为初步鉴定并挖掘出猪戊型肝炎病毒(Hepatitis E virus,HEV)ORF3蛋白影响HepG2细胞核黄素代谢信号通路的lncRNA-mRNA调控网络,本试验通过构建腺病毒过表达载体,制备高滴度过表达腺病毒,介导猪 HEV-ORF3在HepG2细胞中实现过表达。Western blotting检测ORF3蛋白过表达成功后,运用lncRNA高通量组学测序,筛选出差异表达的lncRNA并进行靶向差异基因预测,对lncRNA靶向差异基因进行GO功能和KEGG通路富集分析,初步鉴定出与核黄素代谢信号通路相关的lncRNA-mRNA调控网络。Western blotting结果显示,在约为12 ku处出现目的条带,说明成功实现腺病毒介导猪HEV-ORF3在HepG2细胞中过表达。lncRNA高通量组学测序结果显示,共发现102个显著差异表达lncRNAs表达量上调,80个显著差异表达lncRNAs表达量下调。GO功能和KEGG通路富集分析显示,初步挖掘出lncRNA(MSTRG.13995.2)和lncRNA(MSTRG.1960.1)可能是与核黄素代谢信号通路相关的显著差异表达lncRNA,分别通过顺式调控其靶向基因APC-5和FLAD1来影响核黄素代谢信号通路。本试验初步鉴定出lncRNA(MSTRG.13995.2)-APC5和lncRNA(MSTRG.1960.1)-FLAD1可能是影响HepG2细胞核黄素代谢信号通路的lncRNA-mRNA调控网络。  相似文献

In this study, the effects of mixing process parameters (degree of vacuum, water addition, and mixing time under vacuum) on the cooking and sensory quality properties of Chinese white noodles were investigated by using one commercial‐scale noodle production line and one typical commercial wheat flour. Noodle appearance, firmness, elasticity, smoothness, and total quality scores were significantly improved as the degree of vacuum increased from 0 to 0.06 MPa, although lower sensory scores and larger cooking losses occurred when noodles were mixed at 0.08 MPa. Noodles with a water addition of 35% had the highest total score and the highest scores for each sensory factor. As mixing time increased, the sensory score of cooked noodles increased initially and then decreased. With a mixing time of 7 min, the sensory score was the highest and cooking loss was the lowest. The results of response surface methodology indicated that fresh noodle quality was most affected by the water addition, followed by vacuum degree. Added water was a more important source of variation for appearance, firmness, stickiness, smoothness, total score, and cooking loss than degree of vacuum and mixing time, whereas degree of vacuum was the predominant source of variation for color and elasticity. The interactions between the factors had little effect on sensory and cooking properties. The optimal mixing conditions were determined to be as follows: degree of vacuum, 0.06 MPa; added water, 35.6%; and mixing time, 7.25 min. Furthermore, vacuum mixing produced a more even, coherent, and closed microstructure for the sheeted dough than nonvacuum mixing.  相似文献

以经过栽培的木耳菌棒为试验材料,在万能试验机上对不同含水率的菌棒试样进行剪切、压缩的力学性能试验。试验结果表明:含水率是影响菌棒剪切强度、抗压强度的重要自然因素。平均含水率分别为11.75%,21.16%,27.68%的菌棒其平均剪切强度分别为29.4k Pa、20k Pa、14.5k Pa。平均含水率分别为14.3%,24%的菌棒,其平均抗压强度分别为144.7k Pa、77.7k Pa。菌棒的含水率与剪切强度,抗压强度均呈幂指数关系。这些都为研制菌棒粉碎机械、选择粉碎参数提供了设计依据。  相似文献

山药收获机下支撑支架是根据主旋转支撑装置的离地高度和开沟深度设计的,在实际作业过程中,链刀切削阻力、螺旋刀切割阻力、输送机构支撑力、油缸的推拉力最终都由下支撑支架承载,作用力较为复杂,容易发生变形和断裂;且链刀切入土壤产生的振动、链节和链轮齿的啮合会给链传动带来工作的不平稳性和有规律的振动,可能会引起共振。为此,在进行山药收获机工作装置设计时,首先采用有限元方法对下支撑支架进行静力学分析,并在静力学分析的基础之上对其进行预应力模态分析,以评估设计的合理性,对结构进行优化。  相似文献

