Spermatophore deterioration in pond-reared Penaeus vannamei was studied, and spermatophore quality was evaluated and improved. Sperm production of males collected from grow-out ponds was increased by two techniques: eyestalk ablation using a fresh frozen maturation diet (13% body weight); and laboratory culture using a diet of the formulated feed Nicovita Plus (3% body weight/ d) and frozen squid (2%). Findings are complementary to previous reports that eyestalk ablation improves quality of spermatophores in young (25.7 g) males. The timing of eyestalk ablation for activation of the endocrine mechanism, leading to improved spermatophore quality was also explored. After eyestalk ablation, performed between postmolt and intermolt stages, 26 g males required a minimum of three spermatophore regenerations or 42 d to significantly increase spermatophore size and sperm count. On the other hand, the laboratory culture (2.5 mo) technique improved the quality of spermatophores in successive regenerations for non-ablated males. In the present study, subadult P. vannamei produced spermatophores which, if not transferred or manually ejaculated, gradually deteriorated (successive stages are described), while a new compound spermatophore was being synthesized.  相似文献

Transmission electron microscopy was applied to sperm removed from males and females belonging to Litopenaeus vannamei, L. stylirostris and L. occidentalis. It was discovered that a region named filamentous meshwork (FM), located between the nucleus and the hemispherical cap, develops differently in these three closely related species. In L. vannamei, the FM is synthesized in the male reproductive system, but seems to complete its formation after mating. In L. stylirostris, the FM region was not present in spermatophores collected from males or in sperm from the thelycum. In L. occidentalis, the FM region is fully developed in male sperm. It is suggested that completion of the FM is required for acrosome maturation, and the process continues after mating in some species of Litopenaeus. In vitro induction of the acrosome reaction in sperm from males and females of L. occidentalis demonstrated for the first time that reactivity is significantly superior in sperm cells that have been attached to the open thelycum for some hours, as compared to sperm in males (prior to transfer). This finding suggests that matured sperm cells of L. occidentalis become capacitated to react against egg water after mating.  相似文献

White Spot Syndrome Virus (WSSV) has decimated the shrimp aquaculture around the world. Breeding efforts to generate resistant stocks are necessary but there is a lack of basic information on challenge test strategies focused on genetic selection. Infection routes and developmental stages were evaluated on Penaeus vannamei as a first step in a strategy to select white spot virus (WSSV)-resistant stocks. Mortalities could not be induced before the PL30 stage. The impact of infection by immersion and blended tissue was intermediate on mortalities when compared to the minced tissue treatment on PL30. Blended and minced tissue treatments produced the highest mortalities on PL40 while immersion was intermediate. A general tendency towards higher susceptibility associated with older stages was detected. Additionally, juveniles of 1 g average weight from three local breeding programs were challenged. There were no differences in survival between the programs, although two of them derived their progenies from survivors of strong WSSV events. The implications of these results to the WSSV epidemiological characteristics and breeding programs are discussed.  相似文献

Drip irrigation systems are prone to changes in flow rate (FR) and increasing coefficients of variation (CV) when fed with treated wastewater, due to fouling inside the drippers. A model system was designed to measure the FR and fouling accumulation in laterals and drippers under different treatment conditions. A novel approach was taken to compare the different maintenance treatments. A comparison of chlorination and acidification strategies showed that daily chlorination and periodic acidification may prolong proper functioning of the drippers by maintaining a normal FR [(up to ±7 %) of nominal FR] and CV (<7 %) index in correlation with low fouling accumulation in the pipeline (<0.01 mg deposit/cm pipe 2 ). Current recommendations for the frequency of conventional treatments were found to be insufficient. Chemical analyses of the fouling inside the dripper and accumulated on the pipe wall showed that biofilm can survive inside the dripper under harsh environmental conditions, even when the pipeline stays clean. These results shed light on biofilm growth and survival mechanisms inside the dripper and may pave the way to developing new treatments or improving dripper design.  相似文献

  1. The worldwide reduction in wetlands has led to the large‐scale decline of wetland‐dependent species. In Australia, to redress some of the decline, partial restoration of the hydrology of a small number of wetlands has been attempted using allocations of environmental water.
  2. A common goal of the watering is the maintenance and enhancement of native fish communities, which historically have included populations of the salt tolerant Murray hardyhead (Craterocephalus fluviatilis), a small, short‐lived fish, endemic to the lower Murray–Darling Basin.
  3. Despite the addition of environmental water to several sites at which the species is known to persist, populations continue to decline. This decline is, at least in part, suspected to be a consequence of salinities that conflict with the breeding ecology and survival of early life stages.
  4. Here the effect of salinity on egg hatch rate and the upper salinity tolerance of larval and juvenile Murray hardyhead was determined under laboratory conditions. It was found that eggs were vulnerable to elevated salinities, whereas juveniles were capable of tolerating salinities up to 105 ppt.
  5. Based on the results of the experiment, brackish wetlands managed for Murray hardyhead should be maintained, where possible, between 12 and 45 ppt. Such a salinity regime will necessitate less intensive management of salinity, and a reduced volume of environmental water, providing both environmental and fiscal benefits. The research highlights the benefits of investment in targeted research.

The relation between growth, feed utilization and feed ration was assessed during the first 15 feeding days in Colossoma macropomum larvae fed with Artemia nauplii. Maintenance, optimum and maximum feeding rations (1.23; 5.48 and 27.2% body weight/d, dry feed, respectively) and feed conversions (0.4 and 0.78 optimum and maximum, respectively) were determined. At maximum feeding the larvae grew 2.6 times faster that at optimum feeding, but the feeding costs increased 1.94 times. Two commonly used growth rates (specific growth rate, SGR and coefficient G) were compared for the modelling of larval growth. The maximum specific growth rate (SGR, growth rate at maximum feeding) remained relatively constant during the first 15 d of feeding and is thus much more adequate to model the growth of tambaqui larvae, than the coefficient G, which increased more than four times in this period.  相似文献

The objective of this study was to evaluate dexmedetomidine as a premedicant in dogs prior to propofol-desflurane anaesthesia, and to compare it with medetomidine. Six healthy dogs were anaesthetized. Each dog received intravenously (i.v.) five preanaesthetic protocols: D1 (dexmedetomidine, 1 microg/kg, i.v.), D2 (dexmedetomidine, 2 microg/kg, i.v.), M1 (medetomidine, 1 microg/kg, i.v.), M2 (medetomidine, 2 microg/kg, i.v.), or M4 (medetomidine, 4 microg/kg, i.v.). Anaesthesia was induced with propofol (2.3-3.3 mg/kg) and maintained with desflurane. The following variables were studied: heart rate (HR), mean arterial pressure, systolic arterial pressure, diastolic arterial pressure, respiratory rate (RR), arterial oxygen saturation, end-tidal CO2, end-tidal concentration of desflurane (EtDES) required for maintenance of anaesthesia and tidal volume. Arterial blood pH (pHa) and arterial blood gas tensions (PaO2, PaCO2) were measured during anaesthesia. Time to extubation, time to sternal recumbency and time to standing were also recorded. HR and RR decreased significantly during sedation in all protocols. Cardiorespiratory variables during anaesthesia were statistically similar for all protocols. EtDES was significantly different between D1 (8.1%) and D2 (7.5%), and between all doses of medetomidine. Desflurane requirements were similar for D1 and M2, and for D2 and M4 protocols. No statistical differences were observed in recovery times. The combination of dexmedetomidine, propofol and desflurane appears to be effective for induction and maintenance of general anaesthesia in healthy dogs.  相似文献
