Colonisation by root endophytes can be beneficial to plants growing on acid, nutrient-poor soils. Arbuscular mycorrhizal (AM) fungi can supply herbs with nutrients and may give protection against aluminium toxicity. Two other root colonising fungi, fine endophytes (FE) and dark septate fungi (DSE), are less well known but are potentially of benefit to their host plant. AM fungi are the most prevalent symbionts in herbs at neutral to acidic soil pH. At extremely low pH, fungal growth can be limited and AM colonisation is usually rare. Fine and dark septate endophytes, on the other hand, have been observed more often under these conditions. In order to relate endophyte colonisation to a gradient in soil pH, we investigated root colonisation by AM, FE and DSE in Maianthemum bifolium, Galium odoratum, Mercurialis perennis and Stellaria nemorum, from a range of acidic beech forests. With decreasing pH, colonisation by AM decreased, whereas the other two endophytes increased. AM and FE colonisation were inversely correlated in Maianthemum bifolium. We compared changes in root colonisation with those in chemical composition of soil and leaf samples and found a positive correlation between leaf magnesium concentrations and the presence of DSE in Galium odoratum. Aluminium concentration in Maianthemum bifolium tended to be lower when FE colonisation was high, suggesting a possible role for the fungi in plant protection against Al. We suggest that FE and DSE may replace AM fungi in herbaceous vegetation at extremely low pH, counteracting some of the negative effects of high soil acidity on plants.  相似文献

Phosphorus (P) concentrations in needles and leaves of forest trees are declining in the last years in Europe. For a sustainable forest management the knowledge of site specific P nutrition/availability in forest soils is vital, but we are lacking verified simple methods for the estimation of plant available P. Within this study, four soil P extraction methods [water ( ), double‐lactate (Plac), citric acid (Pcit), and sodium bicarbonate ( )], as well as total P content of the soil (Ptot) were tested to investigate which method is best correlated with foliar P concentrations of spruce [Picea abies (L.) H. Karst.] and beech [Fagus sylvatica (L.)]. Mineral soil samples from 5 depth levels of 48 forest sites of the Bavarian sample set of the second National Forest Soil Inventory (BZE II) were stratified according to tree species (spruce and beech) and soil pH (pH < 6.2 and > 6.2), covering the whole range of P nutrition. The extractable amount of P per mass unit of soil increased in the order << Plac < < Pcit, decreased with soil depth, and was higher in soils with pH < 6.2. Citric acid extracted up to 10% of Ptot in acidic soils. Whereas Pcit delivers adequate regression models for P nutrition in the case of spruce (R2 up to 0.53) and beech (R2 up to 0.58) for acidic soils, shows good results for spruce growing on acidic soils (R2 up to 0.66) and for beech on soils with pH > 6.2 (R2 up to 0.57). Plac produces adequate models only for beech on high pH soils (R2 up to 0.64), while did not produce acceptable regression models. Ptot seems suitable to explain the P nutrition status of beech on acidic (R2 up to 0.62) and alkaline soils (R2 up to 0.61). Highest R2s are obtained mostly in soil depths down to 40 cm. As and Pcit showed good results for both investigated tree species, they should be considered preferentially in future studies.  相似文献

Under‐ as well as overfertilization with nitrogen (N) will result in economic loss for the farmer due to reduced yields and quality of the products. Also from an ecological perspective, it is important that the grower makes the correct decision on how much and when to apply N for a certain crop to minimize impacts on the environment. To aggravate the situation, N is a substance that is present in many compartments in different forms (nitrate, ammonium, organic N, etc.) in the soil‐plant environment and takes part in various processes (e.g., mineralization, immobilization, leaching, denitrification, etc.). Today, many N‐recommendation systems are mainly based on yield expectation. However, yields are not stable from year to year for a given field. Also the processes that determine the N supply from other sources than fertilizer are not predictable at the start of the growing season. Different methodological approaches are reviewed that have been introduced to improve N‐fertilizer recommendations for arable crops. Many soil‐based methods have been developed to measure soil mineral N (SMN) that is available for plants at a given sampling date. Soil sampling at the start of the growing period and analyzing for the amount of NO ‐N (and NH ‐N) is a widespread approach in Europe and North America. Based on data from field calibrations, the SMN pool is filled up with fertilizer N to a recommended amount. Depending on pre‐crop, use of organic manure, or soil characteristics, the recommendation might be modified (±10–50 kg N ha–1). Another set of soil methods has been established to estimate the amount of N that is mineralized from soil organic matter, plant residues, and/or organic manure. From the huge range of methods proposed so far, simple mild extraction procedures have gained most interest, but introduction into practical recommendation schemes has been rather limited. Plant‐analytical procedures cover the whole range from quantitative laboratory analysis to semiquantitative “quick” tests carried out in the field. The main idea is that the plant itself is the best indicator for the N supply from any source within the growth period. In‐field methods like the nitrate plant sap/petiole test and chlorophyll measurements with hand‐held devices or via remote sensing are regarded as most promising, because with these methods an adequate adjustment of the N‐fertilizer application strategy within the season is feasible. Prerequisite is a fertilization strategy that is based on several N applications and not on a one‐go approach.  相似文献

