In the case of grapevine (Vitis vinifera L.), leaf blade and petioles at different sampling times are used for nutritional diagnosis and planning fertilization practices. However, reference values for each nutrient depend on the scion‐rootstock combination. In this sense, the current study aimed at determining the reliability of leaf blade and petiole for diagnosis at different phenological stages and, also, of the period of validity, around flowering and veraison, in which the nutrient concentrations obtained could be compared with reference values. The study was carried out in three vineyards (two planted with cv. Mencía and one with cv. Sousón) within the Ribeira Sacra Designation of Origin (NW Spain). Blades and petioles were collected throughout a growth season (2014) and total concentrations of ten essential elements were determined in both tissues. In general, petioles showed greater variability between replicates for P, K, Ca, Mg, and Zn, while N, Fe, Mn, Cu, and B presented greater variability in blades. Differences between consecutive sampling dates suggest that references for flowering were valid until fruit‐set. Furthermore, blade and petiole references at the beginning of veraison were valid until the advanced ripening stage for most nutrients, with the exception of N, P, and Zn. The seasonal variation of the concentrations in both tissues confirmed the need for reference values for each phenological stage and tissue.  相似文献

The effect of n-3 fatty acid-enriched diets (in the form of 0.5% linseed oil with either 1.5% sunflower oil or 1.5% olive oil) and alpha-tocopheryl acetate supplementation (200 mg/kg feed) on lipid oxidation (thiobarbituric acid-reactive substances, TBARS) and cholesterol oxide products (COPS) in cooked pork was investigated. Longissimus muscle was studied. Meat from pigs fed 0.5% linseed oil-enriched diets had a higher proportion of n-3 fatty acid than meat from pigs in other dietary groups in neutral (P < 0.0001) and polar lipids (P < 0.0001), and a 20% reduction in the n-6:n-3 ratio was observed. Alpha-tocopheryl acetate supplementation increased (P < 0.05) monounsaturated fatty acids in polar lipids and increased (P = 0.0001) alpha-tocopherol levels in muscle. Alpha-tocopherol concentration in muscle was affected by dietary fat (P < 0.05). Groups receiving diets enriched with sunflower oil had significantly higher alpha-tocopherol levels (P < 0.05) in muscle than those groups receiving olive oil-enriched diets. Numbers of TBARS were significantly (P < 0.05) lower in the group fed supplemental olive oil than in those fed sunflower oil. Dietary linseed oil increased (P < 0.05) lipid oxidation principally at the initial period of storage in cooked pork. Overall, dietary alpha-tocopheryl acetate supplementation significantly increased (P < 0.001) lipid stability and decreased (P < 0.05) total COP production across the dietary groups. Alpha-tocopherol was a more effective antioxidant for decreasing TBARS values in cooked meat when adding sunflower oil to the diets instead of olive oil.  相似文献

Pythium group F is a minor pathogen which induces symptomless infections that occur frequently and results in yield losses in tomato soilless cultures. To elucidate the mode of action of this microorganism, the influence of culture filtrates of Pythium group F on tomato growth was investigated and compared to that of the pathogen Pythium ultimum. Depending on metabolite production by the fungus, marked differences were observed in plant response. Pythium group F crude culture filtrates or low molecular weight fractions (< 500) caused swelling behind the root tip and reduced root growth; the cohesion and adherence of cells within the cortical area were also affected. These symptoms were similar to those observed on plants treated with indole-3-acetic acid. By contrast, P. ultimum filtrates caused a marked distortion of cell shape accompanied with folding of host cell walls, particularly in the cortical area. These symptoms were characteristic of the activity of toxic compound(s) on host cells. Chemical analysis of the filtrates demonstrated that indole-3-acetic acid and tryptophol were produced by Pythium group F and P. ultimum. However, Pythium group F took up and metabolized more indole-3-acetic acid precursors, especially tryptophan, a key amino acid in the pathways leading to indole-3-acetic acid synthesis. The fact that Pythium group F and P. ultimum transformed tryptamine and indole-3-acetaldehyde into indole-3-acetic acid and tryptophol confirms the existence of a tryptamine pathway within both fungi. These results support the hypothesis that auxins facilitate Pythium group F infections. On the other hand, toxin(s) and hydrolytic enzymes are likely involved in P. ultimum pathogenesis.  相似文献

ABSTRACT Three forms of tobacco leaf curl (termed classes I, II, and III, based on symptomatology) recently have been described in southern Africa. Numerous attempts to isolate virus particles responsible for a nongeminivirus-induced leaf curl disease (class I) of tobacco in South Africa have been unsuccessful. Recently, 12 dsRNA segments were isolated from tobacco exhibiting class I leaf curl symptoms, suggesting a possible reovirus genome. The objective of our study was to confirm whether the dsRNA segments are associated with a reovirus. Isolation of icosahedral particles with an outer core 60 to 65 nm in diameter and an inner core 40 to 45 nm in diameter was achieved. Twelve distinct nonpolyadenylated dsRNAs were isolated from purified virions, and the total molecular masses of the dsRNAs ranged from 17.86 to 18.40 x 10(6) Da in polyacrylamide and agarose gels, respectively. Using hybridization analysis, dsRNAs were identified as non-homologous distinct segments. Comparisons with other known reoviruses revealed a unique banding pattern that was most similar to the wound tumor virus (WTV), the type species of the genus Phytoreovirus. Hybridizations of WTV cloned DNA probes (segments S4 and S6 to S9) and dsRNAs from infected tobacco indicated no significant sequence similarity, whereas indirect enzyme-linked immunosorbent assay with a polyclonal antiserum to WTV showed strong positive cross-reactivity to tobacco virions. Our results indicate a virus with features consistent with those of phytoreoviruses. This is the first report of a plant reovirus in tobacco, the first record in Africa, and the second example of a field-isolated dicot phytoreovirus.  相似文献

