Repair of injured soft and hard tissues in horses can benefit greatly from the use of regenerative therapies with mesenchymal stem cells (MSC). Vitamin-C and platelet-rich-plasma had been used for in vitro differentiation of MSC. This study was aimed to evaluate the effect of vitamin-C, platelet-rich-plasma and their combination on the in vitro differentiation of adipose horse MSC. We isolated MSC from horse fat and differentiated them in vitro into osteogenic and chondrogenic lineages, as demonstrated by specific staining and RT-qPCR of selected genes. Combining vitamin-C and plasma-rich-platelet positively affected the ability of MSC to differentiate in vitro into mesodermal lineages during 14 days of culture; this effect was not as marked when differentiation was attempted for 21 days. This provides valuable information on the effect of combined use of these molecules in regenerative therapies and their potential application along stem cells for lesions of musculoskeletal tissue in sport horses.  相似文献

Eight helminth taxa were recovered from the necropsy of four stray domestic cats from Colonia Miguelete, county of Colonia, Uruguay. Two of them are recorded for the first time for domestic cats in that country: Alaria alata (Goeze, 1782) from the small intestine (which is also the first trematode species found in domestic cat in Uruguay), and Lagochilascaris major Leiper, 1910 from the pharynx. The remaining helminth species found were Toxocara mystax (Zeder, 1800) and Spirometra sp. from the small intestine, Trichuris serrata (von Linstow, 1879) from the caecum, Eucoleus aerophilus (Creplin, 1839) from the trachea, and Pearsonema feliscati (Diesing, 1851) from the urinary bladder. Moreover, four female specimens of an unidentified Spiruroidea were collected from the stomach and small intestine of one host.  相似文献

Colletotrichum kahawae is an emerging fungal pathogen, which has recently undergone a speciation process from a generalistic ‘C. gloeosporioides species complex' background by acquiring the unique capacity to infect green coffee berries, thus causing coffee berry disease. This is a severe and widespread disease in Africa and an imminent threat to Arabica coffee cultivation in Asia and America, if the pathogen enters those continents. Genetic diversity within C. kahawae is low but notorious differences in pathogen aggressiveness have been described. This work characterized two cytogenomic traits (genome size and minichromosome profiles) of a collection of C. kahawae isolates, representing the breadth of its genetic diversity and distinct aggressiveness classes, along with closely related taxa. The results obtained constitute the first flow cytometry‐based genome size estimation in the genus Colletotrichum and show a c. 8 Mb genome size expansion between C. kahawae (79·5 Mb on average) and its closest relatives (71·3 Mb), corroborating evidence indicating that C. kahawae (i.e. the coffee berry disease pathogens) should remain as a distinct species. Results have also shown the presence of two to five minichromosomes in C. kahawae, suggesting a positive relationship between the number of minichromosomes and the level of aggressiveness of the different isolates analysed, while no correlation could be established between aggressiveness and whole genome size. Overall, these results may be the basis for the identification of pathogenicity/aggressiveness‐related factors in such minichromosomes, and may provide clues to the characterization of specific markers for aggressiveness classes.  相似文献

This study evaluated the effects of Quebracho tannin extract (QTE) on in vitro ruminal fermentation, chemical composition of rumen microbes, ruminal degradation and intestinal digestibility of crude protein (iCPd). Three treatments were tested, the control (basal diet without QTE), the basal diet with 15 g QTE/kg dry matter (DM) and the basal diet with 30 g QTE/kg DM. The basal diet contained (g/kg DM): 339 grass silage, 317 maize silage and 344 concentrate. In vitro gas production kinetic was determined using the Hohenheim gas test (Experiment 1). The Ankom RF technique, a batch system with automatic gas pressure recordings, was used to determine in vitro production of short‐chain fatty acids (SCFA) and ammonia–nitrogen concentration (NH3‐N), as well as nitrogen and purine bases content in liquid‐associated microbes (LAM) and in a residue of undegraded feed and solid‐associated microbes (Feed+SAM) (Experiment 2). Ruminal degradation and iCPd were determined using the nylon bag technique and the mobile nylon bag technique, respectively (Experiment 3). Gas production (Experiment 1), total SCFA and NH3‐N (Experiment 2) decreased with increasing QTE levels. Microbial mass and composition of LAM were not affected by QTE, but total mass of Feed+SAM linearly increased, likely due to decreased substrate degradation with increasing QTE levels. The total amount of N in microbial mass and undegraded feed after the in vitro incubation increased with increasing QTE levels, suggesting a potential greater N flow from the rumen to the duodenum. In contrast to in vivo studies with the same QTE, no effects were detected on ruminal effective degradability and iCPd, when using the nylon bag techniques. Based on the in vitro procedures, QTE increased the supply of N post‐rumen; however, some evidence of a decreased fibre degradation were also observed. Therefore, the benefit of adding QTE to diets of cattle is still questionable.  相似文献

