Mycobacterium avium subsp. paratuberculosis (MAP) strains with two new IS900 restriction fragment length polymorphism (RFLP) BstEII types intermediate suspected to belong to the MAP Type III group were isolated from migrating sheep in Germany. Such strains have only been sporadically identified in a few studies. For a better understanding of the genomic diversity of MAP with regard to specific host associations, geographic origin, and the discussed classification into Type I, Type II and Type III, these isolates were further characterized.Using IS900-RFLP, the isolates showed unique fingerprint patterns after BstEII-, PstI-, PvuII- and BamHI-digestion which had not been published before. Additionally, using gyrB-PCR-restriction endonuclease analysis (PCR/REA) and mycobacterial interspersed repetitive unit (MIRU)-PCR, the two strains showed differences to known patterns of the Type I as well as the Type II group. Unique genotypes were also obtained with multilocus short sequence repeat (MLSSR) sequencing and MIRU-variable-number tandem-repeat (VNTR) typing.As expected, genomic profiles identical to the Type I and different from the Type II group were detected by IS1311-PCR/REA, IS1311 sequencing as well as by Large Sequence Polymorphism analysis (LSPA 8, 17, 20, 4-II, and 18).In addition to distinct growth characteristics, the unique genotypes of the studied sheep strains support their affiliation to the assumed third group within the MAP subspecies and suggest the existence of different genotypes within this Type III group. The results could serve as further evidence that Type I and Type III groups are more closely related to each other than to the bovine Type II group.  相似文献

In pot experiments with rye-seedlings, it was investigated how the size of non-rooted artificially made spherical aggregates influences the development of water potential gradients in the aggregates and the utilization of water. The experiments were carried out with constant low (40%) and high (70%) relative humidity, and with changing low (40–60%) and high (70–90%) relative humidity. Material from a clay soil (52%<2μm) was used. The mean gradient of water potential developing in the aggregates during the course of drying increased with decreasing aggregate size. For constant relative humidity, loss of water from the aggregates increased with increasing mean gradient of water potential. For changing relative humidity, loss of water from the aggregates was independent of the mean gradient of water potential and hence from aggregate size. It is discussed that the change in relative humidity allows a certain redistribution of water in the aggregates which makes the water in the centre of large aggregates accessible to roots.  相似文献

The objective of the present study was to determine whether substrate-induced priming effects in soils are sensitive to increasing levels of Cu and Zn. Soils were collected from ten plots of two Australian field experiments (Spalding and Avon) where increasing amounts of Cu or Zn had been added 2?years prior to sampling, reaching maximum values of 5,880?mg?kg?1 for Cu and 7,400?mg?kg?1 for Zn. In a 21-day incubation experiment, the effect of uniformly 14C-labeled fructose and alanine on the mineralization of the soil organic carbon (SOC) was investigated. With increasing heavy metal content, the initial peak of soil respiration after substrate addition was retarded, indicating that the microorganisms utilizing these substrates were inhibited in soils highly contaminated with heavy metals. Both substrates strongly changed the mineralization of the soil organic matter (SOM), i.e., priming effects were induced. In the soil samples with high Cu concentrations from Spalding, fructose induced a stronger additional mineralization of the SOC than in the lower contaminated samples. In the samples with the highest Zn contamination level, negative priming effects, i.e., a reduced mineralization of SOM, were observed. In contrast, heavy metal effects in the Avon soil (pH?7.6) were less pronounced since substrate mineralization and priming effects were not directly related to the increasing heavy metal content. Apart from direct toxic heavy metal effects, the tested microbial activity parameters were also indirectly affected through the toxic heavy metal effects on plant growth. At the highest heavy metal contaminations, no fresh biomass inputs occurred during the past 2?years so that microorganisms in these soils were highly substrate-limited. As a consequence, complex interactions between different levels of heavy metal contamination, the microbial activity, and the input of SOC via plant biomass have to be considered.  相似文献

Purpose

The present study focuses on a compositional characterization of the humic acid (HA) fraction of several wetland soils using solid-state 13C NMR spectroscopy. The data were analysed using the molecular mixing model (MMM), based on an empirical approach by Nelson and Baldock. The compositional data from HAs obtained with this model were used to obtain a wider assessment of the process of humification from comparison of total soil wetland organic matter composition and HA composition.

