In previous studies, Al extracted by acid ammonium acetate (Ala) or Na-pyrophosphate (Alp), rather than silt or clay content and climate conditions, was the most important factor that controls organic matter (OM) levels in volcanic soils. Here, the hypothesis was tested that Ala is a comparable method (as much as CuCl2) to quantify the proportion of Al bound to OM in allophanic soils. As far as we know, there are no previous antecedents in which selective dissolution method has been compared with this extractant. Secondly, we examine the effects of (a) Al, (b) silt plus clay content (particles size 0-53 µm) and (c) clay mineralogy on the control of organic carbon (OC) level in Chilean volcanic soils. This was achieved by sampling 16 soils series (11 Andisols, one Alfisol and four Ultisols, USDA classification) including 48 soil pedons up to 0.4 m depth. Soils were analyzed for Ala, Alp, oxalate (Alo, Sio and Feo), cold NaOH (Aln) and un-buffered salts, CuCl2 (AlCu), LaCl3 (AlLa) and KCl (Alk). We also measured the Al-humus as soluble C fraction after pyrophosphate extraction and the C associated to the silt plus clay fraction after sonication and gravity decantation. The statistical package (S)MATR was used to examine bivariate linear regressions among soil properties by computing the standardized major axis (SMA). Our results indicate that Ala had a good correspondence with Alp (R2 = 0.76) in the top soil with Ala/Alp ratio of 0.19 and both extractans presented significant and positively relationship with soil OC (R2 > 0.62). Acid ammonium acetate was as effective as AlCu to determine the Al-OM in allophanic soils. It is cheaper than AlCu and Alp and 0.5 h shaking is required compared to 2 h of AlCu and 16 h of Alp. The efficiency of the extraction was: Aln ≥ Alo > Alp > AlCu ≥ Ala > AlLa > Alk. We also found that allophane content (estimated by Al/Si ratio) was strongly correlated (R2 = 0.82) with the OC in the fine silt plus clay and that Al-humus together with C in the finest particles explained (R2 > 0.60) the largest proportion of variation of soil OC across studied soils.  相似文献

REASONS FOR PERFORMING STUDY: Clostridium difficile has been associated with acute colitis in mature horses. OBJECTIVES: To survey C. difficile colonisation of the alimentary tract with age, occurrence of diarrhoea and history of antibiotic therapy; and to study the occurrence and survival of C. difficile in the environment and antimicrobial susceptibility of isolated strains. METHODS: A total of 777 horses of different breeds, age and sex were studied. Further, 598 soil samples and 434 indoor surface samples were examined. Antimicrobial susceptibility of 52 strains was investigated by Etest for 10 antibiotics. RESULTS: In horses that developed acute colitis during antibiotic treatment, 18 of 43 (42%) were positive to C. difficile culture and 12 of these (28%) were positive in the cytotoxin B test. Furthermore, C. difficile was isolated from a small number of diarrhoeic mature horses (4 of 72 [6%]) with no history of antibiotic treatment, but not from 273 healthy mature horses examined or 65 horses with colic. An interesting new finding was that, in normal healthy foals age < 14 days, C. difficile was isolated from 1/3 of foals (16 of 56 [29%]). All older foals (170) except one were negative. Seven of 16 (44%) nondiarrhoeic foals treated with erythromycin or gentamicin in combination with rifampicin were also excretors of C. difficile. On studfarms, 14 of 132 (11%) outdoor soil samples were positive for C. difficile in culture, whereas only 2 of 220 (1%) soil samples from farms with mature horses were positive for C. difficile (P = < 0.001). By PCR, it was demonstrated that strains from the environment and healthy foals can serve as a potential reservoir of toxigenic C. difficile. The experimental study conducted here found that C. difficile survived in nature and indoors for at least 4 years in inoculated equine faeces. The susceptibility of 52 strains was investigated for 10 antibiotics and all were susceptible to metronidazole (MIC < or = 4 mg/l) and vancomycin (MIC < or = 2 mg/l). CONCLUSIONS: C. difficile is associated with acute colitis in mature horses, following antibiotic treatment. Furthermore, C. difficile was isolated from 1 in 3 normal healthy foals age < 14 days. POTENTIAL RELEVANCE: Strains from healthy foals and the environment can serve as a potential reservoir of toxigenic C. difficile.  相似文献

Samples of organs from 50 lambs, 3-45 days old, brought to this laboratory from 44 different flocks for post mortem examination, were examined for Listeria monocytogenes. Various diseases were found as cause of the death. The bacterium was isolated from 3 lambs (6%) which had died from E. coli-septicaemia. Polyamine and phenolic acid levels as well as activities of some enzymes of their biosynthetic metabolism were examined in two sugarcane (Saccharum officinarum) cultivars differing in susceptibility to leaf scald, a disease caused by the bacterium, Xanthomonas albilineans. Juice obtained from both infected cultivars showed significantly increased levels of free putrescine and ornithine decarboxylase activity. However, the pathogen induced different changes in the two cultivars in subsequent metabolic steps. Whereas acid insoluble conjugated spermidine completely disappeared from the highly susceptible cv. C 439-52, an increase in acid insoluble conjugated polyamines was observed in the moderately susceptible cv. L 55-5. Phenolic acid metabolism also differed in the two cultivars. Since total phenolic acid content and phenylalanine ammonium lyase activity was greater in both cultivars after infection, distribution of phenolic acids between free or conjugated forms diverted into different pathways. The level of susceptibility of the two cultivars is discussed in terms of changes in these compounds.  相似文献

Zusammenfassung In Freiland- und Laborversuchen wurde die Wirkung des Pflanzenschutzmittels Sevin auf Regenwürmer untersucht. Dieser Stoff ist in Pulverform und als Suspension für diese Organismen außerordentlich gefährlich. Schon der Kontakt mit sehr geringen Mengen bewirkt schwere Lähmungserscheinungen und irreversible histopathologische Schäden.
Summary Experiments in the field as well as in the laboratory showed up the effect of the pesticide Sevin on rainworms. In the form of powder as well as a suspension this material is extremely toxic for them. Contact with smallest quantities even causes severe paralysis and irreversible histopathologic damages.

