The effect of 1.35% isotonic sodium bicarbonate solution (ISB) administered intravenously on acid-base equilibrium was examined in 18 acidemic Japanese black beef calves with spontaneous diarrhea. The infusion volumes of ISB were decided based on the first half volumes of base needed. In 72.2% (13/18) of calves, improvement of acidemia was detected. There was good correlation (r=0.693, p<0.01) between infused volume of ISB and changes in base excess (y=1.097x + 4.762). Infusion volumes of ISB were 7.5, 10.2, 12.9 and 15.7 ml/kg, respectively, enough to correcting the first half of 5, 10, 15 and 20 mEq/l of base deficit in acidemic calves. Our finding suggested that ISB could be used to correct metabolic acidosis without altering electrolyte concentrations in calves. 相似文献

The role of CD4(+) or CD8(+) T cells in the immune response of BALB/c mice against Neospora caninum infection was examined by using anti-CD4 and/or anti-CD8 monoclonal antibodies (mAbs). Mice were intraperitoneally inoculated with anti-CD4 and/or anti-CD8 mAbs before and after infection with N. caninum and observed for 30 days after infection. Most of the anti-CD4 mAb-treated mice and all of the anti-CD4 and anti-CD8 mAbs-treated mice died within 30 days post-infection (p.i.). In contrast, 100% of PBS-treated mice and 70% of anti-CD8 mAb-treated mice survived more than 30 days. When compared with phosphate-buffered saline (PBS)-treated mice, the weight of mice treated with mAbs tended to decrease. From these results CD4(+) T cells, but not CD8(+) T cells, have an important role for protection of mice against N. caninum infection. Serum antibody levels to N. caninum in infected-mice treated with anti-CD4 mAb or a mixture of anti-CD4 and anti-CD8 mAbs were lower than those in the infected mice treated with anti-CD8 mAb or PBS. The mice treated with anti-interferon-gamma (IFN-gamma) mAb produced high antibody levels to N. caninum, but all mice died within 18 days p.i. These results indicated that IFN-gamma is an important cytokine for protection against N. caninum infection at the early stage of infection. However, since CD4(+) T cells against N. caninum were essential to the production of specific antibody, these antibodies might have important roles in host protection at the later stage of infection. 相似文献

Oocytes were recovered from bitch ovaries at various stages of the estrous cycle by the slicing method. The proportion of Grade A oocytes (darkly pigmented and surrounded in part, or whole, by dense layers of cumulus cells) were counted. Only Grade A oocytes were cultured in TCM199 supplemented with 5% fetal calf serum for evaluation of meiotic competence. There were no significant differences in the total number of oocytes or the proportion of Grade A oocytes that were recovered from bitches at various stages of the estrous cycle. Only 11% of the oocytes reached metaphase II (MII) at 72 hr after initiation of maturation culture. The regional variation in the intraepithelial lymphocytes (IELs) in the small intestine was examined in BALB/c male and female mice and C3H/He and C57BL/6 male mice. The small intestines were taken from 11 to 12-week-old mice and divided equally into 3 parts (the proximal, middle and distal parts). IELs were isolated from each part of the intestine and analyzed with flow cytometer. The number of IELs was highest in the proximal part and lowest in the distal part. The distribution of IEL subsets was markedly different between the proximal and the distal parts, and that in the middle part showed the intermediate pattern. The percentage of alphabeta T cells were higher in the distal part. In alphabeta T cell subset, the percentage of CD8alphaalpha T cells was higher in the proximal part, whereas those of CD4 and CD4CD8alphaalpha double positive T cells were higher in the distal part. In gammadelta T cell subset, no regional variations were found. The regional variations in the number and subsets of IELs showed almost the same patterns between male and female BALB/c mice and similar patterns among three strains of mice. The entire genome of bacteriophage OP1, lytic for Xanthomonas oryzae pv. oryzae causing bacterial leaf blight of rice, and the partial genomes of related phages were sequenced and analyzed. The OP1 genome comprises double-stranded, 4785-bp long DNA with 51.1% G + C content. Fifty-nine open reading frames (ORFs) were detected.
ORF25 had similarity with the tail fiber gene of phages, whose product is related to host specificity. The ORF25 regions were
amplified from four host-range mutants (OP1h, OP1hC, OP1h2, and OP1h2C) by polymerase chain reaction, and their deduced amino acid sequences were compared. Three mutants (OP1hC, OP1h2, OP1h2C) had duplications of a small domain in the N-terminal portion, although there were slight differences in the position of the duplicated sequences. One mutant OP1h had substituted amino acids in the duplication region. New mutants isolated in the laboratory (OP1hC and OP1h2C from OP1 and OP1h2) acquired the ability to lyse strain N5874 belonging to phagovar (lysotype) C. However, they rapidly lost this lytic ability
when incubated with other phagovars. This loss was always accompanied by a loss of the characteristic repeats, suggesting
that the host range of OP1-related phages changed mainly through duplication and deletion of a small domain in ORF25.

