The calcium-calmodulin-dependent protein kinase II is a major component of brain synaptic junctions and has been proposed to play a variety of important roles in brain function. A complementary DNA representing a portion of the smaller 50-kilodalton subunit of the rat brain enzyme has been cloned and sequenced. The calmodulin-binding region has been identified and a synthetic analog prepared that binds calmodulin with high affinity in the presence of calcium. Like the 50-kilodalton kinase polypeptide, the concentration of the messenger RNA varies both neuroanatomically and during postnatal development of the brain. The broad tissue and species cross-reactivity of the complementary DNA suggests that the 50-kilodalton subunit found in rat brain is evolutionarily conserved and is the product of a single gene.  相似文献

We compared the effects of dipotassium ethylenediaminetetraacetic acid (EDTA) and lithium heparin on hematologic values of green iguanas (Iguana iguana). Thirty-two privately owned sibling iguanas had blood drawn, and the sample was divided into three components: an EDTA tube, a heparin tube, and a nonanticoagulated blood smear. A full reptilian complete blood count was performed on each anticoagulated sample, and white blood cell (WBC) and leukocyte differential counts were performed on the whole-blood smears. Heparin and EDTA samples differed significantly in absolute values of thrombocytes, WBC, heterophils, and monocytes. The EDTA had no significant effect on the packed cell volume or plasma protein values, and the white blood count and differential counts produced with EDTA were more similar to those of the nonanticoagulated blood smear than were the counts produced with heparin.  相似文献

OBJECTIVES: To characterise the diet of First Nations in north-western Ontario, highlight foods for a lifestyle intervention and develop a quantitative food-frequency questionnaire (QFFQ). DESIGN: Cross-sectional survey using single 24 h dietary recalls. SETTING: Eight remote and semi-remote First Nations reserves in north-western Ontario. SUBJECTS: 129 First Nations (Oji-Cree and Ojibway) men and women aged between 18 and 80 years. RESULTS: The greatest contributors to energy were breads, pasta dishes and chips (contributing over 20 % to total energy intake). 'Added fats' such as butter and margarine added to breads and vegetables made up the single largest source of total fat intake (8.4 %). The largest contributors to sugar were sugar itself, soda and other sweetened beverages (contributing over 45 % combined). The mean number of servings consumed of fruits, vegetables and dairy products were much lower than recommended. The mean daily meat intake was more than twice that recommended. A 119-item QFFQ was developed including seven bread items, five soups or stews, 24 meat- or fish-based dishes, eight rice or pasta dishes, nine fruits and 14 vegetables. Frequency of consumption was assessed by eight categories ranging from 'Never or less than one time in one month' to 'two or more times a day'. CONCLUSION:We were able to highlight foods for intervention to improve dietary intake based on the major sources of energy, fat and sugar and the low consumption of fruit and vegetable items. The QFFQ is being used to evaluate a diet and lifestyle intervention in First Nations in north-western Ontario.  相似文献

Aeromonas hydrophila are known for being opportunistic pathogens, harboring various virulence factors and triggering lesions and death in fish. The disease caused by bacteria can make fish inappropriate for human consumption, besides representing a risk to public health. The pathogenesis can be influenced by environmental variables, affecting fish productivity and mortality. The present study aimed to determine whether A. hydrophila harbor the virulence genes aerolysin, hydrolipase, elastase, lipase, cytotonic enterotoxin (ast), lateral flagellum (laf), and polar flagellum (fla) and to evaluate the influence of environmental variables on in vitro growth, in vivo virulence and expression of some of these genes. Polymerase chain reaction (PCR)-based screening for the presence of these virulence genes was performed on 35 isolates. Six isolates containing different profiles of virulence genes were tested for in vitro growth under different conditions of pH, temperature, and ammonia and for in vivo virulence under these same environmental conditions. RT-qPCR was used to quantify the expression of aerolysin, lipase, and fla genes. All the tested environmental factors influenced the growth of A. hydrophila, while pH and ammonia concentrations influenced the bacterial virulence. The expression of the fla gene increased when bacteria were grown in higher ammonia concentration. The mortality established by Aeromonas is influenced by several environmental factors pinpointing the importance of its control in fish farming to avoid higher economic loses associated to bacterial disease outbreaks.  相似文献

Chondrilla juncea is one of the most serious introduced weed species in Australia. Successful biological control has been achieved by the release of the rust fungus Puccinia chondrillina. Although P. chondrillina is thought to reduce the competitive ability of C. juncea by reducing plant size, the precise mechanism by which the pathogen controls its host is poorly understood. In this experiment, we examined the effect of P. chondrillina TU 788 infection on individual plant size, and size variability, in two genotypes (one resistant, one susceptible) of C. juncea. Exposure to P. chondrillina significantly reduced the average size of susceptible C. juncea plants, a reduction apparent in both roots and shoots. Plant size variability of the susceptible C. juncea genotype was also reduced by P. chondrillina. Plant size, and plant size variability of the resistant genotype were unaffected by exposure to rust, or association with rust‐affected plants. The effect of P. chondrillina on plant size in rust‐susceptible C. juncea plants shows how effective bio‐control of this weed may occur in field populations.  相似文献

Context

Efficient restoration of longitudinal river connectivity relies on barrier mitigation prioritization tools that incorporate stream network spatial structure to maximize ecological benefits given limited resources. Typically, ecological benefits of barrier mitigation are measured using proxies such as the amount of accessible riverine habitat.

Objectives

We developed an optimization approach for barrier mitigation planning which directly incorporates the ecology of managed taxa, and applied it to an urbanizing salmon-bearing watershed in Alaska.

Methods

A novel river connectivity metric that exploits information on the distribution and movement of managed taxon was embedded into a barrier prioritization framework to identify optimal mitigation actions given limited restoration budgets. The value of ecological information on managed taxa was estimated by comparing costs to achieve restoration targets across alternative barrier prioritization approaches.

Results

Barrier mitigation solutions informed by life history information outperformed those using only river connectivity proxies, demonstrating high value of ecological information for watershed restoration. In our study area, information on salmon ecology was typically valued at 0.8–1.2 M USD in costs savings to achieve a given benefit level relative to solutions derived only from stream network information, equating to 16–28% of the restoration budget.

Conclusions

Investing in ecological studies may achieve win–win outcomes of improved understanding of aquatic ecology and greater watershed restoration efficiency.

The purpose of this study was to create a light responsive nanostructured liquid crystalline matrix using a novel alkylated spiropyran photochromic molecule (spiropyran laurate, SPL) as a light activated drug delivery system. The liquid crystal matrix, prepared from phytantriol, responds reversibly to changes in photoisomerism of SPL on irradiation, switching between the bicontinuous cubic and the reversed hexagonal liquid crystal structures, a change previously shown to dramatically alter drug release rate. In contrast, the non-derivatized spiropyran and spirooxazine photochromic compounds do not sufficiently disrupt the matrix on isomerization to induce the phase change. Thus, novel alkylated spiropyran has the potential to be an effective agent for use in liquid crystalline systems for reversible 'on-demand' drug delivery applications.  相似文献