Résumé Aussi bien dans la nature que au laboratoire nous avons étudié l'activité de l'insecticide Sevin sur les vers de terre. Ce produit est en poudre ou en suspension extrémement dangereux pour ces organismes. Le seul contact avec des quantités minimes de ce produit provoquent des phénomènes de paralysie et des troubles histopathologiques irreversibles.

Understanding the recruitment variability of the Atlantic herring North Sea stock remains a key objective of stock assessment and management. Although many efforts have been undertaken linking climatic and stock dynamic factors to herring recruitment, no major attempt has been made to estimate recruitment levels before the 20th century. Here, we present a novel annually resolved, absolutely dated herring recruitment reconstruction, derived from stable carbon isotope geochemistry (δ13C), from ocean quahog shells from the Fladen Ground (northern North Sea). Our age model is based on a growth increment chronology obtained from fourteen shells. Ten of these were micromilled at annual resolution for δ13C analysis. Our results indicate that the anthropogenically driven relative depletion of 13C, the oceanic Suess effect (oSE), became evident in the northern North Sea in the 1850s. We calculated a regression line between the oSE‐detrended δ13C results (δ13C?) and diatom abundance in the North Sea, the regression being mediated by the effect of phytoplankton on the δ13C of the ambient dissolved inorganic carbon. We used this regression to build an equation mediated by a nutritional link to reconstruct herring recruitment using δ13C?. The reconstruction suggests that there were five extended episodes of low‐recruitment levels before the 20th century. These results are supported by measured recruitment estimates and historical fish catch and export documentation. This work demonstrates that molluscan sclerochronological records can contribute to the investigation of ecological baselines and ecosystem functioning impacted by anthropogenic activity with implications for conservation and stock management.  相似文献

Coagulase-negative staphylococci (CNS) are often associated with bovine mastitis. Knowledge about the relative importance of specific CNS species in different types of mastitis, and differences in antimicrobial resistance among CNS species is, however, scarce. Therefore, the aims of this study were to compare prevalence and antimicrobial susceptibility of CNS species in clinical and subclinical mastitis using material from two national surveys. Overall, Staphylococcus chromogenes and Staphylococcus epidermidis were the most common CNS species found followed by Staphylococcus simulans and Staphylococcus haemolyticus. S. epidermidis was significantly more prevalent in subclinical than in clinical mastitis, and a similar trend was observed for Staphylococcus saprophyticus, while Staphylococcus hyicus was significantly more common in clinical mastitis. The prevalence of β-lactamase producing isolates varied markedly between CNS species, and was significantly higher in S. epidermidis and S. haemolyticus (~ 40%), than in S. simulans and S. chromogenes where none or a few of the isolates produced β-lactamase. Resistance to more than one antimicrobial substance occurred in 9% and 7% of the clinical and subclinical isolates, respectively. In conclusion, the distribution of CNS species differed between clinical and subclinical mastitis indicating inter-species variation of pathogenicity and epidemiology. Overall, the prevalence of antimicrobial resistance was low, but some variation between CNS species was observed.  相似文献

We determined the effects of 16 wk of storage at low temperature (4 C) in darkness on the viability, growth, photosynthetic parameters, and biochemical composition of four diatom cultures. Significant differences in cell density and proximal composition were observed for all diatoms throughout storage. Cell density increased with time of storage for all diatoms. Protein content increased for Navicula incerta, Nitzschia laevis, and Navicula sp., whereas lipid content increased during storage in only N. incerta. When the stored diatoms were used as inocula in fresh medium, they increased their viability, generating a lag phase for Nitzschia thermalis var. minor, N. incerta, and Navicula sp. cultures. There were noted species‐specific modifications in proximal composition, ash‐free dry weight, and photosynthetic parameters in response to storage. We conclude that N. thermalis, N. incerta, N. laevis, and Navicula sp. can be stored at 4 C for 16 wk and are viable in new cultures.  相似文献

Persimmon fruit polyphenol oxidase (PPO) was partially purified using a combination of phase partitioning with Triton X-114 and ammonium sulfate fractionation between 50 and 75%. The enzyme, which showed both monophenolase and diphenolase activities, was partially purified in a latent form and could be optimally activated by the presence of 1 mM sodium dodecyl sulfate (SDS) with an optimum pH of 5.5. In the absence of SDS, the enzyme showed maximum activity at acid pH. SDS-PAGE showed the presence of a single band when L-DOPA was used as substrate. The apparent kinetic parameters of the latent enzyme were determined at pH 5.5, the V(m) value being 15 times higher in the presence of SDS than in its absence, whereas the K(M) was the same in both cases, with a value of 0.68 mM. The effect of several inhibitors was studied, tropolone being the most active with a K(i) value of 0.45 microM. In addition, the effect of cyclodextrins (CDs) was studied, and the complexation constant (K(c)) between 4-tert-butylcatechol (TBC) and CDs was calculated using an enzymatic method.
