A new real-time quantitative polymerase chain reaction (qPCR) test was used to diagnose Campylobacter fetus subsp. venerealis infection associated with dramatic reproductive losses in a commercial cow-calf herd. The results were verified with repeated culture, phenotypic characterization of the organism and DNA sequencing. This case demonstrates the need for a practical field test for C. fetus subsp. venerealis and the importance of considering this organism as a potential cause of pregnancy failure in beef herds.  相似文献

ME-5343 (afidopyropen) is a new and promising insecticide with an unknown mechanism of action that is effective against sucking insects. ME-5343 was highly toxic to pea aphids (Acyrthosiphon pisum), being more toxic than six other widely used insecticides. In contrast, ME-5343 was practically non-toxic to eight other species of insects we tested. ME-5343 was not toxic to German cockroaches (topical application) or American cockroaches (injection), suggesting that lack of toxicity in these species is not due to lack of cuticular penetration. House flies are insensitive to ME-5343 by topical, residual and feeding exposure. Addition of synergists did not change this result, suggesting that insensitivity to ME-5343 in house flies is not due to rapid detoxification nor is it dependent on the method of bioassay used. ME-5343 did not cause firefly lanterns to glow, nor did it prevent the octopamine stimulated lantern glow. Extracellular recordings of action potentials from a tonically active motor nerve of crayfish in situ showed no effects of ME-5343 at concentrations up to 10−5 M. These results suggest that the target site of ME-5343 is not the voltage gated sodium channel, voltage gated potassium channel, GABA gated chloride channel, nicotinic acetylcholine receptor, acetylcholinesterase, octopamine receptor or glutamate receptor. ME-5343 injected into crayfish caused flextion of the legs and tail, similar to the symptoms induced by 5-HT (serotonin). We evaluated the effect of ME-5343 on 5-HT2-like receptors with intracellular recordings of excitatory post-synaptic potentials from the peripheral neuromuscular junction of the crayfish and found no effect of ME-5343. Thus, ME-5343 was neither an agonist nor antagonist of 5-HT2 receptors, did not affect neurotransmitter release and did not affect glutamate receptors. We conclude that ME-5343 is highly toxic to aphids and that this is due to a unique, and currently undefined mechanism of action.  相似文献

Select mushrooms were analyzed for proximate constituents and carbohydrate profiles either raw or cooked and at different stages of maturity. White button mushrooms (Agaricus biporus) contained high concentrations of ash (12.5 and 11.9% for immature and mature mushrooms, respectively). Starch and total dietary fiber (TDF) concentrations were higher in maitake (Grifola frondosa) and shiitake (Lentinus edodes) mushrooms. Crude protein (CP) and acid-hydrolyzed fat (AHF) were highest in crimini (Agaricus bisporus) and white button and maitake mushrooms, respectively. Chitin concentrations were highest in portabella (Agaricus bisporus) and enoki (Flammulina velutipes) mushrooms (8.0 and 7.7%, respectively). Oligosaccharides were found in low concentrations in some mushrooms. CP and TDF accounted for 86.4 and 49.3% of mushroom organic matter. Cooking increased starch, TDF, and AHF but decreased CP and chitin concentrations. The chitin concentration increased with mushroom maturity. These results detail the complete carbohydrate profile of several important mushroom varieties.  相似文献

Several genetically modified (GM) cultivars are registered in Canada although they are not currently in commercial production. The GM cultivars can be distinguished from the non-GM and other GM cultivars by analyzing the DNA nucleotide sequence at the insertion site of the transgene corresponding to a single transformation event in the plant genome. Techniques based on modified polymerase chain reaction (PCR) strategies were used to generate sequence information from the plant genome flanking the insertion site of transgenic DNA for specific GM potato events. The plant genome sequence adjacent to the transgenic insertion was used to design PCR primers, which could be used in combination with a primer annealing to one of the nearby inserted genetic elements to amplify an event specific DNA fragment. The event specific PCR fragments generated were sequenced to confirm the specificity of the method.  相似文献

