The tumor suppressor programmed cell death protein 4 (PDCD4) inhibits the translation initiation factor eIF4A, an RNA helicase that catalyzes the unwinding of secondary structure at the 5' untranslated region (5'UTR) of messenger RNAs (mRNAs). In response to mitogens, PDCD4 was rapidly phosphorylated on Ser67 by the protein kinase S6K1 and subsequently degraded via the ubiquitin ligase SCF(betaTRCP). Expression in cultured cells of a stable PDCD4 mutant that is unable to bind betaTRCP inhibited translation of an mRNA with a structured 5'UTR, resulted in smaller cell size, and slowed down cell cycle progression. We propose that regulated degradation of PDCD4 in response to mitogens allows efficient protein synthesis and consequently cell growth.  相似文献

We describe the genome sequence of the protist Trichomonas vaginalis, a sexually transmitted human pathogen. Repeats and transposable elements comprise about two-thirds of the approximately 160-megabase genome, reflecting a recent massive expansion of genetic material. This expansion, in conjunction with the shaping of metabolic pathways that likely transpired through lateral gene transfer from bacteria, and amplification of specific gene families implicated in pathogenesis and phagocytosis of host proteins may exemplify adaptations of the parasite during its transition to a urogenital environment. The genome sequence predicts previously unknown functions for the hydrogenosome, which support a common evolutionary origin of this unusual organelle with mitochondria.  相似文献

DNA replication in eukaryotes requires nucleosome disruption ahead of the replication fork and reassembly behind. An unresolved issue concerns how histone dynamics are coordinated with fork progression to maintain chromosomal stability. Here, we characterize a complex in which the human histone chaperone Asf1 and MCM2-7, the putative replicative helicase, are connected through a histone H3-H4 bridge. Depletion of Asf1 by RNA interference impedes DNA unwinding at replication sites, and similar defects arise from overproduction of new histone H3-H4 that compromises Asf1 function. These data link Asf1 chaperone function, histone supply, and replicative unwinding of DNA in chromatin. We propose that Asf1, as a histone acceptor and donor, handles parental and new histones at the replication fork via an Asf1-(H3-H4)-MCM2-7 intermediate and thus provides a means to fine-tune replication fork progression and histone supply and demand.  相似文献

Biostimulants are substances promoting plant growth, quality and stress resistance. The present work aimed to investigate whether soluble hydrolysates from biowaste performed as biostimulants. Hibiscus (Hibiscus moscheutos L. subsp. palustris) plants were subjected to four treatments: standard fertilization, low fertilization (LF), and LF with added soluble digestate or soluble compost. Plant performance indicators were biomass accumulation, biometric parameters, leaf gaseous exchanges and elemental composition, and nitrogen (N)-use efficiency. LF negatively affected most of the investigated parameters. However, plants treated with biowaste-derived products performed better than untreated low-fertilized plants (+21 to 145% for biomass accumulation and biometric parameters, and carbon assimilation rate) and for many parameters reached values comparable to those showed by standard-fertilized plants or even higher, as in the case of Potassium (K), Silicon (Si), and Molybdenum (Mo) leaf content, and N use efficiency. Therefore, the tested soluble hydrolysates demonstrated to have biostimulant properties in hibiscus grown under nutritional stress.  相似文献

Purpose

The aim of this work was to assess the concentrations of potentially toxic elements and to evaluate the soil quality of a typical Prosecco Denomination of Controlled and Guaranteed Origin vineyard of the Veneto region, NE Italy.

Materials and methods

Soil samples and leaves of Taraxacum officinale and Vitis vinifera were collected during spring–summer 2014. Element determination (Al, Cd, Cr, Cu, Fe, Mg, Mn, Ni, P, Pb, V, and Zn) were performed with ICP-OES after microwave digestion of samples. Soil quality was assessed via the biological soil quality (BSQ-ar) index. Lipid peroxidation test was performed to evaluate the vegetation oxidative stress, based on malondialdehyde (MDA) content via spectrophotometer.

Results and discussion

High concentrations of Al, Mg, and P were identified in soil, while high contents of Al, Cu, Fe, and Zn were found in V. vinifera leaves. The high concentrations in soil are probably due to agricultural activities, whereas those in leaves are probably due to atmospheric deposition and repeated use of foliar sprays in viticulture. The bioconcentration factor showed an effective transport of Cu, P, and Zn, from soil to leaf. The BSQ-ar values registered were similar to those obtained in preserved soils; hence, the biological class (VI) of these soils is high. The MDA content in T. officinale and V. vinifera leaves was below the reference value for T. officinale (2.9?±?0.2 μM), suggesting that the metal content did not stress the vegetation in the investigated site.

Conclusions

The MDA value for V. vinifera (1.1?±?0.7 μM) could be adopted as another control value for soil quality, which in our case is of “good quality.” Moreover, our results suggest that high concentrations of elements detected in the analyzed samples do not influence negatively the quality of soil, but a better agronomic management could improve soil quality in the studied area.

