Background

The branched chain amino acid leucine is a potent stimulator of insulin secretion. Used in combination with glucose it can increase the insulin response and the post exercise re-synthesis of glycogen in man. Decreased plasma amino acid concentrations have been reported after intravenous or per oral administration of leucine in man as well as after a single per oral dose in horses. In man, a negative correlation between the insulin response and the concentrations of isoleucine, valine and methionine have been shown but results from horses are lacking. This study aims to determine the effect of repeated per oral administration with a mixture of glucose and leucine on the free amino acid profile and the insulin response in horses after glycogen-depleting exercise.

Methods

In a crossover design, after a glycogen depleting exercise, twelve Standardbred trotters received either repeated oral boluses of glucose, 1 g/kg body weight (BW) at 0, 2 and 4 h with addition of leucine 0.1 g/kg BW at 0 and 4 h (GLU+LEU), or repeated boluses of water at 0, 2 and 4 h (CON). Blood samples for analysis of glucose, insulin and amino acid concentrations were collected prior to exercise and over a 6 h post-exercise period. A mixed model approach was used for the statistical analyses.

Results

Plasma leucine, isoleucine, valine, tyrosine and phenylalanine concentrations increased after exercise. Post-exercise serum glucose and plasma insulin response were significantly higher in the GLU+LEU treatment compared to the CON treatment. Plasma leucine concentrations increased after supplementation. During the post-exercise period isoleucine, valine and methionine concentrations decreased in both treatments but were significantly lower in the GLU+LEU treatment. There was no correlation between the insulin response and the response in plasma leucine, isoleucine, valine and methionine.

Conclusions

Repeated post-exercise administration with a mixture of leucine and glucose caused a marked insulin response and altered the plasma amino acid profile in horses in a similar manner as described in man. However, the decreases seen in plasma amino acids in horses seem to be related more to an effect of leucine and not to the insulin response as seen in man.  相似文献

Background

Seminomas in the dog have traditionally been assumed to resemble human spermatocytic seminomas, based on their low malignancy and high occurrence in old individuals. However, recently published studies indicate that canine seminomas can be classified as classical and spermatocytic seminomas in a similar way as in man, and that classical seminomas comprise a substantial proportion of seminomas in the dog. These two factors both contribute to increasing the potential of canine seminoma as a relevant model for human testicular cancer. The aim of the present study was to characterise seminoma in Norwegian dogs using morphology and immunohistochemistry, and determine whether these tumours are comparable with human classical seminoma.

Methods

By applying diagnostic criteria from human pathology, 45 seminomas from the Norwegian Canine Cancer Register were examined histologically with hematoxylin and eosin (HE) and periodic acid-Schiff (PAS) stains. All sections were stained immunohistochemically with antibodies against human placental alkaline phosphatase (PLAP) and the transmembrane receptor c-KIT.

Results

Although two of the seminomas showed immunohistochemical staining characteristics indicative of classical seminoma (PLAP+/c-KIT+), all 45 examined seminomas were morphologically consistent with spermatocytic seminoma.

Conclusions

The value of canine seminoma as a model for SE in man remains unclear. Among the 45 investigated tumours from Norwegian dogs, none were classified as classical seminoma based on morphological criteria consistent with human seminomas. Regional or breed differences in the occurrence of classical seminoma in the dog, as well as the lack of uniform diagnostic criteria, might explain the discrepancy between the findings in the current study and the results presented by other authors.  相似文献

Background

Staphylococcus aureus is one of the most common causes of intramammary infections in dairy cows at dry off. Reliable identification is important for disease management on herd level and for antimicrobial treatment of infected animals. Our objective was to evaluate the test characteristics of PathoProof ™ Mastitis PCR Assay and bacteriological culture (BC) in diagnosing bovine intramammary infections caused by S. aureus at dry off at different PCR cycle threshold (Ct)-value cut-offs.

Methods

Sterile quarter samples and non-sterile composite samples from 140 animals in seven herds were collected in connection with the dairy herd improvement (DHI) milk recording. All quarter samples were analyzed using BC whereas all composite samples were analyzed with PathoProof ™ Mastitis PCR Assay. Latent class analysis was used to estimate test properties for PCR and BC in the absence of a perfect reference test. The population was divided into two geographically divided subpopulations and the Hui-Walter 2-test 2-populations model applied to estimate Se, Sp for the two tests, and prevalence for the two subpopulations.