− from isolated pea cell walls was enhanced by a 70-kDa glycoprotein elicitor but was suppressed by mucin-type glycopeptide
suppressors (supprescins A and B) prepared from pycnospore germination fluid of Mycosphaerella pinodes, causal agent of Mycosphaerella blight of pea. Here, we show that superoxide dismutase (SOD) in the apoplast fluid/cell wall
of pea seedlings responds to the fungal elicitor and suppressor molecules. In a pharmacological study and with internal amino
acid sequencing, the apoplastic SOD in a pea cultivar Midoriusui was found to be a Cu/Zn type SOD. We cloned a full-length
cDNA of the Cu/Zn-SOD and designated it as PsCu/Zn-SOD1. An increase in PsCu/Zn-SOD1 mRNA and the PsCu/Zn-SOD1 protein was induced by treatment with the elicitor more intensively
than by wounding. Such induction by the elicitor or wounding, however, was inhibited by the concomitant presence of supprescins.
The SOD activity of recombinant PsCu/Zn-SOD1 was regulated directly by these signal molecules in a manner similar to their
effect on the SOD activity in the apoplastic fluid and in the cell wall-bound proteins. Based on these findings, we discuss
a role for PsCu/Zn-SOD1 in the pea defense response.

The timing of parturition was recorded for a total of 56 beef cattle (Japanese Black × Holstein Friesian) on different dietary treatments. The rate of calving during daylight hours in cows night‐fed (18.00 hours) with a roughage diet was significantly higher than that in cows night‐fed with a high concentrate diet (79.2% vs 38.5%, P < 0.05). Subsequently, the vaginal temperature (VT) of these cows was analyzed using a cosinor method. When the feeding schedule was changed from twice daily (08.30 and 15.30 hours) to night feeding, the periodicity, the acrophase and the bathyphase, which were the parameters of the cosine curve, were unstable from the first day of night feeding until after day 6 (P < 0.05). Prior to parturition, the midline‐estimating statistic of rhythm (MESOR) and the amplitude for the cows that were fed a high‐roughage diet at night and that calved at night‐time were lower and larger, respectively, than that for the other treatments (P < 0.01). Based on these results, the time of parturition in most of the beef cows was influenced by feeding time and diet composition. Those cows that calved at night‐time in spite of night feeding had lower vaginal temperatures. 相似文献

We induced various flower morphologies in torenia (Torenia fournieri Lind.) by the application of forchlorfenuron (CPPU). Those morphologies were the combination of four basic morphological changes, the development of serrate petals, incised petals, a paracorolla, and an increased number of floral organs. These morphological changes occurred systematically depending on the floral stage at the time of CPPU application. Serrate petals were induced when CPPU was applied during the stages of corolla development, whereas application at younger stages induced petal incision. The serrate petal margin resulted from preferential proliferation of cells around the vascular bundles, whereas petal incision likely resulted from the lateral outgrowths of petal. A paracorolla was induced at the adaxial petal face when CPPU was applied between the sepal development stage and early corolla development. The paracorolla appears to have arisen from the lateral outgrowths of the stamen. The numbers of stamens, petals, and sepals increased when CPPU was applied at and before the differentiation of sex organs and the corolla. Enlargement of the floral meristem probably caused this increase. Application of N6-benzylaminopurine and zeatin did not induce these morphological changes. 相似文献

Root and stem rot with wilt of above ground parts of cultivated chrysanthemums was first found in Ibaraki, Toyama and Kagawa
prefectures, Japan in 2002 and 2003. Pythium species were isolated from the diseased tissues and identified as P. dissotocum, P. oedochilum, P. sylvaticum, P. ultimum var. ultimum and asexual strains of P. helicoides based on their morphologies and sequences of rDNA-ITS region. All the Pythium species were strongly pathogenic to chrysanthemums in pot conditions and were reisolated from the inoculated plants. Because
Pythium root and stem rot of chrysanthemum has never been reported in Japan, we propose that this is a new disease that can