Skins from different hazelnut samples were characterized for total polyphenol content, total antioxidant capacity (TAC), and their content in specific polyphenolic compounds. The main polyphenolic subclass, identified and quantified by means of HPLC-MS/MS, comprised monomeric and oligomeric flavan-3-ols, which accounted for more than 95% of total polyphenols. Flavonols and dihydrochalcones were 3.5% while phenolic acids were less than 1% of the total identified phenolics. The TAC values of the skin samples ranged between 0.6 and 2.2 mol of reduced iron/kg of sample, which is about 3 times the TAC of whole walnuts, 7-8 times that of dark chocolate, 10 times that of espresso coffee, and 25 times that of blackberries. By describing the profile of polyphenols present in hazelnut skins, this study provides the basis to further investigate the potential health effects of hazelnut byproduct.  相似文献

The use of municipal solid waste compost (MSWC) as soil organic amendment is of an economic and environmental interest. However, little is known about the effectiveness of MSWC application on agricultural soil in northern Africa arid climate. We assessed the impact of five years' applications of different organic and mineral fertilizers on wheat grain yields and soil chemical and microbial characteristics. Soils were treated with MSWC at rates of 40 (C1) and 80 (C2) Mg ha?1, farmyard manure at a rate of 40 Mg ha?1 (M), chemical fertilizers (Cf) and the combinations (C1Cf, C2Cf, MCf). Wheat grain yield was enhanced with all amendments. Parallel increases of heavy metal levels and faecal coliform were also recorded except for Cf treatments. Based on wheat grain yield, heavy metal and faecal coliform data, we determined the treatment effectiveness index (Exx), calculated by dividing the pollutant increase ratio by the grain yield increase ratio. The treatment effectiveness index EC1 indicated lower faecal and heavy metal pollution with positive gains in wheat yields. Despite polluting effects on soil determined by the different treatments, no significant differences between treatments were observed in total bacterial count and soil bacterial community structure, as shown by 16S rRNA gene PCR-denaturing gradient gel electrophoresis banding patterns and 16S rRNA gene Length Heterogeneity-PCR analysis. According to the collected data, the use of MSWC at a rate of 40 Mg ha?1 might be recommended.  相似文献

The 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation (ABTS(*)(+)) assay was adapted to a flow injection (FI) system to obtain a sensitive and rapid technique for the monitoring of antioxidant activity of pure compounds and complex matrixes, such as beverages and food extracts. The FI system includes a HPLC pump that flows the mobile phase (a solution of ABTS(*)(+) in ethanol) through a 20 microL loop injector, a single bead string reactor filled with acid-washed silanized beads, a delay coil and a photodiode array UV-visible detector. The technique was very sensitive, with limits of detection and of quantification of 4.14 and 9.29 micromol of Trolox/L, respectively, and demonstrated high repeatability and reproducibility. The proposed technique was then applied to the evaluation of the antioxidant activity of some pure compounds, demonstrating good agreement with published data obtained by the original spectrophotometric ABTS(*)(+) assay. Finally, the total antioxidant activity of 10 beverages was determined by both the proposed and the original method. The values ranged from 0.09 mmol L(-)(1) for cola to 49.24 mmol L(-)(1) for espresso coffee and did not result significantly different from those obtained by the original spectrophotometric ABTS(*)(+) assay (Student's paired t-test: t = 1.4074, p = 0.1929). In conclusion, the proposed FI technique seems suitable for the direct, rapid and reliable monitoring of total antioxidant activity of pure compounds and beverages and, due to the ability to operate in continuous, it allows the analysis of about 30 samples h(-)(1) making the assay particularly suitable for large screening of total antioxidant activity in food samples.  相似文献

This study was conducted on an irrigated area of southern Italy to analyze the current operation of a large-scale irrigation delivery system and the effects of the operation procedures on crop irrigation management and aquifer salinity increase. The area is characterized by relatively high levels of groundwater salinity in the summer that are probably due to intensive groundwater pumping by farmers during periods of peak irrigation demand, with the resulting seawater intrusion. Two alternative delivery schedules, namely the rotation delivery schedule and the flexible delivery schedule, referred to as RDS and FDS, respectively, were simulated using a soil-water balance model under different combinations of crop, soil and climatic conditions. The first set of simulations concerned the farm irrigation management constrained by the rotational delivery used by the local water management organization. The second scenario simulated the farm irrigation schedule most commonly used by growers in the area for maximizing crop yields. Based on crop irrigation management under RDS and FDS, two alternative operational scenarios were also developed at the scheme level and then compared for evaluation. Winter and summer salinity maps of the aquifer were developed by interpolating salinity measurements of the groundwater samples collected during the 2006 irrigation season. From these maps, a close relationship can be inferred among delivery schedule, aquifer exploitation and salinity increase, which justifies the need for implementing FDS that might reduce the groundwater demand for irrigation.  相似文献

Thirty-nine sea beet [Beta vulgaris L. ssp. maritima (L.) Arcang.] accessions of the Adriatic coast were screened genetically and for their adaptive morpho-functional root traits in order to identify new sources of abiotic resistances for sugar beet breeding programs. Genetic diversity was evaluated with 21 microsatellites markers that identified 44 polymorphic alleles. Sea beets grouped into two main clusters: the West and the East Adriatic coast groups, with the latter showing higher genetic diversity. Among sea beet accessions with desirable root traits, four accessions have proved to be interesting for sugar beet [B. vulgaris (L.) ssp. vulgaris] breeding aimed to improve tolerance to nutritional stresses. Lastovo (ID 29) and Zut (ID 34) accessions were characterized by the highest values of RER, TRL, FRL and RSA still maintaining a high value of RTD, while Grado (ID 21) an Portic (ID 23) accessions were characterized by the highest RTD, but with low values of RER, TRL, FRL and RSA parameters.  相似文献