The experiment aimed to find out the effects of dietary crude protein levels on growth performance, digestion and metabolism and serum biochemical indexes of Super Merino lamb after weaning.According to the dietary crude protein level, 64 Super Merino weaned lambs were divided into four groups, which the crude protein levels were 13.25%(group Ⅰ), 14.33%(group Ⅱ), 15.53%(group Ⅲ) and 16.60%(group Ⅳ), respectively.The experiment lasted for 60 days, 30 days for the early stage and 30 days for the later stage;And on the 30th and 60th day, blood samples were collected from one head sheep chosen from each replication, and at the end of the feeding test, 3 lambs were randomly selected from each experiment group for a 15 days digestion and metabolism experiment.The results showed that ADFI and ADG in the early and total trial period were significant differences(P<0.05), ADFI and ADG of group Ⅲ were higher than others.The apparent digestibility of nutrients and ME/DE were not significantly changed(P>0.05).In the detection of serum biochemical indexes, there was no significant difference in the earlier stage(P>0.05);At the end of the test, PK and CK showed significant difference between groups(P<0.05).PK presented a rising trend as the protein level improved, CK of group Ⅰ was higher than other groups.In Super Merino lamb early weaning diet, 15.53% protein level in the diet could significantly improve ADG, the apparent digestibility of nutrients, metabolic energy digestibility.At the end of the test, PK and CK were affected by dietary crude protein level significantly.  相似文献

This experiment was conducted to investigate the effects of complex nutritional regulation additives on growth performance, slaughter performance and serum biochemical indexes of grazing sheep in cold season.A single factor random block design was used and 48 grazing ewes with the average weight (28.01±1.70) kg and 7 months old were randomly divided into 4 groups:Group Ⅰ without supplement;Group Ⅱ with concentrate;Group Ⅲ with concentrate+complex formulation 1 (0.30% malic acid, 0.06% cysteamine, 0.08% saccharicterpenin);Group Ⅳ with concentrate+complex formulation 2 (0.60% malic acid, 0.12% cysteamine, 0.16% saccharicterpenin)with 3 replicates per group and 4 grazing ewes per replicate.The results showed that:①The final weight of group Ⅲ was the highest and higher than group Ⅱ and Ⅳ by 6.26% (P>0.05)、3.69% (P>0.05), respectively.The average daily gain of group Ⅲ was highest, and higher than group Ⅱ and Ⅳ by 26.32% (P<0.01)、5.74% (P>0.05) respectively.②Comparing with group Ⅱ, the slaughter rate of group Ⅲ was increased by 4.68% (P>0.05).Compared with groupⅠ, Ⅱ and Ⅳ, the meat percentage of group Ⅲ was increased 18.46% (P<0.05), 10.97% (P>0.05) and 4.40% (P>0.05), while the net meat weight of carcass in group Ⅲ was increased by 49.74% (P<0.01), 13.22% (P>0.05) and 18.27% (P<0.05), respectively.Compared with groupⅠ, Ⅱ and Ⅳ, the net meat rat of carcass in group Ⅲ was increased 10.42% (P<0.01)、6.00% (P>0.05) and 3.37% (P>0.05), respectively.③There was a rising trend as the order of groupⅠ, Ⅱ, Ⅳ and Ⅲ in serum glucose, Ca, P, HDL, LDL, GPT and IgG, while A/G and TG showed a decreasing trend.Based on the above results, in this trial, supplementing complex nutritional regulation additives had good effects on growth performance, slaughter performance and serum biochemical indexes of grazing sheep in cold season.And supplementing complex formulation group 1 (group Ⅲ) was the best.  相似文献

