Dickeya strains isolated in Israel in 2006–2010 were characterized by dnaX sequence analysis, pulsed‐field gel electrophoresis (PFGE), biochemical assays and pectolytic activity, and found to be homogeneous: most of them could be classified as ‘Dickeya solani’. Of the 34 strains isolated from imported seed tubers or potato plants grown from imported seed, 32 were typed as ‘D. solani’ and only two were characterized as Dickeya dianthicola. Biovar typing indicated that all ‘D. solani’ strains were biovar 3. ‘Dickeya solani’ strains were most closely related to Dickeya dadantii subsp. dieffenbachiae according to PFGE and dnaX analyses and both species exhibited high pectolytic activity. Expression levels of two putative virulence genes, pelL (encoding a pectic enzyme) and dspE (encoding a type III effector) were significantly induced in ‘D. solani’ strains isolated from potato plants or tubers grown in hot climates such as the Negev region in Israel, compared to those isolated from seed tubers imported from the Netherlands, France or Germany. Results of this study support the hypothesis that ‘D. solani’ strains isolated in Israel are also clonal; however, they appear to be more virulent than strains isolated in Europe.  相似文献

Considered a generalist species among the coccinellids, the multicolored Asian lady beetle Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae) feeds upon small arthropods and non-prey plant products, a practice which might enhance its ability to colonize various ecosystems but carries the potential to expose it to prey quality variation and insecticide use. Thus, we examined the effect of four different diets on the development, reproduction, and response to the insecticide lambda-cyhalothrin in this species. Three groups of larvae received laboratory-reared diets: (i) eggs of Anagasta kuehniella (Zeller) (Ak); (ii) eggs of Sitotroga cerealella (Olivier) (Sc); and (iii) eggs of A. kuehniella + S. cerealella at the proportion 1:1; and one group received wild-caught cotton aphids, Aphis gossypii Glover (Ag). Notwithstanding the generalist feeding behavior of H. axyridis, development and reproduction results were superior with diets of Ag or Ak compared with the diet combining 1:1 Ak + Sc or the diet of Sc eggs only. Furthermore, the Sc diet did not result in satisfactory development and reproduction. Despite this significant variation in biological responses, insecticide susceptibility studies using adults that had been reared on these diets showed no statistically significant differences in susceptibility to the lambda-cyhalothrin.  相似文献

Clonal propagation and sexual reproduction have diametrically opposite effects on the genetic diversity of crops, permitting, respectively, the conservation of existing genotypes or the creation of new diversity. Oxalis tuberosa Mol. (“oca”) exhibits a heteromorphic self-incompatibility system and is traditionally propagated clonally but is capable of sexual reproduction. In this study we investigated the influence of sexual reproduction on the in situ genetic diversity of a vegetatively propagated crop, taking oca as a model plant. The occurrence of sexual reproduction in natural conditions and its use by farmers were studied through surveys and interviews, while the effect of sexual reproduction on the in situ genetic diversity was evaluated using microsatellite analysis. Plantlets issuing from sexual reproduction were encountered in five of the nine visited communities. The interviews indicated that six out of the nine interviewed farmers were likely, albeit unconsciously, incorporating clonal lineages issuing from sexual reproduction into their varieties. The microsatellite analysis indicated a strong effect of such incorporations on the genetic diversity of the plant varieties. Assignment tests confirmed the allogamy of oca. “Complex varieties”, made up of individuals exhibiting genetically differentiated genotypes, were most likely the result of recent crossing. Unconscious incorporation of new genotypes into the cultivated oca germplasm is therefore highly likely, raising the question of its potential occurrence in other vegetatively propagated crops. These findings could have an important effect on the adaptive potential of oca, especially in light of the genetic erosion that threatens the crop.  相似文献

Purpose

Endogeic earthworms play a significant role in biogeochemical cycles due to the large amount of soil they ingest, and because after transit through their guts, casts usually show differences in nutrient contents and microbial populations with bulk soil. Here, we studied how three endogeic earthworm species, Postandrilus majorcanus, Postandrilus sapkarevi and Postandrilus palmensis, inhabiting soils in Majorca island (Balearic Islands, W Mediterranean), modify nutrient pools and microbial communities of soil.

Materials and methods

To do this, we analysed C, N and P pools, microbial biomass (phospholipid fatty acids, PLFA) and microbial activity (fluorescein diacetate hydrolysis, FDA) in paired samples of bulk soil and fresh casts.

Results and discussion

The mineral and organic N contents were generally enhanced in casts produced by all three earthworm species. However, inorganic P and organic C contents were only higher in P. sapkarevi (32 %, only P) and P. majorcanus casts (100 % for both soil nutrient pools) than in bulk soil. Bacterial and fungal biomass were only higher than in bulk soil in P. majorcanus casts (65 and 100 %, respectively), but without effects on microbial activity, that was lower in P. palmensis casts (26 %). Earthworm gut transit strongly influenced the soil microbial community structure, resulting in differences between casts and soils.

