Present acid forming emissions to the atmosphere have the potential to alter significantly the chemistry of rain, snow, and surface water of weakly buffered lakes in the Upper Midwest. Average precipitation pH from field measurements during 1979–1983 declined from west to east from 4.8, 4.6, and 4.3 along a cross-section of sites in Minnesota, Wisconsin, and Michigan respectively where 990 lake and stream sampling sites were studied. Measurements of weakly buffered lakes show a parallel decline in lake water pH with the lowest values measured, 5.1, 4.6 and 4.4, respectively in the same regions. Correspondingly, the percentage of lakes sampled with little or no acid neutralizing capacity (ANC) was found to increase from 0 to 4 and 13%, respectively. The geographic patterns in ionic composition of airborne acids and bases, and the resultant surface water concentrations are compared. The acid forming capacity (AFC) from airborne inputs is calculated using mass balance and in-lake processes. Stoichiometric acid-base reactions are used to balance the observed chemical differences between airborne inputs and surface water composition considering nitrification, denitrification, other oxidation-reduction reactions, and the evaporation concentration process. Microbial activity in surface water can result in a net decrease in ionic strength from the conversion of most of the ammonium and nitrate to neutral compounds and biomass, but only a partial reduction of about 20% of the sulfate inputs to weakly buffered lakes. The resulting AFC of airborne inputs are calculated to range from 30 to 60, 50 to 90, and 80 to 130 μeq H+L-l, respectively, in northeastern Minnesota-Ontario, northcentral Wisconsin and northern Michigan-Ontario. Short‐rotation forestry (SRF) on arable soils has high potentials for biomass production and leads to long‐term no‐tillage management. In the present study, the vertical distributions of soil chemical and microbial properties after 15 y of SRF with willows and poplar (Salix and Populus spp.) in 3‐ and 6‐year rotations on an arable soil were measured and compared to a pertinent tilled arable site. Two transects at different positions in the relief (upper and lower slope; transect 1 and 2) were investigated. Short‐rotation forestry caused significant changes in the vertical distribution of all investigated soil properties (organic and microbial C, total and microbial N, soil enzyme activities), however, the dimension and location (horizons) of significant effects varied. The rotation periods affected the vertical distribution of the soil properties within the SRF significantly. In transect 1, SRF had higher organic‐C concentrations in the subsoil (Bv horizon), whereas in transect 2, the organic‐C concentrations were increased predominantly in the topsoil (Ah horizon). Sufficient plant supply of P and K in combination with decreased concentrations of these elements in the subsoil under SRF pointed to an effective nutrient mobilization and transfer from the deeper soil horizons even in the long term. In transect 1, the microbial‐C concentrations were higher in the B and C horizons and in transect 2 in the A horizons under SRF than under arable use. The activities of β‐glucosidases and acid phosphatases in the soil were predominantly lower under SRF than under arable use in the topsoil and subsoil. We conclude, that long‐term SRF on arable sites can contribute to increased C sequestration and changes in the vertical distribution of soil microbial biomass and soil enzyme activities in the topsoil and also in the subsoil. 相似文献

We have shown previously that riboflavin is stored in chloragosomes of chloragocyte-derived earthworm eleocytes what may have an adaptive value for worms vulnerable to soil-derived pathogen invasion. Thus, it is important to determine whether riboflavin stores are also present in earthworm species with low eleocyte counts, especially in the chloragocytes of their chloragogen tissue that surrounds the coelomic site of the intestine, and find out its putative roles in earthworm immunity. Experiments were performed on adult worms from 6 species. Freely floating coelomocytes (consisting of amoebocytes plus species-specific numbers of eleocytes) were extruded by mild electric shock and then chloragocytes from the same anaesthetized worm were mechanically detached into buffer. Both cell suspensions were analyzed by spectrofluorometry for riboflavin content. It turned out that riboflavin is stored in species-specific quantities in all investigated species. Riboflavin storage predominates in free coelomocytes of eleocyte-rich species (Eisenia andrei, Dendrobaena veneta, Allolobophora chlorotica) while in chloragocytes of species with few eleocytes (Lumbricus terrestris, L. rubellus, and Aporrectodea caliginosa). Upon a massive microbial impact, the coelomocytes (both amoebocytes and eleocytes) are involved in the formation of multicellular bodies encapsulating soil-derived pathogens, what is connected with enhanced coelomocyte mobility. Thus the second aim of the present investigation was to check if riboflavin can participate in coelomocyte accumulations. Tests performed in a 48-well chemotaxis chamber revealed that riboflavin behave as chemoattractant for coelomocytes of all investigated earthworm species, which may be one of mechanisms underpinning its immunomodulatory functions. 相似文献

