Crop rotation adoption in no‐tillage systems (NTS) has been recommended to increase the biological activity and soil aggregation, suppress soil and plant pathogens, and increase the productivity aiming at the sustainability of agricultural areas. In this context, this study aimed to assess the effect of crop rotation on the arbuscular mycorrhizal fungi (AMF) community and soil aggregation in a soil cultivated for nine years under NTS. Treatments consisted of combinations of three summer crop sequences and seven winter crops. Summer crop sequences consisted of corn (Zea mays L.) monoculture, soybean (Glycine max L. Merrill) monoculture, and soybean–corn rotation. Winter crops consisted of corn, sorghum (Sorghum bicolor (L.) Moench), sunflower (Helianthus annuus L.), sunn hemp (Crotalaria juncea L.), pigeon pea (Cajanus cajan (L.) Millsp.), oilseed radish (Raphanus sativus L.), and millet (Pennisetum americanum (L.) Leeke). Soil samples were collected at a depth of 0–0.10 m for analyses of soil chemical, physical, and biological attributes. Spore abundance, total glomalin, and soil aggregate stability index were higher in the soil under corn monoculture. The highest values of aggregate mean weight diameter were observed in the soybean–corn rotation (3.78 mm) and corn monoculture (3.70 mm), both differing from soybean monoculture (3.15 mm), while winter crops showed significant differences only between sorghum (3.96 mm) and pigeon pea (3.25 mm). Two processes were identified in the soil under summer crop sequences. The first process was observed in PC1 (spore abundance, total glomalin, easily extractable glomalin, pH, P, and Mg2+) and was related to AMF; the second process occurred in PC2 (aggregate mean weight diameter, soil aggregate stability index, K+, and organic matter) and was related to soil aggregation. The nine‐year no‐tillage system under the same crop rotation adoption influenced AMF abundance in the soil, especially with corn cultivation in the summer crop sequence, which promoted an increased total external mycelium length and number of spores of AMF. In addition, it favored an increased soil organic matter content, which is directly related to the formation and stability of soil aggregates in these managements.  相似文献

The objective was to identify a fat-to-protein ratio (FPR) cut-off to diagnose subclinical ketosis (SCK) and to evaluate the effect of propylene glycol (PPG) treatment of cows with high FPR. The optimized cut-off was > 1.42; sensitivity (Se) = 92%; specificity (Sp) = 65%. A cut-off > 1.5 was selected for the PPG trial for balanced Se-Sp. Fat-to-protein ratio cut-offs > 1.25, 1.35, 1.50, 1.60, and 1.70 resulted in Se-Sp of 100% to 49%, 96% to 59%, 75% to 78%, 33% to 90%, and 8% to 96%, respectively. The proportions of cows with FPR > 1.25, 1.35, 1.42, 1.50, 1.60, and 1.70 were 60%, 50%, 44%, 30%, 14%, and 6%, respectively. Incidences of clinical ketosis and milk yield were similar between cows that received 400 mL of PPG (n = 34) and control cows (n = 38). Prevalence of SCK at enrollment was 29.2%; therefore, FPR > 1.5 is not indicated for treatment. Lower cut-offs should be used for screening.  相似文献

Brucella melitensis is the Brucella species most frequently associated with brucellosis in humans. It is also the causative agent of the disease in goats and other ruminants. Although significant aspects of the pathogenesis of infection by this intracellular pathogen have been clarified, several events during invasion of host cells remain to be elucidated. In this study, infections of human macrophages from the THP-1 monocyte cell line were conducted with B. melitensis Bm133 wild-type strain and a strain of Salmonella serovar Enteritidis as a control. A multiplicity of infection of 100 was used in trials focused on defining the relative expression of syntaxin 4 (STX4), a soluble N-ethylmaleimide-sensitive factor attachment protein receptor, in the early events of phagocytosis (at 15, 30, 45, and 60 min). Immunoblot assays were also done to visualize expression of the protein in cells infected with either bacterial strain. The expression of STX4 was not significantly different in cells infected with B. melitensis strain Bm133 compared to that observed in cells infected with S. Enteritidis. When the expression of STX4 mRNA was inhibited with short or small interfering, or silencing, RNA in the THP-1 cells, the survival of B. melitensis was significantly reduced at time 0, when gentamicin treatment of cultures was begun (after 1 h of phagocytosis), and also at 2 h and 12 h after infection.  相似文献

