Forty-eight growing pigs were randomly assigned to five dietary groups and penned individually. They received a diet based on barley, wheat, corn and soya bean meal according to requirement. The experimental groups were supplemented with 400% or 800% of vitamins B(2), B(6) and pantothenic acid, or 400% or 800% of biotin, while all other vitamins were administered according to requirement. Growth performance, carcass characteristics, aspartate aminotransferase (AST), and content of vitamins in blood, liver and muscles were recorded. Growth performance showed no influence of supplementation, while backfat thickness in the group with 800% B(2)/B(6)/pantothenic acid was significantly higher. Content of B(2) in blood, liver and muscle was similar in all groups. Content of B(6) in blood and liver showed significant differences according to supplementation. The content of vitamin B(6) in muscle in the experimental groups was significantly higher than that in the control group. The content of pantothenic acid in blood and muscle in the experimental groups was significantly higher, while in liver all groups were significantly influenced by the supplementation level. Biotin content in liver showed no influence, but the content in plasma was significantly higher in the experimental groups and the content in muscle was significantly higher according to supplementation. The activity of AST showed no significant influence of the dietary vitamin level, but it was obviously decreased in the groups supplemented with biotin. The findings indicate that the dietary supplementation of vitamin B(2), B(6), pantothenic acid and biotin could not improve performance, but the contents in blood, liver and muscle.  相似文献

OBJECTIVE: To demonstrate the effect of anesthesia and surgery on serum ionized magnesium and ionized calcium concentrations in clinical canine and feline patients. ANIMALS: 37 client-owned dogs, ASA PS I-III and 10 client-owned cats, ASA PS I, all receiving anesthesia for elective or emergent surgery at a Veterinary Teaching Hospital. MATERIALS AND METHODS: Plasma ionized and serum total magnesium, and plasma ionized calcium were measured prior to and after a group-standardized anesthetic protocol. RESULTS: Regardless of pre-operative medication (hydromorphone or butorphanol), anesthetic induction (thiopental or lidocaine/hydromorphone/diazepam (LHD) and propofol combination), or type of surgical procedure (peripheral surgery or laparotomy), post-operative plasma ionized calcium concentration decreased in all groups of dogs, while post-operative plasma ionized magnesium increased in all groups, although the changes were not always significant. The dogs who were induced with an LHD and propofol technique had a greater increase in ionized magnesium (0.36 +/- 0.07 to 0.42 +/- 0.07 mmol L(-1)) than the group in which anesthesia was induced with thiopental (0.41 +/- 0.07 to 0.42 +/- 0.07 mmol L(-1), p = 0.009). The cats showed similar changes in ionized magnesium and ionized calcium, and also had a significant increase in serum total magnesium (2.17 +/- 0.20 to 2.31 +/- 0.25 mg dL(-1), p = 0.009) CONCLUSIONS, CLINICAL RELEVANCE: A post-operative decrease in ionized calcium was demonstrated in healthy animals, as well as an increase in ionized or total magnesium after various anesthetic protocols and surgeries. These changes, while statistically significant, do not appear to be clinically significant, as values remained within reference ranges at all times.  相似文献

Black bears hibernate for 5 to 7 months a year and, during this time, do not eat, drink, urinate, or defecate. We measured metabolic rate and body temperature in hibernating black bears and found that they suppress metabolism to 25% of basal rates while regulating body temperature from 30° to 36°C, in multiday cycles. Heart rates were reduced from 55 to as few as 9 beats per minute, with profound sinus arrhythmia. After returning to normal body temperature and emerging from dens, bears maintained a reduced metabolic rate for up to 3 weeks. The pronounced reduction and delayed recovery of metabolic rate in hibernating bears suggest that the majority of metabolic suppression during hibernation is independent of lowered body temperature.  相似文献

