OBJECTIVE: To evaluate safety and efficacy of LDI-100, a preparation containing human chorionic gonadotropin (hCG) and bacillus Calmette-Guerin (BCG), in the treatment of dogs with mast cell tumors and to compare results with those from a control group receiving single-agent vinblastine. ANIMALS: 95 dogs with measurable grade II or III mast cell tumors. PROCEDURES: Dogs were randomized to receive either LDI-100 (1.35 ng of BCG and 2 units of hCG, SC, q 24 h) or vinblastine (2 mg/m(2), IV, q 1 wk) for 6 weeks. Tumors were measured at baseline and day 42, and dogs were monitored for signs of toxicosis. Clinical performance scores were recorded at each visit. Differences in host factors (sex, weight, and age), clinical performance score, tumor response, and adverse events were analyzed. RESULTS: 46 dogs received LDI-100, and 49 dogs received vinblastine. No significant differences were found between the 2 treatment groups with regard to host factors or clinical performance score. Tumor response (>or=50% reduction) rates were similar between the LDI-100 and vinblastine group (28.6% and 11.7%, respectively). Dogs in the LDI-100 group had significantly less neutropenia than the vinblastine group. CONCLUSIONS AND CLINICAL RELEVANCE: hCG and BCG have immunomodulatory and antitumor effects against a variety of malignancies in humans and dogs. In this study, LDI-100 provided clinical responses comparable to single-agent vinblastine chemotherapy but without myelosuppression. LDI-100 is a promising new agent that should be further investigated for multimodality therapy of mast cell tumors in dogs.  相似文献

Foot pathology is a common and important health concern in captive rhinoceroses worldwide, but osteopathologies are rarely diagnosed, partly because of a lack of radiographic protocols. Here, we aimed to develop the first radiographic protocol for rhinoceros feet and describe the radiographic anatomy of the white rhinoceros (Ceratotherium simum) hind foot (pes). Computed tomographic images were obtained of nine cadaver pedes from seven different white rhinoceroses and assessed for pathology. A single foot deemed free of pathology was radiographed using a range of different projections and exposures to determine the best protocol. 3D models were produced from the CT images and were displayed with the real radiographs to describe the normal radiographic anatomy of the white rhinoceros pes. An optimal radiographic projection was determined for each bone in the rhinoceros pes focusing on highlighting areas where pathology has been previously described. The projections deemed to be most useful were D60Pr‐PlDiO (digit III), D45Pr45M‐PlDiLO (digit II), and D40Pr35L‐PlDiLO (digit IV). The primary beam was centered 5–7 cm proximal to the cuticle on the digit of interest. Articular surfaces, ridges, grooves, tubercles, processes and fossae were identified. The radiographic protocol we have developed along with the normal radiographic anatomy we have described will allow for more accessible and effective diagnosis of white rhinoceros foot osteopathologies.  相似文献

The biology and chemistry of three northeastern Pennsylvania lakes was studied from summer 1981 through summer 1983 to evaluate lakes with different sensitivities to acidification. At the acidified lake (total alkalinity ≤ 0.0 μeq L?1) there were fewer phytoplankton and zooplankton species than at the moderately sensitive lakes. The most numerous plankton species in all three lakes are reportedly acid tolerant. Among the benthic macro- invertebrates (BMI) there were more acid tolerant Chironomidae at the acidified lake, but more acid intolerant Ephemeroptera and Mollusca and a higher wet weight at the least sensitive lake. There were no differences among the lakes' BMI mean total numbers or mean number of taxa. The fish community at the acidified lake was dominated by stunted Lepomis gibbosus, but L. machrochirous were most abundant in the other lakes. Principal component analysis suggested a shift in all three lakes over the sampling period toward combined lower pH, alkalinity, specific conductance, Ca and Mg and higher Al and Mn. Such chemical changes have been associated with acidification. The rate and extent of acidification appeared to be controlled by geological and hydrological characteristics of the drainage basins.  相似文献

