Canine atopic dermatitis (CAD) is an allergic skin disease associated with IgE and IgG antibodies (Ab) to environmental allergens. The aim of this study was to determine which other factors influence serum Ab levels in CAD-affected and non-affected dogs as this has only been poorly investigated in dogs so far. Total and allergen-specific IgE levels and Dermatophagoides farinae (DF)-specific IgG1 and IgG4 were measured by ELISA in sera of 145 CAD-affected and 271 non-affected Labrador- and Golden retrievers. A multivariable logistic regression analysis including the factors age, breed, gender, castration, clinical CAD status and allergen-specific immunotherapy (ASIT) was performed. Golden retrievers had more frequently total (OR=1.87, 95% CI=1.26-2.87, p<0.01) and specific IgE levels above the threshold value than Labrador retrievers, suggesting that genetic factors influence IgE levels in dogs. Castration was generally associated with low Ab levels (OR=0.43-0.65, p<0.05). Surprisingly, dogs with CAD did not have increased odds for high IgE against any of the allergens tested. ASIT with DF was associated with high DF-specific IgG1 (OR=4.32, 95% CI 1.46-12.8, p<0.01) but was not associated with DF-specific IgG4 or decreased IgE levels. Further studies are needed to understand the role of allergen-specific IgE in CAD and of IgG1 in ASIT.  相似文献

Vertebral column pathologies requiring surgical intervention have been described in pet ferrets, however little information is available on the normal vertebral formula and congenital variants in this species. The purpose of this retrospective study was to describe vertebral formulas and prevalence of congenital vertebral anomalies in a sample of pet ferrets. Radiographs of 172 pet ferrets (96 males and 76 females) were included in this retrospective study. In 143 ferrets (83.14%), five different formulas of the vertebral column were recorded with normal morphology of vertebrae (rib attachment included) but with a variable number of thoracic (Th), lumbar (L), and sacral (S) vertebrae. The number of cervical (C) vertebrae was constant in all examined animals. Observed vertebral formulas were C7/Th14/L6/S3 (51.74%), C7/Th14/L6/S4 (22.10%), C7/Th14/L7/S3 (6.98%), C7/Th15/L6/S3 (1.74%), and C7/Th15/L6/S4 (0.58%). Formula C7/Th14/L6/S4 was significantly more common in males than in females (P < 0.05). Congenital spinal abnormalities were found in 29 ferrets (16.86%), mostly localized in the thoracolumbar and lumbosacral regions. The cervical region was affected in only one case. Transitional vertebrae represented the most common congenital abnormalities (26 ferrets) in the thoracolumbar (13 ferrets) and lumbosacral regions (10 ferrets) or simultaneously in both regions (three ferrets). Other vertebral anomalies included block (two ferrets) and wedge vertebra (one ferret). Spina bifida was not detected. Findings from the current study indicated that vertebral formulas may vary in ferrets and congenital abnormalities are common. This should be taken into consideration for surgical planning.  相似文献

The purpose of the study was to investigate whether acute strenuous exercise (1600- to 2500-m race) would elicit an acute phase response (APR) in Standardbred trotters. Blood levels of several inflammatory markers [serum amyloid A (SAA), haptoglobin, fibrinogen, white blood cell count (WBC), and iron], muscle enzymes [creatinine kinase (CK) and aspartate transaminase (AST)], and hemoglobin were assessed in 58 Standardbred trotters before and after racing. Hemoglobin levels increased and iron levels decreased 12 to 14 h after racing and haptoglobin concentrations, white blood cell counts, and iron levels were decreased 2 and/or 7 d after racing. Concentrations of CK, AST, SAA, and fibrinogen were unaltered in response to racing.Acute strenuous exercise did not elicit an acute phase reaction. The observed acute increase in hemoglobin levels and decreases in haptoglobin and iron levels may have been caused by exercise-induced hemolysis, which indicates that horses might experience a condition similar to athlete’s anemia in humans. The pathogenesis and clinical implications of the hematological and blood-biochemical changes elicited by acute exercise in Standardbred trotters in the present study warrant further investigation.  相似文献

