Promoter anchored amplified polymorphism based on random amplified polymorphic DNA (PAAP-RAPD) in cotton 总被引:1,自引:0,他引:1
Non-coding sequences account for a majority of the higher plant genome, some of which have important effects in gene regulation
and plant development. In an effort to develop molecular marker systems to search for polymorphisms associated with high fiber
yield and quality in cotton, we have developed a methodology that could specifically target the regulatory regions of the
cotton genome. In this study we designed 10-nucleotide degenerate promoter primers based on conserved core promoter sequences
and tested their applicability in PCR amplifications in combination with 10-mer random amplified polymorphic DNA (RAPD) primers.
The amplified markers are called promoter anchored amplified polymorphism based on RAPD (PAAP-RAPD). Forty cotton genotypes
with diverse genetic and geographical backgrounds were used to test the PAAP-RAPD system using polyacrylamide gel electrophoresis.
Based on PAAP-RAPD markers amplified from 12 primer combinations, the 40 genotypes were classified into five distinctive groups:
two Upland cotton (Gossypium hirsutum) groups from China, another two Upland cotton groups from the USA, and one group from American Pima cotton (G. barbadense). The groupings are in general consistent with their genetic and geographical origins. Thirty-six PAAP-RAPD and RAPD fragments
were cloned and four of them were further subjected to sequence analysis. Signal scanning using software PLACE confirmed that
they contained an array of cis-regulatory sequences in addition to the core promoter sequences. The results demonstrate the
potential application of PAAP-RAPD as a new marker system specifically targeting regulatory regions of the plant genome. 相似文献

The Flowering Locus T (FT)-like genes of angiosperms are highly conserved. The FT-encoded proteins include a phosphatidylethanolamine-binding domain that is involved in the control of the shoot apical meristem identity and flowering time. In the present study, FT genes were investigated in 20 bamboo species that are grouped into sympodial, mixed and scattered bamboos based on their morphology. All examined orthologous FT genes consisted of four exons and three introns. Their encoded protein sequences contained the critical amino acid residues Tyr85, Glu109, Leu128, Tyr134, Trp138, Arg139, Gln140 and Asn152, of which each possesses a biological function. The DNA sequences were rich in single nucleotide polymorphism (SNP) sites. The SNP frequency was 1 SNP/16.8 bp, and the nucleotide diversity (π) equaled 0.265. Some SNPs altered restriction enzyme sites or resulted in changes in amino acid contents. The correlation analysis showed that several SNPs were informative in relation to the underground rhizome types of bamboos. Therefore, FT polymorphisms could be used as a tool to identify the underground rhizome types of bamboos. The phylogenetic tree constructed based on the FT gene sequences showed that the obtained clustering was consistent with the underground rhizome types. The SNP markers developed in the present study will provide information on the genetic diversity of bamboos and they can aid taxonomic study as well. 相似文献

A straightforward approach was proposed to modify cotton fabric for oil/water separation based on musselinspired reaction. The poly(DMA-Octadecyl acrylate) was designed to contain key chemical constituents present in mussel adhesive proteins by free radical polymerization of dopamine hydrochloride and octadecyl acrylate, which strongly adsorbed to fabric substrates, providing a special surface for fabric. The chemical structure, surface topography, and surface wettability of the fabric were characterized. The results showed that as-prepared cotton fabric displayed a high CA of >150° when dripped water droplets were on the modified fabric surface, and the oil contact angle (OCA) was close to 0°, it had excellent potential to be used in practical applications and has created a new method of fabric modification for oil/water separation. 相似文献

