With the capacity for rapid self-renewal and regeneration, the intestinal epithelium is stereotypical of stem cell-supported tissues. Yet the pattern of stem cell turnover remains in question. Applying analytical methods from population dynamics and statistical physics to an inducible genetic labeling system, we showed that clone size distributions conform to a distinctive scaling behavior at short times. This result demonstrates that intestinal stem cells form an equipotent population in which the loss of a stem cell is compensated by the multiplication of a neighbor, leading to neutral drift dynamics in which clones expand and contract at random until they either take over the crypt or they are lost. Combined with long-term clonal fate data, we show that the rate of stem cell replacement is comparable to the cell division rate, implying that neutral drift and symmetrical cell divisions are central to stem cell homeostasis.  相似文献

After disappointing results from all efficacy trials conducted to date, the field of microbicides research now faces substantial challenges. Poor coordination among interested parties and the choice of nonvalidated scientific targets for phase III studies have hampered progress and created mistrust about the use of microbicides as a method to prevent HIV-1 sexual transmission. Although new promising strategies are available, there will need to be serious reappraisals of how decisions are made to advance the next generations of candidates into clinical trials, and the use of appropriate animal models in this process will be critical.  相似文献

As concerns over the negative social and environmental impacts of industrial agriculture become more widespread, efforts to define and regulate sustainable agriculture have proliferated in the US. Whereas the USDA spearheaded previous efforts, today such efforts have largely shifted to Tripartite Standards Regimes (TSRs). Using a case study of the Leonardo Academy’s initiative to develop a US sustainable agriculture standard, this paper examines the standards-development process and efforts by agribusiness to influence the process. Specifically, we analyze how politics operate in seemingly “depoliticized” TSRs, and how agribusiness and the USDA use “framing practices” and procedural complaints to influence the standard-development process. We contend that although governance mechanisms are a potentially powerful tool for advocates of alternative agrifood, their efficacy may be constrained by science-based requirements, an agribusiness countermovement, and a captured state.  相似文献

Echium oil (EO) is a vegetable oil in which percentages of stearidonic acid (STA, 18:4n‐3) often exceed those of its n‐6 series equivalent γ‐linolenic acid (GLA, 18:3n‐6). Stearidonic acid is elongated to 20:4n‐3 in fish cell cultures, suggesting that EO could be included in diets for marine fish to increase tissue 20:4n‐3 and 20:3n‐6 and, thereby, modulate eicosanoid metabolism. Thus, the present study aimed to test the hypotheses that dietary EO would increase tissue 20:4n‐3 and 20:3n‐6 and modulate immune function and eicosanoid production in juvenile Atlantic cod (Gadus morhua L.) fed a diet where fish oil (FO) was replaced by EO. Duplicate groups of juvenile cod (initial weight ca. 4 g) were fed for 18 weeks on fish meal‐based diets (55% protein and 16% lipid) that differed in oil source (FO or EO). There were no significant differences in growth and feed efficiency, hepato‐somatic index, percentages of liver and flesh lipids and lipid class compositions for cod fed FO and EO. Percentages of 18:4n‐3, 18:3n‐6 and 20:3n‐6 in the total lipids of flesh and liver were higher, and percentages of 20:5n‐3 and 20:4n‐6 were both lower in fish fed EO than in those given FO. In flesh, the increased 18:3n‐6 and 18:4n‐3 were primarily located in phosphatidylcholine and, to a lesser extent, phosphatidylethanolamine, whereas 20:3n‐6 concentration was highest in phosphatidylinositol. Desaturation of 18:3n‐3 (to tetraene products) and 20:5n‐3 to 22:6n‐3 in hepatocytes was very low but was increased by dietary EO. Echium oil significantly decreased the production of prostaglandin F from gill cells stimulated with calcium ionophore A23187, and reduced head kidney macrophage activity, but had no effect on serum lysozyme activity or basic haematology. In conclusion, dietary EO may have beneficial effects on some immune parameters including eicosanoid metabolism in marine fish although this may be primarily because of decreased 20:4n‐6 rather than increasing tissue levels of 20:3n‐6 or 20:4n‐3.  相似文献

