Neosporosis is a major cause of abortion in cattle all over the world. Congenital transmission as well as horizontal transmission by ingestion of oocysts has been described. The detection of Neospora caninum DNA in bull semen warrants the investigation of possible transmission through the use of contaminated semen. In this experiment four cows were artificially inseminated with frozen-thawed semen contaminated in vitro with viable N. caninum tachyzoites (group A) and four control cows were inseminated with tachyzoites-free frozen-thawed semen, from the same bull (group B). Serum samples were collected 15 days before the artificial insemination (AI) and at days 10, 14, 21, 28, 45, 60 and 75 post-insemination. All sera samples were tested for neosporosis by direct agglutination test (DAT). Three of the cows from group A had negative DAT titers (< or =1:20) in all of the samples, while the fourth cow from this group had a low titer of antibodies (1:80) at day 10, and became negative at day 45, suggesting a stimulation of the immune system by the tachyzoites placed in uterus, rather than the induction of an infection. All of the cows from group B had negative DAT titers (< or =1:20) in all of the samples. These results suggest that transmission of neosporosis by artificial insemination with frozen-thawed semen is an unlikely event. 相似文献

Canine leishmaniosis, caused by Leishmania infantum, is a systemic disease with variable clinical signs and a progressive evolution. This disease is characterized by impaired T cell-mediated immune response, which has been associated with disease chronicity and high mortality. Protective immunity against leishmaniosis is thought to be mediated by T cell and cytokine production. The T cell activation requires a primary signal delivered by the major histocompatibility complex (MHC) molecules present on the surface of antigen presenting cells, and a non-specific signal generated by co-stimulatory molecules. To characterize canine immune responses in the presence of L. infantum parasites or their antigens, in vitro cell cultures of canine macrophages and lymphocytes were established, and the macrophages presenting MHC class II molecules were evaluated as well as the expression of IL-12 and CD80-86 co-stimulatory molecules and nitric oxide production. The results showed for the first time the up-regulation of MHC class II molecules on the surface in canine peripheral blood monocyte-derived macrophages during L. infantum infection in the presence of lymphocytes. In addition, a lack of co-stimulatory expression and a reduced release of nitric oxide were observed, suggesting a loss of T cell function and consequently an inactivation of the macrophage oxidative burst which, in turn, favors the survival of Leishmania. These results constitute a new contribution for the understanding of the interactions between L. infantum and the canine immune system. 相似文献

This study aims to evaluate the effect of the Tifton 85 hay (Cynodon dactylon L. Pers) replacement by Orelha de Elefante Mexicana spineless cactus (Opuntia stricta [Haw.] Haw) on the nutrient intake and digestibility, growth performance, microbial protein synthesis, and efficiency of dietary nitrogen compound utilization in the diets of Girolando heifers. Twenty-four (3.5 months and 100 kg ± 3.5 kg) Girolando heifers (5/8 Holstein x Gyr) were used and arranged in a completely randomized design. Dry matter, organic matter, crude protein, and neutral detergent fiber decreased linearly as a function of the levels of Tifton hay (TH) replacement by Orelha de Elefante Mexicana spineless cactus (OEM). The non-fiber carbohydrate intake and coefficient of dry matter digestibility increased, while the neutral detergent fiber (NDF) digestibility decreased linearly with the OEM inclusion. After evaluating the growth performance by observing weight gain and morphometric measurements, there were no significant alterations. The urinary volume, nitrogen balance as well as the efficiency of nitrogen retention had increased, while the urinary and plasma urea concentrations had decreased linearly with the OEM inclusion. The results obtained in this study show that Orelha de Elefante Mexicana spineless cactus should replace Tifton hay in post-weaned heifer diets. 相似文献

Clementines are especially appreciated for their delicious flavor, and recent years have seen a great increase in the consumption of clementine juice. In previous decades, antioxidant compounds have received particular attention because of widely demonstrated beneficial health effects. In this work, the organoleptic, volatile flavor, and antioxidant quality of clementine juice were studied with regard to the influence on them by different juice extraction systems: plug inside fruit and rotating cylinders. The results showed that juice extracted by the former method presented higher yields and hesperidin content, which was related to higher antioxidant activity, demonstrated by ORAC and LDL assays. The organoleptic quality was not affected by the processing technique, whereas there were significant differences in the chemical flavor profile. There are important differences in chemical and functional quality between juice extraction techniques, which must be taken into account when employing processing systems to produce high-quality products. 相似文献

