Eight Beagle dogs were anesthetized and were imaged using a single channel helical CT scanner. The contrast medium used in this study was iohexol (300 mg I/ml) and doses were 0.5 ml/kg for a cine scan, 3 ml/kg for an enhanced scan. The flow rate for contrast material administration was 2 ml/sec for all scans. This study was divided into three steps, with unenhanced, cine and enhanced scans. The enhanced scan was subdivided into the arterial phase and the venous phase. All of the enhanced scans were reconstructed in 1 mm intervals and the scans were interpreted by the use of reformatted images, a cross sectional histogram, maximum intensity projection and shaded surface display. For the cine scans, optimal times were a 9-sec delay time post IV injection in the arterial phase, and an 18-sec delay time post IV injection in the venous phase. A nine-sec delay time was acceptable for the imaging of the canine hepatic arteries by CT angiography. After completion of arterial phase scanning, venous structures of the liver were well visualized as seen on the venous phase.  相似文献

Components of silk including silk fibroin have long been used as anti-diabetic remedies in oriental medicine. However, detailed mechanisms underlying these anti-diabetic effects remain unclear. In this study, we examined the anti-diabetic activity of silk fibroin hydrolysate (SFH) in C57BL/KsJ-db/db (db/db) mice, a well-known animal model of non-insulin dependent diabetes mellitus. When the db/db mice were administered SFH in drinking water for 6 weeks, hyperglycemia in the animals gradually disappeared and the level of glycosylated hemoglobin decreased, indicating that SFH plays important role in reducing the symptoms of diabetes. In addition, SFH-treated db/db mice exhibited improved glucose tolerance with increased plasma insulin levels. Immunohistochemical and morphological analyses showed that SFH up-regulated insulin production by increasing pancreatic β cell mass in the mice. In summary, our results suggest that SFH exerts anti-diabetic effects by increasing pancreatic β cell mass in a non-insulin dependent diabetes mellitus mouse model.  相似文献

Antibiotics have traditionally been used for growth promotion in the pork industry; however, their use in animal feed has recently been limited because of human health concerns. The intestinal microbiota plays an important role in mediating many physiological functions such as digestion and animal growth. It was hypothesized that use of antibiotics as growth promotants and subsequent variations in intestinal microbiota induce significant changes in the intestinal glycoconjugate composition, which ultimately affects animal growth and disease susceptibility. The aim of this study was to characterize the lectin binding profiles of the ileum of weanling pigs in response to the absence of intestinal microbiota and to the use of the antibiotic chlortetracycline as growth promotant. Eighteen half-sib piglets obtained by cesarean section were divided into 3 treatment groups (n = 6) and maintained as control, antibiotic-fed, and gnotobiotic piglets until 5 wk of age. The glycoconjugate composition of the ileal tissues was examined by lectin histochemistry. Lycopersicon esculentum lectin, Jacalin, Pisum sativum agglutinin, Lens culinaris agglutinin (LCA), and Sambucus nigra lectin showed significant differences (P < 0.05) in binding intensities on the dome and villous epithelium between the treatment groups. Griffonia simplicifolia lectin I, Glycine maxi agglutinin, and Arachis hypogea agglutinin exhibited differences (P < 0.05) between treatment groups in lectin binding on goblet cells. Triticum vulgaris agglutinin, Pisum sativum agglutinin, and LCA showed significant differences (P < 0.05) in binding intensities on dome, corona, and follicular regions of the ileum among treatment groups of animals. Overall, ileal tissues from gnotobiotic piglets expressed significantly weaker (P < 0.05) lectin binding for many lectins compared with control and antibiotic groups. This suggests that the intestinal microbiota plays an important role in the expression of sugar moieties in the intestine. Lectins LCA, Phaseolus vulgaris Leucoagglutinin, and Maackia amurensis lectin II showed significant differences (P < 0.05) in lectin bindings between control and antibiotic-fed piglets. This indicates that chlortetracycline as a growth promotant induces biologically relevant changes in the lectin binding profile of the ileum. These findings will help in further understanding the role of the gut microbiota and the mechanisms of action of antibiotics as growth promotants in pigs.  相似文献

Typhoid is a distinct gastrointestinal disease that largely affects the public by consumption of inadequately or partially cooked eggs from contaminated laying hen farms. This has led the research on laying hens to focus on controlling the contamination by an effective anti-Salmonella spp. agent in the intestine. The treatments included, control, without challenge; PC, Salmonella typhimurium challenged (STC); BP5, 5 ppm bacteriophage/kg + STC; BP10, 10 ppm bacteriophage/kg + STC, on Salmonella shedding, body organs inflammatory reactions, and expression of toll-like receptor (TLR), pro-inflammatory cytokines, and heat shock protein (HSP) in the jejunum, liver,and thigh muscle in the STC laying hens. The RT-PCR method was used to enumerate the number of Salmonella typhimurium in the organs. The birds in the STC groups exhibited the increased population of Salmonella spp. in the excreta (p < .01). In the STC groups, the BP5 and BP10 laying hens exhibited a lower (p < .01) population of Salmonella spp. in the excreta at d 7 after STC. Supplementation of bacteriophage significantly decreased (p < .01) the colonization of S. Typhimurium in the spleen, oviduct, caecum and excreta. Among the STC treatments, the BP10 laying hens showed lower (p < .01) mRNA expression of interferon-γ (IFNγ) and TLR-4 in the jejunum compared with the PC treatment. After the STC, dietary supplementation with BP5 or BP10 decreased (p < .01) the mRNA expressions of IFNγ, HSP-27 and tumour necrosis factor-α in the liver compared with the PC treatment. These results suggest that bacteriophage can be used as an effective agent to decrease S. Typhimurium contamination in laying hens and possibly lower S. Typhimurium transfer to foods.  相似文献

