Genes for epilepsy mapped in the mouse   总被引:22,自引:0,他引:22  
The neurological mutant mouse strain E1 is a model for complex partial seizures in humans. The inheritance of epileptic seizures with seven conventional chromosomal markers and over 60 endogenous proviral markers was studied by means of back-crosses of E1 with two seizure-resistant strains, DBA/2J and ABP/LeJ. The major gene responsible for this epileptic phenotype (El-1) was localized to a region distal with respect to the centromere on chromosome 9. At least one other gene, El-2, linked to proviral markers on chromosome 2, also influences the seizure phenotype. In addition, a potential modifier of seizures was detected in the DBA/2J background. The location of El-1 on distal chromosome 9 may allow identification of an epilepsy candidate gene in humans on the basis of conserved synteny with human chromosome 3.  相似文献

OBJECTIVE: To determine clinical, radiographic, and pathologic abnormalities in dogs with multiple epiphyseal dysplasia (MED). DESIGN: Retrospective case series. ANIMALS: 19 dogs with MED from 10 litters. PROCEDURES: The diagnosis was made on the basis of radiographs of the shoulder region and vertebral column. Ten dogs underwent necropsy. RESULTS: There were 11 Hygenhund, 6 Dunker, 1 Golden Retriever, and 1 English Pointer. Most dogs were examined because of lameness that developed at 5 to 8 months of age. The most common radiographic abnormality was a deficiency in ossification of the epiphyses, apophyses, and cuboidal bones of the appendicular skeleton and the epiphyses of the vertebrae; ossification of the metaphyses and the diaphyses typically were normal. Disease severity was consistent among littermates, but varied among dogs from different litters. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that MED is a rare condition in dogs characterized by a deficiency in ossification of the epiphyses of the long bones, the epiphyses of the vertebrae, the cuboidal bones, and the apophyses. Radiographic abnormalities were evident in dogs examined as young as 8 weeks of age, and most dogs had developed severe lameness by 5 to 8 months of age. The condition most likely had an autosomal recessive mode of inheritance, although genetic studies of mode of inheritance could not be performed.  相似文献

Despite the importance both in an ecological and management context, much uncertainty remains about the underlying factors controlling spawning ground use in marine fish. Here, we investigate how spawning ground use of Northeast Arctic (NEA) haddock (Melanogrammus aeglefinus) has changed over time. By combining data from a Soviet‐Russian egg survey conducted over a 35 year period (1959–1993) with a biophysical drift model, we estimate annual median spawning latitudes of NEA haddock. Furthermore, we test for statistical association between the median latitude of spawning and covariates that are candidates for shaping the spawning distribution of haddock. The results indicate no association with temperature, as measured at the Kola transect, or demographic structure, as measured by the mean weight of spawners. However, we find a positive and significant association between spawning latitude and spawning stock biomass, indicating that density‐dependence may be more important in shaping spawning location than other factors such as temperature and demographic structure. A potential mechanism for the reported effect of spawning stock biomass may act through the distribution at the feeding grounds, which is reported to expand north‐ and eastwards at high population density.  相似文献

The effect of adding carotenoids from various sources to feed of juvenile freshwater crayfish Cherax quadricarinatus on pigmentation, growth and survival was examined under laboratory conditions. Juvenile crayfish were fed diets fortified with carotenoids at a level of 100 μg g−1 for a period of either 49 or 55 days. Carotenoid sources were: dried algal cells prepared from Dunaliella salina in which the main carotenoid is β-carotene; a synthetic carotenoid, astaxanthin (Carophyll Pink 8%, manufactured by Hoffman–La Roche) and alfalfa meal. Crayfish receiving feeds fortified with carotenoids exhibited better body colouration than those in the control group, which were fed a diet to which no carotenoids were added. Growth and survival of the crayfish were not affected by the addition of carotenoids to their diet.  相似文献

In this study, we demonstrate how an event that is initially frightening to Atlantic salmon is turned to a positive stimulus through habituation and associative learning. The study was carried out in four commercial sized tanks (5 m3) with near industry densities (>550 fish, 16 kg m−3), using a delay conditioning procedure with an aversive flashing light as the conditioned stimulus and food reward as the unconditioned stimulus. By using video image analysis of the distribution of the fish in the tanks, the changes in behaviour from trial to trial could be documented in great detail. The current study documents the change in behaviour across the individual conditioning trials, clearly showing the step-by-step nature of the transition. The salmon needed more than 26 trials to become fully habituated to the flashing light but showed clear anticipatory behaviour already after about 19 trials. This demonstrates that the learning process is a combination of habituation and associative learning.  相似文献

