Background

The aim of this study was to fabricate the poly caprolactone (PCL) aligned nanofiber scaffold and to evaluate the survival, adhesion, proliferation, and differentiation of rat hair follicle stem cells (HFSC) in the graft material using electrospun PCL nanofiber scaffold for tissue engineering applications.

Methods

The bulge region of rat whisker was isolated and cultured in DMEM: nutrient mixture F-12 supplemented with epidermal growth factor. The morphological and biological features of cultured bulge cells were observed by light microscopy using immunocytochemistry methods. Electrospinning was used for production of PCL nanofiber scaffolds. Scanning electron microscopy (SEM), 3-(4, 5-di-methylthiazol- 2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay, and histology analysis were used to investigate the cell morphology, viability, attachment and infiltration of the HFSC on the PCL nanofiber scaffolds.

Results

The results of the MTT assay showed cell viability and cell proliferation of the HFSC on PCL nanofiber scaffolds. SEM microscopy images indicated that HFSC are attached, proliferated and spread on PCL nanofiber scaffolds. Also, immunocytochemical analysis showed cell infiltration and cell differentiation on the scaffolds.

Conclusion

The results of this study reveal that PCL nanofiber scaffolds are suitable for cell culture, proliferation, differentiation and attachment. Furthermore, HFSC are attached and proliferated on PCL nanofiber scaffolds.Key Words: Nanofiber, Electrospinning, Stem cells, Tissue engineering  相似文献

Lentil (Lens culinaris Medik.) is an important grain legume crop, mostly grown in semi-arid environments and often faces intermittent drought spells during different growth stages, which severely hamper its yield. This study, comprising of three separate experiments, was conducted to evaluate the potential of seed priming with CaCl2 in improving drought tolerance in lentil. In the first experiment, lentil seeds were hydroprimed (water) or osmoprimed with 0.5 and 1% CaCl2; while non-primed seeds were taken as control. In the second and third experiments, lentil seeds were subjected to pre-optimized osmopriming (1% CaCl2) and hydropriming followed by surface drying or re-drying of primed seeds to original weight. The first two experiments were conducted in petri plates, while, in experiment 3, seeds were planted in plastic pots containing peat moss, maintained at 75% water holding capacity (WHC; well-watered) or 50% WHC (water deficit). Hydropriming and osmopriming improved seed germination, seedling growth, biomass production, chlorophyll intensity, sugar accumulation and reduced the oxidative stress in lentil under water deficit. However, osmopriming (1% CaCl2) was more effective than the hydropriming in improving the lentil growth, biomass production, Ca accumulation and sugar metabolism under both well-watered and water deficit conditions. Seed surface drying, after priming, was more beneficial in improving the lentil performance, under both well-watered and water deficit conditions, than re-drying to original weight. Osmopriming (1% CaCl2) increased the seeding dry weight (67%), SPAD value (140%), leaf Ca concentration (56%), α-amylase activity (55%), total soluble sugars (48%) and reduced malanodialdehyde content (35.9%) and total antioxidant activity (29.2%) than un-primed seeds under water deficit. In conclusion, osmopriming improved the lentil performance under optimal and water deficit conditions through early and synchronized emergence, better sugar and Ca accumulation which reduced the oxidative damage and resulted in better seedling growth and biomass production.  相似文献

AIM: To evaluate the relationship between dietary diversity score (DDS) and cardiovascular risk factors in Tehranian adults. METHODS: In this population-based cross-sectional study, a representative sample of 581 subjects (295 males and 286 females) aged over 18 years, residents of Tehran, participated. Hypercholesterolaemia, hypertriglyceridaemia and low high-density lipoprotein cholesterol were defined according to the Adult Treatment Panel III guidelines of the National Cholesterol Education Program. Diabetes was defined as fasting plasma glucose concentration > or = 126 mg dl(-1) or 2-h post challenge glucose concentration > or = 200 mg dl(-1). Hypertension was defined on the basis of the sixth report of the Joint National Committee on Prevention, Detection, Evaluation and Treatment of High Blood Pressure. Dietary diversity was defined according to the Diet Quality Index revised. RESULTS: Mean (+/- standard deviation) DDS was 6.15 +/- 1.02. The probability of having diabetes (odds ratio (OR) among quartiles: 1.45, 1.26, 1.11 and 1.00, respectively; P for trend = 0.04) and hypertriglyceridaemia (OR = 1.41, 1.23, 1.05 and 1.00, respectively; P for trend=0.04) decreased with increasing quartile of the diversity score for whole grains. The probability of having obesity (OR among quartiles: 1.39, 1.06, 1.03 and 1.00, respectively; P for trend = 0.03), hypercholesterolaemia (OR = 1.46, 1.28, 1.11 and 1.00, respectively; P for trend = 0.03), hypertension (OR = 1.32, 1.17, 1.13 and 1.00, respectively; P for trend = 0.03) and high low-density lipoprotein cholesterol (LDL-C) (OR = 1.25, 1.12, 1.07 and 1.00, respectively; P for trend = 0.04) decreased with increasing quartile of the diversity score for vegetables. The probability of having hypercholesterolaemia, high LDL-C, hypertension and diabetes decreased with quartile of the DDS. But the probability of being obese increased with quartile of the DDS (P for trend = 0.03). CONCLUSION: DDS was inversely associated with cardiovascular risk factors in this cross-sectional study. Increased diversity scores of diets, to increase the variety score for vegetables, may be emphasised in programmes attempting to bring about changes in lifestyle.  相似文献

