Based on low 18:0 contents observed in milk fat of cows fed cactus cladodes (CC), we hypothesized that including Opuntia stricta cladodes in a soybean oil (SO)-supplemented diet would promote incomplete rumen biohydrogenation of supplemental PUFA, leading to increased trans-11 18:1 and cis-9, trans-11 CLA contents in milk. Twelve Holstein cows were used in a two-period study: (a) Baseline: all cows received a total mixed ration (TMR) composed of sorghum silage (SS) and a concentrate containing no SO for 14 days; (b) Treatment: cows received one of the following SO-supplemented diets for 21 days: (1) SS-TMR: a TMR composed of SS and a SO-enriched concentrate, (2) CC-TMR: a TMR containing CC as a partial substitute for SS plus the SO-enriched concentrate, and (3) CC-PMR: same diet as in treatment 2, but CC were mixed with the SO-enriched concentrate and fed as a partial mixed ration (PMR). Both CC diets increased relative abundances of trans-11 18:1, cis-9, trans-11 CLA, and 18:2 n-6 in milk fat, whereas opposite effects were observed on 18:0 and cis-9 18:1. Proportion of 18:2 n-6 increased, and cis-9, trans-11 CLA tended to increase with CC-PMR as compared to CC-TMR, whereas 18:3 n-3 was higher with CC-PMR than with SS-TMR. Proportions of several odd- and branched-chain fatty acids, certain 18:1 isomers, and trans-9, cis-11 CLA changed with CC diets, notably with CC-PMR. Milk yield and intake of most nutrients (except fibre) increased or tended to increase with the CC diets, whereas gross milk composition was unaltered. Stearoyl-CoA desaturase-1 index for C18 (SCD18) was higher with CC-PMR than with SS-TMR, and milk n-6:n-3 FA ratio and apparent transfer of 18:2 n-6 to milk increased with CC diets. These results indicate that Opuntia stricta cladodes can be a valuable feed ingredient for improving the nutraceutical value of milk fat.  相似文献

The high Andes region of south Ecuador is characterised by intense land use changes. These changes affect particularly the páramo, which is a collection of high altitudinal grassland ecosystems. In this region, the interaction between airborne volcanic ashes and the cold and wet climate results in very typical soils, with an elevated organic C contents. The physical soil properties are closely related to the high and reliable base flow in rivers descending from the páramo, which makes them important for the socio-economic development of the region. In this study, we analyse the regional variability of the soils in the south Ecuadorian rio Paute basin. In a first part of the study, data from soil profiles along north–south transects are used to determine the soil properties, and to relate the spatial variability of these properties to the major trends in parent material, volcanic ash deposits and climate. The profiles are Histic Andosols and Dystric Histosols devoid of allophane, with very high amounts of organic matter. Significant differences between the western and central mountain range are observed, as well as a general decrease in Andic properties from north to south, coinciding with the decrease in volcanic influence. Finally, the impact of human activities on the soil properties is assessed in a case study in the Machangara valley. Data from 5 profiles, located in an area with natural grass vegetation and a low degree of human impact are compared with 4 profiles in a heavily disturbed, intensively drained cultivated area. Despite the intensity of the land use, very few significant differences are found.  相似文献

Summary In order to introduce currently-available genes with agronomical value into banana, two genetic transformation protocols have been optimized.Firstly, regenerable protoplasts isolated from embryogenic cell suspensions of the cultivar Bluggoe have been used for the introduction of several chimaeric uidA gene constructs by electroporation. With the inclusion of polyethylene glycol and heat shock, the frequency of transiently expressing protoplasts reached 1.8% as shown by an in situ -glucuronidase assay. A duplicated 35S promoter with an alfalfa mosaic virus leader sequence (pBI-426) induced the highest expression rate among the constructs tested.Embryogenic cell suspensions of cv. Bluggoe have also been bombarded with accelerated particles coated with a high expression uidA gene construct (pEmuGN) using a biolistic gun. After a partial optimization of the procedure, transient GUS assays reproducibly demonstrated the presence of 400 blue foci in 30 l of settled cell volume (approximately 25 mg cells).   1. Geospatial models are used to predict the distribution of terrestrial and marine species, according to their ecological and ethological habits. The bottlenose dolphin is a cosmopolitan marine top predator, inhabiting most of the ocean, with the exception of polar and subpolar waters. This wide distribution is associated with a remarkable plasticity in ecological and behavioural habits, which makes it difficult to model and predict its distribution.
  2. This study proposes a ‘multi‐type approach’ to predict the presence and distribution of the bottlenose dolphin in the Pelagos Sanctuary, a Specially Protected Area of Mediterranean Importance located in the north‐west Mediterranean Sea. A multi‐type model based on random forest regression was developed, analysing the distribution habits of two geographical units living in the Pelagos area.
  3. When compared with a classical single‐type model, the multi‐type model performed much better in a prediction test (true skill statistics, TSS = 85% vs. 52%), confirming the value of this experimental approach. This work suggests that wild species should not be considered as one single‐type entity, as local specialization may change and shape their distribution habits.