This study comprised the results of three different seawater trials using unique combination of techniques to study protease digestive efficiency and growth performance quality to illustrate the effects of light regimes and vaccine types in Atlantic salmon (Salmo salar L.). Fish with higher growth had higher trypsin (T) and chymotrypsin (C) specific activities with higher T/C ratio or slope T/C ratio [calculated from the regression between trypsin (y) and chymotrypsin (x) specific activities] in the pyloric caeca. The T/C ratios indicated fish growth rates over a period of 1–2 months, while the slope T/C ratios indicated fish growth rates at sampling. Adaptation period for adjustment to the new environment of continuous light was 70 days, indicated by the differences in trypsin specific activities and the crossing of slope T/C ratio regressions following with the changes in growth rate directions between the control and the treated group. Vaccine types affected fish vertebral growth, and additional continuous light enhanced the impact of vaccines on fish growth during springtime, indicated by differences in slope T/C ratios. Continuous light stimulated fish growth during winter to spring, when the natural day length was short, without significantly changing white muscle and oocyte qualities in the fish of about 500 g, except for significantly increased white muscle RNA concentration. Continuous light also reduced fish growth rate later during summer, when the natural day length was long, by precedently decreasing the T/C ratio in late spring. Interestingly, plasma levels of free lysine related to tryptic digestion were correlated with trypsin specific activity levels. Continuous light caused higher levels of most free amino acids (FAA) involved in nitrogen metabolism, higher incorporation of essential FAA for protein synthesis, and higher protein turnover rate (free hydroxyproline levels) in both plasma and white muscle. However, continuous light did not affect higher protein content, intracellular buffering capacity and RNA levels in the white muscle of the fish of about 1 kg, probably due to limitation of FAA available for protein synthesis. It is therefore suggested that enhancing fish growth by continuous light stimulation should be accompanied by increasing availability or content of dietary protein (and probably minerals), which in turn would improve the quality of fish growth performance through increasing fillet protein concentration, strengthening vertebral growth, and delaying oocyte development.  相似文献

The potential of marine benthic cyanobacteria as a source of anticancer drug candidates was assessed in a screen for induction of cell death (apoptosis) in acute myeloid leukemia (AML) cells. Of the 41 marine cyanobacterial strains screened, more than half contained cell death-inducing activity. Several strains contained activity against AML cells, but not against non-malignant cells like hepatocytes and cardiomyoblasts. The apoptotic cell death induced by the various strains could be distinguished by the role of caspase activation and sensitivity to the recently detected chemotherapy-resistance-associated prosurvival protein LEDGF/p75. One strain (M44) was particularly promising since its activity counteracted the protective effect of LEDGF/p75 overexpressed in AML cells, acted synergistically with the anthracycline anticancer drug daunorubicin in AML cells, and protected cardiomyoblasts against the toxic effect of anthracyclines. We conclude that culturable benthic marine cyanobacteria from temperate environments provide a promising and hitherto underexploited source for novel antileukemic drugs.  相似文献

We evaluated whether bearing tetrodotoxin (TTX) affects salinity stress in the juvenile tiger puffer Takifugu rubripes. Juveniles of hatchery-reared non-toxic T. rubripes [body weight (BW): 1.7?±?0.2 g, n?=?120] were divided into six tanks and acclimatized to salinity (8.5 ppt) that is equivalent to blood osmolality. Fish in three tanks were fed non-toxic diet, and those in the other three tanks were fed a TTX-containing diet (356 ng/g diet) three times a day until satiation. In each diet treatment, salinity of one tank was kept at 8.5 ppt, and the other two tanks were adjusted to either 1.7 or 34.0 ppt, and fish were reared for another 33 days. Then, we compared survival, growth, TTX accumulation, plasma osmolality, plasma cortisol, and glucose levels among treatments. We detected TTX only in the fish in the TTX-diet groups. Survival was highest at 8.5 ppt (70%) and lowest at 1.7 ppt in the TTX-diet group (20%). The BW was greater at 8.5 ppt, and plasma osmolality was significantly higher at 34.0 ppt than at any other salinities. Plasma cortisol level was significantly higher but glucose level was lower at 1.7 ppt. Possessing TTX at a low salinity may be lethal to tiger puffer juveniles.