In order to evaluate whether ovarian volume, presence and diameter of the corpus luteum (CL) have effects on the number and quality of bovine recovered oocytes, 110 ovaries were obtained from the slaughterhouse. Cumulus oocytes complex were aspirated and evaluated under stereomicroscope. Oocytes were counted and classified according to their quality (Grades I, II, III and IV). Ovarian volume was weakly correlated to the number of good quality oocytes (P < 0.05). Ovaries with CL showed greater numbers of good quality oocytes than ovaries without CL (P < 0.05). Further, presence of CL and its diameter positively influenced the probability of recovering good quality oocytes (P < 0.05). In conclusion, ovarian volume is not a good parameter itself to predict important ovarian characteristics; moreover, analysis of CL, its presence and diameter, may be a good tool to improve efficiency on in vitro embryo production programs.  相似文献

In wheat, grain number is considered as the product of spike dry weight (SDW) and the number of grains per unit of SDW, that is an indicator of spike fertility (SF). The aim of this study was to determine the heritability of SF and the effect of early selection for high SF on its relationship with other spike yield components. Two field experiments were conducted in the south‐eastern Pampas (Argentina) with 400 F2 and F2:3 families obtained from two crosses between varieties with contrasting SF (PIG/SSN and B10/KCJ). Heritability estimates in PIG/SSN and B10/KCJ were, respectively, 0.60 and 0.51 by variance component analysis, 0.43 and 0.43 by F2 : F3 parent–offspring regression and 0.30 and 0.28 by realized heritability analysis. The existence of transgressive segregation (i.e. the occurrence of families with SF values that were more extreme than those of the parents) was observed. The top 25% F3 families with the highest SF had 12% more grains per spike, despite a 13% and 5% decrease in SDW per spike and weight per grain, respectively, than the remaining families. These results give support to the application of early selection for high SF.  相似文献

Swine slurry is a common agricultural fertilizer in many countries. However, its long‐term use in large amounts can cause excess nutrient accumulation, alter soil compounds, and potentially influence critical microbial populations such as arbuscular mycorrhizal fungi, which have important roles in plant nutrition and soil sustainability. This work determined if arbuscular mycorrhizal status, external mycelium, and glomalin‐related soil protein content were affected by long‐term swine slurry application to different soil tillage systems. The experiment was conducted on a clayey oxisol, in southern Brazil. Swine slurry (0, 30, 60, 90, and 120 m3 ha−1 y−1) was applied for 15 years to conventional tillage and no tillage soil prior to the summer (soybean or maize) and winter (wheat or oats) crop seasons. Swine slurry decreased mycorrhizal root colonization, spore number, and total external mycelium. Swine slurry increased active external mycelium and both easily extractable and total glomalin‐related soil protein. No‐tillage soil had more glomalin‐related soil protein than conventional tillage soil. The most significant response variables were root colonization, easily extractable glomalin‐related soil protein, and total external arbuscular mycorrhizal mycelia. Long‐term application of swine slurry in this environment, even at high rates, did not adversely affect crop yield but did influence arbuscular mycorrhizae fungi and their products in the soil environment. Benefits of swine slurry application for crop nutrition must be weighed against potential adverse consequences for the size, activity, and benefits of the mycorrhizal community to subsequent annual crops. Soil bacterial communities have significant influence on soilborne plant pathogens and, thus, crop health. The present study focuses on ribotyping soil bacterial communities in different peanut-cropping sequences in Alabama. The objective was to identify changes in microbial assemblages in response to cropping sequences that can play a role in managing soilborne plant pathogens in peanut. Four peanut-cropping sequences were sampled at the Wiregrass Research Station, Headland, AL in 2006 and 2007, including continuous peanut, 4 years of bahiagrass followed by peanut, peanut-cotton, and peanut-corn-cotton. Soil sampling was done at early and mid-season and at harvest. Bacterial community structure was assessed using ribosomal intergenic spacer analysis (RISA) combined with 16S rRNA cloning and sequencing. RISA results indicated >70% dissimilarities among different cropping sequences. However, 90% similarities were noticed among replicated plots of the same cropping sequences. Cropping sequences and time of soil sampling had considerable effect on soil microbial community structure. Bahiagrass rotation with peanut was found to have the highest bacterial diversity, as indicated by a high Shannon Weaver Diversity index. Overall, higher bacterial diversity was observed with bahiagrass and corn rotations compared with continuous peanut. The bacterial divisions Proteobacteria, Acidobacteria, Firmicutes, Bacteroidetes, and Actinomycetes were the predominant bacterial phyla found in all peanut-cropping sequences. The Proteobacteria taxa in these soils were negatively correlated with the abundance of members of division Firmicutes but, conversely, had a significant positive correlation with Gemmatimonadetes taxa. The prevalence of the division Actinomycetes was negatively correlated with the relative abundance of members of division Verrucomicrobia. These results indicate complex interactions among soil bacteria that are important contributors to crop health.  相似文献
