An accidental release of non-toxic waste from decommissioned wood pulp industry in the River Numedalslågen, Southern Norway, occurred in the upper part of the accessible stretches for anadromous fish during a study of migration behaviour of radio tagged Atlantic salmon (Salmo salar, n=32, body length 51–99 cm). The fish had completed the migration phase and initiated the resident phase characterised by little movement until spawning. When the wooden fibres and pulp were released, 16 of 32 (50%) salmon showed an immediate avoidance response. Six (19%) salmon moved upstream and 10 (31%) salmon moved downstream. Of the salmon moving downstream, eight (25%) moved all the way to sea (average 14.8 km). Four re-entered the River Numedalslågen, two entered a neighbouring river and two were not recorded later. Fish moving downstream but without moving to the sea, moved on average 5.3 km during the episode, whereas those moving upstream moved on average 6.7 km. All fish recorded after the episode (n=30) survived until the spawning season. The study demonstrates that fish in nature may show an evident avoidance response, even to non-toxic contaminants. For Atlantic salmon, the size of the spawning population may be reduced by fish leaving the river (13% left the river and never came back). Moreover, the dispersal of salmon to other rivers may increase, and the distribution of the spawning population within the contaminated river may shift.  相似文献

A rapid liquid chromatography electrospray ionization tandem mass spectrometry with negative ion detection method was developed and validated to determine cocoa flavonoid metabolites in human plasma and urine after the intake of a standard portion of a cocoa beverage. A chromatographic run time of only 9 min provided clear separation of all metabolites and internal standards. Samples were analyzed in a product-ion scan of m/z 289, 369, and 465 to identify the metabolites and in multiple reaction monitoring acquisition mode to quantify (-)-epicatechin ((-)-Ec) (289/ 245), (-)-epicatechin-glucuronide ((-)-EcG) (465/289), and (-)-epicatechin-sulfate ((-)-EcS) (369/289). One (-)-Ec-G and three (-)-Ec-S were identified and confirmed in urine as the major metabolites, and one (-)-Ec-G was the only metabolite present in plasma volunteers (n = 5) at a mean concentration of 625.7 +/- 198.3 nmol/L at 2 h after consumption of a cocoa beverage containing 54.4 mg of (-)-Ec.  相似文献

Effects of diethyl suberate (DESU), diethyl sebacate (DES), dibutyl suberate (DBSU), dibutyl sebacate (DBS), and tributyl phosphate (TBP) on diffusion of 14C-2,4-dichlorophenoxy butyric acid (2,4-DB) across cuticular membranes (CM) was studied. Astomatous CM were isolated enzymatically from Stephanotis floribunda Brongn. leaves, and diffusion was measured at 20 degrees C. The alkyl-substituted dicarboxylic acids constitute a homologous series with carbon numbers increasing from C12 to C18. Molecular weights increased only moderately from 230.0 (DESU) to 314.5 (DBS), while partition coefficients varied over orders of magnitude from 92 (DESU), to 1213 (DES), to 15,988 (DBSU), to 210,762 (DBS). All the above compounds turned out to be accelerators as they increased 2,4-DB mobility by up to 40-fold with accelerator concentrations in the CM ranging from only 9.2 to 105 g kg(-1). Efficacy (2,4-DB mobility in the presence/mobility in the absence of accelerators) increased with increasing concentrations of accelerators in CM or in reconstituted cuticular waxes. Plotting efficacy vs accelerator concentration in the CM resulted in straight lines, and their slopes increased in the order DBS (0.14), DBSU (0.31), DES (0.51), and DESU (0.85). Hence, DESU was the most powerful accelerator in this series as it increased 2,4-DB mobility in the CM about 6 times more than DBSU. Waxes constitute the major barrier in plant cuticles, and plots of efficacy vs accelerator concentration in Stephanotis wax were also linear, but compared to CM slopes were steeper by factors of 3.20 (DBS), 2.97 (DBSU), 2.70 (DES), and 1.62 (DESU). TBP was similarly effective as DESU, but plots of efficacy vs concentration were not linear, and curves approached a plateau at 60-80 g kg(-1). These data are discussed with regard to suitability of these accelerators for formulating systemic pesticides.  相似文献

To prevent bone loss that occurs with increasing age, nutritional and pharmacological factors are needed. Traditional therapeutic agents (selective estrogen receptor modulators or SERMs, biphosphonates, calcitonin) may have serious side effects or contraindications. In an attempt to find food components potentially acting as SERMs, we submitted four plant aqueous extracts derived from Greek flora (Sideritis euboea, Sideritis clandestina, Marticaria chamomilla, and Pimpinella anisum) in a series of in vitro biological assays reflective of SERM profile. We examined their ability (a) to stimulate the differentiation and mineralization of osteoblastic cell culture by histochemical staining for alkaline phosphatase and Alizarin Red-S staining, (b) to induce, like antiestrogens, the insulin growth factor binding protein 3 (IGFBP3) in MCF-7 breast cancer cells, and (c) to proliferate cervical adenocarcinoma (HeLa) cells by use of MTT assay. Our data reveal that all the plant extracts studied at a concentration range 10-100 microg/mL stimulate osteoblastic cell differentiation and exhibit antiestrogenic effect on breast cancer cells without proliferative effects on cervical adenocarcinoma cells. The presence of estradiol inhibited the antiestrogenic effect induced by the extracts on MCF-7 cells, suggesting an estrogen receptor-related mechanism. In conclusion, the aqueous extracts derived from Sideritis euboea, Sideritis clandestina, Marticaria chamomilla, and Pimpinella anisum may form the basis to design "functional foods" for the prevention of osteoporosis.  相似文献