Results

The Se for PCR increased with increasing Ct-value cut-off, accompanied by a small decrease in Sp. For BC the Se decreased and Sp increased with increasing Ct-value cut-off. Most optimal test estimates for the real-time PCR assay were at a Ct-value cut-off of 37; 0.93 [95% posterior probability interval (PPI) 0.60-0.99] for Se and 0.95 [95% PPI 0.95-0.99] for Sp. At the same Ct-value cut-off, Se and Sp for BC were 0.83 [95% PPI 0.66-0.99] and 0.97 [95% PPI 0.91-0.99] respectively. Depending on the chosen PCR Ct-value cut-off, the prevalence in the subpopulations varied; the prevalence increased with increasing PCR Ct-value cut-offs.

Conclusion

Neither BC nor real-time PCR is a perfect test in detecting IMI in dairy cows at dry off. The changes in sensitivity and prevalence at different Ct-value cut-offs for both PCR and BC may indicate a change in the underlying disease definition. At low PCR Ct-value cut-offs the underlying disease definition may be a truly/heavily infected cow, whereas at higher PCR Ct-value cut-offs the disease definition may be a S. aureus positive cow.  相似文献

East Coast fever (ECF) causes considerable mortality and production losses in the Tanzania smallholder dairy sector and limits the introduction of improved dairy breeds in areas where the disease is present. The infection and treatment method (ITM) was adopted by smallholder dairy farms for ECF immunisation in Hanang and Handeni districts of Tanzania. This study recorded incidence rates for ECF and other tick-borne diseases (TBDs) for ECF-immunised and non-immunised cattle between 1997 and 2000. Approximately 80% of smallholder households from both sites (n?=?167) participated in this longitudinal study, with immunisations carried out at the request of the livestock owners. Efficacy of ITM for preventing ECF cases in these crossbred dairy cattle was estimated at 97.6%, while that for preventing ECF deaths was 97.9%. One percent of the cattle developed clinical ECF as a result of immunisation. Since ECF immunisation permits a reduction in acaricide use, an increase in other TBDs is a potential concern. Sixty-three percent of farmers continued to use the same acaricide after immunisation, with 80% of these reducing the frequency of applications. Overall, 78% of farmers increased the acaricide application interval after immunisation beyond that recommended by the manufacturer, resulting in annual savings in the region of USD 4.77 per animal. No statistical difference was observed between the immunised and non-immunised animals in the incidence of non-ECF TBDs. However, immunised animals that succumbed to these diseases showed fewer case fatalities. ITM would therefore appear to be a suitable method for ECF control in Tanzania's smallholder dairy sector.  相似文献

The aim of this study was to establish the relationship between the stiffness of scantlings, approaching structural size materials, and small specimens of 6-year-old Eucalyptus urophylla?×?grandis wood using a resonance technique. The correlation between the elastic modulus (E) of the scantlings in longitudinal vibration and small specimens in flexural vibration was r?=?75 when comparing the scantling values with the averaged values of the specimens per scantling. However, when the E of each single specimen was compared with its respective scantling, the coefficient of correlation decreased to r?=?0.64 in the longitudinal tests and r?=?0.61 in the flexural tests. A roughly linear correlation (r?=?0.59) between specific modulus and loss tangent was obtained for the small specimens of Eucalyptus. In short, the resonance technique rapidly provided a large, accurate data set of mechanical wood properties as required for high-throughput phenotyping in recent genetic studies.  相似文献

In temperate Europe alley cropping systems which integrate strips of short rotation coppices into conventional agricultural fields (ACS) are receiving increasing attention. These systems can be used for crops and woody biomass production at the same time, enabling farmers to diversify the provision of market goods. Adding trees into the agricultural land creates various additional benefits for the farmer and society, also known as ecosystem services. However, tree-crop interactions in the temperate region have not been adequately substantiated which is identified as a drawback to the practical implementation of such systems. In order to bridge this gap, the current paper aims to present a comprehensive overview of selected ecosystem services provided by agroforestry with focus on ACS in the temperate region. The literature indicates that compared with conventional agriculture ACS have the potential to increase carbon sequestration, improve soil fertility and generally optimize the utilization of resources. Furthermore, due to their structural flexibility, ACS may help to regulate water quality, enhance biodiversity, and increase the overall productivity. ACS are shown as suitable land use systems especially for marginal sites. Based on the available data collected, we conclude that ACS are advantageous compared to conventional agriculture in many aspects, and therefore suggest that they should be implemented at a larger scale in temperate regions.  相似文献

