The indirect fluorescent antibody (IFA) test for Theileria equi was evaluated to assess test's suitability for the serological diagnosis of equine piroplasmosis, to provide performance parameters for the purpose of test validation, and to compare it with the complement fixation (CF) test. Using a protocol that included Evan's blue, the specificity of the IFA test was estimated at 99.0% for T. equi by the classical method of analysis, and 96.6% by the Bayesian method. The use of Evan's blue in the test protocol increased test specificity and contributed to an excellent test agreement between two collaborating laboratories (kappa = 0.96). Using Bayesian analysis, the sensitivity estimate for the IFA test was 89.2%. The CF test sensitivity and specificity estimates for T. equi were 63.1 and 96.4%, respectively, as determined by Bayesian analysis. The IFA test was more sensitive than the CF test but the specificity estimates were similar.  相似文献

A study of the aquatic fate of the triethylamine salt of triclopyr (3,5,6‐trichloro‐2‐pyridinyloxyacetic acid) was conducted in three bays of Lake Minnetonka, Minnesota. Triclopyr is under review by the US Environmental Protection Agency as a selective aquatic herbicide. The primary purpose of this study was to determine dissipation rates of the parent active ingredient, triclopyr, and its major metabolites, 3,5,6‐trichloropyridinol (TCP) and 3,5,6‐trichloro‐2‐methoxypyridine (TMP) in selected matrices including water, sediment, plants, finfish and shellfish. Two 6.5‐ha plots dominated by the weedy species Eurasian watermilfoil (Myriophyllum spicatum L) were treated with triclopyr‐triethylammonum at a rate of 2.5 mg AE liter−1 (2.5 ppm) on 21–23 June 1994. A third 6.5‐ha plot was established as an untreated reference. Water and sediment samples were collected from within the plots and at selected locations up to 1600 m outside of the plots through six weeks post‐treatment for chemical residue analysis. In addition, residue samples were collected from the target and non‐target plants and other non‐target matrices, including game and rough fish, clams and crayfish. All test animals were sequestered in cages located in the center of each plot and samples were collected through four weeks post‐treatment. Half‐lives for dissipation of triclopyr and TCP in water ranged from 3.7 to 4.7 days and from 4.2 to 7.9 days, respectively, with trace amounts of TMP found. Peak triclopyr sediment values ranged from 257 to 335 ng gram−1, with a mean half‐life of 5.4 days, while peak TCP sediment levels ranged from 27 to 65 ng gram−1 (mean half−life = 11.0 days). Trace levels of TMP were detected at one treatment site at one sampling event. Triclopyr and TCP accumulated and cleared from animal tissues proportionately to concentrations in the water (triclopyr dissipation half‐lives <11 days, TCP < 14 days). TMP levels were two to three times higher than those of the other compounds, particularly in visceral tissue. In all cases, residues of these compounds were higher in the inedible portions of the animals, and were usually higher in bottom‐feeding fish species. © 2000 Society of Chemical Industry  相似文献

The first evidence of proliferative kidney disease (PKD) in an Austrian river (the River Kamp) was documented in 2016, and no information on the PKD infection status of trout in other rivers was available. Since then, brown trout (Salmo trutta fario) and rainbow trout (Oncorhynchus mykiss) have been collected from rivers in Upper and Lower Austria for different diagnostic purposes. In this study, we summarize the recent findings of a first survey concerning the distribution of Tetracapsuloides bryosalmonae, the causative agent of proliferative kidney disease (PKD), from these samples. Between September 2015 and October 2017, a total of 280 brown trout and 39 rainbow trout were collected from 21 rivers in the provinces of Upper and Lower Austria. T. bryosalmonae was detected by PCR of kidney tissue in 17 of 21 sampled rivers and in 138 of 280 brown trout as well as in 11 of 39 rainbow trout. Pathological signs of PKD (e.g., hypertrophy of the kidney) were observed in 33 analysed brown trout and six rainbow trout samples. No correlations between fish infected by Tbryosalmonae and the parameters size and age class, condition factor, geological origin of the streams and distribution within the river course were found, while positively tested fish are significantly increased at sampling sites exceeding water temperatures of 15°C for median periods of 115 days. The prevalence within the affected streams or stream sections is highly variable, and in single rivers, infection rates of up to 90% are confirmed.  相似文献

Ammonia (NH3) volatilization is an important N loss pathway in intensive agriculture of the North China Plain (NCP). Simulation models can help to assess complex N and water processes of agricultural soil–crop systems. Four variations (Var) of a sub‐module for the deterministic, process‐based HERMES model were implemented ranging from simple empirical functions (Var 3 and 4) to process‐oriented approaches (Var 1 and 2) including the main processes of NH3 volatilization, urea hydrolysis, nitrification from ammonium‐based N fertilizer, and changes in soil solution pH. Ammonia volatilization, plant growth, and changes in ammonium and nitrate pools in the soil over several winter wheat–summer maize double‐crop rotations at three locations in the NCP were simulated. Results were calibrated with two data sets (Dongbeiwang 1, Shunyi) and validated using two data sets (Dongbeiwang 2, Quzhou). They showed that the ammonia volatilization sub‐module of the HERMES model worked well under the climatic and soil conditions of N China. Although the simpler equations, Var 3 and 4, showed lower deviations to observed volatilization across all sites and treatments with a mean absolute error (MAE) of 1.8 and 1.4 in % of applied N, respectively, compared to process‐oriented approaches, Var 1 and 2, with a MAE of 2.2 and 1.9 in % of applied N, respectively. Environmental conditions were reflected better by the process‐oriented approaches. Generally, simulation results were satisfying but simulated changes in topsoil pH need further verification with measurements.  相似文献