The chemical composition of the volatile fractions from leaves of three Olea europaea L. cultivars (Leccino, Frantoio, and Cipressino) harvested at two different times of the year were examined by GC and GC-MS. The results showed a high content of aliphatic aldehydes in the three cultivars during both harvesting periods and an increase of (E)-2-hexenal (an aldehyde with high antimicrobial properties) percentage from July to November.  相似文献

Turnover of C and N in an arable soil under Free Air Carbon Dioxide (FACE) experiment was studied by the use of 13C natural abundance and 15N-labeled fertilizers. Wheat was kept four growing seasons under ambient and elevated CO2 concentrations and fertilized for three growing seasons. Density fractionation of soil organic matter (SOM) allowed to track 13C and 15N in free particulate organic matter (fPOM; <1.6 g cm−3), particulate organic matter occluded within aggregates with two densities (oPOM 1.6, oPOM 1.6-2.0 g cm−3), and in mineral-associated organic matter (>2.0 g cm−3) fractions. Elevated CO2 and N fertilization did not significantly affect C and N contents in the bulk soil. Calculated mean residence time (MRT) of C and N revealed the qualitative differences of SOM density fractions: (i) the shortest MRTC and MRTN in fPOM confirmed high availability of this fraction to decomposition. Larger C/N ratio of fPOM under elevated vs. ambient CO2 indicated an increasing recalcitrance of FACE-derived plant residues. (ii) There was no difference in MRT of C and N between lighter and heavier oPOMs probably due to short turnover time of soil aggregates which led to oPOM mixing. The increase of MRTC and MRTN in both oPOMs during the experiment confirmed the progressive degradation of organic material within aggregates. (iii) Constant turnover rates of C in the mineral fraction neither confirmed nor rejected the assumed stabilization of SOM to take place in the mineral fraction. Moreover, a trend of decreasing of C and N amounts in the Min fraction throughout the experiment was especially pronounced for C under elevated CO2. Hence, along with the progressive increase of CFACE in the Min fraction the overall losses of C under elevated CO2 may occur at the expense of older “pre-FACE” C.  相似文献

In Lombardy region (northern Italy) agricultural systems are mostly based on cereals and forage crops, and are normally intensively cultivated. To monitor and analyse the most important agri-environmental issue of this area (water pollution by nitrates and plant protection products, non-renewable fossil energy exploitation, weed dynamics), we conducted an integrated agronomic, environmental and economic assessment of arable and dairy farming using indicators.The structural and management data about farming and cropping systems were collected by periodic interviews over a 3-year period in seven (three arable and four dairy) representative farms.The nitrogen surplus calculated at field scale ranged from low (27 kg N ha?1) to high (339 kg N ha?1) values, depending by the amount of chemical and organic fertilisers applied. Fossil inputs ranged from 11.2 to 46.0 GJ ha?1; the highest values were due to the high use of machinery and chemical inputs. The efficiency factor in energy transformation (an indicator of the dependence of food and feed production on non-renewable energy) ranged from 5.0 to 12.2. Large variability was also observed for economic performance (gross margin; from -364 to 1078  ha?1). The lowest values were observed where the total costs of production (fixed and variable) were elevated. The simplification of cropping systems was responsible for a high probability of weed populations development. The values of Load Index, an indicator describing the potential toxicity of plant protection products on non-target organisms, showed an elevated use of active substances due to the dominance of maize.The indicator-based assessment, founded on a relatively simple data collection procedure, described crop management by combining different aspects into few quantities and highlighted its critical environmental issues. The work represents a starting point for improving cropping system management in the study area; the same procedure could be re-applied if data about improved management scenarios were available.  相似文献

A new method for the evaluation of beer aging, based on a voltammetric analysis of beer distillates, is described. By measuring the current of both acetaldehyde and sulfite voltammetric peaks it is possible to distinguish between fresh, naturally aged, and artificially aged beers. The results obtained for the ratio of acetaldehyde and SO(2) currents correlate well with those given by an expert sensory panel. The kinetics of the combining reaction of sulfite with acetaldehyde was followed for different acetaldehyde/SO(2) molar ratios by using a programmed voltammetric procedure. The formation of an acetaldehyde-sulfite adduct is rapid, and the reaction equilibrium is reached after 30 min, which is in accordance with the results previously obtained by other methods. This voltammetric-based approach seems to be a new attractive tool for detecting chemical equilibria of the addition of sulfite to carbonyls in beer model systems.  相似文献

In order to be able to fertilize oocytes, human sperm must undergo a series of morphological and structural alterations, known as capacitation. It has been shown that the production of endogenous sperm reactive oxygen species (ROS) plays a key role in causing cells to undergo a massive acrosome reaction (AR). Astaxanthin (Asta), a photo-protective red pigment belonging to the carotenoid family, is recognized as having anti-oxidant, anti-cancer, anti-diabetic and anti-inflammatory properties and is present in many dietary supplements. This study evaluates the effect of Asta in a capacitating buffer which induces low ROS production and low percentages of acrosome-reacted cells (ARC). Sperm cells were incubated in the presence or absence of increasing concentrations of Asta or diamide (Diam) and analyzed for their ROS production, Tyr-phosphorylation (Tyr-P) pattern and percentages of ARC and non-viable cells (NVC). Results show that Asta ameliorated both sperm head Tyr-P and ARC values without affecting the ROS generation curve, whereas Diam succeeded in enhancing the Tyr-P level but only of the flagellum without increasing ARC values. It is suggested that Asta can be inserted in the membrane and therefore create capacitation-like membrane alteration which allow Tyr-P of the head. Once this has occurred, AR can take place and involves a higher numbers of cells.  相似文献
