We have quantified observed differences in the microstructure and rheology of creaming emulsions stabilized by protein and low molecular weight surfactants. In this study, we made two sets of emulsions from a single parent emulsion, which differed only in their interfacial composition (i.e., either protein or surfactant). The protein studied was whey protein isolate. The zeta potential of the surfactant-stabilized emulsion was controlled by mixing anionic (SDS) and nonionic (Brij 35) surfactants to match the zeta potential of the protein-stabilized emulsion. Despite this, ultrasonic creaming measurements and confocal microscopy showed that the structures within the cream layers were different between the two sets of emulsions. The protein-stabilized emulsions appeared to slow or arrest the packing within the cream, leading to a lower density network of emulsion droplets, whereas the surfactant emulsion droplets rearranged more quickly into a well-packed, concentrated cream layer. Rheological analysis of the creams showed that despite the protein-stabilized emulsions having a lower dispersed phase volume fraction, their elastic modulus was approximately 30 times greater than that of a comparable surfactant-stabilized emulsion. These differences were caused by the ability of the protein to form a highly viscoelastic interfacial network around the droplets which may include intermolecular covalent cross-links. At close range the adhesive nature of the interaction between the layers contributes to the microstructure and rheology of concentrated emulsions. This is the first time that such well-defined emulsion systems have been studied in detail both noninvasively to look at the impact on creaming and also invasively to look at the impact on bulk rheological properties.  相似文献

Contamination of ground and surface water puts pressure on the use of pesticides. Pesticide contamination of water can often be linked to point sources rather than to diffuse sources. Examples of such point sources are areas on farms where pesticides are handled and filled into sprayers, and where sprayers are cleaned. To reduce contamination from these point sources, different kinds of bioremediation system are being researched in various member states of the EU. Bioremediation is the use of living organisms, primarily microorganisms, to degrade the environmental contaminants into less toxic forms. The systems available for biocleaning of pesticides vary according to their shape and design. Up till now, three systems have been extensively described and reported: the biobed, the Phytobac and the biofilter. Most of these constructions are excavations or different sizes of container filled with biological material. Typical overall clean-up efficiency exceeds 95%, realising even more than 99% in many cases. This paper provides an overview of the state of the art of these bioremediation systems and discusses their construction, efficiency and drawbacks.  相似文献

Bovine abortion represents a major animal welfare issue and a cause of substantial economic loss yet the rate of successful diagnosis remains low. Chlamydia-related organisms including Parachlamydia have recently emerged as putative cattle abortifacients. Placental tissue samples and fetal lung from bovine abortion submissions across Scotland in Spring 2011 were investigated by histopathology for the presence of suspect Chlamydia-related organisms. Evidence of Chlamydia-related organisms was observed in 21/113 (18.6%) placenta samples. Thirteen of the suspect cases and 18 histopathology negative cases were analysed by molecular and immunohistochemical methods. All samples were PCR positive for Parachlamydia but sequencing revealed high homology between identified environmental 16S sequences in all but three cases. Parachlamydial antigen was detected in 10/31 placental samples (32.2%) with pathology consistent with chlamydial infection. This work supports the need for further surveillance investigations and experimental studies to determine the role of Parachlamydia in bovine abortion.  相似文献

This study was designed to evaluate the cats' acceptance and compliance of the owners and cats towards an extemporaneously prepared palatable compounded atenolol (paste and suspension) formulation in comparison to the commercially obtained tablet, in a randomised, cross-over study design.The three formulations were prescribed twice daily for 6 days to 13 healthy privately-owned cats of 13 different owners, with varying levels of experience in medicating cats. Daily compliance was evaluated via an owner-completed diary, completed after each dose administered. Owner's experience and preference of the formulation was evaluated via questionnaires given prior to, at the end of each treatment protocol, and upon completion of the study. Although compounded suspension was association with fewest missed doses, the majority of cat owners expressed a preference for the divided tablet. Atenolol tablets, compounded paste and suspension acceptance and compliance were comparable. Further work is now required to assess the amount and stability of the active ingredient and the robustness of the paste and suspension formulations prior to any bioavailability comparisons between the formulations.  相似文献

This study tests a rapid, user-friendly method for assessing changes in erosion risk, which yields information to aid policy development and decision-making for sustainable natural resources management. There is currently a lack of timely, up-to-date and current information to support policy development on sustainable natural resources management in Uganda. The study was carried out in the Ngenge watershed, a typical catchment in the Ugandan Highlands, characterised by deforestation in favour of subsistence agriculture without adequate soil and water conservation measures. The watershed is experiencing soil erosion, sedimentation and flooding problems which are threatening agricultural productivity and food security. Sustainable management of environmental resources is needed to ensure a livelihood for the rural population which is dependent on the land. Historical erosion risk was evaluated in three steps using multi-temporal satellite data. First, current erosion risk was assessed by combining slope and vegetation cover during periods of high intensity rainfall. The data used for the assessment was obtained from public (free) satellite images. Erosion risk was then linked to land use and finally to the change in vegetation cover over the years 1980-2000. The analysis of erosion risk using rainfall, slope and NDVI (Normalised Difference Vegetative Index) as a proxy for vegetation cover gives an indication of the current erosion risk in the area. The results of historical vegetation cover change analysis indicate an overall increase in areas under erosion risk in the study area from 1980 to 2000. This method of erosion risk mapping provides a quick and straightforward means for identifying priority areas for interventions for soil and water resource management. Considering that resources are limited, the interventions to be appropriate have to be focused mainly on areas affected by degradation.  相似文献