Aflatoxins are a group of highly toxic fungal secondary metabolites that occur in Aspergillus species and may contaminate foodstuffs and feeds. Two different anti-aflatoxin B(1) antibodies were examined to develop a surface plasmon resonance (SPR)-based immunoassay to aflatoxin B(1). A conjugate consisting of aflatoxin B(1)-bovine serum albumin (BSA) was immobilized on the dextran gel surface. Competition between immobilized aflatoxin B(1) conjugate and free aflatoxin B(1) in solution for binding to antibody injected over the surface formed the basis for the assay. Regeneration of the antibody from the immobilized conjugate surface is essential for the development of such an inhibitive immunoassay. Problems were encountered with the regeneration of the sensor surface, due to the high-affinity binding of the antibodies. Conventional regeneration solutions consisting of low concentrations of NaOH and HCl worked to a degree, but regeneration was at the expense of the integrity of the immobilized conjugate. A polyclonal anti-aflatoxin B(1) antibody was produced and was found to be regenerable using an organic solution consisting of 1 M ethanolamine with 20% (v/v) acetonitrile, pH 12.0. This combined high ionic strength and extreme pH, as well as chaotrophic properties and allowed the development of an inhibitive immunoassay. The assay had a linear range of 3.0-98.0 ng mL(-1) with good reproducibility.  相似文献

Extracts from wild blueberry (Vaccinium angustifolium Ait.) were separated into proanthocyanidin-rich fractions using liquid vacuum and open column chromatography on Toyopearl and Sephadex LH-20, respectively. Fractions were characterized using analytical tools including mass spectrometry and NMR spectroscopy; fraction composition was correlated with bioactivity using antiproliferation and antiadhesion in vitro assays. There was a significant positive correlation between proanthocyanidin content of different fractions and biological activity in both the antiproliferation and antiadhesion assays. Two fractions containing primarily 4-->8-linked oligomeric proanthocyanidins with average degrees of polymerization (DPn) of 3.25 and 5.65 inhibited adhesion of Escherichia coli responsible for urinary tract infections. Only the fraction with a DPn of 5.65 had significant antiproliferation activity against human prostate and mouse liver cancer cell lines. These findings suggest both antiadhesion and antiproliferation activity are associated with high molecular weight proanthocyanidin oligomers found in wild blueberry fruits.  相似文献

Competitive exclusion of intestinal pathogens by administration of beneficial and defined cultures of normal intestinal microflora is a safe and effective means of reducing the incidence and severity of chick infections with Salmonella and other intestinal pathogens. It is important that competitive exclusion cultures not carry genetic material (e.g., plasmids), which could transfer antibiotic resistance to other microflora, including pathogens. As such, safe and effective competitive exclusion cultures must be sensitive to commonly used antimicrobial agents. By necessity, intentional or accidental exposure of these beneficial microflora to antibiotics will reduce or eliminate the protection provided by competitive exclusion culture establishment. As antibiotic residues can be present from embryonic, hatchling, or maternal administration, a rapid and sensitive assay for the nonspecific detection of residues, which could interfere with competitive exclusion culture establishment, is needed. This study was conducted to develop a rapid and inexpensive bioassay to detect multiple antimicrobial residues in egg yolk and neonatal yolk sacs. Aerobic bacterial strains with known sensitivity to several antibiotics used by the poultry industry were selected and individually compared for sensitivity to enrofloxacin, gentamicin, tetracycline, ceftiofur, and tylosin concentrations in egg yolks. This assay was found to be relatively sensitive for the detection of these antimicrobials, and detection of residues was associated with reduced competitive exclusion culture (PREEMPT) establishment in one experiment. Importantly, this assay can be implemented with minimal training or equipment under commercial hatchery practices and could be used to determine embryo groups, in advance of hatch, that are not suitable candidates for competitive exclusion treatment in the hatchery.  相似文献

The organic acids present in several samples of quince fruit (pulp and peel) and quince jam (homemade and industrially manufactured) were analyzed by HPLC. The sample preparation was simple, involving only extraction with methanol (40 degrees C) and filtration through a Sep-pack C18 cartridge. The chromatographic separation was achieved using an ion exclusion column, Nucleogel Ion 300 OA (300 x 7.7 mm), in conjunction with a column heating device at 30 degrees C. An isocratic elution with H(2)SO(4) 0.01 N as the mobile phase, with a flow rate of 0.1 mL/min, and UV detection at 214 nm were used. These analyses showed that all samples presented a similar profile composed of at least six identified organic acids: citric, ascorbic, malic, quinic, shikimic, and fumaric acids. Several samples also contained oxalic acid. This study suggests that the organic acids levels and ratios may be useful for the determination of percent fruit content of quince jams. The citric acid value can also be used in the differentiation of the type of manufacture of the commercial quince jams (homemade or industrially manufactured).  相似文献