In vitro regeneration of complete plants from nodal single-bud segments of 2-year-old Australian Cedar (Toona ciliata) trees were obtained under defined nutritional and environmental conditions. Explants were dissected from plants obtained by germination of seeds and growth in pots in a greenhouse. The best medium for shoot regeneration was that of Murashige and Skoog at 1/4 strength with 3% sucrose (1/4 MS), supplemented with 0.1 mg/l IBA and 0.5 mg/l BAP. Rooting of regenerated shoots was observed in MS medium with 0.1 mg/l IBA. Using mature tree material was more difficult. Forced flushing was used to induce shoot development on branches of a 10-year-old tree. Nodal segments of these epicormic shoots formed shoots in vitro on 1/4 MS + 0.01 mg/l IBA + 5 mg/l BAP, but rooting was never observed.  相似文献

This study compared needle-free and needle-based injection devices for vaccination of calves against Clostridium chauvoei in warm and cold conditions. Both devices elicited comparable antibody responses in calves. Needle-free injection devices can be used to vaccinate calves provided appropriate precautions are taken in cold weather.  相似文献

The adsorption of paraquat (1,1′‐dimethyl‐4,4′‐bipyridilium dichloride) and atrazine (6‐chloro‐N 2‐ethyl‐N 4‐isopropyl‐1,3,5‐triazine‐2,4‐diamine) from aqueous solution onto two montmorillonite‐(Ce or Zr) phosphate cross‐linked compounds at different temperatures (288 K and 308 K) has been studied using batch experiments. The adsorption isotherms obtained for paraquat on both adsorbents may be classified as H‐type of the Giles classification, which suggests that paraquat molecules are strongly adsorbed on the samples. For the adsorption of atrazine, L‐type isotherms were obtained for both montmorillonite‐(Ce or Zr) phosphate compounds, which suggests that these compounds have a medium affinity for this herbicide. The increase in temperature from 288 K to 308 K did not have any clear effect on the adsorption process of paraquat on either adsorbent whereas atrazine adsorption decreased slightly as temperature increased, possibly due to a mainly physical process. Fourier transform infrared (FTIR) spectroscopic studies revealed that at the pH generated by the adsorbents, the cationic herbicide interacted to a greater extent with the negatively charged surface of the adsorbents than did atrazine. For both herbicides, the Ce‐montmorillonite adsorbent showed a higher adsorption capacity than the Zr‐montmorillonite adsorbent. © 2000 Society of Chemical Industry  相似文献

To describe the effect and the distribution of zinc in different tissues of a representative marine fish species, gilthead seabreams (Sparus aurata Linnaeus, 1758) were fed different diets containing zinc in different sources (organic, inorganic, encapsulated, blood‐rich diet or fishmeal). The effect was monitored by histology in the lens, liver and intestine. In addition, the anterior and posterior intestine was studied by means of autometallography, a histochemical silver‐based staining method to determine the zinc flow and distribution. The histology of lens did not improve by dietary zinc as there was no occurrence of cataracts. In the liver, the experimental diets were correlated with different changes in the tissue architecture. In the intestine, no histological changes were found using haematoxylin and eosin stain. However, with the use of autometallography stain, it was possible to visualize and describe the route that zinc follows through the enterocytes and the lamina propria. Organic and inorganic zinc diet produced the richest silver deposition in the anterior intestine.  相似文献

Daily growth of Atlantic juvenile hake from Northwest Iberia has been estimated employing a new approach combining analyses of transversal and sagittal sections of the otoliths along the ventral radius. Age of juvenile hake ranging from 3 to 25 cm collected during a spring 2002 survey was estimated. Somatic growth followed a power fit: Fish size (TL) = 3.3254*age0.7336 (r2 = 0.87, p < 0.001, n = 76), yielding an average individual growth rate of 0.66 mm/day (±0.06). The growth model indicates that after a year's life a juvenile can reach 25 cm. Otolith ventral radius ranged from 401 to 1842 μm and daily increments were between 104 and 387. Fish growth and otolith growth were closely related (r2 = 0.92 p < 0.001, n = 76). These first results of daily growth rates for the Southern stock corroborate the fast-growth hypothesis of this species. The evolution of increment widths from hatch dates onwards reveals important seasonal growth peaks during July–August and October–November. A comparison with prior data and discussion is also presented in the light of recent work on hake juveniles and tagging-recapture experiences.  相似文献

‘Samantha’ roses (Rosa hybrida L.) potted in sand or soil mixture were grown under natural or HPS supplementary irradiation and fertilized with 0.05, 0.25, 0.5, 2.5 or 5.0 ppm Cu in each irrigation. Foliar and root Cu contents increased relative to the Cu level in solution while leaf and root Fe contents were reduce with higher concentration of Cu applied. Growing medium influenced the pattern of foliar content response to Cu concentration. In sand, the effect of Cu level in solution was evident from the beginning of the experiment whereas in soil mixture the effect was observed only after six months. Plant growth and flower production was reduced only in plants grown in sand and fertilized with 2.5 and 5.0 ppm Cu. No Cu deficiency symptoms were observed even though leaf content in plants grown in soil mixture was as low as 2.2 ppm. High Cu level in solution induced Fe deficiency symptoms, especially in plants grown in sand. Supplementary irradiation did not influence the effect of Cu fertilization level on foliar content or flower production. Most of the Cu applied was retained in the upper 5 cm layer of soil mixture reducing its availability for plant uptake.  相似文献
