A comparative study between conventional methods (EPA 3050B and ISO 11466.3) of metal extraction and a simple low-cost method, using aqua regia, was carried out in this work. Six elements (Mn, Cu, Zn, Pb, Ni, and Cd) were determined by flame atomic absorption spectrometry (FAAS) in a certified sample of sediment (CNS 392). Central composite design (CCD) and response surface methodology (RSM), as well as machine learning, were used to find the optimal conditions for metal extraction. The influence of the parameters—volume of nitric acid in aqua regia (v), time of extraction (t), and temperature (T)—on Mn, Cu, Zn, and Pb recoveries was investigated. The best condition for the recovery of all the metals was v = 2.5 mL of HNO3, t = 2 h, and T = 90 °C. In comparison with the conventional methods, the aqua regia method was found to present better recovery values and lower standard deviations for all the metals studied. 相似文献

‘Microcenters of diversity’ are restricted geographical areas within which a significant diversity of genetic resources is accumulated. Maize genetic variability, one of the largest among cultivated species, particularly in far western Santa Catarina, southern Brazil, has shown a rich store of landraces, conserved on farm by small-scale farmers. Thus, this study aimed to characterize the diversity of Zea mays L. landraces in two municipalities of this micro-region in Santa Catarina, as well as its geographic distribution, developing the census of diversity as a methodological proposal. Diagnosis was made according to the farmers’ knowledge. The field survey was conducted in 70 rural communities involving 2049 farms. The methodology allowed identifying 136 populations of wild relatives, of which some belong to the species Zea luxurians (Durieu and Ascherson) Bird, and 1513 populations of maize landraces, comprising 1078 of popcorn, 337 of common maize, 61 of sweet maize and 37 of flour maize. The identification of 59 morphological groups, the high use, adaptive and agronomic values, and the Shannon index (H′), estimated according to the grain shape (0.79), endosperm type (0.73), size (0.87), color (1.40), and morphological group (3.16), showed at set a considerable diversity distribution conserved on farm in this micro-region. In pigs, the gut microbiota composition plays a major role in the process of digestion, but is influenced by many external factors, especially diet. To be used in breeding applications, genotype by diet interactions on microbiota composition have to be quantified, as well as their impact on genetic covariances with feed efficiency (FE) and digestive efficiency (DE) traits. This study aimed at determining the impact of an alternative diet on variance components of microbiota traits (genera and alpha diversity indices) and estimating genetic correlations between microbiota and efficiency traits for pigs fed a conventional (CO) or a high-fiber (HF) diet. Fecal microbes of 812 full-siblings fed a CO diet and 752 pigs fed the HF diet were characterized at 16 weeks of age by sequencing the V3-V4 region of the 16S rRNA gene. A total of 231 genera were identified. Digestibility coefficients of nitrogen, organic matter, and energy were predicted analyzing the same fecal samples with near infrared spectrometry. Daily feed intake, feed conversion ratio, residual feed intake and average daily gain (ADG) were also recorded. The 71 genera present in more than 20% of individuals were retained for genetic analyses. Heritability (h²) of microbiota traits were similar between diets (from null to 0.38 ± 0.12 in the CO diet and to 0.39 ± 0.12 in the HF diet). Only three out of the 24 genera and two alpha diversity indices with significant h² in both diets had genetic correlations across diets significantly different from 0.99 (P < 0.05), indicating limited genetic by diet interactions for these traits. When both diets were analyzed jointly, 59 genera had h² significantly different from zero. Based on the genetic correlations between these genera and ADG, FE, and DE traits, three groups of genera could be identified. A group of 29 genera had abundances favorably correlated with DE and FE traits, 14 genera were unfavorably correlated with DE traits, and the last group of 16 genera had abundances with correlations close to zero with production traits. However, genera abundances favorably correlated with DE and FE traits were unfavorably correlated with ADG, and vice versa. Alpha diversity indices had correlation patterns similar to the first group. In the end, genetic by diet interactions on gut microbiota composition of growing pigs were limited in this study. Floodplain lakes are ecosystems characterised by annual flood and dry cycles. Fish ecology is influenced by the flood pulse due to the large influx of allochthonous food resources and diversification of habitats during the flood cycle, while during the dry cycle, fishes tend to be confined in reduced habitat. The aim of this study was to evaluate the seasonal variation in trophic niche width and overlap of four species—Mylossoma duriventre, Prochilodus nigricans, Cichla.pleiozona and Serrasalmus rhombeus—in an Amazonian floodplain lake. Stable isotope analyses were used to estimate trophic niche width and overlap during the flood and dry seasons. We hypothesised broader niche width for all species during the flood cycle and a higher degree of overlap between the two piscivorous fishes during the dry cycle. Isotopic niche width was 72% broader for P. nigricans, 61% for S. rhombeus and 54% for C. pleiozona during the dry cycle, which did not support our hypothesis. Core niche width overlaps were not observed between piscivorous species in either flood or dry cycle. The results indicate that seasonal variation in isotopic niche width is specific to feeding habit. Understanding how fish trophic ecology responds to changes in the hydrological regime during the seasons is crucial for sustainable fishery management in a region where many people rely heavily on fish for nutritional and economic purposes. 相似文献

Climate-induced shifts in plankton blooms may alter fish recruitment by affecting the fatty acid composition of early-life diets and corresponding performance. Early-life nutrition may immediately affect survival but may also have a lingering influence on size and growth via experiential legacies. We explored the short- and longer-term performance consequences of different concentrations of polyunsaturated fatty acids (PUFA) for juvenile Walleye (Sander vitreus, Mitchill 1818). For the first 10 days of feeding, juveniles were provided Artemia enriched with: oleic acid (low PUFA), high docosahexaenoic acid and high eicosapentaenoic acid (high PUFA), or high PUFA and a form of vitamin E (high PUFA + E). After 10 days, all fish were fed a high-quality diet and reared for an additional 27 days. Juveniles fed either high PUFA diet were 1.15-fold larger (PUFA mean ± SD = 20.0 ± 3.3 mg; PUFA + E = 19.8 ± 3.3 mg) than those fed the low PUFA (17.3 ± 2.8 mg) diet after 10 days of feeding. After 27 days, juveniles initially fed the high PUFA diet were still 1.10- to 1.20-fold larger (PUFA = 407.0 ± 61.6 mg; PUFA + E = 422.7 ± 58.7 mg) than those initially fed the low PUFA diet (356.5.0 ± 39.5 mg). Our findings demonstrate that fatty acid composition of juvenile Walleye diets has immediate and lingering size effects. As changes in climate continue to alter lower trophic levels, fish management and conservation may need to consider short- and long-term effects of temporal or spatial differences in early-life diet quality. 相似文献
