In a 40 year old pine stand, soil solution was collected continuously with suction cups from 50 and 200 cm depth over a 42-month period following the surface application of lime and K/Mg-fertilizer. K and Mg showed a much higher mobility than Ca, which hardly increased in 200 cm depth even after 42 months. After an initial nitrate peak, concentrations decreased but remained elevated throughout the entire study period. In 200 cm, Al-, Mn, and Cd-concentrations increased due to exchange processes in the top-soil. Continuously elevated sulfate concentrations in 200 cm indicate S-mineralisation from humus decomposition.  相似文献

The sphingolipid composition of food as well as of physiological samples has received considerable interest due to their positive biological activities. This study quantified the total amount of sphingomyelin (SM) in 20 human breast milk samples from healthy volunteers and determined the structures of SM by detailed mass spectrometric studies in combination with enzymatic cleavage. The quantification of SM was performed by hydrophilic interaction liquid chromatography coupled to electrospray ionization-tandem mass spectrometry (HILIC-HPLC-ESI-MS/MS) measuring the characteristic fragment ion of the phosphorylcholine group at m/z 184.2 and by using hexanoylsphingomyelin (C6-SM) and heptadecanoylsphingomyelin (C17-SM) as internal standards. The structures of SM species were identified after enzymatic cleavage with alkaline sphingomyelinase (SMase) to the corresponding ceramides. Structure elucidation of the sphingoid base and fatty acid backbone was performed by reversed-phase HPLC-ESI-MS/MS. The method includes the sphingoid bases dihydrosphingosine (d18:0), sphingosine (d18:1(Δ4)), 4,8-sphingadienine (d18:2(Δ4,8)), 4-hydroxysphinganine (phytosphingosine (t18:0)), and 4-hydroxy-8-sphingenine (t18:1(Δ8)) and fatty acids with even-numbered carbon atoms (C12-C26) as well as their (poly)unsaturated and monohydroxylated analogues. The total amount of SM in human breast milk varied from 3.87 to 9.07 mg/100 g fresh weight. Sphingosine (d18:1) was the predominant sphingoid base, with 83.6 ± 3.5% in human breast milk, followed by 4,8-sphingadienine (d18:2) (7.2 ± 1.9%) and 4-hydroxysphinganine (t18:0) (5.7 ± 0.7%). The main SM species contained sphingosine and palmitic acid (14.9 ± 2.2%), stearic acid (12.7 ± 1.5%), docosanoic acid (16.2 ± 3.6%), and tetracosenoic acid (15.0 ± 3.1%). Interestingly, the fatty acid composition of SM species in this study differs from the total fatty acids in human breast milk, and the fatty acids are not consistently distributed among the different sphingoid bases.  相似文献

The maintenance of plant genetic resources in living plant collections (genebanks) causes costs due to employment of staff, usage of buildings, equipment and consumables. Since this is especially challenging in vegetatively propagated material, studies were performed for the case of garlic, which is one of the major vegetatively maintained crops in the genebank of IPK Gatersleben. Data were recorded to compare various scenarios of the main strategies field maintenance and cryopreservation. A spreadsheet tool was developed to be used for cost assessment and for drawing conclusions concerning the most effective way of maintenance. Field culture is cheaper in the short term, whereas after a break-even point cryopreservation becomes the more efficient storage method in the long term. This break-even point depends on the particular scenario, which is determined by various factors such as field and in vitro multiplication rates of various genotypes, presence of bulbils in a part of the genepool, the sample size of the accessions as well as the number of stored accessions in cryopreservation. The comparative discussion is exemplified for a 1-year field rotation versus cryopreservation using either in vitro plantlets or a combination of bulbils and unripe inflorescence bases as organ sources. For the more expensive use of in vitro plants cryopreservation becomes less costly than field culture only after 13 years, whereas this is the case already after 8–9 years when using a combination of bulbils in winter and inflorescence bases in summer.  相似文献

Lygeum spartum, Zygophyllum fabago and Piptatherum miliaceum are typical plant species that grow in mine tailings in semiarid Mediterranean areas. The aim of this work was to investigate metal uptake of these species growing on neutral mine tailings under controlled conditions and their response to fertilizer additions. A neutral mine tailing (pH of soil solution of 7.1–7.2) with high total metal concentrations (9,100 and 5,200 mg kg?1 Zn and Pb, respectively) from Southern Spain was used. Soluble Zn and Pb were low (0.5 and <0.1 mg l?1, respectively) but the major cations and anions reached relatively high levels (e.g. 2,600 and 1,400 mg l?1 Cl and Na). Fertilization caused a significant increase of the plant weight for the three species and decreased metal accumulation with the exception of Cd. Roots accumulated much higher metal concentrations for the three plants than shoots, except Cd in L. spartum. Shoot concentrations for the three plants were 3–14 mg kg?1 Cd, 150–300 mg kg?1 Zn, 4–11 mg kg?1 Cu, and 1–10 mg kg?1 As, and 6–110 mg kg?1 Pb. The results indicate that neutral pH mine tailings present a suitable substrate for establishment of these native plants species and fertilizer favors this establishment. Metal accumulation in plants is relatively low despite high total soil concentrations.  相似文献

