The 1997-1998 El-Niño Southern Oscillation was the most severe on record and dramatically impacted corals worldwide. However, the effect of this event on the associated community of reef organisms has received much less attention. The composition of the bryozoan assemblage from the coral reefs of Northern Bahia, Brazil were monitored annually from 1995 to 2000, allowing the investigation of the effects of this large-scale stressor on an important, diverse, yet understudied component of the coral reef system. Bryozoan samples (35 replicates/reef) were collected during April/May from four shallow bank reefs (10-40 m depth) located a few kilometres off the coast, together with measurements of the associated environmental parameters. Currently 157 species have been recorded from the study area, but significant reductions in density and diversity were apparent between pre- and post El-Niño Southern Oscillation (ENSO) years, multivariate analysis denoting significant changes in assemblage composition. A total of 61 species were unrecorded following the 1997-1998 ENSO event (22 species from 1997; 25 further species from 1998 and 14 more from 1999). These included several species endemic to Brazil, suggesting that the 1997-1998 ENSO has had a marked influence on the reef bryozoan community, resulting in the local extinction of several species. Bryozoan mortalities were probably initiated by elevated temperatures, but continued disappearance of species for 2 years after ENSO suggests other indirect factors are also influential. In order to reduce nutrient wastes to the environment the supply should be in accordance to the demand for these. Two experiments were conducted to study and quantify the effect of temperature, irradiance, and plant age on the uptake of nitrate (NO3?), ammonium (NH4+), dihydrogen phosphate ion (H2PO4?), potassium (K+), calcium (Ca2+), magnesium (Mg2+), and sulfate (SO42). In the first experiment, various levels of temperature and irradiance were applied to plants in a growth chamber, while in the second experiment the uptake was studied along the crop season under greenhouse conditions. The uptake rates were calculated at 2-hour intervals through sampling the nutrient solution and analyzing it by inductively coupled plasma atomic emission spectrometry (ICP-AES). Increasing light and temperature enhance the uptake rates, while the rates decrease with plant age. Nitrogen absorption was similar during the day as during the night. No differences were found in the absorption of H2PO4?, K+, Ca2+, Mg2+, and SO42? between day and night. The red grapes co-winemaking effect on phenolic fraction and wine color has been studied for the first time, where Monastrell was comacerated and cofermentated with Cabernet Sauvignon and Merlot. Changes in the relative abundance of anthocyanins were observed as well as hyperchromic shifts at 530 and 620 nm; these effects remain constant after aging. Co-winemaking also favored copigmentation, giving way to more stable anthocyanins and facilitating their polymerization. With regard to color evolution, the mixture of Monastrell with Merlot grapes was more appropriate than with Cabernet Sauvignon for aging wines in oak barrels. The extent of copigmentation was more important in young wines than in aged wines. This is mainly due to the self-anthocyanin monomer reactions in the case of young wines, whereas in aged wines copigmentation is mainly due to the reaction between the anthocyanins and other polyphenolic cofactors. Discriminant analysis showed the possibility of differentiating wines according to the aging time and the type of wine, with color parameters (color intensity, OD 620 nm, and OD 520 nm) being the most important discrimination variables in the first case and petunidin-3-glucoside and peonidin-3-glucoside contents in the second case. 相似文献

Sweet peppers (Capsicum annuum L.) cv. Vergasa have been studied at four maturity stages (immature green, green, immature red, and red). The individual phenolics (hydroxycinnamic acids and flavonoids), vitamin C (ascorbic acid and dehydroascorbic acid), and individual carotenoids were characterized and quantified. Five hydroxycinnamic derivatives and 23 flavonoids were characterized and quantified from the pericarp of sweet pepper by high-performance liquid chromatography-diode array detection-electrospray ionization-mass spectrometry. Identification was carried out by their UV spectra, chromatographic comparisons with authentic markers, identification of hydrolysis products, and tandem mass spectrometry analysis. Hydroxycinnamic derivatives, O-glycosides of quercetin, luteolin, and chrysoeriol, and a large number of C-glycosyl flavones have been characterized. Some of these compounds were found for the first time in nature. Clear differences in the individual and total phenolic content were detected between the different maturity stages. Immature green pepper had a very high phenolic content while green, immature red, and red ripe peppers showed a 4-5-fold reduction. Ascorbic acid was the main form of vitamin C, and its content increased as the pepper reached maturity. The red ripe stage had a relevant impact on the carotenoids content. Thus, immature green peppers showed the highest content of polyphenols, while red ripe fruits had the highest content of vitamin C and provitamin A. 相似文献

