OBJECTIVE: To develop a diagnostic test for recurrent exertional rhabdomyolysis (RER) in Thoroughbreds that relied on in vitro contracture of muscle biopsy specimens and determine whether the inheritance pattern of RER diagnosed on the basis of this contracture test was consistent with an autosomal dominant trait. DESIGN: Clinical trial. ANIMALS: 8 adult horses with RER and 16 control adult horses for development of the contracture test; 23 foals for inheritance of RER. PROCEDURE: External intercostal muscle biopsy specimens from the 24 adult horses were tested for contracture in response to halothane and caffeine, and criteria for a positive test result were determined. These criteria were then applied to results for the 23 foals to determine whether they had RER. Simple segregation analysis was performed to determine whether results were consistent with a dominant pattern of inheritance. RESULTS: Results of the contracture test were positive for 5 of the 12 colts and 4 of the 11 fillies. Results of segregation analysis were consistent with an auto-. somal dominant pattern of inheritance. Two sires with RER produced colts with RER, supporting the hypothesis that RER had an autosomal, rather than an X-linked, inheritance pattern. In addition, in 1 instance, an unaffected colt was produced by 2 affected parents, which was not consistent with a recessive mode of inheritance. CONCLUSIONS AND CLINICAL RELEVANCE: Although the expression of the RER trait is influenced by sex, temperament, and diet, among other factors, results from the in vitro muscle contracture test and this breeding trial suggest that RER in Thoroughbreds can be modeled as a genetic trait with an autosomal dominant pattern of inheritance.  相似文献

It has been reported for many soils that maize (Zea mays L.) has a higher soil‐P critical level than soybean (Glycine max L.) and sunflower (Helianthus annuus L). The objective of this work was to compare the rhizosphere P depletion in these three species in order to investigate if they differ in their capacity to acquire soil P. Sequential P fractionation and pH were determined in rhizosphere and nonrhizosphere soil samples from field and greenhouse experiments. Neither sunflower (the species with highest rhizosphere acidification) nor soybean or maize showed a significant relationship between P depletion and rhizosphere pH. The labile P fraction and the NaOH‐Pi fraction had lower values in the rhizosphere than in the bulk soil in 38% and 77% of the studied cases, respectively. Sunflower and especially maize presented a more intense Pi depletion than soybean. The comparison between sunflower and maize revealed that neither of them took a clear advantage over the other in terms of P depletion. Rhizosphere Pi depletion was associated with the amount of P acquired by the plants. We conclude that the accessibility to different P pools does not explain the differences in soil‐P critical levels among the three species.  相似文献

Soybean is one of the most economically important crops in the world. Its production is affected by several fungal diseases, such as those caused by Fusarium spp., causing significant losses in yield and seed quality. Management interventions are limited, costly, and associated with environmental problems. Host resistance provides a more convenient and cost-effective approach. Host-induced gene silencing (HIGS) has been demonstrated to be an alternative strategy to engineer fungus resistance in plants. We have generated transgenic soybean lines with an intron-hairpin construction in order to express siRNA corresponding to the CYP51B gene from Fusarium oxysporum. Results showed the presence of siRNA corresponding to the F. oxysporum CYP51B gene in both leaves and roots of the transgenic lines. Plants (T3 generation) were challenged against F. oxysporum and F. graminearum. Disease severity was evaluated and revealed resistance to F. oxysporum with one line, named 3.22, presenting no symptoms. In addition, transgenic lines presented better plant development (height and root growth) when compared to the nontransgenic line. Moreover, transgenic lines revealed better development when inoculated with F. oxysporum.  相似文献

Unproductive saline and waterlogged (WT) wastelands could be beneficially transformed into agroforestry systems using trees tolerant to these stresses. We studied the salinity and waterlogging tolerance of five Australian tree species (Acacia salicina, Casuarina glauca, Casuarina obesa, Eucalyptus camaldulensis and Eucalyptus occidentalis) during seedling stage and their relationships with root and leaf ion concentrations. 8-month old plants were exposed for 5 months to five irrigation water salinity treatments (EC values between 2 and 22 dS m?1) and two waterlogging treatments (drained or WT). The salinity tolerance of the five species was high, although decreased in WT conditions. Irrespective of salinity, the two Casuarina species were more tolerant and the other three species were less tolerant to WT than drained conditions. In all species, salinity and waterlogging increased leaf Cl? and Na+ and decreased leaf Ca2+, but not leaf K+. Root Cl? and Na+ increased with salinity but not with waterlogging. Salinity tolerance was negatively correlated with Cl? and Na+ leaf accumulation rates per unit increase in salinity. Waterlogging reduced the ability of the seedlings to exclude Cl? and Na+ from the leaves. The two most salt tolerant Casuarina species under both drained and WT conditions showed the highest leaf Cl? and Na+ exclusion and the highest root Cl? and Na+ accumulation, suggesting that sequestration of these toxic ions in their roots was a significant salt-tolerant mechanism. Revegetation of saline and WT wastelands with these tolerant Casuarina species could be profitably used for biomass, biofuel and renewable energy production.  相似文献