Antioxidant capacity of hydrophilic and lipophilic extracts from eight broccoli genotypes was compared using the oxygen radical absorbance capacity (ORAC) assay. Each genotype was analyzed for carotenoid, tocopherol, ascorbic acid, and flavonoid content. Results indicate that the antioxidant capacity of hydrophilic extracts ranged from 65.8 to 121.6 micromol trolox equivalents (TE)/g of tissue, and the capacity of lipophilic extracts ranged from 3.9 to 17.5 micromol TE/g. Ascorbic acid and flavonoid content of the hydrophilic extracts did not explain the total variation in antioxidant capacity of those extracts, suggesting either the presence of other antioxidant components that have yet to be identified or that the known antioxidants are producing synergistic effects. The carotenoids did correlate with antioxidant capacity of the lipophilic extracts and accounted for the majority of the variability in that fraction. The variability in hydrophilic and lipophilic antioxidant capacity found among these genotypes suggests that potential efficacy from antioxidants will vary considerably from genotype to genotype. 相似文献

Fourier transform infrared spectroscopy (FT-IR, 4000-600 cm(-)(1)) was used to discriminate between intact and sonication-injured Listeria monocytogenes ATCC 19114 and to distinguish this strain from other selected Listeria strains (L. innocua ATCC 51742, L. innocua ATCC 33090, and L. monocytogenes ATCC 7644). FT-IR vibrational overtone and combination bands from mid-IR active components of intact and injured bacterial cells produced distinctive "fingerprints" at wavenumbers between 1500 and 800 cm(-)(1). Spectral data were analyzed by principal component analysis. Clear segregations of different intact and injured strains of Listeria were observed, suggesting that FT-IR can detect biochemical differences between intact and injured bacterial cells. This technique may provide a tool for the rapid assessment of cell viability and thereby the control of foodborne pathogens. 相似文献

A wild strain of Agrobacterium rhizogenes was used to regenerate twelve in vitro plant lines from different hairy roots of H. perforatum (St. John's Wort). The production of the main bioactive constituents was observed even though their yields varied in the different plant lines. Two lines were selected for the hyperoside production (4.9-4.6 mg/gdw) while nine were characterized by significant yields of chlorogenic acid (ranged from 0.47 to 1.09 mg/gdw). Furthermore, one out of twelve lines showed a 10-fold higher hypericin content (0.25 mg/gdw) than that reported for the in vitro shoots in the literature. Isolation and determination of alpha-dicarbonyl compounds by RP-HPLC-DAD in green and roasted coffee

Glyoxal, methylglyoxal, and diacetyl formed as Maillard reaction products in heat-treated food were determined in coffee extracts (coffee brews) obtained from green beans and beans with different degrees of roast. The compounds have been reported to be mutagenic in vitro and genotoxic in experimental animals in a number of papers. More recently, alpha-dicarbonyl compounds have been implicated in the glycation process. Our data show that small amounts of glyoxal and methylglyoxal occur naturally in green coffee beans. Their concentrations increase in the early phases of the roasting process and then decline. Conversely, diacetyl is not found in green beans and forms later in the roasting process. Therefore, light and medium roasted coffees had the highest glyoxal and methylglyoxal content, whereas dark roasted coffee contained smaller amounts of glyoxal, methylglyoxal, and diacetyl. For the determination of coffee alpha-dicarbonyl compounds, a reversed-phase high performance liquid chromatography with a diode array detector (RP-HPLC-DAD) method was devised that involved the elimination of interfering compounds, such as chlorogenic acids, by solid phase extraction (SPE) and their derivatization with 1,2-diaminobenzene to give quinoxaline derivatives. Checks of SPE and derivatization conditions to verify recovery and yield, respectively, resulted in rates of 100%. Plant cell walls are the major structural component of fruits and vegetables, which break down to cell wall particles during ingestion (oral mastication) or food processing. The major health-promoting effect of cell walls occurs when they reach the colon and are fermented by the gut microbiota. In this study, the fermentation kinetics of carrot cell wall particle dispersions with different particle size and microstructure were investigated in vitro using porcine feces. The cumulative gas production and short-chain fatty acids (SCFAs) produced were measured at time intervals up to 48 h. The results show that larger cell clusters with an average particle size (d(0.5)) of 298 and 137 μm were more rapidly fermented and produced more SCFAs and gas than smaller single cells (75 μm) or cell fragments (50 μm), particularly between 8 and 20 h. Confocal microscopy suggests that the junctions between cells provides an environment that promotes bacterial growth, outweighing the greater specific surface area of smaller particles as a driver for more rapid fermentation. The study demonstrates that it may be possible, by controlling the size of cell wall particles, to design plant-based foods for fiber delivery and promotion of colon fermentation to maximize the potential for human health. 相似文献

Ellagic acid (EA), a natural polyphenol abundant in fruits and common in our diet, is under intense investigation for its chemopreventive activity resulting from multiple effects. EA inhibits topoisomerase II, but the effects on the human enzyme of urolithins, its monolactone metabolites, are not known. Therefore, the action of several synthetic urolithins toward topoisomerases II was evaluated, showing that polyhydroxylated urolithins, EA, and EA-related compounds are potent inhibitors of the α and β isoforms of human topoisomerase II at submicromolar concentrations. Competition tests demonstrate a dose-dependent relationship between ATP and the inhibition of the enzyme. Docking experiments show that the active compounds bind the ATP pocket of the human enzyme, thus supporting the hypothesis that EA and polyhydroxylated urolithins act as ATP-competitive inhibitors of human topoisomerase II. 相似文献
