The foam stability of beer is one of the important key factors in evaluating the quality of beer. The purpose of this study was to investigate the relationship between the level of malt modification (degradation of protein, starch, and so on) and the beer foam stability. This was achieved by examining foam-promoting proteins using two-dimensional gel electrophoresis (2DE). We found that the foam stability of beer samples brewed from the barley malts of cultivars B and C decreased as the level of malt modification increased; however, the foam stability of cultivar A did not change. To identify the property providing the increased foam stability of cultivar A, we analyzed beer proteins using 2DE. We analyzed three fractions that could contain beer foam-promoting proteins, namely, beer whole proteins, salt-precipitated proteins, and the proteins concentrated from beer foam. As a result, we found that in cultivar A, some protein spots did not change in any of these three protein fractions even when the level of malt modification increased, although the corresponding protein spots in cultivars B and C decreased. We analyzed these protein spots by peptide mass finger printing using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. As a result, all of these spots were identified as barley dimeric alpha-amylase inhibitor-I (BDAI-I). These results suggest that BDAI-I is an important contributor to beer foam stability. 相似文献

Morphology of the modern cetaceans represents the results of adaptation of the ancestral terrestrial mammals to aquatic life through their evolutional processes. Some of the primitive fossil cetaceans are known to have both fore and hind limbs, whereas the pelvic bones of modern cetaceans are, in general, a pair of slender rod-like structures within the abdominal wall muscles just anterior to the anus with no articulations to the axial skeleton in both sexes. It is interesting and important to consider the causes and processes of how the hind limbs were lost and how the pelvis was reduced during the process of adaptation. In the present study, we tried to evaluate the topography and function of rudimentary pelvic bones of the finless porpoise (Neophocaena phocaenoides), one of the members of the odontocete cetaceans, with special references to the structures around the pelvic bones. Some soft tissues such as M. ischiocavernosus relating to the pelvic bone are transformed following the drastic reduction of the pelvis. This transformation tells us that the cetaceans adapted to the aquatic life during evolutional processes chose the tail flukes driven by the powerful trunk muscles for locomotion, instead of modifying the hind limbs into hind flippers as seen in pinnipeds. On the other hand, it is evident that a function of the pelvic bones of the male finless porpoise was supporting the penis as those of terrestrial mammals. Analysis of the effect of slow-releasing N fertilizers on microbial parameters of the rhizosphere soil is essential for their effective use. This investigation revealed the comparative effects of oxamide and ammonium sulfate as different N sources on some selected microbial parameters as well as the pH in the rhizosphere of wheat (Triticum aestivum cv. Nourin No. 61). The study was conducted on a Japanese upland alluvial soil using a rhizobox system. In the central compartment (CC) of the rhizoboxes treated with oxamide and ammonium sulfate, four wheat seedlings were grown for 53 d. In either of the treatments, the pH of the rhizosphere soil increased with the increase in the distance from the CC. In contrast, nitrate production, bacterial and fungal numbers, microbial biomass nitrogen, and β-acetylglucosaminidase activity decreased with the increase in the distance, with the highest values of the respective parameters being recorded in the CC. However, remarkable changes of these microbial parameters were found within a distance of 1–2 mm from the CC to the distant compartments. Thus the results indicated that pH, bacterial number, and nitrate production were high in the oxamide treatment. In contrast, higher fungal number, microbial biomass N, and .B-acetylglucosaminidase activity were associated with the ammonium sulfate treatment. The concentration of amylose, which is synthesised using granule-bound starch synthase, affects the physical properties of food. However, no studies have focused on starch properties and physical characteristics of low-amylose buckwheat. Here, we hypothesised that low-amylose buckwheat would be useful to produce new buckwheat products because low-amylose characteristics change the texture of buckwheat food. In this study, we bred relatively low-amylose buckwheat compared to wild type and investigated the causative genes of the traits, starch properties and physical properties of noodles. In the GBSSa mutant, the amylose concentration was lower than that in the wild type. Compared with the wild type, the mutant exhibited the following traits: Amylose concentration decreased by approximately 2%, setback in the Rapid Visco Analyzer decreased by 30 points and the physical characteristics of noodles in the sensory analysis were soft and sticky. These results suggest that this trait may be useful for changing the texture of foods. In addition, the mutant is promising for producing new foods with physical characteristics that are different from those of the wild type. 相似文献

In October, 1988, bovine ephemeral fever (BEF) occurred in Nagasaki Prefecture and throughout Kyushu island, with the exception Miyazaki Prefecture. The first outbreak occurred in Hirado-shi on October 17. The total number of diseased cattle was 24 in 24 farms in Nagasaki Prefecture. The clinical findings were mainly sudden fever, anorexia, and instability in standing. The serum neutralizing antibodies against BEF virus (BEFV) rose in all infected cattle. Twelve strains of the virus were isolated in HmLu-1 cell cultures made directly from the heparinized blood of 17 infected cattle. The buffy coat was mainly collected from the samples and washed three times with phosphate buffered saline. These isolates were all neutralized by an antiserum against BEFV (Yamaguchi strain). With the aid of an electron microscope, a representative of isolates named Hirado-9 with a length of 150 nm was seen in the sample of infected HmLu-1 cell cultures. Both Hirado-9 and Yamaguchi strains reacted with antisera. The outbreak of BEF in 1988 was the first since 1971 in Nagasaki Prefecture. The result proved that BEFV can be easily isolated in HmLu-1 cell culture from the washed blood cells of infected cattle. 相似文献

The aim of this study was to examine effects of dietary supplementation with Aspergillus awamori and feeding canola seed on the growth and fatty acid profile in broilers. Twenty‐eight chicks (15 days old) were assigned to the following groups: (1) control, fed a basal diet; (2) awamori, fed the basal diet supplemented with 0.05% A. awamori; (3) canola, fed a diet containing 5% canola seed; and (4) canola + awamori, fed the canola diet supplemented with A. awamori (seven birds/group). Body weight gain was increased by A. awamori but not influenced by canola seed. Breast muscle weight was increased in either awamori or canola groups. Although plasma triglyceride and cholesterol were decreased by feeding A. awamori or canola seed, fat content in the breast muscle were increased, accompanied by decrease in saturated fatty acids and increase in unsaturated fatty acids. Moreover, decreased thiobarbituric acid reactive substance and increased α‐tocopherol content in the breast muscle was observed in all experimental groups. In conclusion, these results suggested that feeding canola seed and A. awamori might improve growth performance, and modified muscle fatty acid profile and α‐tocopherol content, suggesting that they may improve meat quality. 相似文献

Antibody response to toxic shock syndrome toxin-1 (TSST-1) of Staphylococcus aureus in dairy cows was examined by enzyme linked immunosorbent assay (ELISA). Serum antibody to TSST-1 was not detected in 39 (76.5%) of 51 calves, which were 1-6 months of age. In contrast, TSST-1 antibody was demonstrated in 1728 (72.6%) of 2380 lactating cows housed on 36 dairy farms. The ELISA values of antibody ranged from 0.2 to 3.0 OD and presented a distribution with the peak at 1.6 OD. The mean ELISA value differed between farms, and it increased slightly along with parturient history. Somatic cell counts of milk from 174 lactating cows was compared with TSST-1 antibody and tst1,000,000 cells per ml. The mean ELISA values in milk were lower than those of sera, but they rose as somatic cells increased. The tst gene of S. aureus detected in 76.0-86.2% of the milk samples containing somatic cells > 500,000 cells per ml, a level which indicates mastitis. The data suggests that many lactating cows may be infected by TSST-1- producing S. aureus. 相似文献