Seven populations collected at different altitudes in northern Italy, two Ladino breeding populations and two control cultivars (AberHerald and Grasslands Huia) of white clover (Trifolium repens L.) were included in a series of experiments analysing: (i) levels of cold tolerance using artificial and field-based methods; (ii) relationships between these measures of cold tolerance; (iii) components of spring yield, various physiological traits, and their relationships with cold tolerance. Rates of seedling and growing point mortality in the populations over winter, assessed in separate field experiments, were closely related (r = 0.85). Grasslands Huia showed the highest death rates, and material originating from high altitudes the lowest. The LT50 value, i.e., the temperature at which 50% of the growing points would die, estimated by an artificial freezing test, was significantly correlated with field-based measures of seedling (r = 0.64) and growing point (r = 0.84) mortality. The existence of these correlations is of potential interest for the development of indirect selection criteria for complex and expensive-to-evaluate traits such as winter survival in field plots. Besides being reliable, in this study the artificial assessment was also sensitive, providing a greater degree of separation of the populations means than field-based measures. Of the several physiological traits (water content,concentrations of water soluble and total non-structural carbohydrate, and water soluble protein content of stolons) measured at a mid-winter sampling date under field conditions, the only character showing significant variation between populations was soluble protein content. There was a slight trend for material with a higher protein content to exhibit greater field-based values of cold tolerance. High altitude populations tended to have low spring yields. The highest spring yield was found in one of the Ladino populations. The study identified two populations which combined, to differing extents, cold tolerance and spring yield characteristics that would be of potential use in breeding for specified agronomic/climatic zones. 三段式心土混层犁为改造白浆土土体构型实现了“上翻20 cm,下混30~40 cm”的农艺要求,白浆层与淀积层的土壤混拌率Mx达到0.7,白浆层向淀积层的土壤转移率TAw→B达到了0.3。0~40 cm土层土壤物理性状明显改善,疏松透水,硬度下降,农田表涝现象得到缓解,效果持久稳定,连续4年测定,耕翻一次后至第4年其土壤硬度仍然明显低于常规耕作田块。1996年~1998年大面积示范试验,使用该犁耕翻地块作物增产达10%~27.9%。该犁作业幅宽50 cm,深度60 cm,总牵引阻力30~35 kN。  相似文献

The aim of the study was to determine the effect of adding two tropical earthworm species, Rhinodrilus contortus and Pontoscolex corethrurus, to mesocosms on the availability of mineral N (NH4 + and NO3 concentrations), soil microbial biomass (bio-N), and the decomposition rates of three contrasting leaf litter species, in a glasshouse experiment. The mesocosms were filled with forest soil and covered with a layer of leaf litter differing in nutritional quality: (1) Hevea brasiliensis (C/N=27); (2) Carapa guianensis (C/N=32); (3) Vismia sp., the dominant tree species in the second growth forest (control, C/N= 42); and, (4) a mixture of the former three leaf species, in equal proportions (C/N=34). At the end of the 97-day experiment, the soil mineral N concentrations, bio-N, and leaf litter weight loss were determined. Both earthworm species showed significant effects on the concentrations of soil NO3 (p<0.01) and NH4 + (p<0.05). Bio-N was always greater in the mesocosms with earthworms (especially with R. contortus) and in the mesocosms with leaf litter of H. brasiliensis (6 µg N g–1 soil), the faster decomposing species, than in the other treatments (0.1–1.6 µg N g–1). Thus, earthworm activity increased soil mineral-N concentrations, possibly due to the consumption of soil microbial biomass, which can speed turnover and mineralization of microbial tissues. No significant differences in decomposition rate were found between the mesocosms with and without earthworms, suggesting that experiments lasting longer are needed to determine the effect of earthworms on litter decomposition rates.  相似文献

Summary Water balance components (effective precipitation, diverted water, actual evapotranspiration and irrigation return flow) and salt balance components (TDS of precipitation, diverted water and irrigation return flow, and sources/sinks in the soil profile) of the Violada irrigation district (3,913 ha) located in the Ebro river basin were measured for the 1982 and 1983 hydrological years. The irrigation district discharges annually 73,000 t of salt into its receiving river. The gypsiferous character of the soils gives a saturated gypsum character to the leachate resulting in a fairly constant TDS of the irrigation return flow (coefficient of variation of 12% for the 1982 hydrological year). The salt load of this area is 18.8 t/ha due to the gypsum content of the soils and improper water management. This results in a mean annual irrigation application efficiency (JAE), calculated as the average actual evapotranspiration/diverted irrigation water ratio, of 0.67 for the 2 years studied. On-farm irrigation system evaluation tests were performed in order to evaluate uniformity of water application and irrigation requirements of the main crops. The results indicate that JAE can be improved by means of better irrigation water management (especially irrigation scheduling) and proper land levelling of the fields.Formerly Instituto Nacional de Investigaciones Agrarias, CRIDA-03  相似文献
