The aim of this study was to characterize agromorphological and molecular relationships between twelve genotypes of roselle (Hibiscus sabdariffa L.) grown in western Mexico. Variables related to crop yield and management at two localities were used for agromorphological characterization. Data were analyzed using principal component analysis. The first four components explained 75.77% of the variation between genotypes. Molecular characterization was conducted using the inverse sequence-tagged repeat marker, which separated the 12 genotypes into two main groups. Three of the genotypes that grouped together at the molecular level had a similar leaf phenotypic characteristic (the number of lobes), which differed from that of the other genotypes. Variation between genotypes was evident in respect of molecular variables, which were used as genetic markers to distinguish between genotypes. Grouping of genotypes according to agromorphological variables, which reflect the response of a genotype to the environment, yielded similar results to molecular grouping. Genetic structure analysis indicated that the China, Fresa, Diamante, and Gigante genotypes probably share a common origin.  相似文献

Pterygoplichthys disjunctivus viscera chymotrypsin was purified by fractionation with ammonium sulfate (30–70 % saturation), gel filtration, affinity, and ion exchange chromatography. Chymotrypsin molecular weight was approximately 29 kDa according to sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), shown a single band in zymogram. Electrofocusing study suggested being an anionic enzyme (pI ≈ 3.9), exhibiting maximal activity at pH 9 and 50 °C, using Suc-Ala-Ala-Pro-Phe-p-nitroanilide (SAAPNA) as substrate. Enzyme was effectively inhibited by phenyl methyl sulfonyl fluoride (PMSF) (99 %), and N-tosyl-l-phenylalanine chloromethyl ketone (TPCK) (94 %). Enzyme activity was affected by the following ions in decreasing order: Hg2+, Fe2+, Cu2+, Li1+, Mg2+, K1+, Mn2+, while Ca2+ had no effect. Chymotrypsin activity decreased continuously as NaCl concentration increased (from 0 to 30 %). K m and V max values were 0.72 ± 1.4 mM and 1.15 ± 0.06 μmol/min/mg of protein, respectively (SAAPNA as substrate). Results suggest the enzyme has a potential application where low processing temperatures are needed, such as in fish sauce production.  相似文献

Wheat flours commercially produced at 74, 80, and 100% extraction rates made from hard white winter wheat (WWF) and hard red winter wheat (WRF) were used to produce tortillas at a commercial-scale level. Flour characteristics for moisture, dry gluten, protein, ash, sedimentation volume, falling number, starch damage, and particle-size distribution were obtained. Farinograms and alveograms were also obtained for flour-water dough. A typical northern Mexican formula was used in the laboratory to test the tortilla-making properties of the flours. Then commercial-scale tortilla-baking trials were run on each flour. The baked tortillas were stored at room and refrigeration temperatures for 0, 1, 2, and 3 days. Maximum stress and rollability were measured every day. Tortilla moisture, color, diameter, weight, and thickness were measured for each treatment. Finally, tortilla acceptability was tested by an untrained sensory panel. Analyses of variance (ANOVA) were performed on the data. WWF had higher protein content, dry gluten, sedimentation volume, and water absorption than the WRF. The WWF was the strongest flour based on farinograph development time and alveograph deformation work. It also produced the most extensible dough measured with the alveograph (P/L). Flour protein and ash contents, water absorption, and tenacity increased directly with the flour extraction rate. Both WWF and WRF performed well in commercial-scale baking trials of tortillas. Tortillas made with both types of flours at 74 and 80% extraction rates had the best firmness and rollability. However, tortillas made with WWF 80% had the best color (highest L value). Tortillas prepared with 100% extraction rate flour were also well accepted by the sensory panel, had good textural characteristics, and became only slightly firm and slightly less rollable after three days of storage at room temperature.  相似文献

To determine if exposure to a sublethal mixture of metals (Cd, Cu, Fe, Mn, Pb and Zn) increases susceptibility to White spot syndrome virus (WSSV) infection, Litopenaeus vannamei juveniles were fed WSSV‐infected shrimp tissues after 21 days of exposure to the metal mixture (WS‐MM treatment). Other treatments consisted of shrimp not exposed to metals and fed infected tissues (WS), and shrimp fed healthy tissues and exposed (MM) or not exposed to metals (C). The presence of viral DNA and inclusion bodies was detected at 32 hr postinfection (hpi) in the stomach epithelium of shrimp from the WS treatment, and eight hours later in shrimp from the WS‐MM treatment, possibly because of an initial negative effect of metals in viral replication. At 40 hpi, the severity of infection represented by the histopathological index increased in both WS and WS‐MM treatments, and values were higher in WS‐MM than in WS shrimp at the end of the experiment. From 56 hpi to the end of experiment, total hemocyte counts were lower in both WS‐MM and WS treatments, and concentrations were particularly low in WS‐MM shrimp. Conversely, phenoloxidase activity was higher in the WS‐MM treatment from 32 to 56 hpi, suggesting a possible role of the prophenoloxidase (proPO) system in the antiviral defense against WSSV. The presence of heavy metals at sublethal concentrations may increase shrimp susceptibility to WSSV; this is supported by a decrease in circulating hemocytes, an increase in the humoral response, and the development of a higher number of WSSV inclusion bodies.  相似文献

