Photodynamic therapy (PDT) using a haematoporphyrin derivative (Photogem®, General Physics Institute and clustes Ltda) as photosensitizer and light emitting diodes (LEDs) as the light source was evaluated in 12 cats with cutaneous squamous cell carcinoma. Lesions were illuminated with LEDs, (300 J/cm for 30 min) 24 h after the administration of the photosensitizer. Clinical responses were classified as complete disappearance of the tumour with total re-epithelialization; partial response (a reduction greater than 50%); and no response (less than 50% reduction). Tumours localized to the pinna treated with one ( n  = 3) or two ( n  = 4) applications of PDT yielded no response. Highly invasive tumours of the nose and nasal planum also showed no response, after two treatments ( n  = 2). A combination of PDT and surgery was performed in three cases. Two cats showed partial response and one complete response with one application of therapy 30 days after nasal surgery. Small and noninfiltrative lesions ( n  = 3) of the nasal planum showed a PR with one application ( n  = 2) and a CR with two applications ( n  = 1). This study shows that PDT using Photogem® and LEDs can provide local control of low-grade feline squamous cell carcinoma. The addition of PDT to surgery in more invasive cases may help prevent recurrence.  相似文献

The present study used a proteomic data set obtained from a 2 × 2 × 2 factorial experiment on longissimus muscle of 24 pigs to identify single or couples of proteins allowing correct classification of pigs according to gender, sire breed, or rearing environment. An actin isoform, a myosin light chain 2 isoform and cytochrome Bc1 allowed each, correct classification of all pigs according to gender. Peroxiredoxin 6 allowed correct classification of 23 pigs according to indoor or outdoor rearing environment, but only if gender was also taken into account. Heat shock protein 73 allowed correct classification of 21 pigs according to sire breed, Duroc or Large White. Results show that proteins may be used as biomarkers for traceability or genetic research. Relationships between treatment factors and intracellular regulatory mechanisms associated with these proteins are discussed.  相似文献

An 11-month-old prepubertal crossbreed female dog was presented with multiple nodular lesions disseminated over the cervical, back, flank, and abdominal regions. The lesions were ulcerated and cauliflowerlike, or nodular and subcutaneous, measuring up to 13 cm in diameter. Cytologic preparations of one of the lesions revealed a uniform population of round to oval cells, with lightly basophilic cytoplasm that contained multiple distinct vacuoles. Frequent mitotic figures and occasional lymphocytes were also observed. The cytologic diagnosis was cutaneous transmissible venereal tumor (TVT) in a progressing growth phase. This was confirmed by histologic and immunohistochemical findings. Vaginal TVT was diagnosed later in the dog's mother. TVT is a contagious neoplasm of sexually mature dogs that usually is transmitted by coitus and affects the genital mucosa. To our knowledge, this is the first report of naturally occurring multicentric TVT in a prepubertal female dog and also is unique in its exclusively cutaneous (no mucosal) involvement. We speculate that transmission of neoplastic cells occurred during cohabitation and social/mothering behavior between the dogs. Despite the atypical clinical presentation, response to chemotherapy with vincristine was excellent, leading to complete regression of the neoplasm without relapse after 6 months.  相似文献

Peroxisome proliferator-activated receptors (PPARs) are involved in the regulation of lipid and carbohydrate metabolism and can be activated either by natural ligands as fatty acids or by synthetic ligands including several environmental chemicals. In this study, two PPARα isoforms (α1 and α2) were analyzed in turbot (Scophthalmus maximus) for a different tissue distribution. PPARα1 was ubiquitously expressed, while the PPARα2 was predominantly expressed in the heart. Following this result, turbot juveniles were exposed by injection to a synthetic selective PPARα agonist, WY-14643, for 14 days. Suppression subtractive hybridization (SSH) was performed with pools of heart samples of control and exposed fish to get insights into PPARα-regulated genes in the heart of juvenile turbot. Four genes were positively identified in the forward-subtracted and 12 genes in the reverse-subtracted cDNA SSH library, corresponding to the down-regulated and up-regulated genes in response to the WY-14643 treatment, respectively. The confirmation of these results in individual samples of juvenile turbot exposed to WY-14643 revealed a statistically significant mRNA induction of two cardiac muscle proteins (myosin light chain 2 and tropomyosin 4), which were shown to be involved in heart contraction and heartbeat regulation in other teleost species. Herewith, we showed for the first time that PPARα2 is predominantly expressed in the heart and that a PPARα agonist can induce the mRNA expression of cardiac muscle proteins in teleosts.  相似文献