Purpose
This study investigated the phosphorus (P) source, mobilisation and transport potential of ditch bed sediments as well as surrounding field and bank soils in two agricultural headwater catchments with contrasting soil drainage capacities. This information is important for discerning the potential for ditches to attenuate or augment transfers of P from upstream sources and thus for developing appropriate management strategies for these features.Materials and methods
Phosphorus sources were characterised using the Mehlich3-P, water-soluble P and total P tests. Phosphorus mobilisation potential was characterised using the Mehlich3-AL/P, Mehlich3-Ca/P and DESPRAL P tests. Phosphorus transport potential was characterised using data collected on the presence/absence of surface water in ditches during field surveys and downstream turbidity data.Results and discussion
Ditch sediments had similar P source contents (Mehlich3-P, water-soluble P and total P) to the surrounding field soils and higher P contents than bank soils. However, calcium contents of sediments in the poorly drained catchment reflected the deep sub-soils rather than the surrounding field and bank soils. Mehlich3-Al/P and Mehlich3-Ca/P contents of ditch sediments in the well (non-calcareous) and poorly (calcareous) drained catchments respectively indicated potential for P retention (above thresholds of 11.7 and 74, respectively). However, sediments were less aggregated than field soils and may mobilise more particulate P (PP) during rain events. Nevertheless, the majority of surveyed ditches dried out from March to September 2011; thus, their potential to mobilise PP may be less important than their capacity to attenuate soluble and PP during this time.Conclusions
In these and similar catchments, soluble P attenuation and particulate P mobilisation should be maximised and minimised, respectively, for example, by cleaning out the sediments before they become saturated with P and encouraging vegetation growth on ditch beds. This study also highlighted the influence of deep sub-soils on soluble P retention in ditches and thus the utility of characterising soils below depths normally included in soil classifications.196.
A cobalt-mediated decrease in ethylene production reduced the inhibition of nodulation by nitrate in Vigna radiata (mungbean). Nitrate increased the ethylene production in 5-day-old seedlings, while it caused a reduction in the nodulation
status (nodule number and nodule weight) and nodule efficiency (acetylene reduction activity) in mungbean plants. The application
of cobalt chloride inhibited nitrate-affected ethylene production and also decreased the inhibitory effect of nitrate on nodulation.
The effect of cobalt was most marked on nodule number.

This paper introduces a combined modelling approach using a simple water budget model (THC-model) and a 3D reservoir sedimentation
model (MOHID Water) to adapt reservoir operation and visualise their effects on the sediment deposition. By this, an effective
combined sediment-water management can be identified under semi-arid conditions for dry, median and wet years. Results are
presented for the reservoirs of the Tuyamuyun Hydro-Complex (THC), which is located in the lower Amu Darya River. The determination
of the actual and usable reservoir storage volume shows that siltation will significantly adversely affect the ability of
the in-stream Channel Reservoir to regulate seasonal demand for both irrigation and municipal water supply. However, modelling
scenarios results confirm the effectiveness of adapted operation rules for the THC reservoirs and show that the operation
of large dams could be modified according to a combined sediment-water management. The experience gained during this study
emphasizes the fact that the concept of a combined reservoir management of sediments as well as water can be an efficient
measure to improve the sustainable long-term use of reservoirs and to contribute towards a safe water supply in water crisis

RNA silencing pathways in filamentous fungi are composed of multiple component proteins and known to be involved in vegetative growth, virulence or sexual reproduction. We found that the tomato wilt fungus, Fusarium oxysporum f. sp. lycopersici (Fol), carries four homologues genes of Qde-2, an argonaute protein gene and one of the main component protein genes in Neurospora crassa. A nitrogen (N) balance digestion trial was conducted to determine the protein requirement of collared peccaries (Pecari tajacu). In a 4 × 4 Latin square design, four captive adult male peccaries were fed four isoenergy diets containing four different levels of N (11.7, 16.3, 22.8, and 26.7 g N/kg of dry matter—DM). After 15 days of adaptation, a total collection of feces and urine was carried out for five consecutive days. Regression analyses between N intake and N in feces and urine allowed to calculate the metabolic fecal nitrogen (MFN = 2.3 g N/kg of dry matter intake—DMI) and daily endogenous urinary N (EUN = 185 mg N/kg0.75). Likewise, by regression analyses between consumption of nitrogen and the nitrogen balance (NB = N ingested ? N excreted, mg N/kg0.75), a daily requirement of 514 mg N/kg0.75 was calculated. Therefore, if food intake is unrestricted, collared peccaries require a minimum in their diet of about 5.4% crude protein on DM basis. These values are almost as low as those found for browsing and frugivorous wild ruminants, which reinforce the proposition that peccaries’ digestive physiology is nearer to that of domestic and wild ruminants than domestic pigs. This relatively low protein requirement of collared peccary and its great ability to digest protein reveal the relevance of the forestomach for the species on nitrogen/protein metabolism and allow the use of diets with lower crude protein levels than the commercial ones used for the domestic pig, which reduces feed costs. 相似文献