A method was established for the identification and quantification of carotenoids including geometrical isomers in fruit and vegetable juices by liquid chromatography with an ultraviolet-diode array detector, using a C(18) Vydac 201TP54 column. The mobile phase used was the ternary methanol mixture (0.1 M ammonium acetate), tert-butyl methyl ether and water, in a concentration gradient, and a temperature gradient was applied. Retinol palmitate was added as an internal standard. An extraction process (ethanol/hexane, 4:3, v/v) was performed, followed by saponification with diethyl ether/methanolic KOH (0.1%, w/v, BHT) (1:1, v/v) for 0.5 h at room temperature. Seventeen different (cis and trans) carotenoids were identified by UV-vis spectra and retention times in HPLC in the juices analyzed. A headspace solid-phase microextraction (HS-SPME) procedure at 30 degrees C with a 100 microm PDMS fiber of a saturated NaCl solution stirred at 1100 rpm combined to GC-ECD for the 2,4,6-trichloroanisol (TCA) determination in wines has been developed. Due to the matrix complexity and ethanol absorption into the fiber, the internal standard selection was crucial to obtain unbiased results. Thus, matrix effects were observed when analyzing different types of Spanish wines (white, early, and vintage red wines) spiked with TCA at low concentration levels (i.e., <40 ng L(-)(1)). In contrast, the use of 2,4,6-tribromoanisole (TBA) as internal standard overcame these matrix effects, whereas the use of 2,4,6-trichlorophenyl ethyl ether led to inconsistent results. The developed HS-SPME-GC-ECD methodology reaches a limit of quantitation for TCA in wine within 2.9-18 ng L(-)(1), with a relative standard deviation of 2.5-13.4%, depending on the TCA concentration level and wine characteristics. This analytical method is comparable to the existing methodologies based on HS-SPME followed by GC-MS in terms of accuracy, precision, length of determination, and length of quantification; however, analysis cost is reduced. 相似文献

Temporal variability is a key factor to understand the structure of belowground communities. Seasonal and annual variations are especially relevant in unpredictable desert ecosystems, where macroinvertebrates are poorly known, despite constituting an important group of soil organisms. In the present study, we analyse the composition and temporal (seasonal and annual) variations of soil macroinvertebrates in an arid area of southern Spain. During two years, macroinvertebrates were sampled in litter and belowground levels by means of soil cores. Results show that the assemblage was dominated by arthropods, especially Formicidae and Coleoptera. The assemblage differed between litter and belowground levels. In litter, detritivores dominated the community, while belowground fauna showed a similar proportion of detritivores and herbivores and a low percentage of predators. Litter and belowground assemblages showed seasonal variations in richness, abundance, biomass and composition, although variations were more marked in litter than belowground. Patterns of seasonal variation also differed between the two study years for both litter and belowground invertebrates. The seasonal and annual variability of the assemblage has potentially important implications for community dynamics in the study system, since the changes in species composition and trophic structure of soil invertebrate assemblages may affect species interactions and food web dynamics over time. Therefore, integrating temporal variability is likely to be crucial to understand soil community dynamics and food webs, especially in heterogeneous, variable systems as deserts. 相似文献

Soils were examined at 2505 m elevation in Haleakala's crater (Maui, Hawai?i) beneath 50 adult Hawaiian silversword plants (Argyroxiphium sandwicense DC.); mean canopy diameter was 42.0 cm. Exposed volcanic Inceptisols (Andic Humitropepts) seem significantly eroded beyond the dense rosette crowns, but remain unaffected below plants. Rosettes are perched on isolated basal soil mounds or pedestals 27–121 mm high (mean: 77.5 mm). Geomorphic field response of soils below rosettes and adjacent (∼ 100 cm apart) bare soils differs. Infiltration rates are higher under plants (mean: 158.7 mm/min) than in exposed control soils (60.0 mm/min). Soils below silverswords also show greater shear strength (146.1 g/cm2) and compressibility (2.795 kg/cm2) than unprotected soils (36.1 g/cm2, and 0.108 kg/cm2, respectively). Soil in the plant mounds contains more organic matter; this has influenced other pedological properties, which also differ substantially between sampling positions. Substrate under plants has a porosity ∼ 53% higher than exposed soil, while bulk density is 62% higher in soil outside the plant crown. The observed microtopographic differences are ascribed to greater soil erosion by rainsplash and runoff outside the silversword canopy. The dense rosette crown effectively intercepts raindrops; soils beneath plants also have a high surface cohesiveness provided by a dense network of fine plant rootlets and partially decomposed organic material. White clover is one of the most important forage legume species worldwide, playing an important role in Southern Brazil temperate
cultivated pastures. This work was aimed to characterize the genetic variability of the USDA white clover core collection
formed by 78 accessions representing 50 countries, together with two very well known cultivars (Huia and Ladino Regal), using
RAPD (Random Amplified Polymorphic DNA) markers to produce genetic fingerprints. There were used DNA bulks formed by the extraction
and mixture of 20 random individuals from each accession. Twenty four primers were used, which revealed from 3 to 29 bands,
forming a total of 371 polymorphic bans and only one monomorphic, ranging from 50 to 3098 bp. The results showed a genetic
similarity among the accessions, ranging from 0.18 to 0.58 (Jaccard’s index), with an average of 0.24, allowing the identification
of each individual accession using just three primers. The results also showed a large genetic variability within the white
clover core collection, probably due to its reproduction mode and ploidy level, which could be used in plant breeding program. 相似文献
