In fish all the pre-requisites to mount a specific immune response are present, but the main differences from the mammalian system are that the secondary response is relatively minor and IgG is not present. In teleosts mainly IgM is present, and IgD has been recently described but its function is, as yet, unknown. However, different forms of fish IgM and its observed flexibility of structure may compensate for a lack of Ig class diversity. The innate immune response of teleosts is highly developed. Multiple forms of key constitutive and inducible components, such as lysozyme, C3, alpha2-macroglobulin and C-reactive protein, are present, and may enhance immune recognition. Low ambient temperature appears to have an impact on all aspects of the immune response, particularly the T-dependent specific immune response due to the non-adaptive lipid composition of T-cell membranes. Temperature effects on the nonspecific immune system are less well characterised, but there is evidence that low temperatures are also suppressive. Knowledge of immune system function becomes essential for disease prevention strategies such as the development of vaccines, selection for increased disease resistance and identification of genes suitable for trangenesis.  相似文献

The objective of the experiment was to determine the optimal dose of copper oxide wire particles (COWPs) to reduce infection of Haemonchus contortus in male lambs. Five to six-month-old hair breed lambs were housed on concrete and fed 450 (L; n = 25) or 675 g (H; n = 25) corn/soybean meal supplement and bermudagrass hay. In July, lambs were inoculated with 10,000 L(3) larvae (97% H. contortus; Day 0). Lambs were administered 0, 2, 4, or 6 g COWP on Day 28. Concentrations of copper in the liver were determined. There were no effects of supplement level on concentrations of copper in the liver and a linear relationship existed between COWP treatment and concentrations of copper in liver (P < 0.001). Least squares means of the 0, 2, 4, 6 g COWP treatments were 62.2, 135.7, 161.1, and 208.4 ppm (P < 0.001). Between Days 0 and 28, PCV declined and by Day 42, PCV of all COWP-treated lambs was markedly higher than control lambs and remained higher (COWP x day, P < 0.05). By Day 21, PCV was greater in the H compared with the L group of lambs (P < 0.001). Within 14 days of COWP treatment FEC declined from more than 8000 eggs/g (epg) to less than 250 epg in all COWP-treated lambs (COWP x day, P < 0.001). The numbers of H. contortus in the abomasum were greatly reduced in all COWP-treated groups of lambs and remaining nematodes were predominantly males. FEC were greater in L versus H supplemented lambs and values decreased to a greater extent in H lambs when treated with COWP (supplement x COWP; P < 0.02). The 2 g COWP was effective in alleviating H. contortus infection and reducing number of egg-laying nematodes in the abomasum with the lowest concentration of copper in the liver of the COWP treatment groups. PCV values were more favorable for lambs fed the higher level of supplement, especially when FEC were greater than 8000 epg.  相似文献

We tested the hypothesis that a small dose of estradiol benzoate (EB) at the midstage of the estrous cycle in cattle would synchronize the subsequent pattern of ovarian follicular development, estrus, and ovulation. Nonlactating Friesian cows received either 1 mg of EB i.m. on d 13 of the estrous cycle (T; n = 12; estrus = d0) or served as untreated controls (C; n = 12). Their ovaries were examined daily with transrectal ultrasonography from d 7, and blood samples were collected 0, 2, 4, 8, 24, and 48 h after treatment on d 13. Plasma concentrations of estradiol-17beta were elevated to 12 pg/mL during the initial 24 h following treatment, compared with a baseline of 1 pg/mL in untreated controls (P < .001). Progesterone concentrations in cows of the T group declined between 24 and 48 h after treatment (-3.2 +/- .5 ng/mL) compared with little change in concentrations of progesterone in cows of the C group at this time (P < .01). This difference was coincident with an earlier time to regression of the corpus luteum in cows of the T group. Disregarding treatment groups, the second dominant follicle of the estrous cycle (DF2) emerged on d 10.6 +/- .3 and was 9.4 +/- .4 mm in diameter on d 13. Further growth of the DF2 was halted by EB treatment on d 13. Cessation of growth occurred irrespective of whether the DF2 was in the early or late growth phase, and a new follicular wave emerged 4.5 +/- .2 d later. The dominant follicle from this wave (DF3) ovulated 5 d after emergence in most cases. During the estrous cycle of every cow in the T group, there were three waves of follicular development (3-wave), whereas the ratio of 2:3 waves of follicular development in cows of the C group was 1:3. Consequently, the interval from emergence to ovulation of the ovulatory dominant follicle in cows of the C group ranged from 3 to 11 d. The dynamics of ovarian follicular wave development during the estrous cycle can be strategically manipulated by treating with a small dose of EB to synchronize proestrous development of the ovulatory follicle.  相似文献

