The objective of this study was to evaluate the effect of dietary supplementation of whole flaxseed on sperm traits and sperm fatty acid profile in aged broiler breeder roosters. Twelve Ross 308 broiler breeder roosters (age: 52 weeks; weight: 4,900 ± 210 g) haphazardly allotted to three dietary treatments (each treatment contained four replicates and one bird in each replicate) for six weeks. Treatments were different levels of flaxseed (0% flaxseed [GFL0], 2% flaxseed [GFL2] and 4% flaxseed [GFL4]). The feed intake quadratically decreased (p < .05) with increasing whole flaxseed levels for the period (58 to 60 weeks). Sperm traits (semen volume and sperm concentration, sperm total and forward motility, sperm viability and morphology, sperm plasma membrane functionality) were evaluated every two weeks (four times), and sperm fatty acid profile was assessed at the end of the experiment. Semen volume, sperm concentration and sperm morphology were not affected by treatments. On week 60, GFL2 group showed a significantly lower percentage of total and progressive sperm motility and sperm membrane functionality in comparison with the control and GFL4 groups. Also, sperm viability was lower in GFL2 group compared with other groups on week 58 (p < .05). In terms of sperm fatty acid profile, GFL2 group significantly reduced the percentage of linoleic acid (C18:2 [n-6]) in comparison with other groups. However, any of the other fatty acids were not affected by dietary flaxseed. In conclusion, dietary supplementation of whole flaxseed could not improve the quality of aged broiler breeder roosters' sperm in this study, nor it could alter the sperm fatty acid profile; thus, it seems necessary to use some antioxidants such as vitamin E in the diet of aged broiler breeder roosters, when supplementing the diets with oils or oilseeds such as flaxseed.  相似文献

Background: Friedreich ataxia (FRDA) is an autosomal recessive disorder caused by guanine-adenine-adenine (GAA) triplet expansions in the FXN gene. Its product, frataxin, which severely reduces in FRDA patients, leads to oxidative damage in mitochondria. The purpose of this study was to evaluate the triple nucleotide repeated expansions in Iranian FRDA patients and to elucidate distinguishable FRDA clinical differences in these patients. Methods: A number of 22 Iranian patients (8 females and 14 males) from 16 unrelated families were studied. DNA was extracted from the peripheral blood of patients. The frequency and length of (GAA)n repeats in intron 1 of the FXN gene were analyzed using long-range PCR. In this study, the clinical criteria of FRDA in our patients and the variability in their clinical signs were also demonstrated. Results: An inverse relationship was observed between GAA repeat size and the age of onset. Although some distinguishable clinical features (such as limb ataxia and lower limb areflexia) were found in our patients, 90-95% of them had extensor plantar response and dysarthria. The results showed only one positive diabetes patient and also different effects on eye movement abnormality among our patients. Conclusion: The onset age of symptoms showed a significant inverse correlation with allele size in our patients (P>0.05). Based on comparisons of the clinical data of all patients, clinical presentation of FRDA in Iranian patients did not differ significantly from other FRDA patients previously reported. Corn (Zea mays L.) is planted in two seasons per year in northern Iran (mid-April as a main crop and mid-June as a second crop). The main objective of this study was to determine whether corn yield response would differ between these two seasons and different plant populations. Two field experiments were conducted at the Agricultural Research Center of Golestan – Iran in 2007 and 2008 at different planting densities. The results showed that the values of grain yield and most traits were significantly lower in the second season. Maximum grain yield was observed at planting densities of 6.5 plants m?2 in the first season, whereas in the second season grain yield was the same for planting densities between 2.5 and 12.5 plants m?2. Based on the second-year experimental results, the following functions were fitted to show the relationship between yield ha?1 (Y) and planting densities (X) for the first and second seasons, respectively: (Y = ?167.6X 2 + 2672.2X + 511.77; R 2 = 0.992) and (Y = 1200.1 ln(X) + 2924.4; R 2 = 0.935). This study found that the optimum plant population was 6.5 plants m?2 under low heat stress, and should be reduced to 2.5–4.5 plants m?2 under heat stress conditions.  相似文献