In plants, zinc is commonly found bound to proteins. In barley (Hordeum vulgare), major storage proteins are alcohol‐soluble prolamins known as hordeins, and some of them have the potential to bind or store zinc. 65Zn overlay and blotting techniques have been widely used for detecting zinc‐binding protein. However, to our knowledge so far this zinc blotting assay has never been applied to detect a prolamin fraction in barley grains. A radioactive zinc (65ZnCl2) blotting technique was optimized to detect zinc‐binding prolamins, followed by development of an easy‐to‐follow nonradioactive colorimetric zinc blotting method with a zinc‐sensing dye, dithizone. Hordeins were extracted from mature barley grain, separated by SDS‐PAGE, blotted on a membrane, renatured, overlaid, and probed with zinc; subsequently, zinc‐binding specificity of certain proteins was detected either by autoradiography or color formation. The dithizone staining method gave similar reproducibility to the radioactive blotting. The detected zinc‐binding protein was identified as B‐hordein by Western blotting.  相似文献

Background

The natural feeding behaviour of the pig is searching for feed by rooting activities throughout the day; self-feeding pigs randomly space their eating and drinking periods throughout the day consuming ten to twelve meals per day. Pigs in conventional fattening pig production are normally fed 2–3 times daily with the feed consumed within 15 minutes. The aim of this study was to determine if more frequent feedings could improve the performance of conventionally kept fattening pigs.

Methods

The experiment was carried out on 360 fattening pigs (27–112 kg live weight), weighed and assigned to pens stratified by weight and sex. Each treatment group consisted of 180 pigs, allocated to 20 pens with nine pigs in each pen. To evaluate how more feeding occasions affects performance and well-being the pigs were divided into two groups and fed three (control group) or nine (treatment group) times daily. The same total amount of liquid feed was fed to each group and the feed ration was correlated to the live weight of the pigs. All weight and slaughter recordings were made individually and recordings of feed consumption were made pen-wise. At slaughter the stomach of each pig was examined for lesions in the pars oesophagea and scored on a scale from 1–6.

Results

Frequent feeding occasions influenced both performance and status of gastric lesions of the pigs adversely. Pigs in the treatment group grew slower compared to pigs in the control group; 697 g/day (± 6.76) versus 804 g/day (± 6.78) (P < 0.001) with no difference in within-pen variation. There was also a lower prevalence of gastric lesions within pigs in the control group (2.4 (± 0.12) compared to 3.0 (± 0.12) (P < 0.01)). There was a positive correlation between gastric lesions in the treatment group and daily weight gain (r = 0.19; P < 0.01).

Conclusion

Increased daily feeding occasions among group housed pigs resulted in a poorer daily weight gain and increased mean gastric lesion score as compared with pigs fed three times daily. This may be a consequence of more frequently occurring competition for feed in the treatment group. The present study does not support increased daily feeding occasions in fattening pigs.  相似文献

Acquired resistance triggered by elicitins in tobacco and other plants   总被引:17,自引:0,他引:17  
Elicitins are a family of proteins excreted byPhytophthora spp. They exhibit high sequence homology but large net charge differences. They induce necrosis in tobacco plants which then become resistant to the tobacco pathogenPhytophthora parasitica var.nicotianae. In stem-treated plants, resistance was not restricted to the site of elicitin application, but could be demonstrated by petiole inoculation at all levels on the stem. Resistance was already maximum after two days and lasted for at least two weeks. It was effective not only towardsP. p. var.nicotianae infection, but also against the unrelated pathogenSclerotinia sclerotiorum. In contrast to dichloroisonicotinic acid, an artificial inducer of systemic acquired resistance, which was increasingly effective with doses ranging from 0.25 to 5mole per plant, the basic elicitin cryptogein exhibited a threshold effect, inducing near total resistance and extensive leaf necrosis above 0.1 nmole per plant. Between 1 and 5 nmole, acidic elicitins (capsicein and parasiticein) protected tobacco plants with hardly any necrotic symptom. Elicitins exhibited similar effects in various tobacco cultivars andNicotiana species, although with quantitative differences, but induced neither necrosis nor protection in other SolanaceÆ (tomato, petunia and pepper). Among 24 additional species tested belonging to 18 botanical families, only some BrassicaceÆ, noticeably rape, exhibited symptoms in response to elicitins, in a cultivar-specific manner. Zoonotic West Nile virus (WNV) circulates in natural transmission cycles involving certain mosquitoes and birds, horses, humans, and a range of other vertebrates are incidental hosts. Clinical infections in humans can range in severity from uncomplicated WNV fever to fatal meningoencephalitis. Since its introduction to the Western Hemisphere in 1999, WNV had spread across North America, Central and South America and the Caribbean, although the vast majority of severe human cases have occurred in the United States of America (USA) and Canada. By 2002-2003, the WNV outbreaks have involved thousands of patients causing severe neurologic disease (meningoencephalitis and poliomyelitis-like syndrome) and hundreds of associated fatalities in USA. The purpose of this review is to present recent information on the epidemiology and pathogenicity of WNV since its emergence in North America.  相似文献