Distillers dried grains with solubles (DDGS), the major coproduct from the corn‐based fuel ethanol industry, is primarily used as livestock feed. Due to high protein, fiber, and energy contents, there is a high demand for DDGS. Flowability of DDGS is often hindered due the phenomenon of caking. Shipping and handling of DDGS has thus become a major issue due to bridge formation between the DDGS particles. The objective of this investigation was to measure flowability characteristics of DDGS samples from five ethanol plants in the north central region of the United States. Carr and Jenike tests were performed and the resulting data were mathematically compared with a previously developed empirical model. The largest particles had an average geometric mean diameter (GMD) of 1.19 mm, while the lowest particle size had an average GMD of 0.5 mm. Soluble solid levels were ≈10.5–14.8% (db). The effective angle of friction (δ) was 43.00–57.00°. Additionally, a few parameters exhibited fairly high linear correlations, including aerated and packed bulk densities (r = 0.97), geometric standard deviation and Carr compressibility (r = 0.71), geometric standard deviation and Hausner ratio (r = –0.70). Overall flowability assessment indicated that the commercial DDGS samples did have the potential for flow problems, although no samples exhibited complete bridging. Quantifying DDGS flowability is a necessary step toward overcoming this logistical challenge facing the fuel ethanol industry.  相似文献

Distillers dried grains with solubles (DDGS) is a widely used animal feed. But transportation of DDGS is often troublesome because of its stickiness. DDGS is formed by combining condensed distillers solubles (CDS) with distillers wet grains (DWG) and then drying. As a first step toward understanding drying behavior, this study's objective was to investigate batch‐drying kinetic behavior of DWG with three CDS addition levels (10, 15, and 20% wb) and three drying‐temperature levels (100, 200, and 300°C). Multiple nonlinear mathematical models were used to fit experimental drying data for moisture content versus drying rate. A new comprehensive model was developed (R2 = 0.89, SEM = 18.60) from a modified Chen and Douglas model to incorporate CDS and drying‐temperature terms. Drying temperature affected drying rate more significantly than did changes in CDS level; thus, drying temperature was the main effect and CDS was a subeffect. Increasing the drying temperature increased the drying rate significantly for all levels of CDS addition. This model can be used for predicting DWG drying behavior under broad operating conditions; it can be used to help the industry produce better DDGS, which may thus result in better DDGS handling and transport characteristics.  相似文献

Neural network (NN) modeling techniques were used to predict flowability behavior of distillers dried grains with solubles (DDGS) prepared with varying levels of condensed distillers solubles (10, 15, and 20%, wb), drying temperatures (100, 200, and 300°C), cooling temperatures (–12, 25, and 35°C), and storage times (0 and 1 month). Response variables were selected based on our previous research results and included aerated bulk density, Hausner ratio, angle of repose, total flowability index, and Jenike flow index. Various NN models were developed using multiple input variables in order to predict single‐response and multiple‐response variables simultaneously. The NN models were compared based on R2, mean square error, and coefficient of variation obtained. In order to achieve results with higher R2 and lower error, the number of neurons in each hidden layer, the step size, the momentum learning rate, and the number of hidden layers were varied. Results indicate that for all the response variables, R2 > 0.83 was obtained from NN modeling. Compared with our previous studies, NN modeling provided better results than either partial least squares modeling or regression modeling, indicating greater robustness in the NN models. Surface plots based on the predicted values from the NN models yielded process and storage conditions for favorable versus cohesive flow behavior for DDGS. Modeling of DDGS flowability using NN has not been done before, so this work will be a step toward the application of intelligent modeling procedures to this industrial challenge.  相似文献

A 15 year-old Thoroughbred mare was examined for lethargy, fever, and inappetence of 1-day duration. A hard-bodied tick was removed from the horse. A complete blood count (CBC) demonstrated leukopenia with lymphopenia and thrombocytopenia. Morulae were visualized in circulating granulocytes. A polymerase chain reaction (PCR) confirmed the identity of these organisms as Anaplasma phagocytophilum. The horse was treated symptomatically for fever and inappetence with flunixin meglumine (1.1 mg/kg [0.5 mg/lb]) and oral electrolyte paste. Oxytetracycline (6.6 mg/kg [3 mg/lb] intravenously every 24 hours) treatment was begun as soon as a definitive diagnosis was determined. The mare responded to treatment, but she was switched to oral doxycycline (10 mg/kg [4.5 mg/lb] every 12 hours) after 5 days because of perivascular swelling at the injection site. Complete resolution of clinical signs was seen. There was no evidence of recurrence 1 year later. No additional horses at the farm were affected. The horse in this report presented for lethargy, inappetence, and fever, with limited other abnormalities. This represents a classical presentation of a mild to moderate case of anaplasmosis, which had not previously been reported in Virginia. The disease may be more widespread than has been previously reported, and it should warrant inclusion on a complete differential diagnosis list in a case of fever of unknown origin.  相似文献