Groundwater contaminated with sulfate (SO4 2‐) at concentrations higher than allowable for drinking water might still be usable for irrigation. Objectives were to determine the growth response and mineral uptake of two forage crops irrigated with waters containing SO4 2‐ at concentrations ranging from 175 to 1743 mg/L, and with electrical conductivities (EC) ranging from 1.2 to 3.6 dS/m. Plants were grown for 12 weeks in 8‐L pots containing a calcareous sandy loam and were harvested at 4, 8, or 12 weeks for plant growth measurements and tissue analysis. Digested leaves, stems, and reproductive tissues were analyzed by inductively coupled plasma (ICP) spectroscopy at each harvest, as were saturated soil paste extracts. Shoot growth of tall fescue (Festuca arundinacea Schreb.) was not affected by irrigation water treatment, whereas shoot growth of alfalfa (Medicago sativa L.) was increased by a moderate level of soil solution SO4 2‐ Sulfur (S), boron (B), magnesium (Mg), sodium (Na), and zinc (Zn) concentrations in shoot tissues of both species showed a tendency to increase with increasing SO4 2‐ content of irrigation water. Shoot tissue concentration of molybdenum (Mo) increased with maturation in both species, while the concentrations of B, potassium (K), manganese (Mn), Na, and Zn decreased. Soil saturated paste extract concentrations of Mg and Na increased with irrigation water Mg and Na concentrations, while Ca and S concentrations in the soil solution became saturated at the higher irrigation water concentrations of these elements.  相似文献

The luminal surface of the gastrointestinal tract is covered by a mucus gel layer that acts to protect gut epithelial cells from the harsh luminal environment. This study investigated the use of two human colonic adenocarcinoma cell lines, HT29-MTX-E12 and HT29, as a model to mimic gut epithelium with and without a mucus gel layer. The effect of adding the tea polyphenols epigallocatechin gallate (EGCG) and epicatechin (EC) to the cells with subsequent examination of cell morphology and viability was assessed. EGCG, at the concentrations tested, was very toxic to the HT29 cells, but less toxic to the HT29-MTX-E12 cells, suggesting that the mucus gel layer on the HT29-MTX-E12 cells can protect the cells against EGCG toxicity. In contrast, EC had no effect on the viability of either the HT29 or HT29-MTX-E12 cells, suggesting that proteins within the mucus gel layer on the apical surface of gut epithelial cells may bind to the galloyl ring of EGCG. The effect of adding food-related ingredients with the ability to complex with EGCG, β-casein and maltodextrin, on cell viability was also examined. The presence of β-casein was very effective in protecting the cells against the toxicity effect of EGCG, but maltodextrin, at the concentration tested, was less effective in protecting against this toxicity. In conclusion, the results demonstrate that the mucus gel layer on HT29 human colonic adenocarcinoma cells may protect these cells against EGCG toxicity. In addition, the data showing reduced toxicity of EC compared to that of EGCG suggest that the cytotoxic effects of high polyphenol levels may be associated with the ability of polyphenols to interact with cellular proteins and mucins.  相似文献

BACKGROUND

Semiochemicals for monitoring, attracting or repelling pest and beneficial organisms are increasingly deployed in agricultural and forest systems for pest management. However, the use of aggregation pheromones and host‐plant attractants for the express purpose of increasing the efficacy of classical biological control agents of weeds has not been widely reported. Therefore, we conducted field‐based assays to determine if a specialized wax‐based matrix impregnated with an aggregation pheromone of the northern tamarisk beetle Diorhabda carinulata (Desbrochers) or host‐plant volatiles could increase the efficacy of D. carinulata.

RESULTS

The aggregation pheromone and host‐plant volatiles were formulated for field application using a wax‐based matrix. Reported release rates suggest that this matrix is a viable formulation for enhancing D. carinulata aggregations under field conditions. Pheromone‐treated saltcedar plants (Tamarix spp.) not only had higher densities of adult and larval D. carinulata, but also sustained greater levels of foliar damage than control plants. Increased damage from the focused feeding of D. carinulata caused an increase in foliar dieback and decrease in live canopy volume of semiochemical‐treated plants.

CONCLUSION

Field deployment of these semiochemical formulations could be useful in directing populations of D. carinulata for increased impact on Tamarix spp. © 2018 The Authors. Pest Management Science published by John Wiley & Sons Ltd on behalf of Society of Chemical Industry.  相似文献
