The combination of runoff‐generating areas (saturated soils) and overland flow concentration in features such as drainage ditches makes sloping farmland vulnerable to soil erosion. The establishment of drainage ditches aims at draining the excess of water from the farmland, particularly in areas where soils are saturated in the rainy season. The hydrogeomorphic impacts on the farmland itself and on downstream areas need however also to be studied. Off site, downstream problems comprise higher peak discharges, leading to gully initiation, an increase in sediment load, and flooding problems. On‐site problems such as the development of the drainage ditches into (ephemeral) gullies are less documented, although they may be important, as illustrated in the Lake Tana Basin (Ethiopia). The similarities and interactions between ephemeral gully channels and drainage ditches have to be considered to better understand all effects of drainage. Drainage ditches are a potential source of conflict between farmers with different interests and power, as well as between upstream and downstream users. A case study on drainage ditches on sloping farmlands in the Lake Tana Basin showed that nine out of ten catchments had drainage densities by ditches ranging from 53 to 510 m ha−1. Drainage ditches were constructed with an average top width of 27 (±9) cm. A significant correlation was found between stone bund density (physical conservation structures) and ditch drainage density (R = −0·72), in line with the Ethiopian government's ban on drainage ditches in farmlands where stone bunds have been constructed. - The presence of threatened or endangered species often strongly influences management and conservation decisions. Within the Murray–Darling Basin (MDB), Australia, the presence of threatened native fish affects the management and allocation of water resources. In New South Wales, these decisions are currently based on traditional fisheries data and a predictive MaxEnt model. However, it is important to verify the model's predictive power given the implication it may have, but this requires methods with a high detection sensitivity for rare species.
- Although the use of environmental DNA (eDNA) monitoring, in particular eDNA metabarcoding, achieves a higher detection sensitivity compared with traditional methods, earlier surveys in the MDB have shown that the highly abundant and invasive common carp (Cyprinus carpio) can reduce detection probabilities for rare species. Consequently, a polymerase chain reaction (PCR) blocking primer designed to block the amplification of carp eDNA could increase the detection probabilities for rare native species while simultaneously reducing the required sampling effort and survey costs. Although PCR blocking primers are often used in ancient DNA and dietary studies, no aquatic eDNA metabarcoding study to date has evaluated the potential benefits of using PCR blocking primers.
- A laboratory and field-based pilot study was used to address this knowledge gap and assess the impact of a blocking primer, targeting cyprinid fishes (including carp), on the detection probabilities of native species and the minimum sampling effort required.
- Results showed that the inclusion of the blocking primer increased the detection probabilities for native species by 10–20% and reduced the minimum required sampling effort by 25–50%. These findings provide important insights into possible methods for optimizing eDNA metabarcoding surveys for the detection of rare aquatic species.

The objective of this research is to determine whether intramammary antibiotics with complementary acupuncture can reduce bovine mammary inflammation due to subclinical mastitis. Lactating cows were selected based on milk with a somatic cell count (SCC) greater than 500,000 cells/ml. Pre- and post-treatment milk samples were collected to determine SCC, aerobic bacterial content, milk ion conductivity, total protein, lactate dehydrogenase (LDH) and N-acetyl-beta-D-glucosaminidase (NAGase) concentrations. Milk serum was prepared from milk samples by double centrifugation. Concentrations of LDH and NAGase were determined using commercial enzyme-linked immunosorbent assays. Cows being treated with intramammary antibiotics were separated by random assignment to the acupuncture group (n = 10) and a no-acupuncture (control) group (n = 9). Both the acupuncture and control group were restrained for 30 min in a head catch 12 hr apart for a total of four times. For front quarters affected by subclinical mastitis, the acupuncture points used were spleen (SP) 12, SP 17, SP 18, SP 21, stomach (ST) 18 and conception vessel (CV) 12. For rear quarters affected by subclinical mastitis, the acupuncture points used were bladder (BL) 30, BL 30-1, BL 49, kidney (KI) 10, conception vessel (CV) 2 and CV 3. All parameters were compared using a Student t test. Significance was defined as p < .05. Compared to control cows, complementary acupuncture treatment reduced NAGase enzymatic activity in quarters of cows with subclinical mastitis. The reduction in NAGase suggests that complementary acupuncture treatment may be associated with healing of the damaged mammary epithelial cells, which are the primary source of NAGase activity in milk serum. 相似文献