This experiment was designed to study the complex of Lactobacillus rhamnosus fermented herbal and Bacillus subtilis on White Feather broiler immunity performance and impact of Escherichia coli infection.360 one-day-old broiler chickens were randomly divided into 3 groups with 4 replicates in each group and 30 chickens per replicate.The pretrial period lasted for 7 d,and the experiment lasted for 35 d.The chickens in the group Ⅰ(positive control group) and group Ⅱ(negative control group)were all only fed a basal diet,group Ⅲ was test group,by additive 1% fermented herbal preparations,groups Ⅱ and Ⅲ were intraperitoneal injection of 1 mL E.coli at 35 d,broiler mortality,immune organ index,cecalmicroflora content,immunoglobulin levels,IL-2 and IL-6 content were tested.The results showed that injection of E.coli caused massive death of chickens,group Ⅱtook up to 75.00%,it was significantly higher than groups Ⅰ and Ⅲ (P < 0.05),the mortality in group Ⅲ was significantly lower than group Ⅱ(P < 0.05),was only 23.33%.Injection of E.coli maked spleen index and thymus index of group Ⅱincreased significantly (P < 0.05),the spleen index and thymus index of groups Ⅰ and Ⅲ were no significant difference (P > 0.05).E.coli counts was significantly decreased after injectionin group Ⅲ (P < 0.05),but the number of intestinal Lactobacilli of group Ⅲ was significantly increased (P < 0.05),and inhibited the propagation of E.coli,the counts of E.coli in groups Ⅰ and Ⅲ were no significant difference (P > 0.05).At 42 d,the sIgA of the intestinal fluid in group Ⅲ were higher than that of groups Ⅰ and Ⅱ with 11.99% and 36.56%,respectively(P < 0.05).The serum IgG concentrations of group Ⅲ was higher than that of groupsⅠand Ⅱwith 14.68% and 28.15%,respectively(P < 0.05).At 42 d,the IL-2 content of group Ⅱ was the lowest,it was significantly lower than group Ⅲ(P < 0.05),the IL-6 of group Ⅲ was significantly lower than group Ⅱ(P < 0.05).  相似文献

In order to understand the security of a new kind of antidiarrheal Chinese herbal medicine compound preparation for livestock,acute and sub-chronic toxicity test were conducted.Acute toxicity test used the largest drug dose method,20 Wistar rats were orally treated with the Chinese medicine compound preparation.In the sub-chronic toxicity test,80 rats were randomly divided into 4 groups with 20 rats in each group and orally given a dose of 3 000,1 500,750 and 0 mg/(kg·BW)of Chinese medicine compound preparation once a day for 30 days.The general clinical status was observed,rats weight were measured and the dose was adjusted every week during the test,after the test measured blood routine index,biochemistry index,and preceded the gross anatomy observation,weighing each major organs and calculated the viscera coefficient,and proceded main viscera histopathological observation between the high dose group and the control group.The acute toxicity results showed that every rat would be alive gavaged with the lethal dose(LD50)of compound preparation larger 5 g/(kg·BW).The sub-chronic toxicity autopsy showed that except heart,lung,and testicles in individual rats appeared mild bleeding in the high dose group,the other dose group organs found no abnormal change.The haematological index showed except mononuclear cell rate(P<0.05),and hematocrit declined significantly(P<0.05)in the high dose group,all the indexes of the other groups were in the normal range,there was no significant difference from the control group.The test suggested the Chinese medicine compound preparation was no toxicity under the condition of this test according to acute toxicity classification standard of exogenous chemicals by WTO,there was no effect on the growth and development of rats in the sub-chronic toxicity test,and there was no chronic toxicity at least 1 500 mg/kg feeding conditions in short-term repeated application.  相似文献

In order to understand the main bacterial pathogen species causing dairy cow mastitis in Liaoning, as well as the characteristics of drug sensitivity of the pathogenic E.coli,the milk samples from 75 dairy cows with clinical manifestations for mastitis in certain large-scale dairy farm in Liaoning were collected.The bacteria in milk were cultured and isolated with biochemical methods and in vitro drug sensitivity tests were processed with the isolated E.coli strains.The results showed that the main bacterial pathogen for dairy cow mastitis were E.coli(separation rate 58.7%),S.aureus(64.0%)and S.agalactiae(54.7%),and multiple infection including double and triple infection were identified.The drug sensitivity tests on the isolated E.coli indicated that the E.coli isolates were highly resistant to sulfonamides(resistance rate>85%)and chloramphenicol(resistance rate>30%),and they were relatively low resistant to ampicillin(9.5%),ciprofloxacin(9.5%),ceftiofur(7.1%)and ofloxacin(4.8%).The results was able to provide reliable theoretical basis for prevention and control of dairy cow mastitis in Liaoning area.  相似文献
