Captive breeding and release programs, widely used to supplement populations of declining species, minimize juvenile mortality to achieve rapid population growth. However, raising animals in benign environments may promote traits that are adaptive in captivity but maladaptive in nature. In chinook salmon, hatchery rearing relaxes natural selection favoring large eggs, allowing fecundity selection to drive exceptionally rapid evolution of small eggs. Trends toward small eggs are also evident in natural populations heavily supplemented by hatcheries, but not in minimally supplemented populations. Unintentional selection in captivity can lead to rapid changes in critical life-history traits that may reduce the success of supplementation or reintroduction programs.  相似文献

The biodegradation kinetics of anionic (sodium laureth sulfate – SLES), amphoteric (disodium cocoamphodiacetate – DSCADA), and nonionic surfactants (polyalcohol ethoxylate – PAE) were assessed in this laboratory study. Similar degradation behavior was observed for all surfactants with only a fraction of the parent compound readily biodegradable. Biodegradation, as estimated by COD removal, was initially (i.e., within 24 h) rapid, however only 40–70% of the surfactant molecules were readily biodegradable. Intrinsic kinetic parameters were successfully quantified for the readily biodegradable component of the surfactant. Inhibition was not observed and microbial kinetics of SLES, DSCADA, and PAE degradation fit the Monod model well. Average μ-S curves were generated for each surfactant. Based on these results, complete degradation of the target surfactants using biological waste treatment would be limited.  相似文献

Modeling the number of uses of discrete habitat types by animals with a multinomial distribution, we illustrate the use of Bayesian methods to estimate selection. An advantage of this approach in assessing selection is the construction of credibility intervals that do not rely on large sample normal theory. In addition, credibility intervals for ranked selection of habitats are easily obtained. Bayes factors and Bayesian p values (posterior predictive values) are used to test the hypothesis of selection for each animal, test selection across all animals and for multiple comparisons among habitats. We compare our method to alternative methods for a real dataset. Freely available WinBUGS software is used to fit the model and test hypotheses.  相似文献

Bovine viral diarrhea virus (BVDV) is an important pathogen of domestic cattle. Serologic, experimental, and individual case studies explored the presence and pathogenesis of the virus in wild ungulates; however, there remain large gaps in knowledge regarding BVDV infection in nonbovine species. Live twins were born from a white-tailed deer (Odocoileus virginianus) doe infected with noncytopathic BVDV during its first trimester of pregnancy. The twins died at 1 day of age from trauma unrelated to the infection, and tissues were collected for histologic and immunohistochemical examination. The most significant histologic abnormality was diffuse depletion of B-lymphocytes in both fawns. The BVDV antigen was distributed widely throughout many tissues and cell types, most notably epithelium and vascular endothelium, consistent with that reported in cattle. In contrast to cattle, lymphocytes exhibited only very rare positive staining.  相似文献

Ascidians are marine invertebrates that have been a source of numerous cytotoxic compounds. Of the first six marine-derived drugs that made anticancer clinical trials, three originated from ascidian specimens. In order to identify new anti-neoplastic compounds, an ascidian extract library (143 samples) was generated and screened in MDA-MB-231 breast cancer cells using a real-time cell analyzer (RTCA). This resulted in 143 time-dependent cell response profiles (TCRP), which are read-outs of changes to the growth rate, morphology, and adhesive characteristics of the cell culture. Twenty-one extracts affected the TCRP of MDA-MB-231 cells and were further investigated regarding toxicity and specificity, as well as their effects on cell morphology and cell cycle. The results of these studies were used to prioritize extracts for bioassay-guided fractionation, which led to the isolation of the previously identified marine natural product, eusynstyelamide B (1). This bis-indole alkaloid was shown to display an IC50 of 5 µM in MDA-MB-231 cells. Moreover, 1 caused a strong cell cycle arrest in G2/M and induced apoptosis after 72 h treatment, making this molecule an attractive candidate for further mechanism of action studies.  相似文献