Sarcoptic mange occurs in many parts of the world and is common in populations of domestic and wild canids, including red foxes (Vulpes vulpes). In recent years, an indirect antibody enzyme-linked immunosorbent assay (ELISA), with higher sensitivity and specificity than traditional diagnostic methods, has been successfully applied in the diagnosis of sarcoptic mange in dogs. The same ELISA has also demonstrated specific antibodies to Sarcoptes scabiei in experimentally infected red foxes. The aim of this study was to evaluate the indirect ELISA when used to detect antibodies to S. scabiei in field sera from Swedish red foxes. One cohort of both infected and non-infected red foxes (cohort 1; n = 88), and one cohort of apparently non-infected foxes (cohort 2; n = 67) were examined for skin lesions and presence of S. scabiei by thorough visual examination at autopsy and skin scrapings. Samples of blood-tinted body liquid from the abdomen or thorax cavity were collected and analysed by the indirect ELISA. The relative sensitivity and specificity of the ELISA at different cut-offs (OD values) were estimated by comparing the test results to the infection status as determined by examination and skin scrapings. The highest combination of relative sensitivity and specificity, calculated based on cohort 1, was 95.4 and 100.0%, respectively. These estimates were constant for cut-offs 0.150-0.225, which included the cut-off based on the mean plus three standard deviations of test results from cohort 2 (0.165). It is concluded that this test can be useful in diagnosis and epidemiological studies of S. scabiei infection in red foxes.  相似文献

There is little information concerning N2O fluxes in the pasture soil that has received large amounts of nutrients, such as urine and dung, for several years. The aims of this study were to (1) experimentally quantify the relationship between mineral N input and N2O emissions from denitrification, (2) describe the time course of N2O fluxes resulting in N inputs, and (3) find whether there exists an upper limit of the amount of nitrogen escaping the soil in the form of N2O. The study site was a grassland used as a cattle overwintering area. It was amended with KNO3 and glucose corresponding to 10–1,500 kg N and C per hectare, covering the range of nutrient inputs occurring in real field conditions. Using manual permanent chambers, N2O fluxes from the soil were monitored for several days after the amendments. The peak N2O emissions were up to 94 mg N2O–N m−2 h−1, 5–8 h after amendment. No upper limit of N2O emissions was detected as the emissions were directly related to the dose of nutrients in the whole range of amendments used, but the fluxes reflected the soil and environmental conditions, too. Thus, in three different experiments performed during the season, the total cumulative losses of N2O–N ranged from 0.2 to 5.6% of the applied 500kg ha−1. Splitting of high nutrient doses lowered the rate of N2O fluxes after the first amendment, but the effect of splitting on the total amount of N2O–N released from the soil was insignificant, as the initial lower values of emissions in the split variants were compensated for by a longer duration of gas fluxes. The results suggest that the cattle-impacted soil has the potential to metabolize large inputs of mineral nitrogen over short periods (∼days). The aim of this study was to investigate antioxidant capacity of nine Fabaceae species collected on the mountains of Serbia and Montenegro. Antioxidant assays with various reaction mechanisms were used, including total phenolic content by Folin-Ciocalteu, DPPH radical scavenging capacity, Trolox equivalent antioxidant capacity (TEAC) values by ABTS radical cation and inhibition of liposome peroxidation. The investigated plants exhibited strong antioxidant capacity in all the tested methods, and among them, Lathyrus binatus, Trifolium pannonicum, and Anthyllis aurea were found to be the most active.  相似文献

Triploid onion, Allium × cornutum Clementi ex Visiani, 1842 (2n = 3x = 24), a vegetatively reproduced garden crop, possess a complex triparental genome organization with three putative parental species, A. cepa L., A. pskemense B. Fedtsch., and A. roylei Stearn. Two of its most studied clones are the Croatian ‘Ljutika’ and the Indian ‘Pran’, which are genetically highly similar. Earlier studies have shown that ‘Pran’ possesses some molecular markers in the chloroplast DNA (cpDNA) identical to those of the unique male-sterile (S) cytoplasm, used for onion breeding. To find out whether ‘Ljutika’ also possesses a S-type of cytoplasm, we analyzed several cpDNA and mitochondrial (mtDNA) molecular markers. The PCR amplification and RFLP analysis of the chloroplast genes accD, atpF, petB and the mitochondrial gene cob, as well as the sequence analysis of the chloroplast matK and atpB-rbcL regions showed that ‘Ljutika’ possesses the male-sterile S-type of cytoplasm. The phylogenetic analysis of the matK and atpB-rbcL sequences of A. × cornutum, its parental species and other Allium species of the section Cepa showed that none of the analyzed species had the identical type of cpDNA as A. × cornutum. Results also suggested that A. pskemense can be excluded as a donor of the S-cytoplasm and a female parent, whereas cpDNA of A. roylei, although not identical to S-cytoplasm, possessed many polymorphisms of S-type. Fluorescent in situ hybridization, using fluorescently labelled parental genomic DNAs as probes in combination with fluorescently labelled 5S and 35S rDNAs enabled simultaneous visualization of the three genomes during meiosis and confirmed their homeologus intergenomic pairing.  相似文献