Purpose

To support scientifically sound water management in dryland environments a modelling system has been developed for the quantitative assessment of water and sediment fluxes in catchments, transport in the river system, and retention in reservoirs. The spatial scale of interest is the mesoscale because this is the scale most relevant for management of water and land resources.

Materials and methods

This modelling system comprises process-oriented hydrological components tailored for dryland characteristics coupled with components comprising hillslope erosion, sediment transport and reservoir deposition processes. The spatial discretization is hierarchically designed according to a multi-scale concept to account for particular relevant process scales. The non-linear and partly intermittent run-off generation and sediment dynamics are dealt with by accounting for connectivity phenomena at the intersections of landscape compartments. The modelling system has been developed by means of data from nested research catchments in NE-Spain and in NE-Brazil.

Results and discussion

In the semi-arid NE of Brazil sediment retention along the topography is the main process for sediment retention at all scales, i.e. the sediment delivery is transport limited. This kind of deposition retains roughly 50 to 60 % of eroded sediment, maintaining a similar deposition proportion in all spatial scales investigated. On the other hand, the sediment retained in reservoirs is clearly related to the scale, increasing with catchment area. With increasing area, there are more reservoirs, increasing the possibility of deposition. Furthermore, the area increase also promotes an increase in flow volume, favouring the construction of larger reservoirs, which generally overflow less frequently and retain higher sediment fractions. The second example comprises a highly dynamic Mediterranean catchment in NE-Spain with nested sub-catchments and reveals the full dynamics of hydrological, erosion and deposition features. The run-off modelling performed well with only some overestimation during low-flow periods due to the neglect of water losses along the river. The simulated peaks in sediment flux are reproduced well, while low-flow sediment transport is less well captured, due to the disregard of sediment remobilization in the riverbed during low flow.

Conclusions

This combined observation and modelling study deepened the understanding of hydro-sedimentological systems characterized by flashy run-off generation and by erosion and sediment transport pulses through the different landscape compartments. The connectivity between the different landscape compartments plays a very relevant role, regarding both the total mass of water and sediment transport and the transport time through the catchment.  相似文献

The chemical conditions of the rhizosphere can be very different from that of bulk soil. Up to now, little attention has been given to the problem of spatial heterogeneity and temporal dynamics of rhizosphere soil solution and little is known about the influence of different tree species on rhizosphere chemistry. In the present study, we used micro suction cups to collect soil solution from the rhizosphere of Norway spruce (Picea abies [L.] Karst.) and European beech (Fagus sylvatica L.) seedlings in high spatial resolution and capillary electrophoresis for the determination of major cations and anions. The results indicate, that in a soil with a base saturation of about 20—25% and a pH of 6.5, growing roots of beech and spruce lower the concentrations of nutrient cations and nitrate in the rhizosphere soil solution and decrease significantly the pH. The H+ release leads to an enhanced mineral weathering as indicated by an increase of CEC and base saturation and to a mobilization of soluble Al, however, on a very low concentration level. In our experiment rhizosphere effects of spruce have been more pronounced than those of beech, indicating, that with respect to below ground activity young spruce trees have a better competitive power than beech.  相似文献

To investigate the value of the BP26 protein in the serological diagnosis of ovine brucellosis caused by Brucella ovis, recombinant BP26 protein was produced in Echerichia coli and purified for use in an indirect enzyme-linked immunosorbent assay (I-ELISA). The majority of the recombinant protein was recovered from the supernatant of sonicated recombinant E. coli cells in a soluble form. This facilitated the purification of the recombinant BP26 protein which was achieved by using ion-exchange chromatography. After one step of purification, the purity of the recombinant BP26 protein was analyzed by using SDS-PAGE, Coomassie blue staining, and Western blot with a monoclonal antibody (MAb) directed against the BP26 protein. The degree of purity appeared satisfactory so that it could be directly used in I-ELISA. Although the recombinant BP26-ELISA appeared less useful than I-ELISA using the B. ovis hot saline (HS) extract as antigen, the high number of sera from B. ovis infected rams found positive (90%) in the recombinant BP26-I-ELISA indicated that the BP26 protein may be an additional suitable antigen for serological diagnosis of B. ovis infection in rams.  相似文献

Purpose

Hydrosedimentological studies conducted in the semiarid Upper Jaguaribe Basin, Brazil, enabled the identification of the key processes controlling sediment connectivity at different spatial scales (100–104 km2).