Forest emissions of biogenic volatile organic compounds (BVOCs), such as isoprene and other terpenes, play a role in the production of tropospheric ozone and aerosols. In a northern Michigan forest, the direct measurement of total OH reactivity, which is the inverse of the OH lifetime, was significantly greater than expected. The difference between measured and expected OH reactivity, called the missing OH reactivity, increased with temperature, as did emission rates for terpenes and other BVOCs. These measurements are consistent with the hypothesis that unknown reactive BVOCs, perhaps terpenes, provide the missing OH reactivity.  相似文献

In a previous study, Giardia infection patterns were studied in newborn dairy calves over a 4-month period. Chronic Giardia infections were observed in all calves with initial cyst excretion occurring at approximately 1 month of age. In the work presented here, the passive immunity and serological immune response associated with these Giardia infections were examined. Colostrum and milk samples were collected from the dams of these calves, and monthly serum samples were collected from each calf. The colostrum, milk and sera samples were analyzed by ELISA and Western blot for the presence of anti-Giardia IgG antibodies. In addition, the in vitro anti-Giardia activity of milk and colostrum was examined using a miniculture adherence assay. When examined by ELISA, mean anti-Giardia antibody titres were found to be significantly higher in colostrum compared to milk. The monthly mean serum antibody titres in the calves were not found to differ significantly at any time point during the study. Western blot analysis revealed that colostrum from the dams reacted strongly with many different Giardia antigens between 205 and 7.5kDa, while milk reacted with few antigens in the same size range. Sera collected from the calves when 30 and 60 days of age reacted with few Giardia antigens, but as the calves aged, IgG antibodies in their sera began to react with antigens of 21, 50, 65, 73 and 79kDa. The miniculture adherence assay demonstrated that colostrum had significantly more anti-Giardia activity in vitro compared to milk. These results suggest that the calves in this dairy did not mount a significant humoral immune response against Giardia following infection. However, colostrum contained a high level of anti-Giardia antibodies and exhibited anti-Giardia activity in vitro. Therefore, colostrum may have the potential to provide initial protection against Giardia infections in calves, but the lack of a strong, specific humoral immune response by these calves could account for the high prevalence and chronic duration of the infections.  相似文献

The objectives of this study were to determine if percutaneous lung biopsy can be used to characterize early pathologic changes in bovine lung associated with bovine respiratory disease (BRD), to determine if specific infectious respiratory pathogens can be identified in association with these changes, and to determine whether pulmonary pathology at arrival and at the time of initial diagnosis are associated with health and production outcomes. One hundred auction-market derived crossbred steer calves from a commercial feedlot in southern Alberta were included in this study. A percutaneous lung biopsy technique was used to obtain lung samples from the right middle lung. Steers were sampled 295 times yielding 283 samples with 210 (74%) containing lung tissue. Overall, histopathological changes were observed in 20 (9.5%) of lung biopsy samples. There were too few samples with pathology to reveal an association between lung pathology and subsequent health events. In general, percutaneous lung biopsy can be done safely on feedlot steers in a commercial feedlot setting with few clinical side effects. This technique did not prove useful as a diagnostic tool or prognostic indicator for early BRD. However, it may be useful for the diagnosis of BRD in targeted populations of commercial feedlot steers.  相似文献

Cytology is the most common and immediately informative diagnostic tool in the practice of dermatology. It is simple to perform and requires equipment usually already present in most practices such as a microscope and slides, cotton-tipped applicators and stains. Cytology interpretation is also easily self taught. With little practice the practitioner can easily become familiar with sample interpretation from inflammatory lesions; including recognition of bacteria, yeast, dermatophyte spores and hyphae, deeper fungal elements and parasites. Additionally, the practitioner should become familiar with the variety of inflammatory cells seen with these conditions, as well as keratinocyte morphology. This article provides an overview and the initial steps on how to become a more proficient cytologist in practice.  相似文献