Mycoplasma synoviae, a major worldwide pathogen in poultry, causes respiratory tract infection and arthritis in chickens and turkeys. Two major surface antigens of M. synoviae are encoded by a single gene, vlhA (variably expressed lipoprotein and hemagglutinin). The gene product is cleaved post-translationally to yield the lipoprotein major surface protein (MSP) B (MSPB) and the hemagglutinin MSPA. The availability of MSPA as an antigen for serodiagnosis was studied by means of a protein chip based on surface plasmon resonance imaging (SPRi). The diagnostic potential of SPRi for measurement of levels of antibody to MSPA was compared with that of a conventional enzyme-linked immunosorbent assay (ELISA) kit. The results from SPRi, a process that took only 1 h, were similar to those from ELISA. Therefore, MSPA can be used as an antigen for serologic studies, and SPRi, a label-free and high-throughput method, may be a valuable tool in avian serodiagnostic studies.  相似文献

Eggs and sperm were obtained from a female (6.3 kg/BW) and a male (8.4 kg/BW) longtooth grouper Epinephelus bruneus following HCG injection in July 2003. The eggs were fertilized artMcially with the sperm and incubated in one of two 50-m3 tanks after washing the fertilized eggs. The fertilized eggs were 830–950 pn (average 900 ± 2 μm) in diameter and the respective fertilization and hatching rates were 97.7 ± 0.6% and 96.8 ± 0.5% at a water temperature of 25.0 ± 0.5 C. With this regime, the survival rate by day 93 was 7.5% in the 50-m3 tank. The elapsed time from hatching to opening the mouth was 3 d at 25 C. The initial mouth size (z) of the larvae was 0.22–0.23 mm. The newly hatched larvae were 2.02 ± 0.02 mm TL; this increased to 4.12 ± 0.09 mm TL by day 11. By day 54, the larvae had metamorphosed into juveniles and reached 41.12 ± 1.20 mm TL, and by day 93 the juveniles reached 93.78 ± 1.98 mm TL. In all, 49.5% of the larvae were malformed and the type of malformation was diverse.  相似文献

Rice hull–sawdust composite boards were manufactured for sound-absorbing boards in construction. The manufacturing parameters were target density (400, 500, 600, and 700?kg/m3) and rice hull content as percent weight of rice hull/sawdust/phenol resin (10/80/10, 20/70/10, 30/60/10, and 40/50/10). Commercial gypsum board and fiberboard were also used as comparative sound-absorbing materials. The average modulus of rupture (MOR) of the board with a density of 700?kg/m3 and rice hull mixing ratio of 10% was 8.6?MPa, and that of the board with a 400?kg/m3 board density and a rice hull mixing ratio of 40% was 2.2?MPa. The MOR increased with increasing board density or decreasing rice hull mixing ratio. The sound absorption coefficients of some boards (400?kg/m3 and 10%, 500?kg/m3 and 30%, and 500?kg/m3 and 40%) were better than those of the commercial 11-mm-thick gypsum board. Thus, it is concluded that rice hull–sawdust composite boards may be implemented as sound-absorbing barriers in construction due to their high sound absorption coefficients.  相似文献

Phytophthora lateralis, an exotic root pathogen, is the primary cause of Port‐Orford‐cedar (Chamaecyparis lawsoniana) mortality throughout its native range in southwest Oregon and northwest California. Most trees in the field are very susceptible, but genetic resistance to this pathogen has been demonstrated. Since the late 1980s, the USDA Forest Service and the Bureau of Land Management in cooperation with Oregon State University have conducted an intensive programme to identify and test resistant trees from the field, and propagate them in a seed orchard with the goal of providing resistant seedlings for regeneration. Susceptible families showed only 0–10% survival using a variety of inoculation techniques. Rooted cuttings of resistant parents are seldom killed, and seedling families of these parents exhibit 25–100% survival, depending on family and inoculation technique. Symptom development on resistant trees, including sunken lesions, and resinosis as well as reduced colonization and re‐isolation success, was consistent with a hypersensitive reaction. In a long‐term field test, five resistant families had 20–80% survival after 16 years, while three susceptible families had 0–8% survival in the same interval.  相似文献