A 2-year field experiment (2012–2013) was conducted to evaluate the yield and water use efficiency (WUE) response of maize (Zea mays L.) to different soil water managements at different sowing dates. The experiment included three sowing dates (22 June, 6 July and 21 July) and four irrigation regimes based on maximum allowable depletion (MAD) of the total available soil water (TAW). The irrigation treatments were marked by I1 to I3 as 40%, 60% and 80% MAD of TAW, respectively, and with no irrigation. The results showed that grain yield reduced when planting was delayed in both years, ranging from 6105 to 4577 kg ha?1 in 2012 and from 7079 to 5380 kg ha?1 in 2013. However, WUE increased when planting was delayed from 22 June until 21 July. Also the highest grain yield was observed in the first irrigation treatment (MAD = 40%) in both years, and the highest WUE was obtained in the second irrigation treatment (MAD = 60%) with 1.64 and 1.61 (kg m?3) in 2012 and 2013, respectively. These findings suggest that delay in planting date and the use of MAD = 60% treatment in Mediterranean-type region such as Golestan, Iran, can be useful in saving water that is highly important in such regions.  相似文献

1. Embryos of the domestic fowl have been partially sterilised by injecting the drug busulphan into 24‐h incubated eggs.

2. Some of these embryos were injected with primordial germ cells (PGCs) after 55 h of incubation to attempt to repopulate the gonads.

3. Primordial germ cells transfected with a defective retrovirus containing the reporter gene lac Z were shown to settle in these sterilised gonads.

4. Quantitative histology of 6‐d embryos showed that busulphan produced 75% sterilisation but that PGCs could repopulate these gonads.

To determine the effect of different dietary Met sources on oxidative status, male Cobb 500 broiler chickens were fed from day of hatch to 26 days of age (d26) a diet deficient in sulfur amino acids (control) or a diet containing 0.22% DL‐Met, 0.22% L‐Met or 0.31% Met precursor, DL‐2‐hydroxy‐4‐(methylthio) butanoic acid (DL‐HMTBA) to meet the Met + Cys requirements. Liver, breast muscle, duodenum, jejunum, and ileum were collected at day 10 (d10) and d26 to assay markers of oxidative stress, including total glutathione (TGSH), oxidized glutathione (GSSG), reduced glutathione (rGSH), protein carbonyls, thiobarbituric acid reactive substances (TBARS) and ferric reducing/antioxidant power (FRAP). In breast muscle, TGSH and rGSH were greater in L‐Met and DL‐HMTBA groups than in the control group (p < 0.05). An interaction of treatment and age was observed for TGSH in ileum (p = 0.01) and jejunum (p = 0.01), for GSSG in jejunum (p < 0.001), and for rGSH in ileum (p = 0.02). The ratios of rGSH to GSSG and GSSG to TGSH, which define oxidative status, were not affected by Met source. Protein carbonyls varied among groups in jejunum (p = 0.05) and breast muscle (p < 0.001), but were in the normal physiological range. No difference among treatment groups was observed for TBARS and FRAP in different tissues. Age effects were observed in all tissues for multiple oxidative stress markers. In conclusion, consuming different sources of supplementary Met did not alter the oxidative status in several tissues of broilers. Met + Cys deficiency did not compromise antioxidant capacity of chickens although growth was retarded.  相似文献

With climate change posing a serious threat to food security, there has been an increased interest in simulating its impact on cropping systems. Crop models are useful tools to evaluate strategies for adaptation to future climate; however, the simulation process may be infeasible when dealing with a large number of G × E × M combinations. We proposed that the number of simulations could significantly be reduced by clustering weather data and detecting major weather patterns. Using 5, 10 and 15 clusters (i.e., years representative of each weather pattern), we simulated phenology, cumulative transpiration, heat-shock-induced yield loss (heat loss) and grain yield of four Australian cultivars across the Australian wheatbelt over a 30-year period under current and future climates. A strong correlation (r2≈1) between the proposed method and the benchmark (i.e., simulation of all 30 years without clustering) for phenology suggested that average duration of crop growth phases could be predicted with substantially fewer simulations as accurately as when simulating all 30 seasons. With mean absolute error of 0.64 days for phenology when only five clusters were identified, this method had a deviation considerably lower than the reported deviations of calibrated crop models. Although the proposed method showed higher deviations for traits highly sensitive to temporal climatic variability such as cumulative transpiration, heat loss and grain yield when five clusters were used, significantly strong correlations were achieved when 10 or 15 clusters were identified. Furthermore, this method was highly accurate in reproducing site-level impact of climate change. Less than 7% of site × general circulation model (GCM) combinations (zero for phenology) showed incorrect predication of the direction (+/−) of climate change impact when only five clusters were identified while the accuracy further increased at the regional level and with more clusters. The proposed method proved promising in predicting selected traits of wheat crops and can reduce number of simulations required to predict crop responses to climate/management scenarios in model-aided ideotyping and climate impact studies.  相似文献

Purpose

Many closed municipal solid waste landfill sites are near urban areas and there are high expectations to improve geotechnical properties of these sites for re-development. Construction on closed landfill sites is generally a challenging task due to complex behaviour of creep, settlement, high amount of moisture content and weak shear strength of waste. This study presents the experimental results for the use of fly ash and quicklime in improvement of the geotechnical properties of municipal solid wastes (MSW).