Most materials shrink laterally like a rubber band when stretched, so their Poisson's ratios are positive. Likewise, most materials contract in all directions when hydrostatically compressed and decrease density when stretched, so they have positive linear compressibilities. We found that the in-plane Poisson's ratio of carbon nanotube sheets (buckypaper) can be tuned from positive to negative by mixing single-walled and multiwalled nanotubes. Density-normalized sheet toughness, strength, and modulus were substantially increased by this mixing. A simple model predicts the sign and magnitude of Poisson's ratio for buckypaper from the relative ease of nanofiber bending and stretch, and explains why the Poisson's ratios of ordinary writing paper are positive and much larger. Theory also explains why the negative in-plane Poisson's ratio is associated with a large positive Poisson's ratio for the sheet thickness, and predicts that hydrostatic compression can produce biaxial sheet expansion. This tunability of Poisson's ratio can be exploited in the design of sheet-derived composites, artificial muscles, gaskets, and chemical and mechanical sensors.  相似文献

A high-resolution crystallographic analysis of the nitrogenase MoFe-protein reveals a previously unrecognized ligand coordinated to six iron atoms in the center of the catalytically essential FeMo-cofactor. The electron density for this ligand is masked in structures with resolutions lower than 1.55 angstroms, owing to Fourier series termination ripples from the surrounding iron and sulfur atoms in the cofactor. The central atom completes an approximate tetrahedral coordination for the six iron atoms, instead of the trigonal coordination proposed on the basis of lower resolution structures. The crystallographic refinement at 1.16 angstrom resolution is consistent with this newly detected component being a light element, most plausibly nitrogen. The presence of a nitrogen atom in the cofactor would have important implications for the mechanism of dinitrogen reduction by nitrogenase.  相似文献

Human mesenchymal stem cells are thought to be multipotent cells, which are present in adult marrow, that can replicate as undifferentiated cells and that have the potential to differentiate to lineages of mesenchymal tissues, including bone, cartilage, fat, tendon, muscle, and marrow stroma. Cells that have the characteristics of human mesenchymal stem cells were isolated from marrow aspirates of volunteer donors. These cells displayed a stable phenotype and remained as a monolayer in vitro. These adult stem cells could be induced to differentiate exclusively into the adipocytic, chondrocytic, or osteocytic lineages. Individual stem cells were identified that, when expanded to colonies, retained their multilineage potential.  相似文献

Simulating the influence of intensive management and annual weather fluctuations on tree growth requires a shorter time step than currently employed by most regional growth models. High-quality data sets are available for several plantation species in the Pacific Northwest region of the United States, but the growth periods ranged from 2 to 12 years in length. Measurement periods of varying length complicate efforts to fit growth models because observed growth rates must be interpolated to a common length growth period or those growth periods longer or shorter than the desired model time step must be discarded. A variation of the iterative technique suggested by Cao [Cao, Q.V., 2000. Prediction of annual diameter growth and survival for individual trees from periodic measurements. Forest Sci. 46, 127–131] was applied to estimate annualized diameter and height growth equations for pure plantations of Douglas-fir, western hemlock, and red alder. Using this technique, fits were significantly improved for all three species by embedding a multi-level nonlinear mixed-effects framework (likelihood ratio test: p < 0.0001). The final models were consistent with expected biological behavior of diameter and height growth over tree, stand, and site variables. The random effects showed some correlation with key physiographic variables such as slope and aspect for Douglas-fir and red alder, but these relationships were not observed for western hemlock. Further, the random effects were more correlated with physiographic variables than actual climate or soils information. Long-term simulations (12–16 years) on an independent dataset using these annualized equations showed that the multi-level mixed effects models were more accurate and precise than those fitted without random effects as mean square error (MSE) was reduced by 13 and 21% for diameter and height growth prediction, respectively. The level of prediction error was also smaller than an existing similar growth model with a longer time step (ORGANON v8) as the annualized equations reduced MSE by 17 and 38% for diameter and height growth prediction, respectively. These models will prove to be quite useful for understanding the interaction of weather and silviculture in the Pacific Northwest and refining the precision of future growth model projections.  相似文献