Intercropping Ginkgo and crop species in southern China is receiving increasing attention because it offers potential advantages for resource utilization, higher economic income to farmers and increased sustainability in crop production, We carried out a 2-year field intercropping system composed of Ginkgo with wheat, broad bean, and rapeseed, respectively, to determine the competitive interactions between the different species, and productivity and the economic yield of each intercropping system. The density of Ginkgo and crop species was varied systematically in a two-way density matrix composed of three monoculture densities and nine intercropping of all possible pairwise combinations of monoculture densities. Intercropping systems were assessed on the basis of several intercropping indices such as land equivalent ratio, relative crowding coefficient, relative competition intensity and vector competition analysis. The results showed that the combined biomass production of the component crop species was significantly greater in the Ginkgo/crop mixtures than in monocultures crops (Ginkgo, broad bean, wheat, and rapeseed). Ginkgo: rapeseed ratio 24:12, Ginkgo: bread bean ratio 24:5, and Ginkgo: wheat ratio 24:200 had the best total biomass production. Ginkgo: rapeseed (and broad bean) ratio 24:5 and Ginkgo: wheat ratio 24:200 in respective Ginkgo/crop mixtures had the maximum economic yield. Vector competition analysis showed that Ginkgo/rapeseed mixture exhibited an antagonistic interaction type and therefore is not suitable for intercropping. Ginkgo/broad bean mixture demonstrated the most beneficial effects among the three intercropping systems.  相似文献

The landscape matrix is suggested to influence the effect of habitat fragmentation on species richness, but the generality of this prediction has not been tested. Here, we used data from 10 independent studies on butterfly species richness, where the matrix surrounding grassland patches was dominated by either forest or arable land to test if matrix land use influenced the response of species richness to patch area and connectivity. To account for the possibility that some of the observed species use the matrix as their main or complementary habitat, we analysed the effects on total species richness and on the richness of grassland specialist and non-specialist (generalists and specialists on other habitat types) butterflies separately. Specialists and non-specialists were defined separately for each dataset. Total species richness and the richness of grassland specialist butterflies were positively related to patch area and forest cover in the matrix, and negatively to patch isolation. The strength of the species-area relationship was modified by matrix land use and had a slope that decreased with increasing forest cover in the matrix. Potential mechanisms for the weaker effect of grassland fragmentation in forest-dominated landscapes are (1) that the forest matrix is more heterogeneous and contains more resources, (2) that small grassland patches in a matrix dominated by arable land suffer more from negative edge effects or (3) that the arable matrix constitutes a stronger barrier to dispersal between populations. Regardless of the mechanisms, our results show that there are general effects of matrix land use across landscapes and regions, and that landscape management that increases matrix quality can be a complement to habitat restoration and re-creation in fragmented landscapes.  相似文献

Spatial graphs in landscape ecology and conservation have emerged recently as a powerful methodology to model patterns in the topology and connectivity of habitat patches (structural connectivity) and the movement of genes, individuals or populations among these patches (potential functional connectivity). Most spatial graph’s applications to date have been in the terrestrial realm, whereas the use of spatially explicit graph-based methods in the freshwater sciences has lagged far behind. Although at first patch-based spatial graphs were not considered suitable for representing the branching network of riverine landscapes, here we argue that the application of graphs can be a useful tool for quantifying habitat connectivity of freshwater ecosystems. In this review we provide an overview of the potential of patch-based spatial graphs in freshwater ecology and conservation, and present a conceptual framework for the topological analysis of stream networks (i.e., riverscape graphs) from a hierarchical patch-based context. By highlighting the potential application of graph theory in freshwater sciences we hope to illustrate the generality of spatial network analyses in landscape ecology and conservation.  相似文献
