Obesity is a serious international health problem that increases the risk of several common diseases. The genetic factors predisposing to obesity are poorly understood. A genome-wide search for type 2 diabetes-susceptibility genes identified a common variant in the FTO (fat mass and obesity associated) gene that predisposes to diabetes through an effect on body mass index (BMI). An additive association of the variant with BMI was replicated in 13 cohorts with 38,759 participants. The 16% of adults who are homozygous for the risk allele weighed about 3 kilograms more and had 1.67-fold increased odds of obesity when compared with those not inheriting a risk allele. This association was observed from age 7 years upward and reflects a specific increase in fat mass.  相似文献

ABSTRACT Genetic control of avirulence in the net blotch pathogen, Pyrenophora teres, was investigated. To establish an appropriate study system, a collection of 10 net form (P. teres f. teres) and spot form (P. teres f. maculata) isolates were evaluated on a set of eight barley lines to identify two isolates with differential virulence on an individual host line. Two net form isolates, WRS 1906, exhibiting avirulence on the cv. Heartland, and WRS 1607, exhibiting high virulence, were mated and 67 progeny were isolated and phenotyped for reaction on Heartland. The population segregated in a 1:1 ratio, 34 avirulent to 33 virulent (chi(2) = 0.0, P = 1.0), indicating single gene control of WRS 1906 avirulence on Heartland. Bulked segregant analysis was used to identify six amplified fragment length polymorphism markers closely linked to the avirulence gene (Avr(Heartland)). This work provides evidence that the P. teres-barley pathosystem conforms to the gene-for-gene model and represents an initial step toward map-based cloning of this gene.  相似文献

Lobster phyllosoma are known to associate with large cnidarian medusae; however, direct quantitative observations are difficult because gelatinous zooplankton are extremely fragile, and the phyllosoma easily detach from their host when sampled by plankton nets. We provide the first large scale quantitative information of the distribution of this association using an in situ imaging system, with synoptic measurements of water column properties. All phyllosoma were identified as slipper lobsters (Scyllarus chacei) and were associated with previously unreported “hosts” such as small hydromedusae, doliolids, and siphonophores in the northern Gulf of Mexico. Along the shelf, phyllosoma were more likely to be present at greater depths and higher salinities. Approximately 30% of the 347 lobster phyllosoma imaged were attached to at least one gelatinous organism, and salinity and depth were positively related to the probability of attachment on 30 October, but did not show a significant relationship for the other days of sampling. Many of the phyllosoma were larger than the gelatinous organisms to which they were attached, and some gelatinous zooplankton showed damage likely from feeding by the phyllosoma. In this coastal environment, gelatinous zooplankton tended to be more abundant in the same offshore region where phyllosoma occurred, so these gelatinous “hosts” may provide a steady food supply in the more oligotrophic waters on the outer shelf. Similar complex interactions among zooplankton may influence the life histories of other species, hindering our ability to forecast ecosystem level processes until the population level consequences of species interactions are fully understood.  相似文献

Understanding trade-offs associated with occupying various aquatic habitats provides a mechanistic understanding of habitat needs that can be used to evaluate the consequences of habitat loss or alteration. We used instream enclosures and field observations to identify how velocity affects the growth rates of four native species in the upper Gila River basin: longfin dace (Agosia chrysogaster) and speckled dace (Rhinichthys osculus), two species of no conservation concern, and loach minnow (Tiaroga cobitis) and spikedace (Meda fulgida), two federally endangered species. Elevated velocity was predicted to increase food delivery through drift or stimulation of benthic primary production. Energetic costs of high-velocity habitat were predicted to vary with morphology and behaviour and be lowest for speckled dace and loach minnow because they are adapted to occupy interstitial spaces of the substrate in riffles. Spikedace and longfin dace should perform best in moderate velocities, where the trade-off between exposure to drifting macroinvertebrates outweighs the energetic costs of maintaining position in the water column. Growth rates of loach minnow and speckled dace increased in higher velocities, but contrary to our initial predictions, spikedace growth rates also increased in high-velocity habitats while longfin dace grew fastest in low-velocity habitats; similar to the locations these species occupied based on field observations. These results indicate that for spikedace, the increased abundance of drifting macroinvetebrates in high-velocity habitats outweighs the energy expenditure, but for longfin dace the energetic costs of occupying moderate to high-velocity habitats outweigh the benefit to increased food availability. Our experiment provides a mechanistic understanding of habitat requirements across species and may inform predictions on how modifications or restoration of riverine ecosystems influence native fish diversity.  相似文献