Two experiments were performed to evaluate the efficacy of a progestin-based estrus synchronization program that incorporated the use of estradiol at the initiation of progestin treatment and at 48 h after progestin withdrawal (Exp. 2). In Exp. 1, cyclic, lactating dairy cows (n = 112) were assigned to receive either 1 (1mg) or 2 (2mg) mg of estradiol benzoate via an i.m. injection on d -9 (d 0 = initiation of the breeding season). All cows received an intravaginal progesterone-releasing insert (IPI; CIDR-B) on d -9. On d -2, the IPI was withdrawn and all cows were administered 500 microg of cloprostenol sodium. Beginning on d 0, cows were bred by AI upon detection of estrus. Estrus was observed in similar proportions of cows in each treatment during the first 6 d of AI (90% across treatments), but the interval to estrus was shorter (P < .05) in 1mg (1.26 +/- .18 d) than in 2mg (1.77 +/- .18 d). Conception and pregnancy rates did not vary among treatments; however, cows in estrus on d 0 tended to be less fertile (P = .11) than those in estrus on d 1. In Exp. 2, 408 cyclic cows from three herds were assigned to receive either no synchrony treatment (Control, n = 214) or the treatments described in Exp. 1 (1mg, n = 100; 2mg, n = 94). Anestrous cows from all herds received an IPI from d -9 to -2 (n = 143; Anestrus). All cows in the 1mg, 2mg, and Anestrus groups, with the exception of those detected in estrus between d -1 and 0, also received 1 mg of estradiol benzoate on d 0. Greater than 90% of cows that received an IPI were in estrus between d -1 and 3, and 92.1% of cows in the Control group were in estrus by d 21. Conception rate to first service in 2mg (61.7%) was similar to Control (57.0%), tended to be higher (P = .06) than 1mg (49.0%), and was greater (P < .05) than Anestrus (39.9%). The mean day of conception was earlier (P < .05) in the 2mg (d 13.1 +/- 2.0) than the Control (d 23.2 +/- 1.6) and Anestrus (d 22.4 +/- 1.9) groups. Conception occurred earlier in 1mg (d 17.4 +/- 2.1) than in Control. The proportion of cows that were pregnant at the end of the breeding season tended (P = .09) to be greater in the 2mg and Anestrus groups. This regimen of estrus synchronization improved reproductive competence in cyclic cows and resulted in similar reproductive performance in anestrous cows and untreated cyclic cows inseminated at a spontaneous estrus.  相似文献

An antigen-capture, monoclonal-antibody-based enzyme-linked immunoassay (ELISA) that detects a broad range of Salmonella serovars in various serogroups was developed and compared with standard culture procedures for detection of Salmonella in 1055 field samples collected from poultry-hatchery environments. The diagnostic sensitivity of the ELISA relative to culture was 99.9% and the diagnostic specificity 99.6%. The extensive culture procedure included nonselective enrichment (NSE) as well as primary selective enrichment (PSE) and delayed secondary enrichment (DSE) with Hajna tetrathionate (TT) and Rappaport–Vassiliadis (RV) selective-enrichment broths. Significantly more Salmonella-positive samples were detected by ELISA and culture at the DSE stage than at the NSE and PSE stages (P < 0.05). Significantly more RV than TT broths were positive for Salmonella by culture and ELISA by the DSE stage (P < 0.05). This ELISA procedure could be a reliable screening test for the detection of Salmonella in hatchery samples.  相似文献

A full 12-hour synthesis at 6-centimeter wavelength with the Very Large Array confirms the major features previously reported for the double quasar 0957+561. In addition, the existence of radio jets apparently associated with both quasars is demonstrated. Gravitational lens models are now favored on the basis of recent optical observations, and the radio jets place severe constraints on such models. Further radio observations of the double quasar are needed to establish the expected relative time delay in variations between the images.  相似文献

An imaging photopolarimeter aboard Pioneer 11, including a 2.5-centimeter telescope, was used for 2 weeks continuously in August and September 1979 for imaging, photometry, and polarimetry observations of Saturn, its rings, and Titan. A new ring of optical depth < 2 x 10(-3) was discovered at 2.33 Saturn radii and is provisionally named the F ring; it is separated from the A ring by the provisionally named Pioneer division. A division between the B and C rings, a gap near the center of the Cassini division, and detail in the A, B, and C rings have been seen; the nomenclature of divisions and gaps is redefined. The width of the Encke gap is 876 +/- 35 kilometers. The intensity profile and colors are given for the light transmitted by the rings. A mean particle size less, similar 15 meters is indicated; this estimate is model-dependent. The D ring was not seen in any viewing geometry and its existence is doubtful. A satellite, 1979 S 1, was found at 2.53 +/- 0.01 Saturn radii; the same object was observed approximately 16 hours later by other experiments on Pioneer 11. The equatorial radius of Saturn is 60,000 +/- 500 kilometers, and the ratio of the polar to the equatorial radius is 0.912 +/- 0.006. A sample of polarimetric data is compared with models of the vertical structure of Saturn's atmosphere. The variation of the polarization from the center of the disk to the limb in blue light at 88 degrees phase indicates that the density of cloud particles decreases as a function of altitude with a scale height about one-fourth that of the gas. The pressure level at which an optical depth of 1 is reached in the clouds depends on the single-scattering polarizing properties of the clouds; a value similar to that found for the Jovian clouds yields an optical depth of 1 at about 750 millibars.  相似文献

The ability to clone large fragments of DNA in yeast artificial chromosomes (YAC's) has created the possibility of obtaining global physical maps of complex genomes. For this application to be feasible, most sequences in complex genomes must be able to be cloned in YAC's, and most clones must be genetically stable and colinear with the genomic sequences from which they originated (that is, not liable to undergo rearrangement). These requirements have been met with a YAC library containing DNA fragments from Drosophila melanogaster ranging in size up to several hundred kilobase pairs. Preliminary characterization of the Drosophila YAC library was carried out by in situ hybridization of random clones and analysis of clones containing known sequences. The results suggest that most euchromatic sequences can be cloned. The library also contains clones in which the inserted DNA is derived from the centromeric heterochromatin. The locations of 58 clones collectively representing about 8 percent of the euchromatic genome are presented.  相似文献

The first crystalline preparation of isocitrate dehydrogenase specific for nicotinamide adenine dinucleotide phosphate has been obtained with enzyme isolated from Escherichia coli.