The ability of nonacidified, refrigerated pickled cucumbers to produce the fresh cucumber flavor impact compounds (E,Z)-2,6-nonadienal and (E)-2-nonenal declined during storage. Production of these compounds decreased as the pH of refrigerated cucumbers was reduced. Despite the fact that the concentrations of (E,Z)-2,6-nonadienal and (E)-2-nonenal generated were over 10(5)-fold greater than the threshold levels, it was possible for a sensory panel to consistently detect differences in the intensity of fresh cucumber flavor, provided the pH difference between samples was 1 unit or greater. The presence of spices did not interfere with the ability of panelists to detect differences in fresh flavor intensity. There was a linear correlation between sensory scores and the amount of (E,Z)-2,6-nonadienal produced by cucumbers equilibrated at different pH levels.  相似文献

Imaging studies in people indicate that x-ray computed tomography (CT) is a more sensitive technique than thoracic radiography for the detection of pulmonary metastasic neoplasia. Systematic studies comparing CT and thoracic radiographic techniques in veterinary patients have not been performed. The present retrospective study was designed to directly compare the efficacy of these 2 techniques in detecting pulmonary nodules in dogs. Eighteen dogs with histologically confirmed pulmonary metastatic neoplasia had contemporaneous thoracic radiographs and pulmonary CT scans compared. Quantitative analyses included estimation of pulmonary nodule size, number, and lobar distribution on thoracic radiographs and CT images. Only 9% of CT-detected pulmonary nodules were identified on thoracic radiographs (P < .003). The lower size threshold was approximately 1 mm to detect pulmonary nodules on CT images and 7-9 mm to reliably detect nodules on radiographs (P < .0001). Additionally, pulmonary nodules were detected in a significantly greater number of lung lobes using CT as compared with thoracic radiographs (P < .0001). These data indicate that CT is significantly more sensitive than thoracic radiography for detecting soft-tissue nodules in dogs. As such, thoracic CT should be considered in any patient with neoplasia that has potential for pulmonary metastasis to more reliably stage the disease, particularly when accurate characterization of the extent and distribution of pulmonary metastatic disease affects therapeutic planning.  相似文献

A 60‐day indoor feeding trial was conducted to evaluate the effects of dietary tryptophan supplementation on growth performances, whole‐body chemical composition, expression of muscle growth‐related genes (MyoD, myogenin and myostatin), and haematological and biochemical responses of juvenile genetically improved farmed tilapia (GIFT). Five corn–soy‐based isonitrogenous and isoenergetic diets were formulated to contain graded levels of dietary tryptophan (2.6, 3.2, 3.7, 4.2 and 4.8 g/kg of diet). Each diet was randomly assigned to triplicate groups of 30 fish (5.3 ± 0.1 g) per experimental unit, which were fed thrice a day (9:00, 13:00 and 17:00 hr). Maximum growth performances and feed utilization were observed in fish fed tryptophan at 3.7 g/kg of diet. There was no significant (p > .05) effect on whole‐body composition and amino acid profile by dietary tryptophan supplementation. However, significant (p < .05) differences were observed in plasma metabolites and the mRNA expression of MyoD, myogenin and myostatin. Serum cortisol level was found significantly lowest in fish fed tryptophan at 3.7 g/kg of diet. Second‐order polynomial regression analysis of weight gain and nitrogen gain against dietary tryptophan levels indicated that the optimum dietary tryptophan requirement for maximum growth and feed utilization of juvenile GIFT tilapia was 3.8 g/kg of diet.  相似文献

Calcium depletion/replacement studies were carried out to examine the role of calcium in contraction of trout intestinal smoot muscle in vitro. Three chemically distinct calcium channel blockers were used to determine whether voltage operated calcium channels (VOCs) were involved in calcium entry with either agonist or depolarization-induced contractions. Contractions induced by depolarizing intestinal smooth muscle with potassium were totally dependent on extracellular calcium, whereas receptor-mediated responses to 5-hydroxytryptamine (5-HT) and carbachol also relied on calcium derived from intracellular stores. The calcium channel blockers, verapamil, nitrendipine, and diltiazem, all shifted the calcium-response curve for potassium to the right, supporting the existence of VOCs in trout intestinal smooth muscle. The calcium-response curve for 5-HT was also shifted to the right, suggesting that 5-HT can induce calcium uptake into the smooth muscle via VOCs, in addition to mobilizing intracellular calcium. Verapamil also appeared to block 5-HT receptors directly. Carbachol-induced contractions were only reduced by diltiazem at low concentrations of calcium (0.1–1 mM), suggesting that diltiazem has some other mechanisms of action than the other calcium channel blockers. Activation of muscarinic receptors may induce calcium entry through channels other than the VOCs, in addition to mobilizing intracellular calcium.  相似文献