Measurements of local environmental conditions, intensity of Fusarium head blight (FHB) in wheat spikes, biomass of Fusarium graminearum, F. culmorum, and F. poae (pathogens causing FHB) and concentration of the mycotoxins deoxynivalenol (DON) and nivalenol (NIV) in harvested wheat grain were obtained in a total of 150 location-years, originating in three European countries (Hungary, Ireland, United Kingdom) from 2001 to 2004. Through window-pane methodology, the length and starting time of temporal windows where the environmental variables were significantly associated with the biological variables were identified. Window lengths of 5 to 30?days were evaluated, with starting times from 18?days before anthesis to harvest. Associations were quantified with nonparametric Spearman correlation coefficients. All biological variables were significantly associated with at least one evaluated environmental variable (P?≤?0.05). Moisture-related variables (e.g., average relative humidity, hours of relative humidity above 80%) had the highest positive correlations with the biological variables, but there also was a significant negative correlation between average temperature and several biological variables. When significant correlations were found, they were generally for all window lengths, but for a limited number of window start times (generally before anthesis for disease index and after anthesis for the toxins and late-season fungal biomasses). Semi-partial Spearman correlation coefficients were used to evaluate the relationship between the environmental variables and the concentration of DON and NIV after the effects of FHB intensity and fungal biomass on the mycotoxins were removed. Significant semi-partial correlations were found between relative humidity variables and DON, and between temperature and relative humidity variables and NIV for time windows that started after anthesis (and not for any earlier time windows). Results confirm that the environment influences disease, fungal biomass, and mycotoxin production, and help refine the time windows where the association is greatest. However, variability in the relationships was high, indicating that no single environmental variable is sufficient for prediction of disease or mycotoxin contamination.  相似文献

The putrescine analogue. (E)-1,4-diaminobut-2-ene (E-BED), synthesized as the dihydrochloride salt, controlled five economically important crop pathogens, Erysiphe qraminis DC f.sp. hordei Marchal. Uromyces viciae-fabae (Pers.) Schroet, Botrytis fabae Sardina, Podosphaera leucotricha (Ell. & Ev.) Salm. and Phytophthora infestans (Mont) De Bary. The Z-isomer. Z-BED. was also fungicidal, although less so than E-BED. Post-inoculation treatment with E-BED gave greater control of powdery mildew infection on barley and rust and chocolate spot on broad bean than did pre-inoculation application. It was also effective in vitro against Botrytis cinerea Pers. ex Fr., Pyricularia oxyzae Br. & Cav. and Pyrenophora avenae Ito & Karibay. When P. avenae was grown in the presence of E-BED dihydrochloride at 81·5 mg litre?1, growth was reduced by 58% and there were significant reductions in soluble ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC) activity. These changes were accompanied by a sevenfold increase in putrescine concentration, a 60% increase in spermine concentration and a 32% reduction in spermidine concentration within the fungal tissue.  相似文献

Eighty-nine bovine viral diarrhoea viruses (BVDV) from Australia have been genetically typed by sequencing of the 5' untranslated region (5'-UTR) and for selected isolates the N(pro) region of the viral genome. Phylogenetic reconstructions indicated that all of the samples examined clustered within the BVDV type 1 genotype. Of the 11 previously described genetic groups of BVDV-1, 87 of the samples examined in this study clustered with the BVDV-1c, while two samples clustered with the BVDV-1a. Based on these analyses there appears to be limited genetic variation within the Australian BVDV field isolates. In addition, the phylogenetic reconstructions indicate that the clustering of Australian BVDV in the phylogenetic trees is not a result of geographic isolation.  相似文献

Previous research has suggested that ducks and songbirds may benefit from prairie landscapes that consist primarily of contiguous grasslands. However, the relative importance of landscape-level vs. local characteristics on mechanisms underlying observed patterns is unclear. We measured effects of grassland amount and fragmentation on upland and wetland songbird and duck density and nest success, and on some nest predators, across 16 landscapes in southern Alberta, Canada. We compared these landscape-level effects with local-scale responses, including distance to various edges and vegetation characteristics. We also evaluated several statistical approaches to comparing effects of habitat characteristics at multiple spatial scales. Few species were influenced by grassland amount or fragmentation. In contrast, distance to edge and local vegetation characteristics had significant effects on densities and nest success of many species. Previous studies that reported effects of landscape characteristics may have detected patterns driven by local mechanisms. As a corollary, results were very sensitive to statistical model structure; landscape level effects were much less apparent when local characteristics were included in the models.  相似文献