The purpose of the study was to evaluate intermediate adrenal steroid hormones (ISH) in neutered dogs with hair cycle arrest (Alopecia X) during treatment with melatonin, and to see if hair re-growth is associated with sex hormone concentrations within the normal ranges. Twenty-nine neutered, euthyroid, and normo-cortisolemic dogs were enrolled in the study (23 Pomeranians, three keeshonds, two miniature poodles, and one Siberian husky). Coat assessment and an ACTH stimulation test were performed pre-treatment and approximately every 4 months for a year post treatment. Melatonin was administered initially at 3-6 mg, every 12 h. Based on clinical progression, each dog was continued on the current dose of melatonin, given an increased dose of melatonin or changed to mitotane. Partial to complete hair re-growth occurred in 14/23 Pomeranians, and partial re-growth in 3/3 keeshond and 1/2 poodle dogs. A Siberian husky dog failed to re-grow hair. Fifteen dogs had partial hair re-growth at the first re-evaluation. Melatonin dosage was increased in eight dogs but only one had improved hair re-growth. On mitotane treatment, partial to complete hair re-growth was seen in 4/6 dogs and no re-growth in 2/6 dogs. No significant decrease in sex hormone concentrations were seen during melatonin or mitotane treatment. Concentrations of ISH in dogs with hair re-growth did not differ significantly from pre-treatment values. At the completion of the study, androstenedione, progesterone and 17-hydroxyprogesterone were still above reference ranges in 21, 64 and 36%, respectively, of dogs with partial to complete hair re-growth. In conclusion, 62% of dogs had partial to complete hair re-growth. However, not all dogs with hair re-growth had concentrations of ISH within the normal range.  相似文献

Purpose

Dicyandiamide (DCD) has been used commercially in New Zealand to reduce nitrate leaching and N2O emissions in grazed pastures. However, there is a lack of information in the literature on the optimum rate of DCD to achieve the environmental benefits while at the same time reducing the cost of the technology. The objective of this study was to determine the effect of DCD application rate on its effectiveness to inhibit ammonia oxidizer growth and nitrification rate in a grazed pasture soil.

Materials and methods

The soil was a Templeton silt loam (Immature Pallic Soil; Udic Haplustepts) collected from Lincoln University Research Dairy Farm with a mixed pasture consisting of perennial ryegrass (Lolium perenne L.) and white clover (Trifolium repens L.) and was incubated alone (control) or with cow urine at 700 kg N/ha with 6 rates of DCD [0, 2.5, 5, 7.5, 10 (applied twice), 15 and 20 kg/ha] in incubation vessels. The incubation vessels were placed randomly in an incubator with a constant temperature of 12 °C. During 112 days of incubation, soil subsamples were taken at different time intervals to measure the concentrations of NO3 ?-N and NH4 +-N and the amoA gene copy numbers of ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA).

Results and discussion

DCD applied at all the different rates inhibited nitrification in urine-treated soils, but the effectiveness increased with DCD application rate. In addition, AOB growth and the amounts of nitrate-N in the soil were significantly related to the application rate of DCD. However, AOA population abundance showed no relationship to the application rate of DCD. The DCD rate at which the AOB growth rate and nitrate-N concentration were halved (effective dosage that causes 50 % reduction in nitrification rate, or ED50) was about 10 kg DCD/ha.

Conclusions

These results suggest that DCD applied at relatively low rates still slowed down the nitrification rate, and the current recommended rate of 10 kg DCD/ha for DCD use in New Zealand grazed pastures would result in a 50 % reduction in nitrification rate in this soil. The actual rate of DCD application used would depend on the cost of the product and the environmental and agronomic benefits that would result from its use.  相似文献

Sixteen crossbred beef heifers were used to determine the efficacy of serum clinical profiles as diagnostic tools for detection of early stages of locoweed toxicity and to test the ability of two mineral supplements for prevention or therapy of toxicosis. Dietary treatments were (DM basis) 1) 100% sorghum sudangrass hay, 2) 80% sorghum sudangrass hay:20% locoweed, 3) 80% sorghum sudangrass hay:20% locoweed plus 100 g of Silent Herder Mineral Mix (a mineral supplement reported to alleviate locoweed toxicity), and 4) 80% sorghum sudangrass hay:20% locoweed plus 100 g of clinoptilolite (a natural zeolite clay). Diets were fed at 1.5% of BW for 28 d, after which heifers had ad libitum access to sorghum sudangrass hay for 14 d. Jugular blood was sampled before feeding every 7 d, and at 2, 4, 6, and 8 h after feeding on d 28. Compared with controls, heifers fed locoweed had elevated (P less than .01) serum alkaline phosphatase activities from d 7 through 35. On d 42, alkaline phosphatase activities in heifers previously fed locoweed were lower (P less than .05) than in control heifers. Serum glutamic oxaloacetic transaminase activities were elevated (P less than .01) in heifers fed locoweed from d 7 through 42 compared with control heifers. Quantitative hepatobiliary scintigraphy using 99mTc-mebrofenin was performed on eight normal cats and on the same cats after induction of experimental cholangiohepatitis by infection with the liver fluke Platynosomum concinnum. Hepatobiliary scintigraphy was performed 3 times at 10 weeks, 4 months and 6 months after infection. In addition, routine biochemical tests, hepatic ultrasound and ultrasound guided hepatic biopsy samples were obtained at the same time points, and the results compared with hepatobiliary scintigraphy. The normal hepatic extraction fraction was determined to be 85%, and the normal hepatic excretion half time (T 1/2) was 14 minutes. There was no significant change in scintigraphic parameters compared to pre-infection values at any time following infection with the liver fluke. No correlation between scintigraphic parameters and histologic scores was found; however, significant correlation was identified between parasite burden and histologic scores 6 months following infection. Despite the presence of severe multifocal histologic abnormalities, minimal clinical, biochemical and scintigraphic derangements were identified using this model of cholangiohepatitis. Based on this study, hepatobiliary scintigraphy appears to be an insensitive test for structural hepatobiliary abnormalities. The role of hepatobiliary scintigraphy in functional hepatobiliary abnormalities of the feline liver has not been determined.  相似文献