Summary European cattle breeds are being introduced into Ethiopia in an effort to improve the productivity of indigenous breeds. The Ethiopian cattle breeds Horro and Boran were compared for tick burdens with their crosses with Friesian, Jersey and Simmental. Horro animals had the lowest tick burdens and the Horro × Friesian the highest. Adaptation to their environment and long-term natural selection for tick resistance in Horro cattle is the most likely explanation.Repeatability of tick burdens in all animals considered as one herd were only statistically significant forBoophilus decoloratus, the most abundant tick species. Statistically significant correlations between burdens of femaleB. decoloratus, Amblyomma cohaerens andRhipicephalus praetextatus were observed and the addition of the males resulted in all inter-species correlations becoming significant.Care should be taken when crossing Ethiopian with more productive European cattle breeds in order not to lower their tick and disease resistance.
Resistencia En Diferentes Razas De Ganado Vacuno A Garrapatas (ACARI: Ixodidae) En Bako Ethiopia
Resumen Ganado de raza europea estan siendo introducidos en Etiopia en un esfuerzo de mejorar la produccion de las razas indigenas. Las razas etiopes Horro y Boran fueron comparadas por sus cargas de garrapatas con sus cruzas con Friesian, Jersey y Simmental. El ganado Horro mostró las cargas mas bajas y las cruzas con Friesian las mas altas. La adaptacion al medio ambiente y la seleccion a largo plazo de este tipo de ganado es la explicacion mas probable de los resultados.La repitabilidad de las cargas de garrapatas en todos los animales agrupados en un rebaño fueron estadistícamente significativas paraB. decoloratus, la garrapata mas abundante. Correlaciones estadísticamente significativas entre las cargas de hembras deBoophilus decoloratus, Amblyomma cohaerens y Rhipicephalus praetextatus fueron observadas y la adicion de los machos resulto en que todas las correlaciones entre especies se volvieron significativas.Se debe tener cuidado cuando las razas de ganado etiopes se cruzan con las mas productivas razas europeas para no reducir su resistencia a las garrapatas y enfermedades.

Resistance De L'hote Aux Tiques (ACARI: Ixodidae) Chez Differentes Races De Betail A Bako, Ethiopie
Résumé Des races bovines européennes sont actuellement introduites en Éthiopie dans le but d'accroître la productivité des races indigènes. Ainsi, les races Horro et Boran ont été comparées du point de vue du taux d'infestation par les tiques avec les produits de levi croisement avec les races Frisonne, Jersiaise et Simmenthal. Les animaux de race Horro avaient le taux d'infestation le plus bas et le croisement Horro × Frisonne le taux le plus elevé. L'explication la plus vraisemblable de ces résultats est l'adaptation du bétail Horro à son environnement en meme temps qu'une sélection naturelle de longue date à la résistance aux tiques. La répétabilité de la pression ixodienne chez tous les animaux considérés comme formant un troupeau unique, n'a été statistiquement significative que pourBoophilus decoloratus, espèce la plus abondante. Les auteurs ont observé des corrélations statistiquement significatives entre les taux de charge relatifs aux femelles deBoophilus decoloratus, Amblyomma cohaerens etRhipicephalus praetextatus. La prise en compte des mâles a eu pour effet de rendre significative toutes les corrélations inter-espèces. Il faut donc prendre des précautions lors de croisement du bétail éthiopien avec des races européennes plus productives afin de ne pas diminuer leur résistance aux tiques et aux maladies.

This work studies the influence of two mulching treatments on soil properties and the field performance of afforested holm‐oak seedlings (Quercus ilex L. subsp. ballota (Desf.) Samp.) nine years after outplanting. Mulching treatments composed of stones, forest debris, and an untreated control were randomly applied to 180 seedlings (n = 60) in January 2001 at the bed level (1 × 1 m) in an abandoned agricultural field in SE Spain. Survival, growth measured by means of leaf area, and nutrient concentrations in leaves and soil were measured. Both mulches provided higher survival and greater leaf‐area growth than the control, but did not differ in leaf‐nutrient concentration. Most of the analysed soil variables were not affected by the mulching treatments at the end of the study period, and, therefore, the soil properties changes by mulching might be slower than expected under semi‐arid conditions.