Materials and methods

Twenty samples of humic acids (HAs) isolated from a Mediterranean semiarid wetland (‘Tablas de Daimiel’, central Spain) were studied using elemental analysis and cross polarization magic angle spinning (CPMAS)13C nuclear magnetic resonance (NMR) spectroscopy. The NMR data were analysed with the molecular mixing model (MMM) considering up to six generic components (carbohydrate, protein, lignin, lipid, char and ‘carbonyl’). HAs are considered a conceptual mixture of these model components, and the MMM determines the proportions of the characteristic biomolecules contributing to HA composition.

Results and discussion

The composition of the HAs under study depends on local factors such as site vegetation and occurrence of fire. Correlations between the proportions of the six generic components and further comparison with those determined for the unfractionated OM (whole sample, WS), gave information on HA origin and humification mechanisms. In particular, the proportions of char and carbohydrate (R 2 0.637) and contents of lignin and protein (R 2 0.471) in the HAs were negatively correlated (P < 0.05). Significant correlations (R 2 0.439) also existed for char contents in whole sample (WS) compared to HA, and for carbohydrates in WS compared to HA (R 2 0.558). Char proportion grew in HA with respect to the WS, and carbohydrates dropped to a half on average in HA compared to WS.

Conclusions

Two different humification mechanisms could be identified for no-fire and fire areas. In the former, HA-char was preserved selectively from char in the sample, whereas in the latter, char was newly formed by fire effect.

In eukaryotes, termination of messenger RNA (mRNA) translation is mediated by the release factors eRF1 and eRF3. Using Saccharomyces cerevisiae as a model organism, we have identified a member of the DEAD-box protein (DBP) family, the DEAD-box RNA helicase and mRNA export factor Dbp5, as a player in translation termination. Dbp5 interacts genetically with both release factors and the polyadenlyate-binding protein Pab1. A physical interaction was specifically detected with eRF1. Moreover, we show that the helicase activity of Dbp5 is required for efficient stop-codon recognition, and intact Dbp5 is essential for recruitment of eRF3 into termination complexes. Therefore, Dbp5 controls the eRF3-eRF1 interaction and thus eRF3-mediated downstream events.  相似文献

We investigated the prevalence of an anatomic variant of the proximal femur, termed the broomstick-like femoral head and neck formation, and its influence on the Féderation Cynologique Internationale (FCI) hip dysplasia score in 294 German Shepherd dogs. One-hundred and eighty (61%) of the 294 dogs in our study had this anatomic variant. The calculated area of the femoral heads in dogs with a broomstick-like conformation was 4.5 +/- 0.6 cm2 on the hip-extended view. In dogs with a normal femoral head, the calculated area of the femoral head was significantly larger at 4.8 +/- 0.6 cm2 (P < 0.05). In the frog-leg view, there was no significant difference in femoral head area between dogs with the broomstick-like conformation and normal dogs. There was no difference in the antetorsion angle between dogs with broomstick-like conformation and normal dogs. There was also no difference in the distraction ratio between the two phenotypes. The official FCI hip score was similar in dogs with and without the broomstick-like conformation. The average heritability of the broomstick-like conformation was 0.3 +/- 0.1, suggesting heritable influence. We conclude that the broomstick-like conformation is a common finding in the German shepherd dog and has genetic base. The broomstick-like conformation does not appear to be associated with the presence of canine hip dysplasia and it can therefore be assumed to be a normal anatomic variant.  相似文献

OBJECTIVE: To determine whether serum concentrations of cardiac troponin I (cTnI) and cardiac troponin T (cTnT) are increased in dogs with gastric dilatationvolvulus (GDV) and whether concentrations correlate with severity of ECG abnormalities or outcome. DESIGN: Prospective case series. ANIMALS: 85 dogs with GDV. PROCEDURE: Serum cTnl and cTnT concentrations were measured 12 to 24, 48, 72, and 96 hours after surgery. Dogs were grouped on the basis of severity of ECG abnormalities and outcome. RESULTS: cTnl and cTnT were detected in serum from 74 (87%) and 43 (51%) dogs, respectively. Concentrations were significantly different among groups when dogs were grouped on the basis of severity of ECG abnormalities (none or mild vs moderate vs severe). Dogs that died (n = 16) had significantly higher serum cTnI (24.9 ng/ml) and cTnT (0.18 ng/ml) concentrations than did dogs that survived (2.05 and < 0.01 ng/ml, respectively). Myocardial cell injury was confirmed at necropsy in 4 dogs with high serum cardiac troponin concentrations. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that concentrations of cTnI and cTnT suggestive of myocardial cell injury can commonly be found in serum from dogs with GDV and that serum cardiac troponin concentrations are associated with severity of ECG abnormalities and outcome.  相似文献