Recent studies indicate that plant polyphenols could be pointed as potential prebiotic candidates since they may interact with the gut microbiota, stimulating its growth and the production of metabolites. However, little is known about the fate of brown seaweeds’ phlorotannins during their passage throughout the gastrointestinal tract. This work aimed to evaluate the stability and bioaccessibility of Fucus vesiculosus phlorotannins after being submitted to a simulated digestive process, as well as their possible modulatory effects on gut microbiota and short-chain fatty acids production following a fermentation procedure using fecal inoculates to mimic the conditions of the large intestine. The stability of phlorotannins throughout the gastrointestinal tract was reduced, with a bioaccessibility index between 2 and 14%. Moreover, slight alterations in the growth of certain commensal bacteria were noticed, with Enterococcus spp. being the most enhanced group. Likewise, F. vesiculosus phlorotannins displayed striking capacity to enhance the levels of propionate and butyrate, which are two important short-chain fatty acids known for their role in intestinal homeostasis. In summary, this work provides valuable information regarding the behavior of F. vesiculosus phlorotannins along the gastrointestinal tract, presenting clear evidence that these compounds can positively contribute to the maintenance of a healthy gastrointestinal condition. 相似文献

Evolution by natural selection is driven by the continuous generation of adaptive mutations. We measured the genomic mutation rate that generates beneficial mutations and their effects on fitness in Escherichia coli under conditions in which the effect of competition between lineages carrying different beneficial mutations is minimized. We found a rate on the order of 10(-5) per genome per generation, which is 1000 times as high as previous estimates, and a mean selective advantage of 1%. Such a high rate of adaptive evolution has implications for the evolution of antibiotic resistance and pathogenicity. 相似文献

Despite its low prevalence, pancreatic cancer (PC) is one of the deadliest, typically characterised as silent in early stages and with a dramatically poor prognosis when in its advanced stages, commonly associated with a high degree of metastasis. Many efforts have been made in pursuing innovative therapeutical approaches, from the search for new cytotoxic drugs and other bioactive compounds, to the development of more targeted approaches, including improved drug delivery devices. Marine biotechnology has been contributing to this quest by providing new chemical leads and materials originating from different organisms. In this review, marine biodiscovery for PC is addressed, particularly regarding marine invertebrates (namely sponges, molluscs, and bryozoans), seaweeds, fungi, and bacteria. In addition, the development of biomaterials based on marine-originating compounds, particularly chitosan, fucoidan, and alginate, for the production of advanced cancer therapies, is also discussed. The key role that drug delivery can play in new cancer treatments is highlighted, as therapeutical outcomes need to be improved to give further hope to patients. 相似文献

Giardia duodenalis is a protozoan parasite that causes a broad range of clinical symptoms varying from none--in asymptomatic carriers--to mild recurring diarrhea consisting of soft, light-colored stools to acute severe diarrhea. In different parts of the world this parasite has raised increased interest due to its possible zoonotic transmission. Among domestic animals, dogs can play an important role in environmental contamination. As there is little information on the frequency of giardiosis in dogs from the Metropolitan Area of Curitiba-State of Paraná, Southern Brazil, the aim of the present work was to evaluate the prevalence of G. duodenalis in two dog populations (household and shelter). To attain the proposed aim, we collected fecal samples from 200 dogs and utilized three diagnostic techniques: Faust's technique (Faust et al. 1939), Benbrook's technique (1963) and polymerase chain reaction (PCR). Faust's technique presented the best results, as it was able to detect a larger number of Giardia cases. Taking Faust's technique as the standard, Benbrook's technique presented 66% sensitivity and PCR demonstrated 69% sensitivity. The shelter dog population showed a 24% occurrence of G. duodenalis while the household population showed a 9% occurrence. Other epidemiological aspects like age, sex, environmental conditions and methodological aspects are discussed in the present article. 相似文献

It is already known by the scientific and industrial communities that lignocellulosic substrates are, to a certain extent,
inhibitors of the hydration reaction of cement. The extent to which and how they influence such reactions is still a matter
of debate. Several techniques, such as calorimetry, i.e., the measurement of the heat evolved or obtaining temperature profiles
during the hydration, the determination of extractive contents of lignocellulosic substrates and their relation with the characteristics
of the hydration curves, or even testing of the mechanical properties of the wood-cement composites, have been used in previous
research. This study complements past research using two techniques that have been used in the analysis of cement hydration
but are not usually applied to lignocellulose-cement mixes, namely X-ray diffraction (XRD) and differential thermal analysis
(DTA). The raw materials for this study were three lignocellulosic materials of Portuguese origin: cork (the bark of Quercus suber L.), blue gum (Eucalyptus globulus Labill.), and maritime pine (Pinus pinaster Ait); and Portland cement. The two techniques allowed tracking of the evolution of the main cement constituents during hydration.
It was found that all the lignocellulosic substrates had detrimental effects on cement hydration. The blue gum exhibited the
fastest hydration kinetics in the initial stage of reaction, but was then overtaken by cork, which at the end gave the highest
hydration conversion amongst the three lignocellulosic substrates. Although pine caused the slowest initial hydration kinetics,
with the passage of time its effect approached that of blue gum. At the end of the hydration period, specimens containing
either species had similar quantities of hydration reaction products. The DTA and XRD results were consistent and are in good
agreement with the temperature profiles and compatibility indexes reported in a previous work. 相似文献