The aim of this study was to assess the effect of two rearing conditions: outdoor environment with great volume tanks (500 m3) and low stocking density (~0.4 kg m?3); and indoor environment with smaller volume tanks (10 m3) and higher stocking density (~5 kg m?3), on muscle, liver and ovary lipid composition of Seriola dumerili broodstock born in captivity. The rearing conditions tested seem to affect the pattern of lipid body deposition in broodstock fish of S. dumerili, increasing the muscle and liver triacylglycerides (TG) accumulation of fish kept under indoor conditions, probably due to reduced energy expenditure in swimming, with some variations in the fatty acid profile that may respond to the differences in the culture conditions tested. No significant differences were found for Gonadosomatic Index (GSI) or ovary lipid deposition between groups in this study, which may suggest that the conditions tested do not have a major effect on ovary development. However, one season later the females kept under outdoor conditions released eggs spontaneously, whereas those kept under indoor conditions did not spawn, suggesting that the conditions tested actually have an effect on the broodstock's reproductive fitness. More studies are needed in order to evaluate whether the conditions tested have or not any influence on ovary development.  相似文献

Tree adaptation to environment has been extensively studied. However, little is known about the variations in structure and chemical composition of lignocellulosic biomass (LB) in relation to altitudinal gradient. We wonder, are there significant variations in the LB in the wood across an altitudinal gradient? To answer this, we carried out a study of Abies religiosa. Wood samples were collected from 36 trees, grown between 3000 and 3500 masl, and then subjected to gravimetric and FTIR (Fourier Transform Infrared) spectroscopic analyses. The gravimetric results showed a proportion of 54.81 ± 2.20 % cellulose, 12.37 ± 1.33 % hemicellulose and 24.68 ± 1.16 % of insoluble lignin. Using the principal components analysis with analysis of variance (ANOVA), significant differences were found at 3100 and 3200 masl in two independent components related to both hemicellulose and lignin, through gravimetry as well as the spectroscopic bands assigned to the carbonyl groups of these polymers, respectively. However, the observed changes in chemical composition of LB did not follow a linear relationship with respect to the altitudinal gradient, which suggests that complex environmental interactions could also be playing an important role. Also, there were significant differences (p < 0.05) in two of the empirical indexes calculated from the FTIR analysis.  相似文献

Passing from must to wine produced a loss of low-molecular-weight grape structural glucosyl polysaccharides, and an important gain in yeast mannoproteins (MP) and grape-derived arabinogalactan proteins (AGP), and rhamnogalacturonans-II (RG-II). AGP were more easily extracted than RG-II, and small quantities of RG-II monomers and galacturonans were detected. Postmaceration produced a reduction in all grape polysaccharide families, particularly acute in AGP. The reduction of polysaccharides during malolactic fermentation only affected grape AGP, and MP were continuously liberated during the entire vinification process. Wine oak and bottle aging was associated with a relative stability of the polysaccharide families. AGP were thus the majority polysaccharides in young wines but, contrary to what may be thought, structural glucosyl oligosaccharides dominated in musts and MP in aged wines. Precipitation of polysaccharides was noticeable during vinification, and it mainly affected high-molecular-weight AGP and MP. Hydrolytic phenomena affected the balance of wine polysaccharides during late maceration-fermentation.  相似文献

This paper characterizes wood and charcoal made from Hymenaea aurea, Mimosa scabrella, Tabebuia capitata and Eucalyptus alba by scanning electron microscopy and near infrared for the purpose of developing a technique to identify the source of charcoal samples. Ten test samples were prepared for each species, oriented in transverse, radial and tangential section. Samples were carbonized in a muffle furnace with a final temperature of 450 °C and a heating rate of 1.66 °C/min. The microscopic structures of the wood and charcoal samples were verified by scanning electron microscopy, and spectra were obtained with a spectrophotometer, equipped with an integrating sphere and operating in transmittance mode. The spectra of the wood samples present characteristic bands of lignocellulosic material, while the spectra of the charcoal samples show small or nil absorption in the near-infrared region. The near-infrared spectroscopy technique was efficient in discriminating the four species studied.  相似文献

To investigate the effects of microbial inocula and Erythrina poeppigiana pruning residues on soil K, NO3?, and NH4+ concentrations, a greenhouse trial, a field experiment in an organic farm, and three in vitro tests were conducted. Under controlled conditions, weak, temporary effects (10 %) on maize seedling growth were observed on poor soils (taken from the 10–20 cm layer) in the first 2 weeks after application. Positive effects of pruning residue applications on soil K levels (0.09 cmol kg?1, on average) were detected in both the field and greenhouse study. However, significant effects due to the addition of microbial inocula on soil K concentrations were not detected in the field; thus, microbial applications were ineffective at enhancing nutrient availability under field conditions. In contrast, in the in vitro experiments, CO2 production was 31 % greater than that of untreated soil on the 8th and 15th days of incubation. These results highlight the importance of adding tree pruning residues to support coffee-plant nutrition. Experimental outcome data could be valuable for further studies focused on microbial application dosage and timing.  相似文献