Successful prairie restoration will depend in part on convincing private landowners with agricultural and recreational use goals to implement appropriate rangeland management practices, such as prescribed burning and cattle grazing, to control invasive species and encroachment of woody plants. However, landowners have been slow to adopt appropriate practices in the US Midwest. The purpose of this study was to explore attitudes and behaviors of private landowners toward prescribed burning and moderate stocking as rangeland management tools. A survey was mailed to 193 landowners (response rate 51%) in the Grand River Grasslands region of southern Iowa and northern Missouri. While 68% of landowners viewed grazing as a legitimate land management tool, only half of landowners thought of fire as a legitimate tool. Over 75% of respondents believed that the increase in eastern redcedar and other trees in grasslands was a problem, with 44% considering it a major problem. Although 84% of landowners said that they had taken action to control eastern redcedar, only 25% had participated in a prescribed burn. Income from agriculture and recreational goals were negatively and significantly correlated (-0.252, P = 0.035). While holding recreational goals constant in the analysis, landowners reporting income from agriculture goals as very or extremely important were negatively and significantly associated with reporting environment and grassland factors as very or extremely important. Adoption of prescribed burning by private landowners might be more widespread if proponents focus on the effectiveness of fire for controlling eastern redcedar, which is viewed as a problem by most landowners in the region. Production of Atlantic salmon smolts in recirculation aquaculture systems (RAS) is growing, and novel production protocols using continuous light in RAS are being implemented in the industry. In the present study, Atlantic Salmon parr were exposed to either a traditional protocol (short-day winter signal [12:12 L:D] for 6 weeks) or to continuous light. Both photoperiods were applied in freshwater (FW) and brackish water RAS. Salmon from all treatments were transferred to seawater pens at 200 and 600 g and grown until slaughter size. A control group was smoltified with a 6-week short-day winter signal and kept in FW until sea transfer at 100 g. Continuous light gave a higher growth rate in RAS but reduced feed intake and growth and increased feed conversion ratio during the first 8 weeks in seawater. However, at slaughter, fish exposed to continuous light was bigger than fish given a winter signal because of the higher growth rate in RAS. Slaughter weight was lowest in fish transferred to sea at 600 g, despite having the highest day-degree sum during their life span. The best performing group was the control group transferred at 100 g. All treatments handled transfer to seawater and survival and maturation were not affected by the treatments in RAS. The immune status was examined with a multigene expression assay on BioMark HD platform from parr stage to 5–7 months after seawater transfer. Overall, there was no significant effect of photoperiod or salinity on the expression of the selected immune genes. In sum, the results from this study indicate that using continuous light in RAS may have negative effects on performance shortly after transfer in fish transferred to sea at 200 g, whereas at 600 g, all treatments had reduced growth after transfer irrespective of treatment in RAS. 相似文献

OBJECTIVE: To examine the effect of adjuvant doxorubicin chemotherapy on outcome in dogs with high-grade (grade 3) soft tissue sarcomas (HGSTSs). DESIGN: Retrospective case series. ANIMALS: 39 dogs. PROCEDURES: Medical records of dogs with HGSTSs were reviewed. Dogs treated with surgery alone or receiving single-agent doxorubicin chemotherapy postoperatively were included in the study. Owners and referring veterinarians were contacted for follow-up information. Slides from histologic sections were reviewed to confirm the diagnosis of HGSTSs. Cases in which follow-up examination was not performed and radiation therapy or chemotherapy other than doxorubicin was administered were excluded. RESULTS: 39 dogs met inclusion criteria. Twenty-one dogs received adjuvant doxorubicin. Tumor-, patient-, and treatment-related variables were not significantly associated with measured outcomes including local, metastatic, and overall disease-free intervals as well as survival time. Overall median disease-free interval was 724 days with a median survival time of 856 days for all dogs. CONCLUSIONS AND CLINICAL RELEVANCE: Adjuvant doxorubicin-based chemotherapy did not benefit this population of dogs with HGSTSs. Outcome for visceral HGSTSs was similar to that of nonvisceral HGSTSs in these cases. 相似文献

A 4‐year‐old spayed female American Staffordshire Terrier presented to the U‐Vet Animal Hospital, Werribee, Australia, with a cutaneous mass that had been slowly growing over 12 months. Cytologic evaluation showed cohesive to individualized, vacuolated spindled cells often arranged in a perivascular pattern. The mass was completely excised, and the histopathologic examination demonstrated sheets of vacuolated spindled to round cells expanding the full thickness of the dermis. The cells demonstrated both Iba1 and CD18 antibody binding, leading to an initial interpretation of histiocytic sarcoma. Given the discordance with the clinical presentation, further immunohistochemistry (IHC) was performed. The cells demonstrated strong CD204 antibody binding and did not bind E‐cadherin antibody, consistent with a dermal macrophage origin. Ki‐67 antibody binding was regionally variable from <5% to 25%, with more regions that had low Ki‐67 expression. A fasted serum biochemistry panel revealed hypertriglyceridemia and persistent hypercholesterolemia. Based on clinical, microscopic, biochemical, and IHC results, the final interpretation was an indolent dermal histiocytic proliferation of macrophage origin, with a preference for cutaneous xanthoma or reactive dermal fibrohistiocytoma. 相似文献