Conclusions

The increased nutrient mineralization (6-, 1.3- and 1.4-fold for N, C and P, respectively) in casts produced by these earthworm species is of particular importance because of the amount of casts released and the seasonal variations in earthworm activity, which may favour plant growth.  相似文献

A population of the wild potato S. stoloniferum form fendleri (PI 660270) was collected as botanical seeds in the Santa Rita Mountains near Green Valley, Arizona, USA in fall 2010. Original seeds planted for multiplication at the genebank produced two plants with extra whorls of petals, sometimes fused with anthers, and, most remarkably, successive whorls of petals, anthers and carpels nested inside the primary carpel. This mutant was named matryoshka after the similarly nested Russian dolls. Floral morphology of mutants varies. It can have nearly normal male and female fertility in some individuals. Crossing studies indicate that the mutant form is dominant. Expression of the mutant may vary over the flowering cycle of the plant, with earlier flowers appearing mutant and later flowers appearing normal. Tests for pathogens were negative. Flower development mutants are of interest considering the potential for manipulating interspecific crossability, apomixis, and virus elimination in potato, and their usefulness may be extended to the important closely-related fruit crops of tomato, pepper, and eggplant. Matryoshka could also be useful in studies of potato adaptation in the wild: For example, seedless matryoshka fruit may serve as decoys to suppress the seed-eating larvae of Odenicarena fruit flies and Cecidomyiid gall flies which are especially prevalent in the geographic area where the mutant originates.  相似文献

Cyclic imines are lipophilic marine toxins that bioaccumulate in seafood. Their structure comprises a cyclic-imino moiety, responsible for acute neurotoxicity in mice. Cyclic imines have not been linked yet to human poisonings and are not regulated in Europe, although the European Food Safety Authority requires more data to perform a conclusive risk assessment for consumers. This work presents the first detection of pinnatoxin G (PnTX-G) in Spain and 13-desmethyl spirolide C (SPX-1) in shellfish from Catalonia (Spain, NW Mediterranean Sea). Cyclic imines were found at low concentrations (2 to 60 µg/kg) in 13 samples of mussels and oysters (22 samples analyzed). Pinnatoxin G has been also detected in 17 seawater samples (out of 34) using solid phase adsorption toxin tracking devices (0.3 to 0.9 µg/kg-resin). Pinnatoxin G and SPX-1 were confirmed with both low and high resolution (<2 ppm) mass spectrometry by comparison of the response with that from reference standards. For other analogs without reference standards, we applied a strategy combining low resolution MS with a triple quadrupole mass analyzer for a fast and reliable screening, and high resolution MS LTQ Orbitrap® for unambiguous confirmation. The advantages and limitations of using high resolution MS without reference standards were discussed.  相似文献

Global economic activity is networked through cross‐national linkages between firm headquarters, branches, and subsidiaries. Brokerage emerges as a key territorial function of this network, with some places acting as gateways or intermediaries for flows of global knowledge, information, or trade. This function is particularly salient for small states and territories leveraging the benefits of borrowed size by offering global professional services, warehousing, logistics, shipping, and finance to wealthy nations or high net individuals. Nonetheless, to date our understanding of how small states and territories facilitate wealth accumulation is limited to broad concepts of their role as “gateways” or “brokers.” Drawing on a typology of brokerage and a network analysis applied to the ties between approximately 700,000 firm headquarter and subsidiary locations of 13 of the world's largest stock exchanges, we explore the brokerage role of small states and territories through case studies of Luxembourg, Singapore, Hong Kong, and Panama. Brokerage is found to play an important role in the economy of all four. We argue that each of these small states and territories is uniquely positioned as a broker in global corporate networks, but that this role differs according to geo‐economic and political positionality.  相似文献

Urban rat infestations have multifactorial causes and may result in severe public health and environmental problems, as well as heavy economic losses. The identification of putative environmental determinants of urban rat infestations and the mapping of areas prone to experiencing such infestations (hot spots) are crucial for effectively addressing intervention efforts. We investigated the associations between a selection of environmental factors and the occurrence of rat infestations in the city of Madrid, Spain. This was done by modelling 10,956 citizen-reported rat sightings from 2002 to 2008 using generalized additive models, both at municipality (Madrid) and district (Latina) levels. Increased age and density of housing, and decreased distance to vegetated areas, markets and cat feeding stations were factors associated with an increased risk of rat infestations. Risk maps for rat infestations were also developed and recurrent hot spots of rat activity were identified. Although a better fit to the data was obtained in the model for the smaller scale and possibly more environmentally homogenous study area of Latina, modelling the spatial distribution of rat sightings was useful for identifying factors associated with an increased risk of urban rat infestations, as well as for identifying hot spots of rat activity, providing local authorities with a practical tool for effectively targeting intervention efforts to high-risk situations based on the local environmental contexts.  相似文献
