Diacronema sp. was cultured and its sterols were separated by column chromatography on silica gel. The new sterol 24-ethyl-4alpha-methyl-cholestane-3,20-diol (1) was characterised by NMR and MS spectrometry, as well as (22E)-24-ethyl-4alpha-methyl-5alpha-cholest-22-en-3beta-ol (2) and beta-sitosterol, the major components of the sterol fractions. The aim of this study was to assess the effects of feeding Atriplex halimus (AH) silage treated with two developed enzyme cocktails to sheep on feed intake, nutrient digestibility and ruminal fermentation. The AH silage was treated without or with 2 L of ZAD1® or ZAD2®/1000 kg with 5% molasses and ensiled for 30 days. Barley grain (300 g/head/day) was fed as an energy supplement once daily at 10.00 hours and AH silage with or without enzyme treatment was offered ad libitum to animals twice daily at 09.00 and 16.00 hours. Sheep were fed on four experimental forage diets comprised of AH silage and barley (D1), AH silage treated with ZAD1® and barley (D2), AH silage treated with ZAD2® and barley (D3) and AH silage treated with a combination of ZAD1® and ZAD2® (1:1) and barley (D4). Ensiling AH with enzymes reduced its contents of neutral detergent fiber and acid detergent fiber. The dry matter intake of AH of D2, D3 and D4 decreased (P < 0.001) as compared to D1. However, enzyme‐treated diets had greater total digestible nutrients intake (P < 0.001) as compared to D1. The nutrients digestibility for D2, D3 and D4 were higher than those for D1 (P < 0.001), and were higher for D3 as compared to both D2 and D4. Sheep fed on D3 had highest (P < 0.001) ruminal total volatile fatty acids concentration, ammonia nitrogen concentration and microbial protein yield. It could be concluded that AH silage treated with ZAD1® or ZAD2® improved digestibility and rumen fermentation in sheep. 相似文献

In the present work sub- and supercritical extraction conditions using carbon dioxide were studied in order to obtain extracts with different compositions from the green microalgae Dunaliella salina. Different compositions of beta-carotene isomers were identified in the extracts by using HPLC-DAD. Also, antioxidant activity of the extracts was measured using a TEAC assay. An experimental design was applied considering two factors, extraction pressure and temperature, in a wide range of values, trying to maximize the extraction yield. Higher yields were obtained at high pressures and low temperatures, that is, at higher CO2 densities. Attempts were made to correlate the antioxidant activity of the extracts with their chemical composition by means of principal component analysis. A certain relationship was found between their antioxidant activity and the isomeric composition of beta-carotenes. As a result, an original equation is proposed to predict the antioxidant activity of extracts from D. salina in terms of the ratio 9-cis-beta-carotene/all-trans-beta-carotene, the concentration of alpha-carotene, and, especially, the concentration of 9-cis-beta-carotene. 相似文献

Trichobilharzia regenti is a neurotropic bird schistosome,causing cercarial dermatitis in humans. In this study, ZAP cDNA expression library from Radix peregra s. lat. hepatopancreases containing intramolluscan stages of T. regenti was constructed and screened using PCR with specific and degenerate primers, designed according to previously described serine and cysteine peptidases of other parasite species. Full-length sequences of cathepsins B1 and L, and two serine peptidases, named RpSP1 and RpSP2, were obtained.The protein-protein BLAST analysis and parallel control reactions with template from hepatopancreases of T. regenti non-infected snails revealed that only cathepsin B1 was of parasite origin. The remaining sequences were derived from the snail intermediate host, which implies that the initial source of parasite mRNA was contaminated by snail tissue.Regardless of this contamination, the cDNA library remains an excellent molecular tool for detection and identification of bioactive molecules in T. regenti cercariae. 相似文献

The objective of the present study was to quantify the marketing margins of cold sheep carcass and barbacoa meat in the southern state of Mexico. Data were obtained from the production chain links (production, industrialization, and marketing) where the marketing margins were calculated along with the benefit/cost (B/C) ratio. In the absolute marketing margin of the final consumer price per kilogram of carcass meat, the producer obtained US$2.7/kg (47 %) of the utilities, while the intermediaries obtained US$3.1/kg (53 %). Considering the final cooked product in barbacoa (typical dish), the margin was US$6.3/kg (29 %) for the producer and US$15.2/kg (71 %) for the intermediaries. The B/C ratio was 1.0, 1.1, 2, and 1.3 for the producer, stocker, butcher, and barbacoa seller, respectively. It is concluded that the best marketing channel for the producer was the producer-stocker-processor (butcher and barbacoa seller). The highest marketing margin was for the intermediaries followed by the producer. The order of importance of the B/C kilogram ratio of meat was for the butcher first, then barbacoa seller, and lastly stocker and producer. 相似文献

