An indirect solid-phase microradioimmunoassay (IRIA) was developed for detection and quantitation of antibodies to pseudorabies virus (PRV) in swine serum. Qualitative results of the IRIA compared closely with results of the serum neutralization test (NT) and the microimmunodiffusion test (MIDT). The IRIA was more sensitive than the NT for detection of antibodies to PRV in swine serum. The IRIA result is expressed numerically. With the IRIA and NT, antibody to PRV was first detectable in 3 experimentally infected pigs at 9 days after inoculation. With MIDT, antibody was detected in the 3 experimentally infected pigs at 9 days after inoculation. With the MIDT, antibody was detected in the 3 experimentally infected pigs at 7, 8, and 9 days after inoculation. 1. Four rearing temperature regimes (15, 20, 25, 30 °C) and three feeding schedules (ad libitum, restricting to the ad libitum intake of the 12th week and feeding 70% of this rate) were carried out with layer replacement pullets to 170 d of age. From this age, during lay, birds were kept at either 21 °C or on a 24‐h cycle of 21 for 18 h and 28 °C for the 6 h before lights out. Both a white and a brown egg‐laying strain were used.
2. Body weight at 169 d of age varied from, on average, 1409 g (15 °C, ad libitum) to 943 g (30 °C, 70% schedule) for the white strain and 1947 to 1250 g for the same treatments respectively for the brown strain. Sexual maturity was considerably delayed by the 70% feeding schedule, only slightly by rearing at 30 °C.
3. Rearing at 30 °C tended to depress subsequent egg output. The 70% feeding schedule at least maintained egg output compared with birds fed ad libitum in rearing.
4. There was a highly significant effect of temperature treatment during lay on food intake. The reduction in food intake due to the 21–28 °C cycle, however, appeared small. 相似文献
This study examines the possible predictive value of the LIVE/DEAD fluorescence viability assay for evaluation of survival of cryopreserved human ovarian tissue. Ovarian tissue from ten patients was examined by LIVE/DEAD viability staining before and after cryopreservation and after freezing in a -20 C freezer (negative control). After cryopreservation with a slow freezing protocol and cryoprotectant the LIVE/DEAD assay showed 86% viable follicles (an intact oocyte and at least more than 50% of the granulosa cells alive), whereas after freezing at -20 C the survival rate was 67%. The healthy follicular loss after cryopreservation was 4%, whereas with freezing at -20 C, it was 25%. Although this assay overestimates the survival rate of cryopreserved primordial follicles, if the LIVE/DEAD assay yields greater than approximately 85% viable follicles, it can be assumed that the follicles in the cryopreserved tissue have maintained their developmental potential and that the tissue is suitable for retransplantation. 相似文献
An enzyme-linked immunosorbent assay (ELISA) was developed for detecting antibody to type A avian influenza (AI) virus. The sensitivity and group specificity of the AI-ELISA were compared with those of the agar-gel-precipitin test (AGPT) and the hemagglutination-inhibition (HI) test under conditions of both controlled and field exposure. During the course of temporal experimental infection (0-76 days) of specific-pathogen-free (SPF) chickens with AI subtype Hav9N2, the AI-ELISA was able to detect specific AI antibody as early as 8 days postinoculation (PI), and it measured rising levels of antibody through 35 days PI, at which time the chickens were re-exposed to AI virus. Conversely, AGP tests were negative through 35 days PI, and HI tests began to detect low levels of AI antibody only at 21 days PI. Following a secondary infection at 35 days PI with the same AI subtype, all tests measured rising levels of AI-specific antibody (35-76 days PI). However, the AGP test was positive at only the 7- and 14-day samplings postsecondary immunization. Under field conditions, the AI-ELISA was able to detect serum AI antibody in flocks from which highly pathogenic AI was isolated, but the AGP tests of these sera were negative. 相似文献
The enteric nervous system in the small intestine of cattle during Schistosoma bovis infection was studied by histological stains and immunohistochemical methods. Lesions due to migration of schistosoma eggs were located mainly in the mucous and the submucous layer overlaying the submucous vascular arcades. Granulomas destroyed ganglia, neurons, nerves fibre strands and nerve fibres. Ganglia situated within or near granulomas were infiltrated by mast cells, eosinophils, lymphocytes, globule leukocytes, neutrophils and macrophages. Mast cells were in close contact with degenerating neuronal perikarya. Whereas vasoactive intestinal peptide-like immunoreactivity in the nerves and neurons in the ganglia within and around granulomas was increased, the neurofilament-like immunoreactivity was reduced. Compared to the myenteric and external submucous plexuses, the internal submucous and mucous plexuses were the most damaged. These changes imply reduced functional capacity in the nervous tissue which might cause reduced motility, malabsorption and partly account for the loss of body weight and condition and failure to thrive which occur in schistosomosis.Biotinylated affinity purified swine anti-rabbit and mouse anti-rabbit immunoglobulins reacted nonspecifically with a subset of mast cells. The reaction revealed many mast cells in early forming granulomas and around schistosome egg tracts and infiltration of mast cells into the ganglia of intestinal nerve plexuses. The observation shows a localized, Type I hypersensitivity reaction suggesting for the release of mast cell-derived chemical mediators in the intestinal reaction to trap or evict S. bovis eggs and to cause diarrhoea. 相似文献
A method to determine ascorbic acid at any stage of potato processing was developed. Potato samples are frozen in dry ice, lyophilized and analyzed using a modified spectrophotometric method. The milk yields obtained from 845 herd-years within the ICI Recorded Farms Scheme have been correlated, by multiple regression analysis, with the N fertilizer supplied to the grassland, the quantities of concentrates fed and the stocking rates. The large quantity of data gave highly significant correlations which indicated that, over the whole period, the supply of an extra 1 kg N from fertilizer was associated with an average extra yield of 9·9 1 milk. 4·9 1 were associated with the extra N independently of any simultaneous changes in either stocking rate or concentrate feeding, and 4-5 1 were ascribable to a simultaneous increase in stocking rate. There was a substantial variation in the ‘milk yield per kg N’ response from year to year, but the average value over 6 years agreed closely with that observed in the period 1949–52. 相似文献