It is well known that the addition of easily available substrates to soils can affect microbial activity and thus the mineralization of soil organic carbon (SOC). Up to now, little is known about the processes leading to these priming effects and which fractions of organic matter (OM) are affected. The objectives of this study were to determine if SOC associated with isolated soil size fractions showed different susceptibility to priming effects, whether these pools are easily depleted, or whether the amount of substrate addition affects the extent of priming effects. In an incubation experiment, the effect of the uniformly 14C‐labeled substrates fructose and alanine on the mineralization of the SOC of a Bs horizon of a Haplic Podzol was investigated. The soil sample was fractionated into the three soil size fractions sand, silt, and clay by a mild sonication followed by sieving and sedimentation. Additionally, nonfractionated soil of the horizon was included in the experiment. Every soil sample received four substrate additions repeated at weekly intervals with 3.325 μg substrate‐C (mg SOC)–1 and a final addition of 13.3 μg substrate‐C (mg SOC)–1 after 4 weeks. The respiration was determined hourly and 14CO2 was analyzed every 2, 4, and 7 d after the respective substrate addition. After 56 d, between 42% and 58% of the added substrates had been mineralized. Both substrates strongly increased the mineralization of the OM in all fractions (positive priming effects). The priming effects were always higher after the addition of the high substrate dose than during the first 4 weeks when four small doses were added. In general, the priming effects increased with decreasing particle size. Alanine generally caused higher priming effects than fructose in the soil size fractions (up to 280% vs. 231%, respectively). This indicates that alanine serves not only as an energy substrate but also as a N source and, thus, also promotes microbial growth. The strong priming effects in the silt and clay fraction (133% and 125% with fructose, 172% and 168% with alanine) showed, that not only the labile pool of OM is affected, but also a more stable pool characterized by higher 14C ages. We assume that the stability of the OM in these fractions is not only due to recalcitrance or to interactions with the minerals, but that it may also be caused by a substrate limitation of the degrading microorganisms.  相似文献

Applications of polyphosphate‐based fertilizers have been reported to have a positive impact on crop yields as compared to orthophosphate sources. Since plants take up P mainly as orthophosphate, hydrolysis rates of polyphosphates into orthophosphates will determine their fertilizer ability. Laboratory and soil incubation experiments were performed to evaluate hydrolysis rates of pyrophosphate (PP), tripolyphosphate (TP), and trimetaphosphate (TMP) in water as well as in two soils having different P‐fixing capacities. P availability was characterized by measuring the orthophosphate (ortho‐P) and polyphosphate (poly‐P) concentration in soil solution as well as the calcium‐acetate‐lactate (CAL)‐extractable amounts of both forms. In water, PP was completely hydrolyzed within 15 d, whereas TMP was hydrolyzed only to about 30% after 90 d. In the two soils, polyphosphates hydrolyzed during the incubation period increasing ortho‐P concentration in soil solution as well as in CAL extract. At the end of the incubation, no significant differences in ortho‐P concentration in soil solution and CAL extract were found in the sandy soil, whereas in the silty‐loam soil, polyphosphate applications resulted in higher soil‐solution ortho‐P concentration. Although polyphosphate hydrolysis is mainly affected by the soil‐specific enzymatic activity, it seems that polyphosphates and/or hydrolysis products are preferentially adsorbed/precipitated compared to ortho‐P in the silty loam, thereby influencing the P availability from polyphosphate sources.  相似文献

Information about disease management in winter wheat (Triticum aestiva) in eight European countries was collated and analysed by scientists and extension workers within the European Network for the Durable Exploitation of Crop Protection Strategies (ENDURE). This included information about specific disease thresholds, decision support systems, host varieties, disease prevalence and pathogen virulence. Major differences in disease prevalence and economic importance were observed. Septoria tritici blotch (Mycosphaerella graminicola) was recognized as the most yield reducing disease in countries with intensive wheat production, but also rust diseases (Puccinia striiformis and Puccinia triticina), powdery mildew (Blumeria graminis) and Fusarium head blight (Fusarium spp.) were seen as serious disease problems. Examples of current integrated pest management (IPM) strategies in different countries have been reported. Disease management and fungicide use patterns showed major differences, with an average input equivalent to 2.3 full dose rates (TFI) in the UK and a TFI of 0.6 in Denmark. These differences are most likely due to a combination of different cropping systems, climatic differences, disease prevalence, and socio-economic factors. The web based information platform www.eurowheat.org was used for dissemination of information and results including information on control thresholds, cultural practices which can influence disease attack, fungicide efficacy, fungicide resistance, and pathogen virulence, which are all elements supporting 1PM for disease control in wheat. The platform is open to all users. The target groups of EuroWheat information are researchers, advisors, breeders, and similar partners dealing with disease management in wheat.  相似文献

The Neoproterozoic glaciations supposedly ended in a supergreenhouse environment, which led to rapid melting of the ice cover and precipitation of the so-called cap carbonates. If Earth was covered with ice, then extraterrestrial material would have accumulated on and within the ice and precipitated during rapid melting at the end of the glaciation. We found iridium (Ir) anomalies at the base of cap carbonates in three drill cores from the Eastern Congo craton. Our data confirm the presence of extended global Neoproterozoic glaciations and indicate that the duration of the Marinoan glacial episode was at least 3 million, and most likely 12 million, years.  相似文献