For developing nature-based silvicultural practices in a beech forest to promote, abundance, height,vitality, and preferred growth form, regenerated trees growing in gaps were compared with those under closed canopies.A systematic 50×50 m grid was plotted in a beech stand in the Kheyrud Experimental Forest for selecting trees to measure variables in gaps and under closed canopies.Abundance and mean height of regenerated beech trees were significantly higher in closed canopies than in canopy gaps.Beech seedlings with excurrent growth were significantly taller within regeneration patches under closed canopy.Moreover, regenerated trees with high vitality were more abundant in closed canopy areas than in gaps.Thus, beech regeneration should improve under closed canopies; hence, gaps in a near natural forest should be created only after adequate regeneration and appropriate growth under the parent tree in a closed canopy area is ensured.The results of this research have profound implications for the sustainable management of the forest and for ensuring sustainable beech regeneration.The presence of a closed canopy cover likely will reduce potential stresses on oriental beech regeneration.  相似文献

In a context of the global major changes, it is mandatory to enlarge the range of crops supporting food security and pay great attention to neglected and underutilized species. However, basic knowledge of the biology of many neglected and underutilized species is still lacking to increase their yields. In this study, the mating system of white fonio [Digitaria exilis (Kippist.) Stapf], a West African minor and promising cereal, is assessed. Progenies arrays from both homozygous and heterozygous mothers were genotyped with microsatellites markers. The rate of genotyping errors in the experiments was assessed and a likelihood framework was used to determine the probability of different mating systems: outcrossing, self-fertilization and apomixis. The results suggested that white fonio has a highly selfing reproductive system with a possible outcrossing rate of 1.7%. Understanding the reproduction system of white fonio opens up opportunities for more effective breeding programs and a wider use of this cereal for food security improvement.  相似文献

Ameliorative effect of silicon (Si) (2 mM as sodium silicate (Na2SiO3)) was studied in tobacco (Nicotiana rustica L.) plants grown under control at 100% field capacity (FC), mild drought (60% FC), and severe drought (30% FC) conditions. Si-treated plants had higher biomass of particularly above-ground parts both under drought and control conditions. Plants with Si supply had significantly higher net assimilation rates but lower transpiration rates. Silicon supply enhanced osmotic potentials only in the leaves, but not in the roots. A considerable rise in the concentrations of soluble sugars was observed particularly in the leaves under both drought and Si treatments. Soluble proteins, free α-amino acids, and proline concentrations increased in Si-treated plants under all watering treatments. Si enhanced the activity of antioxidative enzymes and decreased hydrogen peroxide (H2O2) concentrations. Results indicate that Si supplementation alleviates drought stress via improvement of water relation parameters, enhancement of photosynthesis, and elevation of antioxidant defenses.  相似文献

A 12-week incubation experiment was conducted to determine the pattern and rate of N mineralized from organic materials. Treatments consisted of sheep manure (SM), cattle manure (CM), poultry manure (PM), sewage sludge (SS) at 1% (W/W) level, and unfertilized treatment with three replications. The concentrations of nitrate (NO3)- nitrogen (N) and ammonium (NH4)-N were determined in day 1 and 1, 2, 4, 8, and 12 weeks after the beginning of incubation. Results indicated that the magnitude of N mineralized during the incubation time periods was in the order of CM (134 mg kg?1) > PM (83 mg kg?1) > SS (56 mg kg?1) > SM (55 mg kg?1), and different management is required for obtaining optimum N-use efficiency. In conclusion, improving N-uptake efficiency in manure- and SS-amended soils depends on the pattern and rate of N mineralization to synchronize N released with crop N demand periods.  相似文献

This study investigated the effects of wood-derived biochar (BC) applied at 1% to a C-poor silty-loam soil in the drought-tolerant (D24) and in the drought-sensitive (P1921) Pioneer Hi-Bred maize hybrids in pot and field trials (NE Italy). D24 had better growth than P1921 under rain-fed conditions without irrigation and soil amendment. The addition of biochar increased root growth in D24 (+38% root area) and decreases it in P1921 (?9%) at the silking stage, while the fraction of finer roots (<250 µm diam.) was reduced in D24 and increased in P1921. This led both hybrids to maintain the maximum transpiration at a lower fraction of transpirable soil water (from 82% to 45% in D24, and from 46% to 22% in P1921). There were no significant variations in plant nutrient contents, productivity and in the protein and starch contents of the grains, whereas the lipid content was reduced by biochar, particularly in P1921 (2.6% vs. 3% DW, ?13%).

We conclude that biochar can be profitably used to enhance drought tolerance in maize, possibly due to improvements in the physicochemical characteristics and the water content of treated soils, although maximum benefits are expected in drought-tolerant hybrids through increased root elongation and transpiration.  相似文献

Bread staling involves a combination of physico-chemical phenomena that leads to a reduction of quality. This study aims at evaluating the impact of baking conditions (280 °C, 8 min; 310 °C, 5.5 min; 340 °C, 4 min), baking type (of fully baked (FB) and part-baked (PB)) and storage temperature (−18, 4 and 20 °C) on the staling of Sangak bread. Results showed that lower baking temperature with longer baking time produced drier bread with higher firmness. In FB Sangak breads, amylopectin retrogradation, amount of unfreezable water and firmness (measured by compression test) increased during storage at positive temperatures but hardness (determined by Kramer shear test) decreased significantly during first day of storage. The recrystallized amylopectin traps the free water resulting in crumb hardening. Water is also absorbed by the dry crust resulting in changes of rheological properties in the crust and crumb, and finally in staling. Storage at 4 °C resulted in increasing melting enthalpy of amylopectin crystallite in comparison with storage at 20 °C. Also it was found that firmness of PB breads due to rebaking was significantly lower than FB breads. There were no significant changes in staling parameters of FB and PB stored at −18 °C.  相似文献
