the integument were used as indicators of sow well-being in a comparison of group housing with electronic sow feeding [GH] and confinement in gestation crates [C] in experiment 1, which included 2 × 4 groups of 20–25 sows. Effects of earlier experience with a gestation system on the indicators of stress was investigated in experiment 2, which included 2 × 3 groups of 20–25 sows. Reaction in a novelty test and reactivity in plasma Cortisol to the procedure of blood sampling were used as indicators of stress. Behaviour in the pen as well as lesions of the integument revealed that both gestation systems were suboptimal in some manner. However, following acclimatization for 1–2 weeks, young GH sows experienced less stress than young C sows as measured by reduced fear in the novelty test. In experiment 2 this interpretation was further supported by lower reactivity in plasma Cortisol. It was concluded that sow well-being was higher for the majority of the gestation period in young group-housed animals. Thus, in spite of the inadequate feeding method, well-being of sows may be improved in group housing systems with electronic sow feeding if the system is well functioning with low levels of aggression and vulva bites. One of the major determinants of vulva biting is suggested to be the mechanism of gate control.  相似文献

Abstract

The objective was to examine behavioural and physiological reactions of cows in pens with slatted floors, when pretreatment was tethering in stalls. Ten cows were kept tethered (C), whereas 10 other cows were kept loose in pens with slatted flooring (S). Cow behaviour was observed prior to treatment, and after 4 and 8 weeks, and behaviour in two open-field tests was recorded. Serial plasma concentrations of ACTH and cortisol were measured on day 23 during a 7.5 h period. On d 24, cortisol response was measured in serial samples drawn 0.5 h prior to and for 5 h after intravenous ACTH administration. S-cows showed reduced lying duration and frequency and spent less time exploring and more time immobile in the first test. S-cows showed a slightly increased response to ACTH-injection, otherwise pituitary-adrenal axis reactions did not differ between treatment groups.  相似文献

A necessary pre-requisite for variable rate application of plant nutrients to manage inputs in a site-specific manner is a technique to detect nutrient stresses in plants in real-time. Spectral reflectance of plant canopy may provide a non-destructive and rapid technique to detect nutrient and water stress in plants. In this study, we investigated the effect of water on nutrient stress discrimination (nitrogen, N; phosphorus, P; and potassium, K) based on the visible and near infrared reflectance of maize leaves grown under controlled greenhouse conditions. The analysis of the spectral data indicated that it was challenging to detect the nutrient deficiencies if the plant water status was unknown. However, if a priori knowledge of water stress existed, such as a well-watered situation, the ability to discriminate nutrient stress improved significantly. The analysis of whole plant behaviour (i.e., mean reflectance data from all leaves within a plant) versus leaves located at a specific location within a plant indicated that knowledge of spatial location of leaves within a plant helped to identify N and P stress and NPK as a multi-stress condition more clearly.  相似文献