Sulfated fucans comprise families of polydisperse natural polysaccharides based on sulfated L-fucose. Our aim was to investigate whether fucan nanogel induces cell-specific responses. To that end, a non toxic fucan extracted from Spatoglossum schröederi was chemically modified by grafting hexadecylamine to the polymer hydrophilic backbone. The resulting modified material (SNFuc) formed nanosized particles. The degree of substitution with hydrophobic chains was close to 100%, as estimated by elemental analysis. SNFfuc in aqueous media had a mean diameter of 123 nm and zeta potential of −38.3 ± 0.74 mV, as measured by dynamic light scattering. Nanoparticles conserved their size for up to 70 days. SNFuc cytotoxicity was determined using the MTT assay after culturing different cell lines for 24 h. Tumor-cell (HepG2, 786, H-S5) proliferation was inhibited by 2.0%–43.7% at nanogel concentrations of 0.05–0.5 mg/mL and rabbit aorta endothelial cells (RAEC) non-tumor cell line proliferation displayed inhibition of 8.0%–22.0%. On the other hand, nanogel improved Chinese hamster ovary (CHO) and monocyte macrophage cell (RAW) non-tumor cell line proliferation in the same concentration range. The antiproliferative effect against tumor cells was also confirmed using the BrdU test. Flow cytometric analysis revealed that the fucan nanogel inhibited 786 cell proliferation through caspase and caspase-independent mechanisms. In addition, SNFuc blocks 786 cell passages in the S and G2-M phases of the cell cycle.  相似文献

This work describes the mechanical and solvent extraction of Sterculia striata seed oil. It was determined that the seeds contain up to 41% in oil, which has an unusual composition. Indeed, up to 50% of the fatty acid contain cyclopropenoid ring. The oil was used as raw material to produce bio-oil and biodiesel and their physical-chemical properties were evaluated. Some of the studied physical-chemical properties of the S. striata biodiesel are in acceptable range for use as biodiesel in diesel engines, showing a promising economic exploitation of this raw material in semi-arid regions. It was also observed that the cyclopropenoid ring remains after transesterification and is decomposed during pyrolysis.  相似文献

Abstract

The L46 (Hypancistrus zebra) stands out as one of the most valuable Amazonian species in the international market for ornamental fish and faces a notable problem: the risk of extinction versus the demand for new specimens for aquariums. Considering that breeding in captivity can be a conservational tool for aquatic species and an alternative source for generating income, the objective of the present paper was to verify the economic feasibility of producing H. zebra in recirculation aquaculture systems (RAS), in different scenarios of rearing: residential breeding (S1), mid-size store production (S2) and large-scale production as a supplier (S3). The main profitability indicators used were: net present value (NPV), internal rate of return (IRR) and payback period (PP). The selling price per unit was fixed in US$260.15 (S1/S2) and US$156.09 (S3). The largest investments were made with the acquisition of broodstocks (S1/S2) and property (S3), while the highest costs were with labor (S2/S3) and taxes (S1). Only S1 demonstrated economic profitability (NPV 8.50%?=?US$28,187.85; IRR?=?74.71%; PP?=?1.80?years). Conclusion: the production in this study appeared to be profitable on smaller scales, however the profitability of large-scale production depends upon reducing expenses and increasing the selling price.  相似文献

This study proposes to evaluate rapid methodologies to estimate oxidative stability and biodegradability for mineral, vegetable and synthetic oil samples, using quick and simple experiments. The synthetic oil sample was obtained from castor oil, so the vegetable oil chosen for this evaluation was a sample of the crude castor oil source. The oxidative stability tests described enabled the comparison of synthetic lubricant samples and showed that their stability was lower than that of petroleum-based oil. However, the physico-chemical properties, such pour points and viscosity index, were improved and potentially interesting for lubricant applications. The biodegradability experiments were carried out using a model of bio-kinetics. These studies proved that synthetic lubricant samples were easily degradable (similar to crude castor oil) and showed half-life significantly lower than those of the mineral oil samples.  相似文献

Knowledge of seed desiccation tolerance is fundamental for conservation and use of forest species. The protocol used for classification of seed desiccation tolerance and storage is time consuming and many times limited by the lack of information about optimum conditions for seed germination and treatments to overcome seed dormancy. This study evaluated 66 Brazilian tree species aiming to correlate seed characteristics with desiccation tolerance. For this purpose, a model was established to explain the relationship of tegument/seed mass ratio (SCR), seed mass, and water content of embryo + endosperm with desiccation tolerance. The principal component analysis showed the establishment of two groups, indicating the interaction between desiccation tolerance and seed characteristics. Recalcitrant seeds are more often associated with the water content of embryo + endosperm and water content of tegument + endocarp, while orthodox seeds are more associated with SCR and number of seeds per kilogram. The classification found using the model proposed was significantly correlated with desiccation tolerance and storage, with 92% confidence for the analyzed species. Seeds morphological characteristics can be used for prediction of desiccation tolerance and storage behavior; however, the use of a model that combines more variables increases the chance of accurate classification.  相似文献

Soil acidity is one of the most important factors limiting crop production. The objective of this work was to evaluate the effects of limestone application on the soil chemical properties, nutrition and yield of mango plants in an orchard under implementation. The design was randomized blocks, with five limestone doses (0; 2; 4; 6 and 8 t ha?1) and four replications. Soil chemical analyses were performed (at 12, 24, 36 and 48 months after the experiment implementation) in the layers 0–20; 20–40 and 40–60 cm deep. Nutrition status and yield were assessed during the first and second crop seasons. The highest fruit yield was associated with the application of 3.9 and 3.8 t ha?1 of limestone in the 2008/09 and 2009/10 seasons, respectively, that is, the dose recommended by the literature for raising base saturation to 80%, as a function of the fertility conditions of the soil initially obtained.  相似文献