The last remains of native laurel forest in the Azores are highly threatened by the spread of invasive plants. Because landslides are very frequent in these islands, conservation of native laurel forest requires knowledge of the patterns of bird-dispersed seed rain into forest gaps. We monitored 78 seed traps over 1 year to investigate (1) the role of perches in attracting avian dispersers into gaps, (2) temporal patterns in the dispersal of exotic and native seeds, (3) how seed rain affects vegetation establishment in gaps at different distances from the native forest and (4) whether the caloric content of fruits could explain the number of seeds dispersed. Perches were highly effective in concentrating avian seed dispersal. While some native fruits are produced all year-round, most exotic plants set fruits during the main peak of the native fruit production (August–November). Most seeds recovered from the traps were native, and native seed rain inside the native forest was higher than in gaps. However, deposition of exotic seeds was not affected by distance from native forest. Seed dispersal frequencies monitored by seed traps and by faecal analysis were correlated with each other, but not with fruit caloric content, suggesting that other factors are more important that the nutritional value in predicting avian fruit choice. Forest restoration activities should take into consideration that seed dispersal decreases sharply beyond 100 m from native forest and the attractive potential of perches to direct natural seed dispersal into forest gaps. 相似文献

Many estimators and algorithms have been developed to infer the genealogical relationships from molecular marker data when there is no pedigree information. Most pairwise methods provide estimates in a continuous range that needs to be converted into genealogical relationships (namely full-sibs, half-sibs and unrelated) if there is a previous knowledge of the population structure. Transformations are usually based on arbitrary thresholds, but the possibility of correcting the coancestry estimates via explicit pedigree reconstruction methods has been suggested. Using molecular data for ten highly polymorphic microsatellite loci on a population of turbot (Scophthalmus maximus) with a known genealogy, the behaviour of four pairwise marker-based coancestry estimators and the molecular coancestry has been evaluated. The population consisted on 138 families with 4 full-sib individuals each and one family with 8 full-sib individuals (up to 15 half-sib families were also present due to the sharing of parents between some of the full-sibs families). Our results suggested that transforming the pairwise estimators and the molecular coancestry to family relationships through the establishment of thresholds performs slightly better than the explicit pedigree reconstruction method, when accuracy is measured in a pairwise basis. However, if joint relationships between more than two individuals were tested at a time, the threshold methods led to a high percentage of incongruous triads of full-sib individuals, with Mendelian incompatibilities appearing in congruous full-sib families (more than 70% and 60% in our study, respectively). The explicit pedigree reconstruction approach, due to its nature, is free from such problems. Therefore, the pedigree reconstruction approach seems to be a valuable tool to provide a congruent and compatible pedigree when the pairwise marker-based coancestry matrices or the molecular coancestry need to be transformed. 相似文献

Abstract.— The aquaculture performance of mutton snapper Lutjanus analis raised in floating net cages was assessed by measuring their growth, survival, and feed conversion rates during a growout trial conducted in a 3.2‐ha saltwater lake in the Florida Keys, Florida, USA. Approximately 10,500 hatchery‐reared finger‐lings were stocked in two circular, high‐density polyethylene (HDPE) net cages of 7‐m diameter × 7‐m deep (300 m2) and 10‐m diameter × 7‐m deep (600 m3) dimensions. Cages were stocked at 25 fish/m3 (3.2 kg/m3) and 5 fish/m3 (0.72 kg/m3), respectively. Fish grew from a mean of 16.5 g to 302.8 g (25.6 cm TL) in 246 days in the former cage and from a mean of 42.3 g to 245.6 g (23.8 cm TL) in 178 d in the latter cage. Growth rates in weight were best expressed by the following exponential equations: cage 1 (high stocking density): W = 20.716 e0.0112x (r2= 0.83); cage 2 (low stocking density): W = 38.848 e0.0118x (r2= 0.81). Length‐weight data indicate that hatcheryraised, cage‐cultured mutton snapper are heavier per unit length than their wild counterparts. There was no significant difference (P < 0.05) between the slopes of the two lines, indicating that fish in the two cages grew at the same rate. The length‐weight relationships for mutton snapper stocked in cages 1 and 2 are expressed, respectively, by the equations W = 0.000009 L 3.11 (r2= 0.99) and W = 0.000005 L 3.22 (r2= 0.97). Overall feed conversion rate for both cages combined was 1.4. Approximately 10% of the fish sampled exhibited some degree of deformity, particularly scoliosis. Overall survival rate was 70%. Results suggest that L. analis has potential for aquaculture development in net cage systems. 相似文献

One of the requirements for the forest restoration of soils disturbed by the oil‐exploitation industry is that saplings be able to endure soil‐adverse conditions. In this study, saplings of 20 species susceptible to be used in reforestation programs were evaluated for their ability to grow on substrates derived from soils disturbed by petroleum extractions in the Ecuadorian Amazon. Seeds of each species were planted in germination trays. Once seedlings reached 5 cm in height they were transplanted to plastic bags with three treatment substrates: two derived from petroleum‐exploitation activity (soils from mud and drill cutting cells and from areas surrounding oil wells) and a control soil. Plant survival rate, stem height, and diameter were measured on a weekly basis until 14 weeks after transplantation, when we harvested the plants and also measured plant biomass and calculated the Dickson quality index for each species. Oil‐exploitation by‐product substrates impaired the performance of many saplings, with the substrate from mud and drill cutting cells being the one that most affected plant performance. Only saplings of five native species in the Amazon basin—Apeiba membranaceae, Cedrelinga cateniformis, Inga densiflora, Myroxylon balsamum, and Pouroma cecropiifolia—exhibited high or similar Dickson quality index values in all soil treatments and performed better than the rest. The use of these five species in remediation of soils disturbed by petroleum extraction in the Amazon basin could prove important because of their high potential to adapt to these disturbed sites.