Materials and methods

Water and sediment fluxes were assessed from discharge, sediment concentrations and reservoir siltation measurements. Additionally, mathematical modelling (WASA-SED model) was used to quantify water and sediment transfer within the watershed.

Results and discussion

Rainfall erosivity in the study area was moderate (4600 MJ mm ha?1 h?1 year?1), whereas runoff depths (16–60 mm year?1), and therefore the sediment transport capacity, were low. Consequently, ~60 % of the eroded sediment was deposited along the landscape, regardless of the spatial scale. The existing high-density reservoir network (contributing area of 6 km2 per reservoir) also limits sediment propagation, retaining up to 47 % of the sediment at the large basin scale. The sediment delivery ratio (SDR) decreased with the spatial scale; on average, 41 % of the eroded sediment was yielded from the hillslopes, while for the whole 24,600-km2 basin, the SDR was reduced to 1 % downstream of a large reservoir (1940-hm3 capacity).

Conclusions

Hydrological behaviour in the Upper Jaguaribe Basin represents a constraint on sediment propagation; low runoff depth is the main feature breaking sediment connectivity, which limits sediment transference from the hillslopes to the drainage system. Surface reservoirs are also important barriers, but their relative importance to sediment retention increases with scale, since larger contributing areas are more suitable for the construction of dams due to higher hydrological potential.  相似文献

Earthworms strongly affect soil organic carbon cycling. The aim of this study was to determine whether deep burrowing anecic earthworms enhance carbon storage in soils and decrease C turnover. Earthworm burrow linings were separated into thin cylindrical sections with different distances from the burrow wall to determine gradients from the burrow wall to the surrounding soil. Organic C, total N, radiocarbon (14C) concentration, stable isotope values (δ13C, δ15N) and extracellular enzyme activities were measured in these samples. Anecic earthworms increased C stocks by 270 and 310 g m?2 accumulated in the vertical burrows. C-enrichment of the burrow linings was spatially highly variable within a distance of millimetres around the burrow walls. It was shown that C accumulation in burrows can be fast with C sequestration rates of about 22 g C m?2 yr?1 in the burrow linings, but accumulated C in the burrows may be mineralised fast with turnover times of only 3–5 years. Carbon stocks in earthworm burrows strongly depended on the earthworm activity which maintains continuous C input into the burrows. The enhanced extracellular enzyme activity of fresh casts was not persistent, but was 47% lower in inhabited burrows and 62% lower in abandoned burrows. Enzyme activities followed the C concentrations in the burrows and were not further suppressed due to earthworms. Radiocarbon concentrations and stable isotopes in the burrow linings showed an exponential gradient with the youngest and less degraded organic matter in the innermost part of the burrow wall. Carbon accumulation by anecic earthworm is restricted to distinct burrows with less influence to the surrounding soil. Contrary to the initial hypothesis, that organic C is stabilised due to earthworms, relaxation time experiments with nuclear magnetic resonance spectroscopy (NMR) did not reveal any enhanced adsorption of C on iron oxides with C stabilising effect. Our results suggest that earthworm activity does not substantially increase subsoil C stocks but burrows serve as fast ways for fresh C transport into deep soil horizons.  相似文献

The paucity of enzymes that efficiently deconstruct plant polysaccharides represents a major bottleneck for industrial-scale conversion of cellulosic biomass into biofuels. Cow rumen microbes specialize in degradation of cellulosic plant material, but most members of this complex community resist cultivation. To characterize biomass-degrading genes and genomes, we sequenced and analyzed 268 gigabases of metagenomic DNA from microbes adherent to plant fiber incubated in cow rumen. From these data, we identified 27,755 putative carbohydrate-active genes and expressed 90 candidate proteins, of which 57% were enzymatically active against cellulosic substrates. We also assembled 15 uncultured microbial genomes, which were validated by complementary methods including single-cell genome sequencing. These data sets provide a substantially expanded catalog of genes and genomes participating in the deconstruction of cellulosic biomass.  相似文献

Ultrasonic dispersion is a prevalent tool for soil fractionation. It is widely ignored that variation in ultrasonic power might lead to significantly different dispersion. We evaluated the effect of power variation with constant energy on the fine fraction mass, its organic C content and quality. All parameters increased significantly with power. The term “stable aggregates” as used in fractionation schemes cannot be defined by ultrasonic energy alone but power needs to be standardized, too.  相似文献