Materials and methods

The waste materials were collected from a closed landfill in the southwest of Sydney. The samples were prepared by integrating MSW with a mixture of fly ash–quicklime with a ratio of 3:1 in percentages of 5, 10, 15 and 20 of fly ash by dry weight of the MSW. An array of experimental tests has been conducted on treated and untreated MSW samples including sieve analysis, Atterberg limits, compaction and consolidated–drained triaxial tests.

Results and discussion

Results of this investigation showed a significant improvement in geotechnical properties of MSW. It has been found that by increasing fly ash–quicklime admixtures from 0 to 26.7 % (0 to 20 % fly ash), the internal friction angle increased from 29 to 39° and the cohesion intercept increased from 11 to 30 kPa. Under an effective confining pressure of 300 kPa, the peak strength, the brittleness index and the Young's modulus at failure increased from 600 to 1,150 kPa, 0.13 to 0.35 and 5.5 MPa to 28 MPa, by addition of 26.7 % fly ash–quicklime admixture, respectively. Moreover, the compression and the secondary compression indices decreased from 0.33 to 0.23 and 0.052 to 0.033, respectively.

Conclusions

It is found that the chemical stabilisation effectively increases the compressive strength, the shear strength parameters, the stiffness and the brittleness index whilst decreases the settlement of the MSW layer. It will be beneficial and effective in re-development of closed landfill sites incorporating chemical treatments. The findings of this study may facilitate the calculations of the bearing capacity and settlement as well as the slope stability analysis of chemically treated closed landfill sites.  相似文献

In order to study the effects of desert dust on yield and yield components of cowpea, two field experiments were conducted in the factorial layout based on randomized complete block design with three replicates in Dezful and Mashhad, Iran in 2015. The experimental treatments were combinations of desert dust in three levels (0, 500 and 1500 µg m?3), numbers of desert dust application (once, twice and thrice) and two types of desert dust. The desert-dust particles were collected during dust storms occurred in Dezful and Zabol, which are the main sources of dust in Iran. The results showed that the biological yield and the grain yield were significantly decreased in both studied sites (Dezful and Mashhad) when the plants exposed to 500 µg m?3 desert dust treatment compared to normal conditions. Total soluble sugar and proline contents in plant leaf tissues increased significantly when they affected by 1500 µg m?3 of desert dust, while the plant height was significantly decreased by increasing the desert-dust concentrations at both sites. Biological yield, grain yield, and 100-seed weight were also significantly affected by the desert dust. Overall, the desert dust has adverse effects on yield of cowpea.  相似文献

Background:

Bax and Bcl-2 are the major members of Bcl-2 family whose play a key role in tumor progression or inhibition of intrinsic apoptotic pathway triggered by mitochondrial dysfunction. Therefore, the balance between pro- and anti-apoptotic members of this family can determine the cellular fate.

Methods:

In this study, the relative level of mRNA expression of Bax and Bcl-2 genes was determined using RNA extraction, cDNA synthesis and RT-qPCR technique from 22 tumoral tissues and adjacent non-tumoral tissues from adenocarcinoma colorectal cancer.

Results:

The potential prognostic and predictive significance of Bax and Bcl-2 gene expression and Bax/Bcl-2 ratio were demonstrated in colorectal cancer. The significant correlation between qPCR data and different clinicopathologic parameters of colorectal carcinoma, including age, gender, tumor size, tumor stage, tumor location, and tumor differentiation was also examined. Interestingly, no significant correlation was seen between Bax and Bcl-2 expressions and clinicopathological parameters of colorectal cancer. However, Bax/Bcl-2 ratio was statistically correlated with age and tumor location. Patients with age above 50 showed decreased levels of Bax/Bcl-2 ratio. Moreover, the Bax/Bcl-2 ratio was significantly lower in tumors resected from colon compared to sigmoid colon, rectosigmoid and rectum tumors.

Conclusion:

This study indicates a significant correlation between age and tumor location with Bax/Bcl-2 expression ratio, suggesting predictive value as a potential molecular marker of colorectal cancer.Key Words: Colorectal cancer, Bax/Bcl-2 ratio, Bax expression, Bcl-2 expression  相似文献
